Standardization and Validation of a Methodology for Assessing Least Reflection Factor and Skin Lipid Replenishment Using Skin Reflectance and Biophysical Measurements.

BACKGROUND: Assessment of skin lipid replenishment and surface appearance requires standardized methodologies capable of measuring both skin-reflecting and barrier-related skin properties. The least-reflection factor (LRF) reflects skin shine and surface lipid presence and is influenced by skin shine and sebum levels. Furthermore, lipid replenishment of the stratum corneum is associated with barrier-related parameters such as skin hydration, transepidermal water loss (TEWL), and surface scaliness. Integrating the reflecting and biophysical parameters may provide a comprehensive approach for ev

Contraceptive use in acne.

Acne vulgaris is an inflammatory disorder of the pilosebaceous follicle. It is well established that androgen hormones play a major role in sebum production and excretion, and are vital in the pathogenesis of acne. Isotretinoin notwithstanding, hormonal therapies such as combined oral contraceptives (COCs) and spironolactone are the only treatments that can affect sebum production and the androgen component of acne. Contraceptives are also used during isotretinoin therapy for pregnancy prevention. It is important for a dermatologist to be familiar with all the available methods of contraceptio

Oily skin: an overview.

Oily skin (seborrhea) is a common cosmetic problem that occurs when oversized sebaceous glands produce excessive amounts of sebum giving the appearance of shiny and greasy skin. This paper overviews the main concepts of sebaceous gland anatomy and physiology, including the biosynthesis, storage and release of sebum, as well as its relationship to skin hydration and water barrier function. We also address how skin oiliness may vary according to diet, age, gender, ethnicity and hot humid climates. The deeper understanding of this skin type provides the opportunity to better guide patients regard

Autophagy-dependent changes in the sebaceous gland and epidermal lipid distribution localized by mass spectrometric imaging.

Sebum is a lipid mixture produced and delivered to the skin surface by appendages called sebaceous glands (SGs) in a holocrine secretion process. Sebum contributes to the skin barrier, lubricates hair and skin, provides antimicrobial protection, and has pheromone functions. During the terminal differentiation of SG cells (sebocytes), macroautophagy, an intracellular process for major metabolic reconfiguration, is activated. Deletion of autophagy affects the hair sebum composition in mice. This study aimed to investigate lipid composition and localization within SG and the epidermis in Atg7 (au

Unsaturated fatty acids induce calcium influx into keratinocytes and cause abnormal differentiation of epidermis.

Abnormal follicular keratinization is involved in comedogenesis in acne vulgaris. We recently demonstrated that calcium influx into epidermal keratinocytes is associated with impaired skin barrier function and epidermal proliferation. Based on these results, we hypothesized that sebum components affect calcium dynamics in the keratinocyte and consequently induce abnormal keratinization. To test this idea, we first observed the effects of topical application of sebum components, triglycerides (triolein), saturated fatty acids (palmitic acid and stearic acid), and unsaturated fatty acids (oleic

Surface lipids as multifunctional mediators of skin responses to environmental stimuli.

Skin surface lipid (SSL) film is a mixture of sebum and keratinocyte membrane lipids, protecting skin from environment. Its composition is unique for the high percentage of long chain fatty acids, and of the polyterpenoid squalene, absent in other human tissues, and in non-human Primates sebum. Here, the still incomplete body of information on SSL as mediators of external chemical, physical, and microbial signals and stressors is revised, focusing on the central event of the continuous oxidative modification induced by the metabolic activity of residential and pathological microbial flora, nat

Hormonal therapy for acne using oral contraceptive pills.

The etiology of acne vulgaris is multifactorial and complex. The four key factors involved in the development of acne include follicular plugging, inflammation, the presence and activity of Propionibacterium acnes, and sebum. Androgen hormones stimulate the sebaceous gland and promote sebum excretion. Therefore, therapies that have an overall antiandrogen effect, like combination oral contraceptive pills, may be useful in the management of acne vulgaris. Numerous combination oral contraceptive pills have been evaluated in the treatment of acne vulgaris and have been found to be effective. With

Evaluation of molecules or extracts modulating seborrhea and its consequences, using normal human culture of sebocytes and keratinocytes, skin explants models and in vivo methods: a case study.

Skin produces sebum through sebocytes. Hyper-seborrhea creates conditions for the development of inflamed cutaneous alterations through bacteria colonization triggering dead cell accumulation and pro-inflammatory mediator release. Study of sebum production, its modulation, and its consequences requires complementary in vitro models in order to evaluate the effect of molecules on cell metabolisms. Clinical studies need to be performed to confirm in vitro results. Effects of phenylpropanoids, obtained by elicitation and purification from plant cell culture of Syringa vulgaris (CCSV), were studie

The Sebaceous Gland: A Key Player in the Balance Between Homeostasis and Inflammatory Skin Diseases.

The sebaceous gland (SG) is an integral part of the pilosebaceous unit and is a very active and dynamic organ that contributes significantly to the maintenance of skin homeostasis. In addition to its primary role in sebum production, the SG is involved in the maintenance of skin barrier function, local endocrine/neuroendocrine function, the innate immune response, and the regulation of skin bacterial colonization. Structural and functional alterations of SGs leading to the dysregulation of sebum production/composition and immune response may contribute to the pathogenesis of inflammatory derma

Possible intermediary role of ghrelin in seborrhea.

Seborrhea, or oily skin, is a very common condition, especially among young people, caused by the increased secretion of sebum by sebaceous glands in the skin. Based on today's knowledge, a regulatory role of various hormones especially androgens is considered for sebaceous gland secretion; but despite significant evidences emphasis on the effects of the gastrointestinal disorders on coetaneous manifestations, the role of gastrointestinal problems in sebum secretion has not been emphasized yet. This study aimed to explain hormonal changes occurring in gastroesophageal reflux disease which may

New targets in the battle against dandruff.

Dandruff is a scalp disorder characterized by flaking skin and itch of an excessive oily scalp skin. It affects 55% of the global youth and adult population. Seborrheic dermatitis is a similar scalp skin disorder with aggravated itchy rashes and flaking. Different factors are identified in the dandruff development: increased sebum production, uncontrolled fungal growth of Malassezia strains and individual reaction to pro-inflammatory environment, and the susceptibility to trigger an immunological response. Using in vitro and ex vivo models, we show that an Epilobium angustifolium extract dose
