Parvovirus B19 genotypes 1 and 2 detection with real-time polymerase chain reaction assays.

BACKGROUND AND OBJECTIVES: Parvovirus B19 (B19V) DNA screening has been introduced to comply with European regulations for certain plasma products. Current commercial and some in-house B19V DNA assays fail to detect or under-quantify the recently identified genotypes 2 and 3. In this report, we describe 2-year experience with B19V DNA screening using the commercial assay from Roche (detecting only genotype 1) combined with an in-house assay (detecting genotypes 1, 2 and 3). This dual testing approach enables the identification of molecular variants of B19V. MATERIALS AND METHODS: Between 2005

Time-resolved spectroscopic study of the reaction Cl + n-C5H12 --> HCl + C5H11 in solution.

We study the hydrogen abstraction reaction from pentane by chlorine radicals using four different experimental approaches. We use two different solvents (CH2Cl2 and CCl4) and two different chlorine atom sources (photodissociation of dissolved Cl2 and two-photon photolysis of the solvent) to investigate their effects on the recombination and reactivity of the chlorine radical. All four experimental schemes involve direct probing of the transient chlorine population via a charge transfer transition with a solvent molecule. In one of the four approaches, photolysis of Cl2 in dichloromethane, we a

Real time polymerase chain reaction: a new powerful tool for the diagnosis of neurobrucellosis.

BACKGROUND/METHODS: We compared the diagnostic yield of a real time polymerase chain reaction (PCR) assay in cerebrospinal fluid (CSF) samples with conventional microbiological techniques for the diagnosis of neurobrucellosis. Following amplification of a 223 bp sequence specific for Brucella genus, melting curve analysis was performed to verify the specificity of the PCR products. RESULTS: All six patients with neurobrucellosis (three meningitis and three meningoencephalitis) had a positive real time PCR assay, whereas CSF cultures and Wright seroagglutination tests were positive in only two

Development of a quantitative real-time polymerase chain reaction assay specific for Orientia tsutsugamushi.

Two specific and sensitive polymerase chain reaction (PCR) assays were developed to detect and quantitate Orientia tsutsugamushi, the agent of scrub typhus, using a portion of the 47-kD outer membrane protein antigen/ high temperature requirement A gene as the target. A selected 47-kD protein gene primer pair amplified a 118-basepair fragment from all 26 strains of O. tsutsugamushi evaluated, but it did not produce amplicons when 17 Rickettsia and 18 less-related bacterial nucleic acid extracts were tested. Similar agent specificity for the real-time PCR assay, which used the same primers and

Effect of Reaction Time and Temperature on the Impurity Profile of Amphetamine in the Leuckart Method.

Illicit amphetamine synthesis is frequently conducted using the well-known Leuckart method, generating complex impurity profiles that provide forensic insights. This study reports the effect of reaction temperature and time during the first step of the Leuckart synthesis on subsequent conversion efficiency and impurity profile. Controlled syntheses were conducted at 120°C, 140°C, and 160°C, with samples collected at defined intervals. Reaction progress was monitored by gas chromatography-flame ionization detection (GC-FID), and impurity profiling followed the European harmonized method for pro

Digitally enabled hemovigilance allows real time response to transfusion reactions.

BACKGROUND: Transfusion carries a risk of transfusion reaction that is often underdiagnosed due to reliance on passive reporting. The study investigated the utility of digital methods to identify potential transfusion reactions, thus allowing real-time intervention for affected patients. METHOD: The hemovigilance unit monitored 3856 patients receiving 43,515 transfusions under the hemovigilance program. Retrospective comparison data included 298,498 transfusions. Transfusion medicine physicians designed and validated algorithms in the electronic health record that analyze discrete data, such

Emerging Object Representations in the Visual System Predict Reaction Times for Categorization.

Recognizing an object takes just a fraction of a second, less than the blink of an eye. Applying multivariate pattern analysis, or "brain decoding", methods to magnetoencephalography (MEG) data has allowed researchers to characterize, in high temporal resolution, the emerging representation of object categories that underlie our capacity for rapid recognition. Shortly after stimulus onset, object exemplars cluster by category in a high-dimensional activation space in the brain. In this emerging activation space, the decodability of exemplar category varies over time, reflecting the brain's tra

Simple reaction times to lateralized visual stimuli in a case of callosal agenesis.

Simple reaction times (RTs) of a young acallosal woman were measured using three different intensity levels of the visual stimulus. It was found that "'crossed" stimulus/hand combinations yielded finger RTs about 20 msec longer, on average, than "uncrossed" combinations. In addition, an inverse relation was found between stimulus intensity and the magnitude of this crossed/uncrossed difference in RT. It was also found that vocal RTs displayed a large advantage (18 msec) for right visual-field stimulation. It is suggested that the stimulus information in crossed finger reactions must cross the

Effect of ethanol and of noise on reaction time in the monkey: variation with stimulus level.

To determine whether the latency-increasing effects of ethanol were differential with respect to the intensity of the stimulus that initiated the response, three rhesus monkeys were trained on a behavioral task in which the latency of a simple motor response was measured following the onset of a pure tone stimulus. Following training, the animals were tested at a number of different tone intensities and functions relating latency to tone intensity were constructed. When these were stable, the animals were given ethanol in doses of 1.0-2.5 g/kg and the effects on response latencies to different

Utility of tolerance assays for real-time management of infliximab infusion reactions.

Failure of infliximab (IFX) therapy is often associated with pharmacokinetic challenges, frequently linked to the development of anti-infliximab antibodies (ATI). ATI formation is a key factor contributing to therapeutic failure and significant safety concerns, as these antibodies can trigger severe infusion-related reactions that may require treatment discontinuation. In Spain, most available ATI detection assays are sensitivity-based, identifying antibodies only in the absence of circulating IFX. In contrast, tolerance assays can detect ATI even in the presence of IFX, offering distinct adva

Effects of unseen stimuli on reaction times to seen stimuli in monkeys with blindsight.

In three macaque monkeys with unilateral removal of primary visual cortex and in one unoperated monkey, we measured reaction times to a visual target that was presented at a lateral eccentricity of 20 degrees in the normal, left, visual hemifield. When an additional stimulus was presented at the corresponding position in the right hemifield (hemianopic in three of the monkeys), it significantly slowed the reaction time to the left target if it preceded it by delays from 100-500 msec. The most effective delay depended on the particular experimental paradigm and perhaps on the experience of the

Slower and more variable reaction times in schizophrenia: what do they signify?

Extensive research has demonstrated that schizophrenic subjects are slower than normal comparison subjects on a range of reaction-time tasks. Some investigators have also observed that schizophrenic patients exhibit larger intraindividual variability in reaction times when performing these tasks than do normal comparison subjects. This study, using a lexical decision choice reaction time (CRT) task, explored the relation of mean CRT and it intra-individual variability (CRT-SD) to psychiatric symptoms and to performance on executive-motor tasks in 26 medication-free schizophrenic out-patients a

A lack of timing-dependent effects of transcranial direct current stimulation (tDCS) on the performance of a choice reaction time task.

Anodal transcranial direct current stimulation (tDCS) can enhance the retention of a previously practiced motor skill. However, the effects of tDCS on the performance of the choice reaction time task are not fully understood. We examined the effects of anodal tDCS over the left primary motor cortex (M1) on the retention of a 4-choice visual-motor reaction time task (4-ChRT). Right-handed healthy participants (n = 100) were randomly assigned to five groups: three groups received anodal tDCS: before (tDCSbefore), during (tDCSduring), or after (tDCSafter) motor practice. In addition, there were t

Static background distance modulates reaction time and interceptive timing for approaching targets in a real three-dimensional environment.

Rapid and accurate processing of approaching objects is critical for everyday activities and sports. Environmental context in three-dimensional (3D) space provides depth that may modulate visuomotor responses to motion in depth, yet its influence on rapid motor reactions and temporal prediction remains unclear. This study examined whether static background distance affects reaction time (RT) and interceptive timing for approaching targets in a real 3D environment. Fourteen participants completed RT and interceptive tasks with backgrounds positioned 0.6 m (near), 4 m (middle), and 8 m (far) fro

[Investigation of Factors Affecting Viral Clearance Time by Real-Time Polymerase Chain Reaction Method in Nasopharyngeal Swab Samples of COVID-19 Patients].

The coronavirus disease-2019 (COVID-19) pandemic continues to threaten the lives of millions of people. Viral shedding through the respiratory tract is the main risk factor for the transmission of the severe acute respiratory syndrome-2 (SARS-CoV-2) virus from sick individuals to healthy individuals. In this study, we aimed to investigate the viral clearance (VC) time in PCR tests of COVID-19 patients and the possible factors affecting this time. Seventy patients older than 18 years of age whose presence of SARS-CoV-2 virus was proven by real-time polymerase chain reaction (Rt-PCR) in nasophar
