Polyunsaturated fatty acid metabolism in the retinal pigment epithelium and its association with outer retinal disease.

Polyunsaturated fatty acids (PUFAs) are essential for photoreceptor (PR) development, structure and function, and their availability in the outer retina is predominantly mediated by the retinal pigment epithelium (RPE), a cellular monolayer juxtaposed to the PR outer segments. Because PRs lack enzymatic machinery to generate critical PUFA intermediates from dietary precursors, they rely on the RPE to coordinate multiple steps of PUFA uptake, metabolism and export. Consequently, disruption in any of these steps perturbs PR homeostasis and compromises overall retinal health. In this review, we s

Retinal Pigment Epithelium Transplantation in Retinal Disease: Clinical Trial Development, Challenges, and Future Directions.

Replacement of the retinal pigment epithelium (RPE) is emerging as a promising approach to treat degenerative retinal diseases, including age-related macular degeneration and Stargardt disease, in which RPE function cannot otherwise be restored. Despite the limitations of existing treatments, advances in cell sourcing and surgical methods have enabled initial human trials of RPE transplantation, with early results indicating potential efficacy. This review comprehensively examines the evolution of RPE transplantation in recent decades, highlighting the advantages and limitations of different c

Retinal Pigment Epithelial Tear and Retinal Detachment Following Hypotony After Trabeculectomy: A Case Report.

Hypotony after trabeculectomy (TLE) can lead to choroidal detachment (CD) and maculopathy. We present a case of retinal detachment caused by extensive retinal pigment epithelial (RPE) tears following hypotony following TLE surgery. A 67-year-old man underwent TLE in the left eye for open-angle glaucoma. Postoperatively, intraocular pressure (IOP) remained stable at approximately 10 mmHg. However, after 50 days, the IOP decreased due to aqueous humor leakage, which required several conjunctival sutures. CD occurred on day 87. On day 108, retinal detachment was detected in the inferior retina of

The toxic effects of polystyrene microplastic/nanoplastic particles on retinal pigment epithelial cells and retinal tissue.

The increasing use of contact lenses, artificial tears, and anti-vascular endothelial growth factor (anti-VEGF) drug injections for age-related macular degeneration has heightened the likelihood of eye exposure to microplastic particles. Extensive research has established that microplastic particles can induce oxidative stress on the ocular surface, resulting in damage. However, the impact of these particles on the retina remains unclear. Therefore, this study investigated whether microplastics/nanoplastics (MPs/NPs) cause retinal damage. In vitro human retinal pigment epithelial (RPE) cells w

New Retinal Pigment Epithelial Cell Model to Unravel Neuroprotection Sensors of Neurodegeneration in Retinal Disease.

Retinal pigment epithelial (RPE) cells sustain photoreceptor integrity, and when this function is disrupted, retinal degenerations ensue. Herein, we characterize a new cell line from human RPE that we termed ABC. These cells remarkably recapitulate human eye native cells. Distinctive from other epithelia, RPE cells originate from the neural crest and follow a neural development but are terminally differentiated into "epithelial" type, thus sharing characteristics with their neuronal lineages counterparts. Additionally, they form microvilli, tight junctions, and honeycomb packing and express di

Retinal detachment with retinal pigment epithelial tear under hypotony after trabeculectomy: A case report.

PURPOSE: To report a case of retinal detachment with retinal pigment epithelium (RPE) tear, under hypotony after trabeculectomy, which immediately resolved upon elevation of intraocular pressure. OBSERVATIONS: A 67-year-old man who had undergone trabeculectomy presented for treatment of a visual field defect in his left eye. His glaucoma surgery was uneventful. At his first visit to our clinic, the patient's visual acuity was 15/20 in the right eye and 20/40 in the left; intraocular pressure was 18 mmHg in the right and 5 mmHg in the left. Fundus examination of the left eye revealed a retinal

Protective effects of human iPS-derived retinal pigmented epithelial cells on retinal degenerative disease.

BACKGROUND: Retinitis pigmentosa (RP) is an inherited retinal disease characterized by progressive loss of photoreceptor cells. This study aim at exploring the effect of retinal pigment epithelium (RPE) derived from human-induced pluripotent stem cell (hiPSC-RPE) on the retina of retinal degeneration 10 (rd10) mice, which are characterized with progressive photoreceptor death. METHODS: We generated RPE from hiPSCs by sequential supplementation with retinal-inducing factors and RPE specification signaling factors. The three-dimensional (3D) spheroid culture method was used to obtain optimal in

Retinal Pigment Epithelium Cell Death Is Associated With NLRP3 Inflammasome Activation by All-trans Retinal.

PURPOSE: Visual (retinoid) cycle anomalies induce aberrant build-up of all-trans retinal (atRAL) in the retinal pigment epithelium (RPE), which is a cause of RPE atrophy in Stargardt disease type 1 and age-related macular degeneration. NLR family pyrin domain containing 3 (NLRP3) inflammasome activation is implicated in the etiology of age-related macular degeneration. Here, we elucidated the relationship between NLRP3 inflammasome activation and atRAL-induced death of RPE cells. METHODS: Cellular toxicities were assessed by MTS or MTT assays. Expression levels of mRNAs and proteins were dete

Polarized retinal pigment epithelium generates electrical signals that diminish with age and regulate retinal pathology.

The transepithelial potential difference (TEP) across the retinal pigment epithelial (RPE) is dependent on ionic pumps and tight junction "seals" between epithelial cells. RPE cells release neurotrophic growth factors such as pigment epithelial derived factor (PEDF), which is reduced in age-related macular degeneration (AMD). The mechanisms that control the secretion of PEDF from RPE cells are not well understood. Using the CCL2/CX3CR1 double knockout mouse model (DKO), which demonstrates RPE damage and retinal degeneration, we uncovered an interaction between PEDF and the TEP which is likely

In vivo response of the rat's retinal pigment epithelium to azide at advanced stages of hereditary retinal dystrophy.

Electrophysiological properties of the retinal pigment epithelium (RPE) were studied in the rat with hereditary retinal dystrophy (rdy). Transocular potential changes evoked by intravenous bolus injections of azide and thiocyanate (SCN-) are the only available indication of RPE state when degeneration of rods is in progress. Also determined were age-dependent decrease in retinal DNA content and in counts of cones that survive after degeneration of rods. The azide response in the pigmented and albino rdy rat was already reduced at the earliest age tested (60 d) and continued to decrease till th

[A hormone-sensitive stage in the development of the retinal pigment epithelium in Hunter rats with hereditary retinal dystrophy].

The sensitivity of the retinal pigment epithelium (RPE) to the melanotropic effects of alpha-MSH and dbcAMP was assayed in an organ culture of the eye scleral part in the Hunter rats with inherited retinal dystrophy. The melanin synthesis was estimated by liquid scintillation on the RPE isolated enzymatically after 48-h cultivation. One eye from every animal was cultivated in a medium without natural components and with the hormone or dbcAMP (experiment), while the other in a hormone-free medium (control). The melanin synthesis was estimated by 14C-thiouracil incorporation. The result was expr

Development of traction retinal detachments following intravitreal injections of retinal Muller and pigment epithelial cells.

We injected varying numbers of retinal Muller glia into the rabbit vitreous in an established model of traction retinal detachment. We used indirect ophthalmoscopy to observe the changes elicited during the following 1 month. Although the severity of the tractional changes increased with increasing numbers of the glial cells, the pathology produced stabilized within the 1st week of injury. Muller glia were less effective at eliciting retinal detachments than retinal pigment epithelial cells (RPE) or mixtures of glia and RPE. Intravitreal tissue membranes derived from the glia differed morpholo

Evolving Congenital Hypertrophy of the Retinal Pigment Epithelium in a Pediatric Patient.

Purpose: To describe the longitudinal evolution of a solitary congenital hypertrophy of the retinal pigment epithelium lesion demonstrating progressive enlargement and increasing border irregularity over time. Methods: A single case was reviewed. Results: A 13-year-old boy presented with an evolving and enlarging congenital hypertrophy of the retinal pigment epithelium lesion over a 7-year period. Fundus images obtained when the patient was 7 years old showed a solitary, circular lesion with a hypopigmented center and mildly irregular, hyperpigmented borders that measured 1.5 mm × 1.5 mm. Molecular underpinnings of induced degenerative heterogeneity in the retinal pigment epithelium.

Cigarette smoking induces epigenetic changes that can cause degenerative heterogeneity with aging and disease. In disease such as age-related macular degeneration (AMD), the leading worldwide cause of blindness among the elderly, retinal pigment epithelial (RPE) cell heterogeneity is a key change. Since smoking is a powerful risk factor for AMD, we hypothesized that smoke induces epigenetic-mediated degenerative RPE heterogeneity. We administered cigarette smoke condensate (CSC) to young and aged mice. Using snRNA-seq and single nuclear ATAC sequencing, we identified distinct healthy and dedif

Stress Inference in Retinal Pigment Epithelium in C57BL/6J Mouse.

PURPOSE: To quantitatively map the spatial architecture of mechanical stress and pressure in the retinal pigment epithelium (RPE) and to test the central hypothesis that the mechanical state of normal mouse RPE is spatially patterned from center to periphery and that this spatial pattern is maintained during adult aging. METHODS: Whole-mount flat images of C57BL/6J mouse RPE spanning multiple age groups were analyzed using a tissue-scale stress inference framework under the assumption of mechanical equilibrium. Individual RPE cells were segmented, intercellular junctions were skeletonized, an

Retinal Pigment Epithelial Detachment Following Topical Travoprost Therapy.

Prostaglandin analogs, including travoprost, are widely used to lower intraocular pressure in primary open-angle glaucoma. While generally safe, rare posterior segment changes have been reported. Retinal pigment epithelial detachment (RPED) associated with travoprost is uncommon and poorly documented. We report three patients who developed serous RPED shortly after initiating topical travoprost, none of whom had preexisting macular disease. Optical coherence tomography (OCT) confirmed the detachments, and complete anatomical and visual recovery occurred following discontinuation of travoprost

Polarized secretion of mitochondrial DNA by retinal pigment epithelial cells.

Mitochondrial DNA (mtDNA) damage is strongly implicated in age-related macular degeneration (AMD), the most frequent cause of age-mediated visual impairment in developed countries. Here, we investigated and compared the fate of acutely induced oxidation damage in primary retinal pigment epithelial (RPE) cells. The individual RPE clones responded heterogeneously to hydrogen peroxide, both with respect to cell sensitivity and mtDNA damage formation. Peroxide-induced mtDNA damage in human RPE (hRPE) was fully repaired within 4 h. In parallel, hRPE cells secreted mtDNA from both apical and basolat

IGFBP5 as a Novel Basolateral Secretion Marker in the Retinal Pigment Epithelium.

The retinal pigment epithelium (RPE) is a highly polarized cell type in the retina which is specialized to support the photoreceptors and choroid. To assess the adequacy of RPE polarity in culture or altered RPE polarity under disease conditions in vivo, the polarized secretion of particular proteins can be monitored. Currently accepted basolateral secretion markers for RPE include the pro-angiogenic factor, VEGF, and extracellular matrix remodeling protein, TIMP-3. However, VEGF secretion is low enough that it often requires ELISA for detection, and TIMP-3 secretion is only modestly polarized
