TGF-β in the Secretome of Irradiated Peripheral Blood Mononuclear Cells Supports In Vitro Osteoclastogenesis.

Osteoclastogenesis required for bone remodeling is also a key pathologic mechanism of inflammatory osteolysis being controlled by paracrine factors released from dying cells. The secretome of irradiated, dying peripheral blood mononuclear cells (PBMCs) has a major impact on the differentiation of myeloid cells into dendritic cells, and macrophage polarization. The impact on osteoclastogenesis, however, has not been reported. For this aim, we used murine bone marrow macrophages exposed to RANKL and M-CSF to initiate osteoclastogenesis, with and without the secretome obtained from γ-irradiated P

Application of rat cryptorchidism model for the evaluation of mesenchymal stromal cell secretome regenerative potential.

Male infertility represents a severe social and medical challenge. In recent years the progress in regenerative medicine promoted the development of novel options to overcome this medical condition. We are elaborating a promising approach to restore spermatogenesis using mesenchymal stromal cell (MSC) secretome components as a novel class of cell-free cell therapy medicinal products for regenerative medicine. However, the choice of the representative in vivo model of spermatogenesis failure to evaluate the effectiveness of regenerative drugs remains challenging. Using the rat model of bilatera

Secretome of in vitro cultured human embryos contains extracellular vesicles that are uptaken by the maternal side.

Communication between embryo and maternal endometrium occurs during a specific time frame in which implantation is possible. Here we demonstrate for the first time that conditioned media from non-manipulated human embryos cultured in vitro for 3 days or up to the blastocyst stage contain extracellular vesicles (EVs) with a diameter of 50 to 200 nm and bearing the traditional microvesicle and exosome marker proteins CD63, CD9 and ALIX. The embryonic origin of these EVs has been confirmed by the presence of stemness gene transcripts and their enrichment in the non-classical HLA-G protein. NANOG

Effects of fluconazole on the secretome, the wall proteome, and wall integrity of the clinical fungus Candida albicans.

Fluconazole is a commonly used antifungal drug that inhibits Erg11, a protein responsible for 14α-demethylation during ergosterol synthesis. Consequently, ergosterol is depleted from cellular membranes and replaced by toxic 14α-methylated sterols, which causes increased membrane fluidity and drug permeability. Surface-grown and planktonic cultures of Candida albicans responded similarly to fluconazole at 0.5 mg/liter, showing reduced biomass formation, severely reduced ergosterol levels, and almost complete inhibition of hyphal growth. There was no evidence of cell leakage. Mass spectrometric

Discovering new bioactive neuropeptides in the striatum secretome using in vivo microdialysis and versatile proteomics.

The striatum, a major component of the brain basal nuclei, is central for planning and executing voluntary movements and undergoes lesions in neurodegenerative disorders such as Huntington disease. To perform highly integrated tasks, the striatum relies on a complex network of communication within and between brain regions with a key role devoted to secreted molecules. To characterize the rat striatum secretome, we combined in vivo microdialysis together with proteomics analysis of trypsin digests and peptidomics studies of native fragments. This versatile approach, carried out using different

The secretome of the filarial parasite, Brugia malayi: proteomic profile of adult excretory-secretory products.

The secretome of a parasite in its definitive host can be considered to be its genome in trans, to the extent that secreted products encoded by the parasite fulfill their function in the host milieu. The 'extended phenotype' of the filarial parasite, Brugia malayi, is of particular interest because of the evidence that infection results in potent down-modulation of the host immune response. We collected B. malayi 'excretory-secretory' (BES) proteins from adult parasites and using a combination of shotgun LC-MS/MS and 2D gel electrophoresis, identified 80 B. malayi and two host proteins in BES,

Proteomic characterization of the whole secretome of Legionella pneumophila and functional analysis of outer membrane vesicles.

Secretion of effector molecules is one of the major mechanisms by which the intracellular human pathogen Legionella pneumophila interacts with host cells during infection. Specific secretion machineries which are responsible for the subfraction of secreted proteins (soluble supernatant proteins [SSPs]) and the production of bacterial outer membrane vesicles (OMVs) both contribute to the protein composition of the extracellular milieu of this lung pathogen. Here we present comprehensive proteome reference maps for both SSPs and OMVs. Protein identification and assignment analyses revealed a tot

HUC-MSC secretome and Nanoemulsion Propolis synergistically modulate inflammatory responses in hyperglycemia-induced calvarial osteolysis.

BACKGROUND: Hyperglycemia-induced osteolysis is a significant consequence of diabetes mellitus, marked by heightened inflammatory responses, augmented osteoclastogenesis, and compromised osteoblast function. Innovative therapy approaches aimed at inflammatory and cytoprotective mechanisms are essential to avert diabetic bone loss. OBJECTIVE: This study investigated the synergistic effects of human umbilical cord mesenchymal stem cell (HUC-MSC) secretome and Nanoemulsion Propolis (NEP) on inflammatory responses and protective protein expression in a rat model of hyperglycemia-associated calvar

Calpain inhibition by calpeptin modulates adipocyte lipid metabolism and secretome-mediated inflammatory crosstalk with hepatocytes.

OBJECTIVE: Calpeptin, a calpain inhibitor with potential for treating inflammatory diseases, reduces extracellular vesicle (EV) secretion. While adipose tissue is a recognized target of calpeptin, its effects on lipid metabolism remain unknown. We investigated calpeptin's impact on fatty acid (FA) profiles and metabolic pathways in human Simpson-Golabi-Behmel Syndrome adipocytes and their EVs. METHODS: Adipocytes were treated with 25 or 50 µM calpeptin, and EVs were isolated from conditioned media (CM) by ultracentrifugation. Immortalized human hepatocytes (IHHs) were pre-treated with 0 or 40

Cell-free regenerative therapies for female infertility: The emerging role of mesenchymal stem cell secretome.

BACKGROUND: Female reproductive disorders are a leading cause of infertility, affecting millions of women worldwide and resulting in significant emotional and social challenges. Despite advances in medical science, current treatment options are often limited in their ability to restore normal reproductive function, particularly in cases involving hormonal imbalances, complex tissue damage, or unexplained infertility. These limitations highlight the urgent need for innovative and more effective treatment strategies to address the underlying causes of female infertility. MAIN BODY: Recent advan

A novel cell-free regenerative therapy for hair loss: human fetal cartilage progenitor cell secretome.

Hair loss, particularly androgenic alopecia, is driven by testosterone-induced apoptosis and impaired dermal papilla (DP) cell function, disrupting epithelial-mesenchymal interactions and hindering hair follicle regeneration. Current non-surgical treatments, such as minoxidil and finasteride, provide limited and temporary benefits. Here, we report the development of a novel, cell-free regenerative therapy using the secretome of human fetal cartilage progenitor cells (ShFCPC), enriched with extracellular matrix (ECM) proteins and bioactive factors critical for tissue remodeling and cell surviva

A Broad-Spectrum Chemokine Inhibitor Drives M2 Macrophage Polarization Through Modulation of the Myometrial Secretome.

The uterine smooth muscle (myometrium) is an immunomodulatory tissue capable of secreting multiple chemokines during pregnancy. We propose that before term labor, chemokines secreted as a result of mechanical stretch of the uterine walls by the growing fetus(es) induce infiltration of maternal monocytes into myometrium, drive their differentiation into macrophages, and induce pro-inflammatory (M1) polarization, leading to labor contractions. This study used high-throughput proteomic mass-spectrometry to investigate the underlying mechanisms and explored the therapeutic potential of a broad-spe

Mesenchymal stem cell secretome alters gene expression and upregulates motility of human endometrial stromal cells.

IN BRIEF: Endometrial stromal cell motility is fundamental to regeneration and repair of this tissue and crucial for successful reproduction. This paper shows a role for the mesenchymal stem cell (MSC) secretome in enhancing endometrial stromal cell motility. ABSTRACT: Cyclic regeneration and repair of the endometrium are crucial for successful reproduction. Mesenchymal stem cells (MSCs) derived from bone marrow (BM-MSC) and umbilical cord (UC-MSC) facilitate tissue repair via their secretome, which contains growth factors and cytokines that promote wound healing. Despite the implication of M

Apoptosis in mesenchymal stromal cells activates an immunosuppressive secretome predicting clinical response in Crohn's disease.

In vivo apoptosis of human mesenchymal stromal cells (MSCs) plays a critical role in delivering immunomodulation. Yet, caspase activity not only mediates the dying process but also death-independent functions that may shape the immunogenicity of apoptotic cells. Therefore, a better characterization of the immunological profile of apoptotic MSCs (ApoMSCs) could shed light on their mechanistic action and therapeutic applications. We analyzed the transcriptomes of MSCs undergoing apoptosis and identified several immunomodulatory factors and chemokines dependent on caspase activation following Fas

Nanoparticles functionalized with stem cell secretome and CXCR4-overexpressing endothelial membrane for targeted osteoporosis therapy.

BACKGROUND: Osteoporosis is a chronic condition affecting patients' morbidity and mortality and represents a big socioeconomic burden. Because stem cells can proliferate and differentiate into bone-forming cells, stem cell therapy for osteoporosis has been widely studied. However, cells as a live drug face multiple challenges because of their instability during preservation and transportation. In addition, cell therapy has potential adverse effects such as embolism, tumorigenicity, and immunogenicity. RESULTS: Herein, we sought to use cell-mimicking and targeted therapeutic nanoparticles to r

Neuroprotective effects of human amniotic fluid stem cells-derived secretome in an ischemia/reperfusion model.

Stem cells offer the basis for the promotion of robust new therapeutic approaches for a variety of human disorders. There are still many limitations to be overcome before clinical therapeutic application, including a better understanding of the mechanism by which stem cell therapies may lead to enhanced recovery. In vitro investigations are necessary to dissect the mechanisms involved and to support the potential development in stem cell-based therapies. In spite of growing interest in human amniotic fluid stem cells, not much is known about the characteristics of their secretome and regarding

Alternatively activated macrophage-derived secretome stimulates ovarian cancer spheroid spreading through a JAK2/STAT3 pathway.

High-grade serous ovarian cancer (HGSOC) metastasizes when tumor spheroids detach from the primary tumor and re-attach throughout the peritoneal cavity. Once the cancer cells have implanted in these new sites, the development of metastatic lesions is dependent on the disaggregation of cancer cells from the spheroids and subsequent expansion across the collagenous extracellular matrix (ECM). As HGSOC progresses an increase in alternatively activated macrophages (AAMs) in the surrounding ascites fluid has been observed and AAMs have been shown to enhance tumor invasion and growth in a wide range
