Development of the human retina in the absence of ganglion cells.

Retinal development was studied in eyes from fetal and neonatal human anencephalic (AnC) and normal age-matched infants to determine the time of retinal ganglion cell (GC) loss and its effect on the development of other retinal neurons. At fetal week (Fwk) 14, GC loss was evident in central retina and by Fwk 19-20 almost all GC were absent, although immunocytochemical labeling for GC markers brain 3, neurofilament M and parvalbumin detected a few GC in the AnC far periphery at older ages. The inner nuclear and inner plexiform (IPL) layers showed variable amounts of thinning but all normal bipo

[A 50-year-old man with metamorphopsia and optic nerve swelling].

We report a 50-year-old male having metamorphopsia associated with retinal edema. He was well until one month prior to the present admission when he developed occipital headache and backache. Three weeks later, he noted a sudden onset of twisting of visual images. On admission, he was in no acute distress with well preserved general conditions. Only neurological abnormalities were bilateral papilledema, retinal edema and horizontal nystagmus with a rotatory component. Routine blood chemistries were unremarkable. The CSF contained 28 cells/cmm with 60% consisting of large atypical cells. Crania

Controlled delivery of tauroursodeoxycholic acid from biodegradable microspheres slows retinal degeneration and vision loss in P23H rats.

Successful drug therapies for treating ocular diseases require effective concentrations of neuroprotective compounds maintained over time at the site of action. The purpose of this work was to assess the efficacy of intravitreal controlled delivery of tauroursodeoxycholic acid (TUDCA) encapsulated in poly(D,L-lactic-co-glycolic acid) (PLGA) microspheres for the treatment of the retina in a rat model of retinitis pigmentosa. PLGA microspheres (MSs) containing TUDCA were produced by the O/W emulsion-solvent evaporation technique. Particle size and morphology were assessed by light scattering and

Retrograde transneuronal degeneration in the retina and lateral geniculate nucleus of the V1-lesioned marmoset monkey.

Retrograde transneuronal degeneration (RTD) of retinal ganglion cells and dorsal lateral geniculate (LGN) neurons are well described following a lesion of the primary visual cortex (V1) in both Old World monkeys and humans. Based on previous studies of New World monkeys and prosimians, it was suggested that these species displayed no RTD following a lesion of V1. In this study of the New World marmoset monkey, 1 year after a unilateral V1 lesion either in adults or at 14 days after birth, we observed ~20 % ganglion cell (GC) loss in adult but ~70 % in infants. This finding is similar to the RT

The receptive field construction of midget ganglion cells in primate retina.

The midget pathway of the primate retina provides the visual system with the foundations for high spatial resolution and color perception. An essential contributor to these properties is center-surround organization, in which responses from the central area of a cell's receptive field are antagonized by responses from a surrounding area. Two key questions about center-surround organization are unresolved. First, the surround is largely or completely due to negative feedback from horizontal cells to cones: how can this feedback be reconciled with the popular difference of Gaussians (DOG) model,

Second and third visual areas of the cat: interindividual variability in retinotopic arrangement and cortical location.

1. The cortical location and the retinotopic arrangement of the second (V2) and third (V3) visual areas in the cat have been investigated with single and multiple unit recordings in anaesthetized and immobilized animals.2. V2 and V3 are arranged side by side anterior and medial to V1 and occupy the lateral gyrus and the postlateral sulcus. In addition, V2 spreads to postlateral parts of the lateral sulcus and, occasionally, to the posterior suprasylvian gyrus. The contralateral lower hemifield is represented on the lateral gyrus, the area centralis and the horizontal meridian are found in most

Retinal ganglion cell population in adult albino and pigmented mice: a computerized analysis of the entire population and its spatial distribution.

UNLABELLED: In adult Swiss albino and C57 pigmented mice, RGCs were identified with a retrogradely transported neuronal tracer applied to both optic nerves (ON) or superior colliculi (SCi). After histological processing, the retinas were prepared as whole-mounts, examined and photographed under a fluorescence microscope equipped with a motorized stage controlled by a commercial computer image analysis system: Image-Pro Plus((R)) (IPP). Retinas were imaged as a stack of 24-bit color images (140 frames per retina) using IPP with the Scope-Pro plug-in 5.0 and the images montaged to create a high-

More insights from Abca4-/- mouse models of recessive Stargardt disease.

Mutations in the ABC transporter ABCA4 (ABC, subfamily A, member 4) are responsible for recessive Stargardt disease 1 (STGD1), a juvenile form of macular degeneration. In preclinical and clinical studies, it has been shown that deficiency in ABCA4 leads to accelerated formation of the toxic bisretinoid fluorophores that form as the product of nonenzymatic reactions of retinaldehyde with phosphatidylethanolamine (2:1 ratio). Here, by comparing photoreceptor cell viability in albino versus agouti Abca4-/- mice and by dark-rearing albino Abca4-/- mice, we show that photoreceptor cell degeneration

"Slow activity transients" in infant rat visual cortex: a spreading synchronous oscillation patterned by retinal waves.

A primary feature of the preterm infant electroencephalogram is the presence of large infra-slow potentials containing rapid oscillations called slow activity transients (SATs). Such activity has not been described in animal models, and their generative mechanisms are unknown. Here we use direct-current and multisite extracellular, as well as whole-cell, recording in vivo to demonstrate the existence of regularly repeating SATs in the visual cortex of infant rats before eye opening. Present only in absence of anesthesia, SATs at postnatal day 10-11 were identifiable as a separate group of long

pH in the vertebrate retina and its naturally occurring and pathological changes.

This review summarizes the existing information on the concentration of H+ (pH) in vertebrate retinae and its changes due to various reasons. Special features of H+ homeostasis that make it different from other ions will be discussed, particularly metabolic production of H+ and buffering. The transretinal distribution of extracellular H+ concentration ([H+]o) and its changes under illumination and other conditions will be described in detail, since [H+]o is more intensively investigated than intracellular pH. In vertebrate retinae, the highest [H+]o occurs in the inner part of the outer nuclea

Is the melatonin receptor type 1 involved in the pathogenesis of glaucoma?

Melatonin in the mammalian eye is synthesized by the photoreceptors and its levels show a clear daily pattern with high levels at night and lower levels during the day. It is synthesized in the ciliary body and secreted into the aqueous humor with a pattern similar to what has been reported for the retina. It acts by interacting with a family of G-protein coupled receptors that are negatively coupled with adenylate cyclase. Melatonin receptor subtypes MT1 and MT2 have been identified in the retina. Both are found in the inner nuclear layer (horizontal and amacrine cells), in the inner plexifor

Approaching a connectome of the human foveal retina.

The foveal retina is a unique primate specialization and a promising target for the first connectome of a human central nervous system structure. In the fovea, neural cells and circuits are miniaturized and compressed to sample the visual image at high spatial resolution, initiating form, color, and motion perception. Here, we report a working-draft human foveal connectome. We used deep learning to segment all cells and their synaptic connections in a piece of human fovea. We classified ~3,000 cells into 51 neuronal types and three glial types based on morphology and connectivity. Some cell ty

Ocular comparative anatomy of the family Rodentia.

There is little information regarding ocular anatomy and histology in many of the rodent species. Histological analyses for morphologic features were performed in 31 globes from 18 rodent species submitted to and archived at the Comparative Ocular Pathology Laboratory of Wisconsin. The following measurements were taken: thickness of the cornea, corneal epithelium, corneal stroma, Descemet's membrane, and retina. H&E sections were evaluated for the following anatomical features: presence of pigmented epithelial cells in the peripheral cornea, presence and location of Schlemm's canal, presence o

Oculomotor deficits in aryl hydrocarbon receptor null mouse.

The Aryl hydrocarbon Receptor or AhR, a ligand-activated transcription factor, is known to mediate the toxic and carcinogenic effects of various environmental pollutants such as 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD). Recent studies in Caenorhabditis elegans and Drosophila melanogaster show that the orthologs of the AhR are expressed exclusively in certain types of neurons and are implicated in the development and the homeostasis of the central nervous system. While physiological roles of the AhR were demonstrated in the mammalian heart, liver and gametogenesis, its ontogenic expression an
