N-(4-diethylamino-2-butynyl)-succinimide, or DKJ-21, is a muscarinic receptor antagonist with a high degree of selectivity for the central nervous system. In the present study of 6 rats maintained under a fixed-interval 50-sec schedule of food reinforcement, atropine and methylatropine reduced responding in a dose dependent manner, while DKJ-21 had little or no effect. Our findings suggest that the suppression caused by atropine and methylatropine may be the result of the dry mouth induced by these agents. Doses of DKJ-21 which had no effect on schedule performance antagonized the rate-lowerin

Both an agonist and its associated prodrug for metabotropic glutamate2/3 (mGlu2/3) receptors demonstrated anxiolytic efficacy in large, randomized, multicenter, double-blind, placebo-controlled trials studying patients with generalized anxiety disorder (GAD). These mGlu2/3 receptor agonists produced robust preclinical anxiolytic-like effects in rodent models. Several different metabotropic glutamate2 receptor positive allosteric modulators have been found to produce antidepressant-like effects on several preclinical screening paradigms, including differential-reinforcement-of-low-rate 72-secon

INTRODUCTION: Patients with moderate or severe traumatic brain injury (TBI) may show impairment in decision- making processes. The Iowa Gambling Task (IGT) has become a widely used neuropsychological research tool to investigate decision-making by simulating real-life decision-making in terms of uncertainty, reward, and punishment.
OBJECTIVE: Investigate decision-making in patients with moderate or severe TBI during performance of the IGT, and study patient's conscious knowledge of the task's reinforcement contingencies (reward/punishment schedule).
PATIENTS AND METHODS: Forty patients with

Similar excitatory or depressant response rate dependent effects on monkeys responding on a variable interval reinforcement schedule were observed following intravenous administration of either thyrotropin releasing hormone (TRH) or thiobarbiturate. However, these agents were mutally antagonistic when given together even though the response rate altering effects of each agent were in the same direction. These findings establish an additional behavioral effect of exogenously administered TRH in primates and suggest that barbiturates might alter behavior in part through an interaction with brain

Athina Markou spent a research period in my laboratory, then in the Department of Anatomy in Cambridge University, in 1991 to help us establish a cocaine-seeking procedure. Thus we embarked on developing a second-order schedule of intravenous cocaine reinforcement to investigate the neural basis of the pronounced effects of cocaine-associated conditioned stimuli on cocaine seeking. This brief review summarizes the fundamental aspects of cocaine seeking measured using this approach and the importance of the methodology in enabling us to define the neural mechanisms and circuitry underlying cond

The transcription factor DeltaFosB accumulates in substance P-dynorphin-containing striatal neurons with repeated cocaine use. Here, we show that inducible transgenic DeltaFosB overexpression in this same striatal cell type facilitates acquisition of cocaine self-administration at low-threshold doses, consistent with increased sensitivity to the pharmacological effects of the drug. Importantly, DeltaFosB also enhances the degree of effort mice will exert to maintain self-administration of higher doses on a progressive ratio schedule of reinforcement, whereas levels of cocaine intake are not al

Clozapine and some other atypical antipsychotics (e.g. quetiapine, olanzapine) have been found to exert a characteristic profile of action on operant behaviour maintained by progressive-ratio schedules, as revealed by Killeen's Mathematical Principles of Reinforcement model of schedule-controlled behaviour. These drugs increase the value of a parameter that expresses the 'incentive value' of the reinforcer (a) and a parameter that is inversely related to the organism's 'motor capacity' (δ). This experiment examined the effects of two further atypical antipsychotics, aripiprazole and amisulprid

The endocannabinoid system interacts with the reward system to modulate responsiveness to natural reinforcers, as well as drugs of abuse. Previous preclinical studies suggested that direct blockade of CB1 cannabinoid receptors (CB1R) could be leveraged as a potential pharmacological approach to treat substance use disorder, but this strategy failed during clinical trials due to severe psychiatric side effects. Alternative strategies have emerged to circumvent the side effects of direct CB1 binding through the development of allosteric modulators. We hypothesized that pharmacological inhibition

Reward uncertainty can sensitize reward pathways, promoting increased reward-seeking and -taking behaviours. This is relevant to human conditions such as pathological gambling, eating disorders and drug addiction. In the context of addiction, preclinical self-administration procedures have been developed to model the intermittency of human drug use. These intermittent-access (IntA) procedures involve intermittent but predictable access to drug during self-administration sessions. However, human drug use typically involves intermittent and unpredictable drug access. We introduce a new procedure

The Ts65Dn mouse displays several phenotypic abnormalities that parallel characteristics found in Down syndrome. One important characteristic associated with Down syndrome is an increased incidence of early-onset Alzheimer's disease. Since Alzheimer's disease is characterized largely by progressive memory loss, it is of interest to study working memory in the Ts65Dn mouse. Previous research in our lab using a titrating, delayed matching-to-position schedule of reinforcement has demonstrated that young, adult male Ts65Dn mice do not display a working memory deficit when compared to age-matched

Previous work suggests a role for dopamine D3-like receptors in psychostimulant reinforcement. The development of new compounds acting selectively at dopamine D3 receptors has opened new possibilities to explore the role of these receptors in animal models of psychostimulant dependence. Here we investigated whether the dopamine D3 partial agonist CJB090 (1-10 mg/kg, i.v) and the D3 antagonist PG01037 (8-32 mg/kg, s.c.) modified methamphetamine (0.05 mg/kg/injection) intravenous self-administration under fixed- (FR) and progressive- (PR) ratio schedules in rats allowed limited (short access, Sh

RATIONALE: Previous work has shown that adult female rats are more sensitive than adult male rats to the reinforcing effects of cocaine, an effect that appears to be due, at least in part, to ovarian hormones.
OBJECTIVE: In this study, we examine sex differences in cocaine self-administration during adolescence, a period of marked hormonal change.
MATERIALS AND METHODS: Adolescent male and female Sprague-Dawley rats were trained to self-administer cocaine (0.75 mg/kg per infusion) under a fixed ratio 1 schedule (i.e., each response was reinforced by an infusion of cocaine) beginning on postn

RATIONALE: We recently found that muscarinic receptor (mAChR) stimulation in the ventral tegmental area (VTA) is involved in the acquisition of a feeding task.
OBJECTIVE: To investigate the involvement of VTA mAChR and nicotinic receptors (nAChR) in the acquisition and performance of a food-rewarded lever-pressing task.
METHODS: In experiment 1 (N=54), rats were trained under a fixed ratio 1 schedule of reinforcement and received bilateral intra-VTA microinjections of scopolamine (0, 2.5 or 5 microg/0.5 microl) or mecamylamine (0, 5 or 10 microg/0.5 microl) before each of the first four sess

Monoamine releasers such as d-methamphetamine (d-MA) can reduce cocaine use in laboratory studies and have been forwarded for the management of cocaine use disorder (CUD). However, the proven abuse liability of d-MA has limited enthusiasm for clinical use. The levorotatory isomer of MA, l-MA, appears to have lesser stimulant effects, possibly due to its preferential norepinephrine-releasing properties compared with dopamine. The present study evaluated the abuse potential of l-MA by comparing its reinforcing effects with known stimulant drugs of abuse in nonhuman primates. Adult rhesus macaque

Key-pecking was maintained under a 30-response fixed-ratio schedule of food presentation and pigeons received 30 mg/kg/day chlorpromazine immediately prior to experimental sessions. After responding stabilized during chronic drug and animals were receiving all available reinforcers, the time available for receiving reinforcers was systematically varied. When that time was decreased, animals initially received fewer reinforcers but responding subsequently increased to an extent that animals once more received all available reinforcers. When that time was again increased, responding once again d

Pausing within multiple fixed-ratio schedules differing in reinforcer magnitude is jointly controlled by both past and upcoming conditions of reinforcement. Abrupt shifts from a just-received large reinforcer to a signaled upcoming small reinforcer (i.e., a negative incentive shift) produce marked disruptions in responding, as indexed by extended pausing. The purpose of this experiment was to determine if reducing the level of food deprivation via prefeeding enhanced these disruptive effects. Five Long Evans rats lever-pressed according to a fixed-ratio schedule. Half of the components ended i

RATIONALE: Performance on progressive ratio schedules has been proposed as a means of assessing the effects of drugs on the value or "efficacy" of reinforcers. A mathematical model affords a basis for quantifying the effects of drugs on progressive ratio schedule performance. According to this model, the relation between response rate and ratio size is described by a bitonic (inverted-U) function. One parameter of the function, alpha, expresses the motivational or "activating" effect of the reinforcer (duration of activation of responding produced by the reinforcer), whereas another parameter,

Operant renewal refers to the relapse of eliminated behavior as a function of context change. Two experiments were performed to assess operant renewal effects in rat subjects with different response location criteria. In both experiments, subjects were trained to respond to a variable-interval schedule on an array of five levers. For Experiment 1, in Context A, only responses on the middle lever were reinforced. For these subjects, response distributions formed a generalization gradient around the middle lever. For Experiment 2, responses on any lever could meet the reinforcement criteria. In

To address the high-rate, interfering stereotypy of three autistic students, a chained schedule for treating stereotypy was combined with skill-based treatment for challenging behavior. Treatment consisted of progressively widening contingencies to differentially reinforce functional communication, toleration, and accurate task completion with escape from instruction to engage in stereotypy. Stimuli were correlated with periods during which instructions were presented and motor stereotypy was redirected (S-) and periods during which escape was provided and motor stereotypy was not redirected (

The rate-dependency principle postulates that "stimulants" tend to increase low baseline rates of response and decrease high baseline responses. One exception to this principle is that stimulants do not typically increase responses that has been extinguished. However, drug self-administration studies showed that stimulant pretreatments reinstated extinguished drug-maintained responses. The goal of this study was to determine the effects of noncontingent cocaine on extinguished food-maintained responding in 3 male cynomolgus monkeys with extensive cocaine self-administration experience.
