Genetic control of immune responses to parasites: selection for responsiveness and non-responsiveness to Trichuris muris in random-bred mice.

Populations of Schofield strain, random-bred mice were shown to have a bimodal variation in ability to bring about immune expulsion of the nematode Trichuris muris. This variation was genetically determined and independent of the size of infection experienced. The proportion of mice unable to achieve worm expulsion (non-responders) was relatively constant in various populations of the strain but was increased by selective breeding from mice of known status. Crosses made between non-responder and responder mice produced progeny that were almost all (92%) of responder phenotype, showing that the

Rapid evolution and genetic basis of spinetoram resistance in Phthorimaea absoluta: evidence from six years of monitoring and laboratory selection.

BACKGROUND: Phthorimaea absoluta (Meyrick) (Lepidoptera: Gelechiidae) is one of the most destructive pests of tomato worldwide and has repeatedly evolved resistance to insecticides. Spinosyn insecticides, particularly spinetoram, are widely used for its control due to their high efficacy and favorable selectivity. This study investigated temporal changes in susceptibility to spinetoram in Brazilian populations of Phthorimaea absoluta over a 6-year monitoring period and characterized the genetic basis, metabolic contribution, and cross-resistance patterns of a resistant population. RESULTS: Di

Genetic diversity and natural selection of cell-traversal protein for ookinetes and sporozoites (CelTOS) in Plasmodium falciparum isolates from Vietnam.

Plasmodium falciparum cell-traversal protein for ookinetes and sporozoites (PfCelTOS) plays a crucial role in the parasite's life cycle at both vector and human liver stages and is a potential vaccine candidate impacting various phases of the parasite's life cycle. However, current knowledge about the genetic diversity and evolutionary trend of this gene, which could impede the development of effective vaccines based on this protein, remains greatly limited. In this study, genetic diversity and natural selection in Vietnamese pfceltos were analyzed. Blood samples were collected from P. falcipa

Identification of Novel Biomarkers for Response to Preoperative Chemoradiation in Locally Advanced Rectal Cancer with Genetic Algorithm-Based Gene Selection.

BACKGROUND: The conventional treatment for patients with locally advanced colorectal tumors is preoperative chemo-radiotherapy (PCRT) preceding surgery. This treatment strategy has some long-term side effects, and some patients do not respond to it. Therefore, an evaluation of biomarkers that may help predict patients' response to PCRT is essential. METHODS: We took advantage of genetic algorithm to search the space of possible combinations of features to choose subsets of genes that would yield convenient performance in differentiating PCRT responders from non-responders using a logistic reg

A New Computer-Aided Diagnosis System with Modified Genetic Feature Selection for BI-RADS Classification of Breast Masses in Mammograms.

Mammography remains the most prevalent imaging tool for early breast cancer screening. The language used to describe abnormalities in mammographic reports is based on the Breast Imaging Reporting and Data System (BI-RADS). Assigning a correct BI-RADS category to each examined mammogram is a strenuous and challenging task for even experts. This paper proposes a new and effective computer-aided diagnosis (CAD) system to classify mammographic masses into four assessment categories in BI-RADS. The mass regions are first enhanced by means of histogram equalization and then semiautomatically segment

Genetic signatures of gene flow and malaria-driven natural selection in sub-Saharan populations of the "endemic Burkitt Lymphoma belt".

Populations in sub-Saharan Africa have historically been exposed to intense selection from chronic infection with falciparum malaria. Interestingly, populations with the highest malaria intensity can be identified by the increased occurrence of endemic Burkitt Lymphoma (eBL), a pediatric cancer that affects populations with intense malaria exposure, in the so called "eBL belt" in sub-Saharan Africa. However, the effects of intense malaria exposure and sub-Saharan populations' genetic histories remain poorly explored. To determine if historical migrations and intense malaria exposure have shape

Differences in the Selection Bottleneck between Modes of Sexual Transmission Influence the Genetic Composition of the HIV-1 Founder Virus.

Due to the stringent population bottleneck that occurs during sexual HIV-1 transmission, systemic infection is typically established by a limited number of founder viruses. Elucidation of the precise forces influencing the selection of founder viruses may reveal key vulnerabilities that could aid in the development of a vaccine or other clinical interventions. Here, we utilize deep sequencing data and apply a genetic distance-based method to investigate whether the mode of sexual transmission shapes the nascent founder viral genome. Analysis of 74 acute and early HIV-1 infected subjects reveal

Genetic diversity in RHD and RHCE genes among a selected Kenyan blood donor population.

BACKGROUND: Serologic typing for ABO and RhD is standard in transfusion services, with extended serology and genotyping performed to reduce red cell alloimmunization risk. In Kenya, RH typing is limited to RhD, and genotyping is unavailable. This study used RHD/RHCE genotyping to predict phenotypes and their distribution in a Kenyan blood donor population. STUDY DESIGN AND METHODS: A total of 191 donors (114 D-, 74 D+, and 3 weak D) from the Kenya National Blood Transfusion Service were selected. Next-generation sequencing was performed on DNA extracts using a targeted blood group sequencing

Whole-genome resequencing reveals population genetic structure and selective sweeps in Bombus waltoni.

BACKGROUND: Bombus waltoni is a typical alpine bumblebee species and an important indicator species of the Qinghai-Tibetan Plateau. However, whole-genome studies of bumblebees from this region remain limited. In this study, we investigated the genetic diversity, population structure, and adaptive genomic variation of B. waltoni, providing essential genomic resources for understanding its population genetic structure and signatures of local adaptation. RESULTS: Based on whole-genome SNP data, genetic diversity was relatively similar across B. waltoni populations, with expected heterozygosity (

Precision patient selection for improved detection of circulating genetically abnormal cells in pulmonary nodules.

Circulating genetically abnormal cells (CACs) have emerged as a promising biomarker for the early diagnosis of lung cancer, particularly in patients with pulmonary nodules. However, their performance may be suboptimal in certain patient populations. This study aimed to refine patient selection to improve the detection of CACs in pulmonary nodules. A retrospective analysis was conducted on 241 patients with pulmonary nodules who had undergone pathological diagnosis through surgical tissue specimens. Utilizing consensus clustering analysis, the patients were categorized into three distinct clust

Brain monoamine levels and behaviour of young and adult chickens genetically selected on feather pecking.

Severe feather pecking (SFP) in chickens is a detrimental behaviour with possibly neurochemical deficits at its base. Recent neurological studies depicted conflicting results on the role of serotonin (5-hydroxytryptamine, 5-HT) and dopamine (DA) in the development and display of feather pecking. We studied brain monoamine levels and behaviour in domestic chickens divergently genetically selected on feather pecking behaviour, the Low Feather Pecking (LFP) and High Feather Pecking (HFP) lines, both at a young age and when adult, to elucidate the role of 5-HT and DA in feather pecking. Also pecki

Genetic disruption of the On visual pathway affects cortical orientation selectivity and contrast sensitivity in mice.

The retina signals stimulus contrast via parallel On and Off pathways and sends the information to higher visual centers. Here we study the role of the On pathway using mice that have null mutations in the On-specific GRM6 receptor in the retina (Pinto LH, Vitaterna MH, Shimomura K, Siepka SM, Balannik V, McDearmon EL, Omura C, Lumayag S, Invergo BM, Brandon M, Glawe B, Cantrell DR, Donald R, Inayat S, Olvera MA, Vessey KA, Kirstan A, McCall MA, Maddox D, Morgans CW, Young B, Pletcher MT, Mullins RF, Troy JB, Takahashi JS. Genetic analysis of metabolic traits in an intercross between body weight-selected chicken lines.

A network of four interacting loci has been reported previously to influence growth in two lines of chickens divergently selected for body weight at 56 days of age. Located on chromosomes 3 (Growth4), 4 (Growth6), 7 (Growth9), and 20 (Growth12), they explained nearly half of the difference in body weight at selection age between the two lines. The original study reported effects on body weight and fat deposition, but no attempts were made to explore the effects of the network on other phenotypes measured in the F(2) population. In this study we conducted further analyses to evaluate the specif

Effects of selected genetic polymorphisms in xeroderma pigmentosum complementary group D on gastric cancer.

DNA repair capacity (DRC) can be altered based on sequence variations in DNA repair genes, which may result in cancer susceptibility. The current study was to evaluate the association between genetic polymorphisms, including associated haplotypes of xeroderma pigmentosum complementary group D (XPD), and individual susceptibility to gastric cancer. Two-hundred-eight patients with gastric cancer and 339 healthy controls were enrolled in this study. Their genomic DNA was extracted from peripheral blood leukocytes. The genotypes at exon 6, 10 and 23 were identified by polymerase chain reaction (PC

Genetic screens and selections for small molecules based on a synthetic riboswitch that activates protein translation.

Genetic selection provides the most powerful method to assay large libraries of biomolecules for function. However, harnessing the power of genetic selection for the detection of specific, nonendogenous small-molecule targets in vivo remains a significant challenge. The ability to genetically select for small molecules would provide a reaction-independent mechanism to clone biosynthesis genes from large DNA libraries and greatly facilitate the exploration of large libraries of mutant enzymes for improved synthetic capabilities including altered substrate specificities and enhanced regio- or st

Dimensions and disorders of adolescent adjustment: a quantitative genetic analysis of unselected samples and selected extremes.

One of the fundamental questions for developmental psychopathology concerns the etiological links between the normal and abnormal. To what extent do disorders differ quantitatively or qualitatively from variation in the normal range? Genetic research on the normal and the abnormal differs in terms of concepts, methods, statistics, and target audiences. An approach, referred to as "DF" analysis, provides a framework for integrating these two worlds of genetic analysis. We applied traditional correlational analyses as well as DF (DeFries & Fulker, 1985) analyses to mother and father ratings of a
