First detection of IncHI2 plasmid carrying blaNDM-5 in Escherichia coli from retail shellfish.

The increasing dissemination of carbapenemase-producing Enterobacterales (CPE) across multiple ecological niches underscores a critical One Health challenge. Despite documentation of CPE contamination in shrimp, the occurrence of CPE within broader aquatic food products remains underexplored. In this study, 262 retail shellfish samples from farmers' markets were screened for blaNDM-producing Gram-negative bacteria. Three Escherichia coli (ST6769, ST10, and ST3871) and five Shewanella algae (unable to assign ST) isolates recovered from eight (3.1%) shellfish samples were identified as carrying

Tumor-Tropic Bacterial-Nanoparticle Hybrids for Metabolic Reprogramming and Cancer Immunotherapy.

Metabolic dysregulation and immune suppression are intrinsically linked within the solid tumor microenvironment, yet effective strategies to simultaneously reprogram tumor metabolism and restore antitumor immunity remain challenging. Here, we present a tumor-tropic bacterial-nanoparticle hybrid (SW@CDFM) that integrates metabolic reprogramming with immune activation for synergistic cancer treatment. The designed biohybrid system is constructed by covalently conjugating Shewanella oneidensis bacteria with tumor cell membrane-camouflaged DOX-Fe/CpG nanoparticles. Leveraging the intrinsic hypoxia

A synthetic protein plug for assembly of bio-nano-hybrids with direct electron transfer powered photobiocatalysis.

A "plug-and-play" biohybrid assembly strategy was established by reprogramming cell membrane proteins into "synthetic protein plugs". This approach covalently confined carbon dots onto Shewanella oneidensis within the optimal electron tunneling regime, thereby achieving efficient direct electron transfer to empower an 81.3-fold enhancement in photocatalytic H2 production.

Self-regenerable bio-hybrid with biogenic ferrous sulfide nanoparticles for treating high-concentration chromium-containing wastewater.

Biogenic ferrous sulfide nanoparticles (bio-FeS) as low-cost and green-synthesized nanomaterial are promising for heavy metals removal, but the need for complicated extraction, storage processes and the production of iron sludge still restrict their practical application. Here, a self-regenerable bio-hybrid consisting of bacterial cells and self-assembled bio-FeS was developed to efficiently remove chromium (Cr(VI)). A dense layer of bio-FeS was distributed on the cell surface and in the periplasmic space of Shewanella oneidensis MR-1, endowing the bacterium with good Cr(VI) tolerance and unus

Oxidized OxyR Up-Regulates ahpCF Expression to Suppress Plating Defects of oxyR- and Catalase-Deficient Strains.

It is well established that in bacteria, such as Escherichia coli, OxyR is a transcriptional regulator that mediates the response to H2O2 by activating the OxyR regulon, which consists of many genes that play vital roles in oxidative stress resistance. In Shewanella, OxyR regulates, however, in both reduced and oxidized states, the production of H2O2 scavengers, including major catalase KatB and NADH peroxidase AhpCF. Here we showed that the oxyR mutant carried a plating defect manifested as division arresting, a phenotype that can be completely suppressed by an OxyR variant constitutively exi

Anaerobic petroleum biodegradation by microbial communities enriched from mangrove sediments.

Mangrove sediments are increasingly exposed to petroleum contamination; however, the anaerobic degradation of petroleum hydrocarbons by sediment-derived microbial communities remains poorly understood. In this study, anaerobic microbial communities were enriched in sediments collected from three mangrove wetlands using petroleum as the sole carbon source and nitrate/sulfate as dual electron acceptors. The enriched communities exhibited a strong capacity for petroleum hydrocarbon degradation, with degradation ranging from 65.20% to 93.20% for total petroleum hydrocarbons and from 64.68% to 99.9

In-situ nano-encapsulation of living cells with polydopamine for enhanced artificial interspecies electron transfer.

Interspecies electron transfer (IET) determines the efficiency of electro-syntrophic metabolism in electroactive microbial communities. Compared with mediated IET (MIET) mediated by diffusible electron shuttles, contact-based direct IET (DIET) can enable more efficient energy transfer. However, engineering DIET is constrained by the challenges of maintaining stable and conductive cell-to-cell interfaces. In this study, we developed a conductive and adhesive interface by in-situ polydopamine (PDA) nano-encapsulation of Shewanella oneidensis MR-1 (SW, electron donating partner) surfaces. The coa

Visualizing Microbial Indirect Extracellular Electron Transfer.

Indirect extracellular electron transfer (IEET) mediated by soluble electron shuttles is a critical pathway for anaerobic microbial respiration, influencing redox transformations and element cycling in natural environments. However, direct spatial visualization of the electron transfer extent has remained limited. Here, employing silver ions (Ag+) as electron traps and photothermal imaging of as-formed Ag nanoparticles, we visually demonstrated that microbes can effectively transfer electrons over centimeter distances. For instance, Shewanella oneidensis MR-1 transferred electrons across 11.5

Biogenic FeS promotes dechlorination and thus de-cytotoxity of trichloroethylene.

Abiotic iron monosulfide (FeS) has attracted growing interests in dechlorinating trichloroethylene (TCE) in anoxic groundwater, but it is still unclear how biogenic FeS affects the dechlorination and thus the cytotoxity of TCE. In this work, a biogenic FeS was synthesized by Shewanella oneidensis MR-1 with addition of ferrihydrite and S0, and it was used for dechlorination of TCE in alkaline environment and the de-cytotoxicity was evaluated by the growth of Synechocystis sp. PCC6803. The results show that the biogenic FeS was of mackinawite, with a loose flower-like mosaic structure. The dechl

Non-photosynthetic microorganisms drive the transformation of bioinert emerging contaminants through photoinduced singlet oxygen production.

Ubiquitous non-photosynthetic microorganisms play a crucial role in the environmental attenuation of contaminants. However, this microbial-driven transformation is often ineffective for bio-inert or bio-toxic emerging contaminants (ECs). Herein, we demonstrated that non-photosynthetic microorganisms such as Escherichia coli, Bacillus subtilis, Shewanella oneidensis and microorganisms from actual waters could produce singlet oxygen (1O2) through photoexcited extracellular metabolites, effectively driving the transformation of ECs such as cimetidine, estriol, and sulfamethoxazole. The extracellu

Microbial succession drives quality deterioration in refrigerated golden pompano (Trachinotus ovatus): regulatory network of dominant spoilage microbiota on characteristic biomarkers.

Elucidating the quality deterioration mechanisms of refrigerated golden pompano (Trachinotus ovatus), this study integrated 16S rRNA gene sequencing and untargeted metabolomics to systematically dissect how microbial ecological succession drives metabolic changes. As critical spoilage indicators, total volatile basic nitrogen (TVB-N) and thiobarbituric acid reactive substances (TBARS) continuously increased during storage, indicating significant deterioration by day 6. Microbial community succession analysis revealed that Psychrobacter, Shewanella, and Pseudomonas gradually became the dominant

Red-light-activated living bacterial electron generator for on-demand drug release in colonic inflammation.

Inflammatory bowel disease demands spatiotemporally precise drug delivery, yet the variable gut redox environment limits stimuli-responsive nanocarriers. Here we report a living biohybrid platform in which optogenetically engineered Shewanella oneidensis MR-1 is electrostatically conjugated with azo-bond covalent organic frameworks (TA-COFs) loaded with anti-inflammatory drugs magnolol or 4-iodobenzoic acid. Under intestinal conditions and non-invasive red-light irradiation (660 nm), light-induced restoration of the metal-reducing pathway promotes extracellular electron transfer, thereby cleav

Microbial Diversity and Hydrocarbon-Oxidizing Bacteria in Coastal Waters and Sands Contaminated by the Fuel Oil Spill in the Black Sea.

In 2024, an accident involving two tankers in the Kerch Strait resulted in the release of approximately 2400 tons of fuel oil into the Black Sea, causing significant contamination of seawater and the coastal zone. This study presents the first microbiological and molecular-ecological assessment of prokaryotic community composition and hydrocarbon-oxidizing bacteria (HOB) in coastal seawater and sand near Anapa (Russian Federation) following the spill. The taxonomic composition of nine samples was analyzed using high-throughput sequencing of 16S rRNA genes (V3-V4 regions), identifying Bacteria

Effects of Ozone Water Combined With Ultra-High Pressure on Quality and Microorganism of Catfish Fillets (Lctalurus punctatus) During Refrigeration.

This study described the quality and microbial influence on ozone water (OW) and ultra-high pressure (UHP) processing alone or in combination with refrigerated catfish fillets. The analysis parameters included total volatile base nitrogen (TVBN), thiobarbituric acid reactive substances (TBARs), chromaticity, microbial enumeration, 16S rRNA gene sequencing, electronic nose (E-nose), and sensory score. The study found that compared with the control (CK), ozone water combined with ultra-high pressure (OCU) delayed the accumulation of TVBN and TBARs. The results of sensory evaluation illustrated t

Gelatin/Chitosan Films Incorporated with Curcumin Based on Photodynamic Inactivation Technology for Antibacterial Food Packaging.

Photodynamic inactivation (PDI) is a new type of non-thermal sterilization technology that combines visible light with photosensitizers to generate a bioactive effect against foodborne pathogenic bacteria. In the present investigation, gelatin (GEL)/chitosan (CS)-based functional films with PDI potency were prepared by incorporating curcumin (Cur) as a photosensitizer. The properties of GEL/CS/Cur (0.025, 0.05, 0.1, 0.2 mmol/L) films were investigated by evaluating the surface morphology, chemical structure, light transmittance, and mechanical properties, as well as the photochemical and therm

Mtr extracellular electron-transfer pathways in Fe(III)-reducing or Fe(II)-oxidizing bacteria: a genomic perspective.

Originally discovered in the dissimilatory metal-reducing bacterium Shewanella oneidensis MR-1 (MR-1), key components of the Mtr (i.e. metal-reducing) pathway exist in all strains of metal-reducing Shewanella characterized. The protein components identified to date for the Mtr pathway of MR-1 include four multihaem c-Cyts (c-type cytochromes), CymA, MtrA, MtrC and OmcA, and a porin-like outer membrane protein MtrB. They are strategically positioned along the width of the MR-1 cell envelope to mediate electron transfer from the quinone/quinol pool in the inner membrane to Fe(III)-containing min

Beta-propeller phytases in the aquatic environment.

Phytate, which is one of the dominant organic phosphorus compounds in nature, is very stable in soils. Although a substantial amount of phytate is carried from terrestrial to aquatic systems, it is a minor component of organic phosphorus in coastal sediments. The ephemeral nature of phytate implies the rapid hydrolysis of phytate under aquatic conditions. Among the four classes of known phytases that have been identified in terrestrial organisms, only beta-propeller phytase-like sequences have been identified in the aquatic environment. A novel beta-propeller phytase gene (phyS), cloned from S

COmmunity and Single Microbe Optimisation System (COSMOS).

Bioprocessing utilises microbial monocultures and communities to convert renewable resources into valuable products. While monocultures offer simplicity, communities provide metabolic diversity and cooperative biosynthesis. To systematically evaluate these systems, we developed COmmunity and Single Microbe Optimisation System (COSMOS), a dynamic computational framework that simulates and compares monocultures and co-cultures to determine optimal microbial systems tailored to a specific environment. COSMOS revealed key factors shaping biosynthetic performance, such as environmental conditions,
