GI-19007, a Novel Saccharomyces cerevisiae-Based Therapeutic Vaccine against Tuberculosis.

As yet, very few vaccine candidates with activity in animals against Mycobacterium tuberculosis infection have been tested as therapeutic postexposure vaccines. We recently described two pools of mycobacterial proteins with this activity, and here we describe further studies in which four of these proteins (Rv1738, Rv2032, Rv3130, and Rv3841) were generated as a fusion polypeptide and then delivered in a novel yeast-based platform (Tarmogen) which itself has immunostimulatory properties, including activation of Toll-like receptors. This platform can deliver antigens into both the class I and c

Genome-wide analysis reveals the vacuolar pH-stat of Saccharomyces cerevisiae.

Protons, the smallest and most ubiquitous of ions, are central to physiological processes. Transmembrane proton gradients drive ATP synthesis, metabolite transport, receptor recycling and vesicle trafficking, while compartmental pH controls enzyme function. Despite this fundamental importance, the mechanisms underlying pH homeostasis are not entirely accounted for in any organelle or organism. We undertook a genome-wide survey of vacuole pH (pH(v)) in 4,606 single-gene deletion mutants of Saccharomyces cerevisiae under control, acid and alkali stress conditions to reveal the vacuolar pH-stat.

Quantitative study of interactions between Saccharomyces cerevisiae and Oenococcus oeni strains.

This study examines the interactions that occur between Saccharomyces cerevisiae and Oenococcus oeni strains during the process of winemaking. Various yeast/bacteria pairs were studied by applying a sequential fermentation strategy which simulated the natural winemaking process. First, four yeast strains were tested in the presence of one bacterial strain leading to the inhibition of the bacterial component. The extent of inhibition varied widely from one pair to another and closely depended on the specific yeast strain chosen. Inhibition was correlated to weak bacterial growth rather than a r

Saccharomyces cerevisiae intervention for relieving flutamide-induced hepatotoxicity in male rats.

The aim of this work was to investigate the protective role of baker's yeast Saccharomyces cerevisiae against the hepatotoxic effect of the drug flutamide that is widely used for treatment of metastatic prostate adenocarcinoma. Administration of flutamide to adult male rats in a dose of 100 mg/kg b.w. daily for 15 days resulted in serious hepatic injury. Highly significant increase in each of serum ALT, ALP, bilirubin, bile acids and cholesterol level, relative to the control group, was observed. Also, a highly significant increase in the serum glutathione-S-transferase isoforms: alpha-GST and

Chronological and replicative lifespan of polyploid Saccharomyces cerevisiae (syn. S. pastorianus).

Chronological lifespan may be defined as the result of accumulation of irreversible damage to intracellular components during extended stationary phase, compromising cellular integrity and leading to death and autolysis. In contrast, replicative lifespan relates to the number of divisions an individual cell has undertaken before entering a non-replicative state termed senescence, leading to cell death and autolysis. Both forms of lifespan have been considered to represent models of ageing in higher eukaryotes, yet the relation between chronologically and replicatively aged populations has not

Anti-Saccharomyces cerevisiae antibodies--a novel serologic marker for Behçet's disease.

OBJECTIVE: To evaluate the prevalence and clinical correlations of antibodies against Saccharomyces cerevisiae (ASCA) among patients with BD. METHODS: Twenty-seven BD patients were studied. Data from medical files and from patients' interviews was collected, regarding the entire spectrum of disease manifestations, and a severity score was calculated for each patient. IgA- and IgG-ASCA levels, determined by ELISA, were studied in all BD patients and in three control groups: patients with recurrent aphthous stomatitis (RAS), systemic lupus erythematosus (SLE) and healthy volunteers. RESULTS: T

Novel peroxisome clustering mutants and peroxisome biogenesis mutants of Saccharomyces cerevisiae.

The goal of this research is to identify and characterize the protein machinery that functions in the intracellular translocation and assembly of peroxisomal proteins in Saccharomyces cerevisiae. Several genes encoding proteins that are essential for this process have been identified previously by Kunau and collaborators, but the mutant collection was incomplete. We have devised a positive selection procedure that identifies new mutants lacking peroxisomes or peroxisomal function. Immunofluorescence procedures for yeast were simplified so that these mutants could be rapidly and efficiently scr

Replication-dependent sister chromatid recombination in rad1 mutants of Saccharomyces cerevisiae.

Homolog recombination and unequal sister chromatid recombination were monitored in rad1-1/rad1-1 diploid yeast cells deficient for excision repair, and in control cells, RAD1/rad1-1, after exposure to UV irradiation. In a rad1-1/rad1-1 diploid, UV irradiation stimulated much more sister chromatid recombination relative to homolog recombination when cells were irradiated in the G1 or the G2 phases of the cell cycle than was observed in RAD1/rad1-1 cells. Since sister chromatids are not present during G1, this result suggested that unexcised lesions can stimulate sister chromatid recombination e

Fluphenazine-resistant Saccharomyces cerevisiae mutants defective in the cell division cycle.

An fls1 mutant of Saccharomyces cerevisiae, which did not grow in the presence of 30 micrograms of fluphenazine per ml, was isolated. Mutants that were resistant to 90 micrograms of fluphenazine per ml and temperature sensitive for growth were obtained from the fls1 mutant. One fluphenazine-resistance mutation, fsr1, was located near the his7 locus on chromosome II. Growth of the fsr1 mutants at 35 degrees C was arrested after nuclear division. The other group of fluphenazine-resistant mutants, carrying fsr2 mutations, showed Ca2+-dependent growth at 35 degrees C. Growth of the fsr2 mutants at

Alleviating the Crabtree effect for high-level production of squalene in Saccharomyces cerevisiae.

Saccharomyces cerevisiae is a widely used biomanufacturing chassis; however, its inherent Crabtree effect severely limits energy yield and diverts carbon flux away from the desired non-ethanol products. To address this challenge, we developed a multidimensional metabolic engineering strategy to alleviate the Crabtree effect, using squalene high-level production as a model. Central metabolism was first modulated through three coordinated interventions: modulation of the SNF1/AMPK energy-sensing system, introduction of an A81D variant of the global transcriptional regulator MTH1, and enhancement

Harnessing natural diversity of Saccharomyces cerevisiae for enhanced resistance to weak acids.

The production of second-generation bioethanol from lignocellulosic biomass is a promising solution for sustainable energy, yet it faces significant challenges also due to the inhibitory effects of weak acids released during biomass pretreatment, particularly acetic, formic and levulinic acids. This review describes the ability of Saccharomyces cerevisiae, with a focus on natural isolates, in overcoming these challenging compounds. Indeed, natural isolates exhibit greater genetic and phenotypic diversity than laboratory and industrial strains, offering unique traits such as enhanced stress tol

ABPM secondary to Saccharomyces cerevisiae mimicking lung cancer in an immunocompetent patient.

SummaryWe present a case of allergic bronchopulmonary mycosis due to Saccharomyces cerevisiae mimicking lung cancer in an immunocompetent woman in her early 70s. She had a 6-week history of dry cough and chest X-ray changes concerning for malignancy. CT initially suggested primary lung cancer. Blood tests showed eosinophilia (4.2×10⁹/L) and moderately elevated total IgE (296 IU/mL). Aspergillus precipitins were mildly raised (140 mgA/L), but no Aspergillus-specific growth or serological sensitisation was found. She developed acute respiratory distress requiring emergency cardiothoracic interve

Raman Imaging of an Amphotericin B Potentiator in Single-Cell Saccharomyces cerevisiae.

Nectriatide, a fungal cyclotetrapeptide, was previously found to enhance the antifungal activity of amphotericin B (AmB), with nectriatide linear derivatives (NLDs) exhibiting even stronger potentiation. Although we showed that NLDs have an affinity for ergosterol in vitro, the precise mechanism underlying these interactions within living fungal cells remained elusive. Here, we demonstrate for the first time a label-free, single-cell Raman microspectroscopic imaging approach to visualize how NLD-1 colocalizes with ergosterol in Saccharomyces cerevisiae. Our results revealed clear evidence of N

Effect of dietary supplementation with Saccharomyces cerevisiae on reproductive parameters of rabbit bucks.

The objective of the present study was to evaluate the effect of dietary supplementation with the prebiotic Saccharomyces cerevisiae yeast on testicular width and semen quality in rabbit bucks. Twenty-four male rabbits were randomly assigned to three groups and their semen was sampled for 8 weeks: a control group (C) fed with a conventional diet without S. cerevisiae, a group supplemented with 0.3 g of S. cerevisiae/kg of feed (T1) and a group supplemented with 1 g of S. cerevisiae/kg of feed (T2). Testicular width was determined using a caliper. Semen was collected from each buck and evaluate

Effect of halo-tolerance gene Hal5 on ethanol tolerance of Saccharomyces cerevisiae.

Hal5 gene is involved in halo-tolerance of Saccharomyces cerevisiae during high salt stress. Ethanol stress and high salt stress have similarities, as both decrease the availability of water for cells and strain the osmotic homeostasis across the cell membrane. The Hal5 over-expression strain of yeast has more ethanol tolerance, but the Hal5 null mutant strain also has more ethanol tolerance than the wild-type strain. Hal5 over-expression in this yeast strain may help in adaptation to ethanol stress by way of directly stabilizing the proteins (trk1-trk2) that are responsible for maintaining os

Mutations of ribosomal protein genes induce overexpression of catalase in Saccharomyces cerevisiae.

Ribosome assembly defects result in ribosomopathies, primarily caused by inadequate protein synthesis and induced oxidative stress. This study aimed to investigate the link between deleting one ribosomal protein gene (RPG) paralog and oxidative stress response. Our results indicated that RPG mutants exhibited higher oxidant sensitivity than the wild type (WT). The concentrations of H2O2 were increased in the RPG mutants. Catalase and superoxide dismutase (SOD) activities were generally higher at the stationary phase, with catalase showing particularly elevated activity in the RPG mutants. Whil

GTP-dependent regulation of heterochromatin fluctuations at subtelomeric regions in Saccharomyces cerevisiae.

In eukaryotes, single cells in a population display different transcriptional profiles. One of the factors regulating this heterogeneity is the chromatin state in each cell. However, the mechanisms of epigenetic chromatin regulation of specific chromosomal regions remain unclear. Therefore, we used single-cell tracking system to analyze IMD2. IMD2 is located at the subtelomeric region of budding yeast, and its expression is epigenetically regulated by heterochromatin fluctuations. Treatment with mycophenolic acid, an inhibitor of de novo GTP biosynthesis, triggered a decrease in GTP, which cau
