T-cell signaling abnormalities in human systemic lupus erythematosus.

Abnormal expression of key signaling molecules and defective functions of T lymphocytes play a significant role in the pathogenesis of systemic lupus erythematosus (SLE). T-cell receptor (TCR)/CD3-mediated stimulation of SLE T cells shows increased protein tyrosine phosphorylation of cellular proteins, with faster kinetics, heightened calcium response, and decreased interleukin (IL)-2 production. The molecular mechanism of T-cell signaling abnormalities in SLE T cells is complex and cannot be explained fully by the current theories of T-cell signaling. Current research on lymphocyte signaling

Human CD3 gamma delta + activated lymphocytes exhibit killer activity in vitro against autologous leukemic cells.

TCR gamma delta expressing T cell clones are able to exhibit in vitro strong cytolytic activity against cultured tumor cell lines as the myeloid K562 or the Burkitt's lymphoma Daudi cell line. We investigate the possibility of developing TCR gamma delta bearing T cell lines from peripheral blood of 2 leukemic patients in complete remission in order to study their ability to kill autologous leukemic cells. T lymphocyte clones are obtained by limiting dilution of lymphocytes from the blood of patients with an acute lymphoblastic leukemia (ALL). The T cell clones are first selected for their CD8-

Species differences influencing the safety assessment of CD3 T-cell engagers.

Bispecific CD3 T-cell engagers (TCE) redirect endogenous T-cells to tumor cells and have demonstrated substantial clinical activity, but their mechanism of action also drives immune-mediated toxicities that are often underpredicted in nonhuman primate (NHP) studies. To investigate the biological basis of this translational gap, we performed a comparative evaluation of human and cynomolgus macaque T-cells using immunophenotyping, functional characterization with TCE, and single-cell transcriptomic profiling. Human and NHP donors showed broadly similar T-cell frequencies, subset distributions, a

Differential Vbeta T-cell receptor usage during chronic experimental schistosomiasis corresponds with distinct pathological presentations.

CBA/J male mice with chronic Schistosoma mansoni infections display either moderate splenomegaly syndrome (MSS) or hypersplenomegaly syndrome (HSS). As MSS and HSS mice differ in several immunologic characteristics, we investigated T-cell receptor Vbeta usage. The groups had significantly different expression of several Vbetas, suggesting a relationship between the T-cell repertoire and schistosomiasis pathology.

Induction of transient murine T cell anergy by a low molecular weight compound obtained from supernatants of human placental cultures is linked to defective phosphorylation of TCR CD3 chain.

PROBLEM: Allopregnancy induces specific transient tolerance to paternal grafts, and we know that a low molecular weight material ("filtrate") present in a human placental supernatant can do so in vitro (specific unresponsiveness) as well as in vivo, such as when preventing graft-versus-host reaction (GVH) produced by A cells injected into irradiated A x B F1s recipient. We also know by studies carried out using specific anti-V beta-specific stimulation as well as secondary and primary mixed lymphocyte reaction in major histocompatibility complex (MHC) only incompatible combinations that the ma

T cell antigen receptor expression by intra-epithelial lymphocytes in oral lichen planus.

The distribution of T lymphocytes expressing the alpha beta or gamma delta heterodimer of the T cell receptor (TCR) was examined in normal oral mucosa (NOM) and reticular oral lichen planus (OLP) using a panel of antibodies specific for CD3, the alpha beta TCR and the gamma delta TCR. Intra-epithelial lymphocytes were counted and epithelial surface length was measured by image analysis. T cells in the lamina propria were not quantified. Total intra-epithelial lymphocytes were increased in OLP compared with NOM (P = 0.0004). The proportions of cells expressing the gamma delta TCR in NOM and OLP

Detection of Epstein-Barr virus genome in benign polyclonal proliferative T cells of a young male patient.

Epstein-Barr virus (EBV) DNA was detected in polyclonal T cells that proliferated transiently in a 21-year-old male (referred to as H.J.) who underwent an apparently benign lymphocytosis (white blood cells, 31 x 10(6)/microL; lymphocyte, 79%) with fever, tonsillar swelling, lymphadenopathy, and hepatosplenomegaly. The symptoms and signs subsided mostly within a month of hospitalization. The major population of the lymphocytes at admission was positive for CD3, CD8 (4/8 ratio, 0.16), WT31, and DR antigen. Eight percent of the leukocytes were too blastoid to be classified as atypical lymphocytes

[Chronic lymphocytic leukemia with peripheral T lymphocytes expressing CD 2+, CD 3+, CD 4-, CD 8-, CD 16+, and CD 56+ and lymph-node lymphocytes expressing CD 2+, CD 3-, CD 4-, CD 8-, CD 16+, CD 38+, and CD 56+].

A 33-year-old man was hospitalized because of thrombocytopenia and severe splenomegaly. On admission 78% of peripheral lymphoid cells were abnormally large, with pale cytoplasm. Flow cytometry of the abnormal lymphocytes showed that they expressed CD 2, CD 3, CD 11, CD 16, and CD 56, but not CD 4 nor CD 8, so they were T-cell large granular lymphocytes (T-LGL). Abnormal lymphocytes obtained from a lymph node expressed CD 2, CD 16, CD 38, and CD 56, but not CD 3, CD 4, and CD 8, so they were natural killer(NK) cells. Splenectomy was performed and the operative specimen showed diffuse infiltrati

T-lymphocyte subgroups and the activity of human natural killer (HNK) cells in low-grade and high-grade malignant cases of non-Hodgkin lymphoma.

T-lymphocyte subgroups and the percentage and activity of Human Natural Killer (HNK) cells were investigated in 24 patients suffering from low-grade and 24 patients with high-grade malignancies of non-Hodgkin lymphoma (NHL). The ratio of CD3, CD4, Leu-7, and HNK cells as well as the release by HNK cells of the 51Cr bound to target cells were found decreased, depending on the pathological stage. The tests were performed with OKT-monoclonal sera. A significant change was observed in the reactivity of bone marrow cells to monoclonal sera; the change was identical in character with that observed w

Comparison of alpha beta and gamma delta expressing CD3+ acute lymphoblastic leukemias.

Analysis of 267 consecutive cases of acute lymphoblastic leukemias (ALL) showed that 67 (25%) of cases were of T cell lineage and that 21/67 (31%) expressed the CD3 antigen. Of 13 CD3+ T-ALLs, 7 expressed gamma delta and 6 alpha beta. Comparison of these 2 groups showed differences in clinical and immunophenotypic parameters. The patterns of rearrangement of TCR gamma and TCR delta genes and their junctional nucleotide sequences also differed between the 2 groups. Further analysis of these patients may allow the distinction of subpopulations of ALL which merit altered management.

The flow cytometric crossmatch and early renal transplant loss.

Data from this retrospective study indicate that a positive two-color T and/or B cell flow cytometric crossmatch (FCXM) is predictive of early renal allograft loss (less than 2 months) in cadaveric kidney donor recipients who had a negative crossmatch by the antihuman globulin complement-dependent cytotoxicity technique. Among 90 cadaveric kidney donor recipients (67 primary, 23 regrafts), 14 (8 primary, 6 regrafts) lost their renal allografts within 2 months, and 10 of the 14 were FCXM positive and HLA sensitized. The remaining 76 allografts survived beyond 2 months, 12 of which were FCXM-pos

Identification of the tumor cells in peripheral T-cell lymphomas by combined polymerase chain reaction-based T-cell receptor beta spectrotyping and immunohistological detection with T-cell receptor beta chain variable region segment-specific antibodies.

Most nodal peripheral T-cell lymphomas (PTCL) originate from alphabeta-T cells, and they often contain reactive T cells that may hamper immunophenotyping. To specifically identify the neoplastic population in immunohistochemically stained slides, we assessed the heterogeneity of the T-cell receptor beta chain variable region (TCRVbeta). This region contains 65 gene segments, of which only one is expressed after rearrangement. To investigate PTCL, we developed a polymerase chain reaction assay to define the clonally rearranged TCRVbeta segment. Detecting the corresponding epitope with segment-s

[The significance of CD3/MIB1 and CD3/CD25 in B cell lymphomas].

OBJECTIVE: To identify the distribution and significance of proliferating T cells (PTC) in normal tonsil and a group of B cell lymphomas. METHODS: Double immunohistochemical staining was used to detect PTC with MIB1 expression. Giemsa staining was used to detect Helicobacter pylori (Hp) in gastric MALT-type lymphoma. Cell culture was applied to investigate the relationship among tumor cells, Hp and T cells. RESULTS: Very few T cells expressed MIB1 in the follicular centre of normal tonsil and in that of follicular lymphoma. PTC were mainly distributed to the extrafollicular and T cell zones.

Decreased alpha/beta heterodimer among CD8 molecules of peripheral blood T cells in Wiskott-Aldrich syndrome.

Cell surface CD8 molecules exist as either alpha/alpha homodimers or alpha/beta heterodimers, and most CD8+ T cells that undergo differentiation in the thymus express alpha/beta heterodimers. CD8 molecules on peripheral blood lymphocytes of patients with Wiskott-Aldrich syndrome (WAS), an X-linked immunodeficiency disease, have now been shown to be composed mostly of alpha/alpha homodimers, while the total number of lymphocytes was maintained. This observation may indicate aberrant differentiation of CD8+ T cells in the thymus of WAS. Decreased CD8alpha/beta heterodimer expression, and the rel

AI-guided CAR designs and targeted pathway modulation to enhance multi-antigen CAR T cell durability and overcome antigen escape.

The persistence of CAR T cells and antigen escape remain major barriers to durable therapeutic success in hematologic malignancies. Our study integrates AI-guided design with targeted protein degradation to overcome these challenges. Utilizing an in-silico library of CAR constructs followed by an in vitro screening, we developed a predictive model, CARMSeD, which forecasts constructs prone to self-activation and dysfunction. Optimized bispecific CD20/CD19 CAR T cells demonstrate superior persistence and anti-tumor efficacy. To further improve durability, the platform incorporates a PROTAC-base

CD3+, CD16+, CD56+ cells: a novel subset of T lymphocytes from patients with Balkanic nephropathy.

Balkanic Nephropathy (BN) is characterized by: an incidence limited to a geographic area; a familial character and a slow progressive evolution towards chronic renal failure associated with the symmetrical reduction of the kidney size. The aim of our study was to find out the immune alterations in BN pathology. The analysis of various subpopulations of peripheral lymphocytes obtained from patients with BN showed no quantitative differences in comparison with healthy subjects and healthy subjects from the endemic area. In the BN patients we identified a novel subset of the CD3+, CD16+, CD56+ ce

Computational Design of a TCR-Based Bispecific Engager Targeting Cancerous KRAS G12V Mutations.

Bispecific T-cell engager (BiTE) therapy is a type of immunotherapy that redirects cytotoxic T cells to target tumor cells. Traditionally, a BiTE structure dual binds to the CD3 receptor on the T-cell surface and to the mutated antigen presented on the surface of the cancer cell as an extracellular protein. An example of this is blinatumomab, an FDA-approved BiTE structure for the treatment of B-cell acute lymphoblastic leukemia (B-ALL). However, traditional BiTE complexes are at risk of off-tumor effects due to their targeting of cell surface antigens, which are not specific to certain cancer

Computational Design of a TCR-like Human VH Domain Antibody That Specifically Recognizes a WT1-Derived Peptide Presented by Class I Major Histocompatibility Complex.

T cell receptor (TCR)-like antibodies that recognize a peptide-major histocompatibility complex (pMHC) can enable precise targeting of cancer cells, but developing single-domain binders with high specificity and affinity is challenging. Here, we report the computational design and experimental validation of a TCR-like human heavy chain variable (VH) domain antibody that specifically recognizes the Wilms' tumor 1 protein (WT1)-derived peptide RMFPNAPYL presented by HLA-A*02:01/β2-microglobulin (RMF/HLA-A*02). Expect the unexpected - Loss of surface CD3 on flow cytometry in hepatosplenic T-cell lymphoma: An eye opener.

Hepatosplenic T-cell lymphoma (HSTCL) is a rare extranodal T-cell lymphoma that shows preferential sinusoidal infiltration of spleen and liver. It usually shows bright expression of surface CD3 (sCD3) with restriction for γδ-T cell receptors (TCR). We present a case of a 34-year-old male who presented with hepatosplenomegaly and B symptoms. His peripheral blood and bone marrow (BM) was involved by atypical lymphoid cells that were CD2+, CD7+, CD56+, cytoplasmic CD3+, and sCD3- on immunophenotyping by flow cytometry. As sCD3 is a lineage marker for T-cell lymphomas, the loss of sCD3 posed a dia
