Diabetes mellitus disrupts lncRNA Malat1 regulation of cardiac mitochondrial genome-encoded protein expression.

Understanding the cellular mechanisms behind diabetes-related cardiomyopathy is crucial as it is a common and deadly complication of diabetes mellitus. Dysregulation of the mitochondrial genome has been linked to diabetic cardiomyopathy and can be ameliorated by altering microRNA (miRNA) availability in the mitochondrion. Long noncoding RNAs (lncRNAs) have been identified to downregulate miRNAs. This study aimed to determine if diabetes mellitus impacts the mitochondrial localization of lncRNAs, their interaction with miRNAs, and how this influences mitochondrial and cardiac function. In mouse

Ionizable mesoporous silica nanoparticle-mediated siAURKB delivery for colorectal cancer therapy.

Colorectal cancer (CRC) is a common malignant tumor with high incidence and mortality worldwide. Conventional therapeutic strategies, such as surgical resection and chemotherapy, which result in improved outcomes for some patients, are greatly hindered by drug resistance and off-target toxicity. Small interfering RNA (siRNA)-mediated RNA interference has provided a breakthrough for precision CRC therapy. However, efficient delivery of siRNA to the tumor site with its complex tumor microenvironment remains challenging. Here, an ionizable mesoporous silica nanoparticle (MSN)-based nanocarrier wi

Rapid divergence and high diversity of miRNAs and miRNA targets in the Camelineae.

MicroRNAs (miRNAs) are short RNAs involved in gene regulation through translational inhibition and transcript cleavage. After processing from imperfect fold-back structures, miRNAs are incorporated into RNA-induced silencing complexes (RISCs) before targeting transcripts with varying degrees of complementarity. Some miRNAs are evolutionarily deep-rooted, and sequence complementarity with their targets is maintained through purifying selection. Both Arabidopsis and Capsella belong to the tribe Camelineae in the Brassicaceae, with Capsella rubella serving as an outgroup to the genus Arabidopsis.

Regulation of tumor angiogenesis: Roles of lncRNAs in molecular signaling, microenvironmental remodeling, and anti-angiogenic therapy.

Angiogenesis is a crucial step in the growth and metastasis of invasive tumors. Tumor-induced angiogenesis is not due to a single factor; it involves activation of oncogenic signaling pathways and abnormal expression of angiogenesis-related genes within the complex tumor microenvironment (TME). Traditional research has primarily focused on the regulation of angiogenesis by protein-coding genes. In recent years, growing evidence has shown that long non-coding RNAs (lncRNAs) play important roles in angiogenesis. In this study, we systematically elucidate the molecular mechanisms by which lncRNAs

PRC2-mediated apoptosis evasion is a therapeutic target of MDS/AML harboring inv(3)/t(3;3) and monosomy 7.

Myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML) harboring both inv(3)/t(3;3) and monosomy 7 (-7) are highly aggressive myeloid cancers of which molecular pathogenesis and therapeutic vulnerability remain elusive. High throughput drug screens, CUT&Tag/RNA sequence, and functional assays using human MDS/AML cells revealed that EZH2 inhibitors efficiently induce apoptosis preferentially in MDS/AML with inv(3)/t(3;3) and -7 through the activation of GADD45γ-p38-p53 axis. EVI1 activated in 3q-rearranged MDS/AML was responsible for GADD45γ silencing by direct binding to its consensu

Epigenomic and Functional Characterization of Junctophilin 3 (JPH3) as a Novel Tumor Suppressor Being Frequently Inactivated by Promoter CpG Methylation in Digestive Cancers.

Junctophilin (JPH) proteins stabilize junctional membrane complexes between plasma membrane and endoplasmic reticulum, also implicated in some human diseases. JPH3 mutations are linked to Huntington's disease-like 2 syndrome. Through epigenomic study of a colon cancer cell line pair (HCT116 and DKO), we identified JPH3 as a methylated novel tumor suppressor gene (TSG) candidate at 16q24. We further studied its epigenetic alterations and functions in digestive tumorigenesis. JPH3 expression at the RNA level was found to be frequently silenced or reduced in colorectal and gastric cancers due to

Involvement of p32 and microtubules in alteration of mitochondrial functions by rubella virus.

The interaction of the rubella virus (RV) capsid (C) protein and the mitochondrial p32 protein is believed to participate in virus replication. In this study, the physiological significance of the association of RV with mitochondria was investigated by silencing p32 through RNA interference. It was demonstrated that downregulation of p32 interferes with microtubule-directed redistribution of mitochondria in RV-infected cells. However, the association of the viral C protein with mitochondria was not affected. When cell lines either pretreated with respiratory chain inhibitors or cultivated unde

Mechanism study of LncRNA PRINS targeting TFAM to regulate mitochondrial dysfunction in human renal tubular epithelial cells under ischemia-reperfusion.

BACKGROUND: Acute kidney injury (AKI) is a significant clinical problem associated with high morbidity and mortality. The psoriasis susceptibility-associated RNA gene (PRINS) is a stress-induced transcript implicated in kidney disease. However, its role in regulating mitochondrial function during AKI remains unclear. METHODS: We measured the expression of LncRNA PRINS and key mitochondrial genes, including mitochondrial transcription factor A (TFAM), NDUFV1, NDUFS1, and CYPD, in peripheral blood and renal tissues from AKI patients and controls. A hypoxia/reoxygenation (H/R) model was establis

Characterization of Viral Interference in Aedes albopictus C6/36 Cells Persistently Infected with Dengue Virus 2.

Arboviruses are an important group of pathogens that cause diseases of medical and veterinary concern worldwide. The interactions of these viruses with their host cells are complex, and frequently, the coexistence of two different viruses in the same cell results in the inhibition of replication in one of the viruses, which is a phenomenon called viral interference. This phenomenon can be exploited to develop antiviral strategies. Insect cell lines persistently infected with arboviruses are useful models with which to study viral interference. In this work, a model of C6/36-HT cells (from Aede

The BIK1-WRKY9 module activates jasmonate signaling to confer apple resistance to Alternaria blotch.

Apple Alternaria blotch, caused by Alternaria alternata f. sp. mali (A. alternata), is a devastating fungal disease threatening global apple production. However, the early signaling pathway underlying apple resistance to A. alternata remains unclear. In this study, through transcriptomic analysis, we identified that MdWRKY9 is a key positive regulator of apple resistance to A. alternata, whose expression is induced in the resistant "Hanfu" while being reduced in the susceptible "Gala" following A. alternata infection. Stable overexpression of MdWRKY9 enhanced apple resistance to A. alternata,

Reversal of C9orf72 mutation-induced transcriptional dysregulation and pathology in cultured human neurons by allele-specific excision.

Efforts to genetically reverse C9orf72 pathology have been hampered by our incomplete understanding of the regulation of this complex locus. We generated five different genomic excisions at the C9orf72 locus in a patient-derived induced pluripotent stem cell (iPSC) line and a non-diseased wild-type (WT) line (11 total isogenic lines), and examined gene expression and pathological hallmarks of C9 frontotemporal dementia/amyotrophic lateral sclerosis in motor neurons differentiated from these lines. Comparing the excisions in these isogenic series removed the confounding effects of different gen

Ankyrin repeat domain 1 regulates innate immune responses against herpes simplex virus 1: A potential role in eczema herpeticum.

BACKGROUND: Atopic dermatitis (AD) is a common inflammatory skin disease. A subset of patients with AD are susceptible to disseminated herpes simplex virus (HSV) infection, a complication termed eczema herpeticum (ADEH+). The immune mechanisms causing ADEH+ remain elusive. Using RNA sequencing, we recently found that ankyrin repeat domain 1 (ANKRD1) was significantly induced in human PBMCs upon HSV-1 stimulation, and its induction in patients with ADEH+ was significantly reduced compared with that seen in AD patients without a history of eczema herpeticum (ADEH-). OBJECTIVE: We sought to vali

Large-Mesopore Covalent Organic Framework Nanoparticles for Small Interfering RNA Delivery toward Cancer Immunotherapy.

Small interfering RNA (siRNA) offers a promising modality for tumor immunotherapy through a precise gene-silencing mechanism, whereas its efficient loading and in vivo delivery rely on specialized carriers because of its polyanionic nature. Porous nanomaterials represent attractive platforms for designing such carriers. However, constructing biocompatible porous organic materials for efficient siRNA loading and in vivo delivery is challenging because structurally stable organic nanoparticles (NPs) with abundant, ordered, and large mesopores have rarely been reported. Herein, a linker-design st

Identification of RISC-associated adenoviral microRNAs, a subset of their direct targets, and global changes in the targetome upon lytic adenovirus 5 infection.

UNLABELLED: Adenoviruses encode a set of highly abundant microRNAs (mivaRNAs), which are generated by Dicer-mediated cleavage of the larger noncoding virus-associated RNAs (VA RNAs) I and II. We performed deep RNA sequencing to thoroughly investigate the relative abundance of individual single strands of mivaRNA isoforms in human A549 cells lytically infected with human adenovirus 5 (Ad5) at physiologically relevant multiplicities of infection (MOIs). In addition, we investigated their relative abundance in the endogenous RNA-induced silencing complexes (RISCs). The occupation of endogenous RI

Role of a Synonymous Mutation at Codon 178 in P1 That Attenuates Potato Virus Y-Induced Tobacco Vein Necrosis and Its Application for Expressing Human Interferon.

Plant virus-based vectors are gaining significant attention as bioreactors for recombinant protein production due to their broad host range, robust expression levels and cost-effectiveness; however, their practical utility is frequently constrained by severe viral pathogenicity. Notably, the utility of potato virus Y (PVY), the type member of the genus Potyvirus, is severely hampered by its elicitation of tobacco vein necrosis (TVN). Here, we demonstrate that a single synonymous substitution (AAA to AAG) at the K178 codon of P1 causes almost no TVN. Mechanistically, the P1-AAG mutation attenua

Trichomonas vaginalis induces IL-1β production in a human prostate epithelial cell line by activating the NLRP3 inflammasome via reactive oxygen species and potassium ion efflux.

BACKGROUND: Trichomonas vaginalis is a sexually transmitted protozoan parasite that causes vaginitis in women, and urethritis and prostatitis in men. IL-1β is synthesized as immature pro-IL-1β, which is cleaved by activated caspase-1. Caspase-1 is, in turn, activated by a multi-protein complex known as an inflammasome. In this study, we investigated the inflammatory response of a prostate epithelial cell line (RWPE-1) to T. vaginalis and, specifically, the capacity of T. vaginalis to activate the NLRP3 inflammasome. METHODS: RWPE-1 cells were stimulated by live T. vaginalis, and subsequent ex

HDAC7-Induced Epigenetic Repression Modulates ATF3 Functional Plasticity in Colorectal Cancer Pathogenesis.

Class IIa histone deacetylase 7 (HDAC7) regulates transcription primarily through scaffolding functions, but its molecular mechanisms in cancer pathogenesis remain incompletely understood. Here, we establish HDAC7 as a key epigenetic regulator in colorectal cancer (CRC). HDAC7 is overexpressed in CRC tumors and correlates with advanced disease stages, lymph node metastasis, and poor patient survival. Mechanistically, HDAC7 scaffolds a repressive complex with HDAC3 and the stress-responsive transcription factor ATF3. This reduces H3K27ac/H3K18ac occupancy and blocks BRD4/RNA polymerase II (Pol
