[Expression changes in microRNA in the retina of retinal degenerative diseases].

Because visual information accounts for 80-90% of sensory information that we get from our circumstance, loss of vision seriously diminishes our quality of life. According to a recent epidemiological study, glaucoma is the first, and retinitis pigmentosa (RP) is the second leading causes of acquired blindness in Japan. Degeneration of the retinal ganglion cells (RGC) and photoreceptor cells causes glaucoma and RP, respectively. Intraocular pressure-lowering therapy is an only effective treatment for glaucoma, and the agents that protect RGC directly against glaucomatous injury have not been av

The Interplay between Peripherin 2 Complex Formation and Degenerative Retinal Diseases.

Peripherin 2 (Prph2) is a photoreceptor-specific tetraspanin protein present in the outer segment (OS) rims of rod and cone photoreceptors. It shares many common features with other tetraspanins, including a large intradiscal loop which contains several cysteines. This loop enables Prph2 to associate with itself to form homo-oligomers or with its homologue, rod outer segment membrane protein 1 (Rom1) to form hetero-tetramers and hetero-octamers. Mutations in PRPH2 cause a multitude of retinal diseases including autosomal dominant retinitis pigmentosa (RP) or cone dominant macular dystrophies.

Suprachoroidal Delivery of Viral and Nonviral Gene Therapy for Retinal Diseases.

Retinal gene therapy is a rapidly growing field with numerous clinical trials underway, and route of delivery is a critical contributor to its success. Subretinal administration, which involves pars plana vitrectomy in the operating room, offers targeted delivery to retinal-pigment epithelium cells and photoreceptors. Due to the immune-privileged nature of the subretinal space, the risk of an immune reaction against viral capsid antigens is minimized, an advantage of subretinal administration in patients with preexisting neutralizing antibodies. Intravitreal administration, with fewer procedur

Retinal oximetry: Metabolic imaging for diseases of the retina and brain.

Retinal oximetry imaging of retinal blood vessels measures oxygen saturation of hemoglobin. The imaging technology is non-invasive and reproducible with remarkably low variability on test-retest studies and in healthy cohorts. Pathophysiological principles and novel biomarkers in several retinal diseases have been discovered, as well as possible applications for systemic and brain disease. In diabetic retinopathy, retinal venous oxygen saturation is elevated and arteriovenous difference progressively reduced in advanced stages of retinopathy compared with healthy persons. This correlates with

Applications of antisense oligonucleotides for the treatment of inherited retinal diseases.

PURPOSE OF REVIEW: Over the last years, antisense oligonucleotides (AONs) have gained attention as a therapeutic tool for the treatment of ocular diseases such as cytomegalovirus retinitis, keratitis-induced corneal neovascularization, and inherited retinal diseases (IRDs). In this review, we summarize the recent key findings, and describe the challenges and opportunities that lie ahead to translate AON-based therapies to the clinic, in particular for IRDs. RECENT FINDINGS: The efficacy of AONs to restore splice defects and cellular phenotypes associated with a common mutation in CEP290 was d

Trophic factors in the pathogenesis and therapy for retinal degenerative diseases.

Trophic factors are endogenously secreted proteins that act in an autocrine and/or paracrine fashion to affect vital cellular processes such as proliferation, differentiation, and regeneration, thereby maintaining overall cell homeostasis. In the eye, the major contributors of these molecules are the retinal pigment epithelial (RPE) and Müller cells. The primary paracrine targets of these secreted proteins include the photoreceptors and choriocapillaris. Retinal degenerative diseases such as age-related macular degeneration and retinitis pigmentosa are characterized by aberrant function and/or

[Pigment epithelium-derived factor and its application in retinal angiogenic diseases].

Retinal neovascularization (RNV) is one of the most important causes of blindness. No efficient treatment for RNV is available. Pigment epithelium-derived factor (PEDF) is a natural inhibitor of angiogenesis. It plays an important role in the modulation of ocular neovascularization. It has been demonstrated recently that the ischemia-induced retinal neovascularization can be dramatically inhibited by the recombinant PEDF protein or local viral vector-mediated delivery of PEDF cDNA, which predicts PEDF to be an optional strategy for intractable retinal angiogenic diseases like diabetic retinopa

[Diagnostic references in optic nerve diseases using retinal nerve fiber photography].

Two patients with optic nerve disease are reported. The first patient was 20 years old and had suffered a closed head injury 1 year earlier. She presented with an incongruous right hemianopsia; retinal nerve fiber layer photography disclosed an infragenicular lesion and descending atrophy of the medial postchiasmal parts of the optic tract. The second patients was 24 years old. He had had depressed visual acuity in his right eye for 7 h before presentation. Perimetry showed a quadrant anopsia of the lower nasal field. The diagnosis made on the basis of fluorescein-angiography was recurrent ret

Tubercular Retinitis and Retinal Vasculitis: Manifestations of a Shared Disease Spectrum.

Isolated retinal involvement in ocular tuberculosis (TB) is not well understood and remains largely underreported. TB retinal lesions can be classified into two broad categories: retinal vasculitis and isolated retinal granuloma. TB retinal vasculitis is the more recognized phenotype and may present with perivascular infiltrates alone or alongside other ocular signs such as subvascular lesions, focal vascular tortuosity, and occlusive vasculitis. These additional signs are crucial for diagnosing TB retinal vasculitis in TB-endemic regions. However, isolated retinal granulomas due to TB are rar

Microvascular Diseases of the Eye: Retinal Vasculature as a Cardiometabolic Biomarker.

Microvascular disease of the eye encompasses both structural and functional changes to the retinal vessels. Cardiometabolic disorders, such as hypertension and diabetes, are associated with early, measurable abnormalities in the retinal microvascular network that often emerge before cardiovascular disease becomes detectable. These features can be directly, noninvasively assessed with a multitude of evolving imaging techniques. Furthermore, these vascular changes can be used to predict future cardiac events, making them promising biomarkers for systemic disease. Early detection is essential, as

Gene-Agnostic Therapeutic Strategies for Inherited Retinal Diseases: Neuroprotection and Immunomodulation.

Background/Objectives: Inherited retinal diseases (IRDs) represent a genetically heterogeneous group of disorders caused by mutations in over 280 genes with more than 3100 identified variants. While gene-specific replacement therapies have achieved landmark success with voretigene neparvovec (Luxturna) for biallelic RPE65-associated retinal dystrophy, developing individual therapies for each genetic subtype remains impractical. This review examines gene-agnostic therapeutic approaches utilizing neuroprotection and immunomodulation that target common pathophysiological mechanisms shared across

Vitreoretinal procedures in inherited retinal diseases: a large retrospective cohort study.

OBJECTIVE: Management of inherited retinal diseases (IRDs) traditionally has been viewed as nonsurgical, but there are known complications of IRDs that may warrant surgical intervention. Patterns of procedural interventions among patients with IRDs remain poorly characterized, given their relatively small representation among individual institutions. The objective of this study is to characterize patterns of surgical diagnoses and procedures in patients with IRD compared with a control cohort. DESIGN: This retrospective cohort study used deidentified aggregated electronic health records from

Application of Nanomaterials in the Diagnosis and Treatment of Retinal Diseases.

In recent years, nanomaterials have demonstrated broad prospects in the diagnosis and treatment of retinal diseases due to their unique physicochemical properties, such as small-size effects, high biocompatibility, and functional surfaces. Retinal diseases are often accompanied by complex pathological microenvironments, where conventional diagnostic and therapeutic approaches face challenges such as low drug delivery efficiency, risks associated with invasive procedures, and difficulties in real-time monitoring. Nanomaterials hold promise in addressing these limitations of traditional therapie

Sialylation as a checkpoint for inflammatory and complement-related retinal diseases.

Sialylation is a modification process involving the addition of sialic acid residues to the termini of glycoproteins and glycolipids in mammalian cells. Sialylation serves as a crucial checkpoint inhibitor of the complement and immune systems, particularly within the central nervous system (CNS), including the retina. Complement factor H (FH), complement factor properdin (FP), and sialic acid-binding immunoglobulin-like lectin (SIGLEC) receptors of retinal mononuclear phagocytes are key players in regulating the complement and innate immune systems in the retina by recognizing sialic acid (Sia

Potential role of endoplasmic reticulum quality control in retinal degenerative diseases.

The endoplasmic reticulum (ER) is the largest organelle in eukaryotic cells, and it plays a crucial role in regulating various biological processes, including protein folding, translation, and structural maturation. Accurate protein modification is essential for maintaining oxidative stress, apoptosis, and cellular senescence in the organism. The regulation of protein homeostasis involves three biological processes: endoplasmic reticulum stress (ERS), endoplasmic reticulum autophagy (ERPA), and endoplasmic reticulum-associated degradation (ERAD). Retinal degenerative disease (RDD) is a blindin

Artificial intelligence-enhanced retinal imaging as a biomarker for systemic diseases.

Retinal images provide a non-invasive and accessible means to directly visualize human blood vessels and nerve fibers. Growing studies have investigated the intricate microvascular and neural circuitry within the retina, its interactions with other systemic vascular and nervous systems, and the link between retinal biomarkers and various systemic diseases. Using the eye to study systemic health, based on these connections, has been given a term as oculomics. Advancements in artificial intelligence (AI) technologies, particularly deep learning, have further increased the potential impact of thi
