Strong evidence indicates that remodeling gut microbiota may be an effective approach to combat Parkinson's disease (PD). Scorpion Venom Heat-Resistant Synthesized Peptide (SVHRSP), a synthesized peptide discovered from scorpion venom, displays potent neuroprotection in multiple PD models. However, the potential mechanisms remain unclear. In this study, we demonstrated that SVHRSP effectively attenuated gastrointestinal function impairments and reinstated the microbiota composition in rotenone-induced PD mouse model. Microbiota depletion and FMT verified that the restored gut microbiota was ne

Pachyrhizus erosus seeds have been reported to have various biological activities, including antifungal, antisecretory, insecticidal, antibacterial, and antispasmodic properties. In this study, we evaluated the hypopigmentation effects of the ethanol extract of Pachyrhizus erosus seeds (PESE), identified rotenone as a representative active metabolite, and proposed a mechanism for inhibiting α-MSH-mediated melanogenesis in B16F10 cells. PESE treatment effectively inhibited melanin synthesis in B16F10 cells stimulated with α-MSH or forskolin. Among the three major metabolites characterized from

Over time, Parkinson disease (PD) develops as a neurological illness. The goal of this study was to see whether ferulic acid has any neuroprotective benefits on the cerebellum of rats that have Parkinson's disease brought on by rotenone poisoning. A total of twenty-four male albino rats, in good condition, weighed between 200 and 250 g and nine to ten weeks old, were employed in the investigation. The control group received 1 ml of sunflower oil intraperitoneally (i.p.) each day. Rats' motor performance was considerably worse when given rotenone than it was in the control group. Rats given Fer

BACKGROUND: We aimed to investigate the effects of whey protein (WP) supplements in a rat model of rotenone-induced locomotor and biochemical features of Parkinson's disease (PD).
MATERIALS AND METHODS: Male Wistar rats were used. Daily injections of rotenone (2 mg/kg; i.p.) for 16 days were used to induce PD. WP or soy protein (SP) at 1, 2, and 4 g/rats were administrated daily by gavage. Motor skills were measured in rats 24 h after the last injection using the bar test, grid test, rearing, and open field tests. In the following, striatum tissue was isolated for biochemical measurements. EL

ETHNOPHARMACOLOGICAL RELEVANCE: In traditional Chinese medicine, scorpion is used to treat diseases with symptoms such as trembling, convulsion and dementia. Our laboratory employs patented technology to extract and purify the active single component from scorpion venom. We then utilize mass spectrometry to determine the amino acid sequence of the polypeptide and synthesize it artificially to acquire the polypeptide with a purity of 99.3%, named SVHRSP (Scorpion Venom Heat-Resistant Peptide). SVHRSP has been demonstrated to display potent neuroprotective efficacy in Parkinson's disease.

Mild hypothermia is a well-established technique for alleviating neurological injuries in clinical surgery. RNA-binding protein motif 3 (RBM3) has been identified as a crucial factor in mediating hypothermic neuroprotection, providing its induction as a promising strategy for mimicking therapeutic hypothermia. However, little is known about molecular control of RBM3 and signaling pathways affected by hypothermia. In the present study, human SH-SY5Y neuroblastoma cells were used as a neural cell model. Screening of signaling pathways showed that cold exposure led to inactivation of ERK and AMPK

Parkinson's disease (PD) is the second most common age-related neurodegenerative disease after Alzheimer's disease, characterized by loss of dopaminergic neurons in substantia nigra pars compacta, accompanied by motor and nonmotor symptoms. The neuropathological hallmarks of PD are well reported, but the etiology of the disease is still undefined; several studies assume that oxidative stress, mitochondrial defects, and neuroinflammation play vital roles in the progress of the disease. The current study was established to investigate the neuroprotective effect of agmatine on a rotenone (ROT)-in

Parkinson's disease (PD) is a major age-related neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra par compacta (SNpc). Rotenone is a neurotoxin that is routinely used to model PD to aid in understanding the mechanisms of neuronal death. Safflower (Carthamus tinctorius. L.) has long been used to treat cerebrovascular diseases in China. This plant contains flavonoids, which have been reported to be effective in models of neurodegenerative disease. We previously reported that kaempferol derivatives from safflower could bind DJ-1, a protein associ

INTRODUCTION: Parkinson's disease (PD), ranking as the second most common neurodegenerative disorders following Alzheimer's disease, involves the progressive accumulation of misfolded proteins in affected neural tissues. This pathological process appears linked to overproduction of reactive oxygen species (ROS) and reactive nitrogen species (RNS). Significantly, more than 30% of proteins aggregated in Lewy bodies exhibit post-translational modifications, particularly RNS-mediated nitration and S-nitrosylation. Experimental evidence suggests that α-synuclein nitration promotes its misfolding an

Parkinson's disease (PD) is the second most common neurodegenerative disorder that is characterized by two major neuropathological hallmarks: the degeneration of dopaminergic neurons in the substantia nigra (SN) and the presence of Lewy bodies in the surviving SN neurons, as well as other regions of the central and peripheral nervous system. Animal models have been invaluable tools for investigating the underlying mechanisms of the pathogenesis of PD and testing new potential symptomatic, neuroprotective and neurorestorative therapies. However, the usefulness of these models is dependent on ho

We investigated the effect of the nucleoside uridine on the activities of key enzymes of oxidative phosphorylation (succinate dehydrogenase, SDH) and glycolysis (lactate dehydrogenase, LDH) in peripheral blood lymphocytes, and on oxidative metabolism in the blood serum of rats in the rotenone and 6-OHDA models of Parkinson's disease. Both models were characterized by increased SDH and LDH activities in lymphocytes and accumulation of lipid peroxides in the blood serum. Injections of uridine, affecting the functioning of mitochondrial K-ATP channel (mitoKATP), to animals at a dose of 30 mg/kg b

Despite years of research, it is still unclear why iron accumulates in the substantia nigra (SN) of Parkinson's disease (PD) animal models and PD patients. To address this, we investigated the changes in serum iron, iron content, transferrin receptor (TfR1), ferroportin 1 (Fpn1), hepcidin, and iron regulatory proteins (IRPs) expression in the SN, and hepcidin in serum and cerebrospinal fluid (CSF) in rotenone-induced PD rat model, as well as iron content and hepcidin in rotenone-induced PD cell models. We demonstrate that rotenone induces PD-like dyskinesia, dopaminergic neuron loss, as well a

INTRODUCTION: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and non-motor dysfunction resulting from dopaminergic neuronal loss. Emerging evidence suggests a role of the renin-angiotensin system (RAS) in neurodegeneration, making it a potential therapeutic target. The present study aimed to evaluate the behavioral effects of captopril, perindopril, and losartan on motor, behavioral, and anxiety parameters in a rotenone-induced model of Parkinson's disease in Wistar albino rats.
MATERIALS AND METHODS: An experimental in vivo study was conducted usi

Oxidative stress and changes in lncRNA expression are key factors in the pathophysiology of Parkinson's disease. This study investigated the protective effects of N-acetylcysteine and S-methylcysteine on the expression of long non-coding RNAs and oxidative stress-related genes in the brain, as well as the activity of antioxidant enzymes in the brain and serum of mice with rotenone-induced Parkinson's disease. In this experimental study, 56 male BALB/c mice were utilized and treated continuously for 10 days. Gene expression of superoxide dismutase, glutathione peroxidase, catalase, Nrf2, Ho-1,

Parkinson's disease (PD) is a neurodegenerative disease characterized by motor and non-motor symptoms, often associated with the accumulation of α-synuclein, neuroinflammation and oxidative stress in the central and peripheral systems. Considering that patients with Parkinson's disease often have gastrointestinal symptoms, this study aimed to investigate whether a rotenone-induced rat model of PD can induce intestinal changes and the neuroprotective effects of a cannabidiol (CBDne) nanoemulsion formulation, focusing on the striatum and intestine. Wistar rats were divided into six groups: contr

ETHNOPHARMACOLOGICAL RELEVANCE: 'Karkataka Taila (KT), an ancient Ayurvedic Rasayana comprising the edible freshwater crab Scylla serrata Forskal flesh, is still used by local traditional practitioners in Kerala state to treat tremors and palsy. In the scientific community, it becomes less exposed due to the lack of adequate scientific validations and brief reports. There has been no published research on the effectiveness of KT in treating Parkinson's disease (PD).
PURPOSE: The purpose of the current research work was to investigate the anti-Parkison's potential of KT against rotenone-induce

Background: Rotenone (ROT) is currently being used in various research fields, especially neuroscience. Separated from other neurotoxins, ROT induces a Parkinson’s disease (PD)-related phenotype that mimics the associated clinical spectrum by directly entering the central nervous system (CNS). It easily crosses through the blood−brain barrier (BBB) and accumulates in mitochondria. Unfortunately, most of the existing data focus on locomotion. This is why the present study aimed to bring novel evidence on how ROT alone or in combination with different potential ant(agonists) might influence the

Although sexual health is affected by Parkinson's disease (PD), the effect on testicular health and/or sperm quality is not well discussed. After 21 days of rotenone lesioning, we observed dopaminergic neuronal degeneration in the substantia nigra and hypothalamus. There were minimal SPACA-1-expressing epididymal spermatozoa with morphological abnormalities, scanty luminal spermatozoa and reduced testicular spermatids and post-meiotic germ cells indicating hypospermatogenesis. Occludin-expressing sertoli cells were dispersed over a wide area indicating compromised blood-testes barrier. Activat
