Plasma concentrations of morphine, morphine-3-glucuronide, and morphine-6-glucuronide after intravenous and oral administration to healthy volunteers: relationship to nonanalgesic actions.

Healthy volunteers were given morphine as an i.v. infusion (10 mg), immediate release (IR) tablets (3 x 10 mg), and as a new controlled release (CR) tablet (30 mg) on separate occasions. Venous blood samples were analyzed for morphine, morphine-3-glucuronide (M3G), and morphine-6-glucuronide (M6G) using high-performance liquid chromatography (HPLC). Pupil size, salivation, and central nervous system (CNS) effects were evaluated serially. Pharmacokinetic parameters, calculated using a two-compartment model, were in accordance with previous results for i.v. administration of morphine. The absolu

Participation of different brain regions in the anti-narcotic withdrawal action of clonidine in the dependent rat.

The naloxone-precipitated withdrawal response in morphine dependent rats was quantitated by measuring the post-withdrawal increase in mean arterial pressure (MAP), heart rate and several characteristic behavioral changes. Pretreatment with intrathecal injection of clonidine produced a significant reduction in most of the autonomic symptoms of withdrawal, including the increase in MAP, salivation, and chromodacryorrhea, but did not effect the behavioral signs, body shakes, escape attempts or teeth chattering. Central i.c.v. injection of clonidine also reduced the autonomic symptoms of withdrawa

Salivary stones: symptoms, aetiology, biochemical composition and treatment.

Salivary stones, also known as sialoliths, are calcified concrements in the salivary glands. Sialoliths are more frequently located in the submandibular gland (84%), than in the parotid gland (13%). The majority of the submandibular stones are located in Wharton's duct (90%), whereas parotid stones are more often located in the gland itself. Salivary stones consist of an amorphous mineralised nucleus, surrounded by concentric laminated layers of organic and inorganic substances. The organic components of salivary stones include collagen, glycoproteins, amino acids and carbohydrates. The major

PERIOPERATIVE CARE OF A CHILD WITH CRISPONI SYNDROME.

Crisponi syndrome is an autosomal recessive disorder characterized by intermittent episodes of muscular contraction of the facial muscles with trismus and excessive salivation simulating a tetanic spasm. These episodes occur in response to tactile stimulation or during crying. Associated physical and constitutional findings include characteristic facial anomalies, camptodactyly, intermittent hyperthermia, and feeding difficulties. We present a 15-month-old girl who required anesthetic care during laparoscopic fundoplication and gastric tube insertion. The perioperative implications of the diso

Suspected dimethoate toxicity in cattle.

Dimethoate, an organophosphorus insecticide, was the suspected cause of toxicosis in a group of young cattle grazing on pasture that had been sprayed 6 weeks before the onset of clinical signs. Affected animals had primarily nicotinic signs, such as muscle twitching, stiffness, weakness and paralysis, though muscarinic signs, such as diarrhea, salivation and pollakiuria, were also observed. Whole blood acetylcholinesterase activity was depressed in 3 animals. The atypical clinical syndrome and poor response to treatment with atropine and other anticholinergics may have been due to coexistent h

Impact of varied oral processing on starch hydrolysis, bolus properties, and sensory texture attributes of mashed potatoes.

BACKGROUND: Oral processing behavior influences starch bioavailability, yet the impact of chewing conditions on starch digestibility remains unclear. This study investigated how varied chewing behaviors affect starch digestibility in a mashed potato (MP) sample. Three chewing conditions were compared until individual swallowing readiness: slow-rate (SR) chewing, squeezing (tongue-palate compression without teeth), and free chewing. RESULTS: Instrumental texture analysis showed pure MP was too soft for chewing; κ-carrageenan (κ-Car) addition increased hardness, enabling mastication in MP-Car.

EEG: Characteristics of drug-induced seizures in rats, dogs and non-human primates.

Seizures are amongst the most frequent neurological issues encountered in pre-clinical safety testing. The objective was to characterize EEG morphologies and premonitory signs in drug-induced seizures in preclinical species. A comparative (inter-species) retrospective analysis for drug-induced seizures recorded by video-telemetry was conducted in rats (n = 53), dogs (n = 195), and non-human primates (n = 234). The most frequent premonitory signs were, in rats, myoclonus (100%), tremors (93%), salivation (75%), partial ptosis (58%) and chewing/bruxism (58%); in dogs, tremors (77%), ataxia/uncoo

Feline Temporal Lobe Epilepsy: Review of the Experimental Literature.

Accumulating evidence suggests that epileptic seizures originating from the temporal lobe (TL) occur in cats. Typically, affected animals have clinically focal seizures with orofacial automatisms including salivation, facial twitching, lip smacking, chewing, licking, and swallowing. Motor arrest and autonomic and behavioral signs also may occur. Many affected cats have magnetic resonance imaging (MRI) changes within the hippocampus or histopathologically confirmed hippocampal sclerosis or necrosis. From the 1950s to the 1980s, cats frequently were used as animal models for neurophysiological e

Comparison of racemic ketamine versus (S+) ketamine when combined with midazolam for anesthesia of Callithrix jacchus and Callithrix penicillata.

Ten common marmosets (Callithrix jacchus) and 10 black-tufted marmosets (Callithrix penicillata) were immobilized to compare the anesthetic effects of racemic ketamine and (S+) ketamine in combination with midazolam. The animals were distributed into four groups: five common marmosets (group CJR) and five black-tufted marmosets (group CPR) received 9.8 +/- 1.4 mg/kg of racemic ketamine, and five common marmosets (group CJS) and five black-tufted marmosets (group CPS) received 10.4 +/- 1.6 mg/kg of (S+) ketamine. All groups received similar dosages of midazolam (1.0 +/- 0.15 mg/kg). During immo

Autonomic dysfunction in the Lambert-Eaton myasthenic syndrome: serologic and clinical correlates.

Autonomic dysfunction is a recognized feature of the Lambert-Eaton myasthenic syndrome (LES). However, the characteristic pattern of dysautonomia has not been clearly documented and its pathophysiologic basis is not known. We therefore abstracted autonomic symptomatology and results of quantitative tests for salivation, and vasomotor, cardiovagal, and sudomotor reflexes from records of 30 LES patients. Dry mouth (77%) and impotence (45% of men) were the most common symptoms. Composite Autonomic Scoring Scale results were abnormal in 93% of patients, and autonomic failure was severe in 20%. The

Serotonin syndrome from venlafaxine-tranylcypromine interaction.

Excessive stimulation of serotonin 5HT1A receptors causes a syndrome of serotonin excess that consists of shivering, muscle rigidity, salivation, confusion, agitation and hyperthermia. The most common cause of this syndrome is an interaction between a monoamine oxidase inhibitor (MAOI) and a specific serotonin reuptake inhibitor. Venlafaxine is a new antidepressant agent that inhibits the reuptake of serotonin and norepinephrine. We report a venlafaxine-MAOI interaction that resulted in the serotonin syndrome in a 23-y-old male who was taking tranylcypromine for depression. He had been well un

Glycopyrrolate Sorbitol Lollipops for Drooling: Two Case Reports.

Drooling is a pathologic condition that is commonly associated with neurodisability and poor quality of life. An elderly dementia patient and an adult cancer patient suffered from persistent drooling, and they were both prescribed glycopyrrolate 0.5-mg sorbitol lollipops, per recommendation of their local compounding pharmacist. The severity and frequency of drooling decreased substantially in both patients, and there were no reported adverse effects. These two case reports demonstrate the safety and efficacy of glycopyrrolate lollipops in managing excessive salivation, as well as the importan

[Effect of autonomic denervation on the secretory function of submandibular glands].

Submandibular gland is innervated by sympathetic and parasympathetic nerves, both of which work synergistically to promote the salivation of the glands. However, denervation does not always produce an inhibitive effect. Regeneration secretion was put forward to describe the situation in which the glands after denervation were found to be supersensitive to the stimulation of several agonists of saliva secretion-related receptors. It is of clinical significance to get a good understanding of the secretory mechanism changes in the autonomically denervated submandibular gland. The present paper ai

The first outbreak of camelpox in Syria.

In this study we report the first outbreak of camelpox in two provinces in Syria. Clinical symptoms started with fever, salivation and general exanthema. The main features were facial and legs oedema, pustules on the mucosa of the lips and a high rate of abortion. Lesions may also occur on the whole body including scrotum and udder. Specimens were investigated by electron microscopy, virus isolation in cell culture and embryonated eggs and by immunohistochemistry. The causative agent was identified as camelpox virus by polymerase chain reaction and sequencing of the hemagglutinin gene.

[Symptomatic treatments in amyotrophic lateral sclerosis].

ALS is a progressive, fatal, degenerative motor neuron disease of unknown cause. Although advances in understanding pathophysiology of ALS have stimulated the development of new therapies, most of them remain few efficient or ineffective and the main management of ALS patient, to improve quality of life by alleviating symptoms, is symptomatic treatment. This article discusses the approaches now in use to manage some of the most common symptoms of ALS including the following: spasticity, cramps, pain, laryngospasm, pseudobulbar syndrome, salivation and drooling, sleep disorders and fatigue, con

Vestibular-induced vomiting after vestibulocerebellar lesions.

Vestibular stimulation, by means of sinusoidal electrical polarization of the labyrinths of decerebrate cats, can produce vomiting and related activity resembling that seen in motion sickness. The symptoms include panting, salivation, swallowing, and retching as well as vomiting. These symptoms can be produced in cats with lesions of the posterior cerebellar vermis. In contrast to a previous proposal by Wang and Chinn in 1956, we suggest that a transcerebellar pathway from the vestibular apparatus through the nodulus and uvula to the 'vomiting center' is not essential for vestibular-induced vo
