Polysaccharides derived from macroalgae exhibit potent immunomodulatory and gut-regulatory activities; however, comprehensive comparisons of their differential bioactivities remain scarce. This study systematically evaluated the immunomodulatory potential of structurally distinct polysaccharides extracted from Sargassum fusiforme (SP) and Ulva lactuca (UP) in a cyclophosphamide (CTX)-induced immunosuppressed mouse model. Structural analyses revealed that SP consisted mainly of fucose (64.5%) and galactose (20.4%) with molecular masses of 95.6 and 1.4 kDa, while UP was primarily composed of glu

The bioactive materials in brown seaweeds hold great interest for developing new drugs and healthy foods. The oil content in brown seaweeds (Saccharina japonica and Sargassum horneri) was extracted by using environmentally friendly supercritical CO2 (SC-CO2) with ethanol as a co-solvent in a semi-batch flow extraction process and compared the results with a conventional extraction process using hexane, ethanol, and acetone mixed with methanol (1:1, v/v). The SC-CO2 method was used at a temperature of 45 °C and pressure of 250 bar. The flow rate of CO2 (27 g/min) was constant for the entire ext

In this study, a marine medicinal brown alga Sargassum cristaefolium-derived fungal strain Scytalidium lignicola SC228 has been isolated and identified. Column chromatography of the extracts from liquid-fermented products of the fungal strain was carried out, and led to the purification of eight compounds. Their structures were characterized by spectroscopic analysis, and the absolute configurations were further established by single X-ray diffraction analysis and modified Mosher's method as four previously undescribed compounds, namely scytabenzofurans A-C (1-3), and (3S,4S)-5-chloro-3,4-dihy

Plants and brown algae avoid photoinhibition (decline in photosystem II efficiency, Fv/Fm) caused by excess light energy and oxidative stress through several photoprotective mechanisms, such as antioxidant xanthophyll production and heat dissipation. The heat dissipation can be measured as non-photochemical quenching (NPQ) and is strongly driven by de-epoxidation of xanthophyll cycle pigments (XCP). Although NPQ is known to increase under high light acclimation and nutrient-deficient conditions, a few studies have investigated the combined effects of the conditions on both NPQ and associated x

We previously demonstrated that diet supplementation with seaweed Sargassum fusiforme (S. fusiforme) prevented AD-related pathology in a mouse model of Alzheimer's Disease (AD). Here, we tested a lipid extract of seaweed Himanthalia elongata (H. elongata) and a supercritical fluid (SCF) extract of S. fusiforme that is free of excess inorganic arsenic. Diet supplementation with H. elongata extract prevented cognitive deterioration in APPswePS1ΔE9 mice. Similar trends were observed for the S. fusiforme SCF extract. The cerebral amyloid-β plaque load remained unaffected. However, IHC analysis rev

The identification of direct molecular targets for bioactive dietary components is critical for precision nutrition intervention in metabolic dysfunction-associated steatotic liver disease (MASLD). Fucoxanthin, a marine carotenoid from Sargassum fusiforme, exhibits potent lipid-lowering effects; however, its precise intracellular targets and upstream regulatory mechanisms remain elusive. Herein, using drug affinity responsive target stability (DARTS) coupled with LC-MS/MS, we identified the endoplasmic reticulum (ER) chaperone glucose-regulated protein 78 (GRP78) as a direct binding target of

Hyperpigmentation disorders of the skin adversely influence the quality of life. We previously demonstrated the hypopigmenting properties of the ethanolic extract from Sargassum serratifolium and identified sargaquinoic acid (SQA) as an active component. The current study aims to investigate the hypopigmenting action of SQA in α-melanocyte stimulating hormone (α-MSH)-stimulated B16F10 cells. SQA attenuated cellular melanin synthesis by inhibiting the expression of the melanogenic enzymes, including tyrosinase (TYR), tyrosinase-related protein 1 (TRP1), and TRP2. SQA also inhibited cellular TYR
