Somnambulistic-like episodes secondary to combined lithium-neuroleptic treatment.

Ten of 114 psychiatric patients undergoing combined lithium-neuroleptic treatment exhibited somnambulistic-like episodes. These episodes are differentiated from nocturnal wanderings and epileptic attacks during sleep; they occurred within two to three hours after sleep onset and were characterized by the patients appearing confused and walking about in a quiet, detached and clumsy manner. Generally, there was amnesia for the event. Since sleepwalking occurs out of slow wave sleep, the increase in slow wave sleep induced by lithium and certain neuroleptics may represent a neurophysiological mec

Sleep homeostasis impairment across obstructive sleep apnea severity: findings from a population-based cohort study.

PURPOSE: Obstructive sleep apnea (OSA) is a sleep disorder that causes recurrent airway obstruction and sleep fragmentation and increases cardiometabolic, cognitive, and mortality risk. Although slow-wave activity (SWA) and sleep homeostasis (SH) are central to restorative sleep and synaptic regulation, the extent to which SH is disrupted across OSA severity remains unclear. METHODS: In this population-based cohort study, we analyzed 945 polysomnography (PSG) studies from adults with mild, moderate, severe OSA, or no OSA. Sleep parameters including apnea-hypopnea index (AHI) and slow wave act

Sleep Disorders in Mild Cognitive Impairment.

We have an increasingly aging population and, therefore, cognitive impairment and dementia are becoming more common. Similarly, sleep disorders are also more common among the older population. There is a bidirectional relationship between mild cognitive impairment and sleep disorders. Additionally, both of these issues are underdiagnosed. By identifying and treating sleep disturbances early, we may delay the onset of dementia. Sleep helps in clearing metabolites like amyloid-beta (A-beta) lipoprotein. Clearance leads to decreased fatigue and proper functioning of the brain. A-beta lipoprotein

Slow wave generation and propagation in a model of brain lesions.

Slow waves (SWs), the hallmark of non-rapid eye movement (NREM) sleep, reflect the periodic occurrence of transient silent periods in cortical neurons (Down states). During NREM, SWs and Down states physiologically disrupt large-scale network interactions. Since early EEG studies, SWs have also been observed in awake patients after brain injury. Emerging evidence indicates that these intrusions of sleep-like activity interfere with ongoing network activity and contribute to motor and cognitive deficits; yet, the mechanisms governing the generation and spread of post-lesional SWs remain unclear

SWS Brain-Wave Music May Improve the Quality of Sleep: An EEG Study.

AIM: This study investigated the neural mechanisms of brain-wave music on sleep quality. BACKGROUND: Sleep disorders are a common health problem in our society and may result in fatigue, depression, and problems in daytime functioning. Previous studies have shown that brain-wave music generated from electroencephalography (EEG) signals could emotionally affect our nervous system and have positive effects on sleep. However, the neural mechanisms of brain-wave music on the quality of sleep need to be clarified. METHODS: A total of 33 young participants were recruited and randomly divided into

Brain white matter damage and its association with neuronal synchrony during sleep.

The restorative function of sleep partly relies on its ability to deeply synchronize cerebral networks to create large slow oscillations observable with EEG. However, whether a brain can properly synchronize and produce a restorative sleep when it undergoes massive and widespread white matter damage is unknown. Here, we answer this question by testing 23 patients with various levels of white matter damage secondary to moderate to severe traumatic brain injuries (ages 18-56; 17 males, six females, 11-39 months post-injury) and compared them to 27 healthy subjects of similar age and sex. We used

Pre-sleep central-occipital alpha-theta dynamics predict NREM2 onset in adult.

Objective.Sleep onset (SO) marks the transition from wakefulness to a restorative state of sleep in the human brain. Recent studies suggest that the sleep onset process (SOP) is a back-and-forth progression that occurs before the brain reaches Non-Rapid Eye Movement Sleep Stage 2 (NREM2) and that its duration is highly variable. Very short sleep onset latencies may increase the risk of accidents, while long SO may indicate poor sleep quality and insomnia.Approach.In this study, we investigated electroencephalography power dynamics before the first NREM2 sleep epoch to identify potential predic

Cortical hyperarousal in NREM sleep normalizes from pre- to post- REM periods in individuals with frequent nightmares.

STUDY OBJECTIVES: Frequent nightmares have a high prevalence and constitute a risk factor for psychiatric conditions, but their pathophysiology is poorly understood. Our aim was to examine sleep architecture and electroencephalographic markers-with a specific focus on state transitions-related to sleep regulation and hyperarousal in participants with frequent nightmares (NM participants) versus healthy controls. METHODS: Healthy controls and NM participants spent two consecutive nights in the sleep laboratory. Second night spectral power during NREM to REM sleep (pre-REM) and REM to NREM (pos

Impaired emotional memory dissipation in insomnia disorder.

BACKGROUND: Insomnia disorder, characterized by chronic sleep disruption, often co-occurs with maladaptive emotional memory processing. However, much remains unknown regarding the evolution of emotional memories and their neural representations over time among individuals with insomnia disorder. METHOD: We examined the electroencephalographic (EEG) activities during emotional memory encoding, post-encoding sleep, and multiple retrieval phases - including immediate post-encoding, post-sleep, and a 7-day delayed retrieval - among 34 participants with insomnia disorder and 35 healthy control par

Effect of Gabapentin on Sleep and Event-Related Oscillations (EROs) in Rats Exposed to Chronic Intermittent Ethanol Vapor and Protracted Withdrawal.

BACKGROUND: Disturbances in sleep architecture, especially reductions in slow-wave sleep (SWS), are symptoms commonly observed in individuals with alcohol use disorders. Recent clinical trials have demonstrated that the anticonvulsant and analgesic drug gabapentin may have therapeutic value in normalizing sleep quality in recovering alcoholics. However, the brain mechanisms underlying this improvement in sleep following gabapentin treatment remain unknown. METHODS: In this study, adult Wistar rats were exposed to 8 weeks of chronic intermittent ethanol [EtOH] vapor (blood EtOH concentrations

Novel murine closed-loop auditory stimulation paradigm elicits macrostructural sleep benefits in neurodegeneration.

Boosting slow-wave activity (SWA) by modulating slow waves through closed-loop auditory stimulation (CLAS) might provide a powerful non-pharmacological tool to investigate the link between sleep and neurodegeneration. Here, we established mouse CLAS (mCLAS)-mediated SWA enhancement and explored its effects on sleep deficits in neurodegeneration, by targeting the up-phase of slow waves in mouse models of Alzheimer's disease (AD, Tg2576) and Parkinson's disease (PD, M83). We found that tracking a 2 Hz component of slow waves leads to highest precision of non-rapid eye movement (NREM) sleep detec

Examining Sleep-Disordered Breathing Events Using Latent Profile Analysis.

The clinical utility of the ratio of the apnea-hypopnea index (AHI) occurring during rapid eye movement (REM) and non-REM (NREM) sleep (AHIREM/AHINREM ratio) has been debated. We investigated the heterogeneity of REM and NREM sleep behaviors to identify unobserved distinct subtypes of sleep-disordered breathing (SDB) and examine their demographic and clinical features. The present study used a sample of 3626 adult patients who underwent diagnostic polysomnography evaluations at the Sleep Disorders Center of King Abdulaziz Medical City in Riyadh, Saudi Arabia. Latent profile analysis was perfor

Somnogenic, pyrogenic, and hematologic effects of experimental pasteurellosis in rabbits.

Previous work has demonstrated that intravenous inoculation of rabbits with various microorganisms induces complex time-dependent alterations in sleep as well as other pathophysiological effects typically associated with infectious disease. To evaluate the effects of bacterial challenge that more closely resembles naturally developing disease, we inoculated rabbits with Pasteurella multocida, a common pathogen of this species, using routes of administration that mimic normal routes of exposure. Biphasic sleep alterations characterized initially by enhanced slow-wave sleep and later by decrease

Incorporation of recent waking-life experiences in dreams correlates with frontal theta activity in REM sleep.

Rapid eye movement (REM) sleep and its main oscillatory feature, frontal theta, have been related to the processing of recent emotional memories. As memories constitute much of the source material for our dreams, we explored the link between REM frontal theta and the memory sources of dreaming, so as to elucidate the brain activities behind the formation of dream content. Twenty participants were woken for dream reports in REM and slow wave sleep (SWS) while monitored using electroencephalography. Eighteen participants reported at least one REM dream and 14 at least one SWS dream, and they, an

Insufficient non-REM sleep intensity in narcolepsy-cataplexy.

STUDY OBJECTIVES: To compare electroencephalogram (EEG) dynamics during nocturnal sleep in patients with narcolepsy-cataplexy and healthy controls. Fragmented nocturnal sleep is a prominent feature and contributes to excessive daytime sleepiness in narcolepsy-cataplexy. Only 3 studies have addressed changes in homeostatic sleep regulation as a possible mechanism underlying nocturnal sleep fragmentation in narcolepsy-cataplexy. DESIGN, SETTING AND PARTICIPANTS: Baseline sleep of 11 drug-naive patients with narcolepsy-cataplexy (19-37 years) and 11 matched controls (18-41 years) was polysomnogr

[U-133, a heat shock proteins inducer, precludes sleep disturbances in a model of the preclinical stage of Parkinson's disease in aged rats.].

Parkinson's disease (PD) is a chronic progressive neurodegenerative disease, closely associated with aging. It is considered incurable due to both late diagnosis and symptomatic treatment, which is able to alter neither molecular mechanisms of sleep disruption nor the neurodegenerative processes, developing with aging and PD progression. In the present study, we assess the therapeutic potential of a novel chaperone inducer U-133 (acetyl 2,3,7-tris-O-glucoside echinochrome) in the preclinical stage of PD modelled in aged rats by the inhibition of the proteasomal system in the brain. U-133 is a

Sleep during experimental trypanosomiasis in rabbits.

Trypanosoma brucei subspecies cause the human condition known as "sleeping sickness." In rabbits, these organisms induce a chronic and ultimately fatal disease characterized by periodic parasitemia. To characterize sleep alterations during a chronic infectious condition and to determine how immune stimulation of the host, as reflected by cyclic parasitemia, is related to altered somnolence, we monitored sleep and other clinical indices in rabbits inoculated subcutaneously with Trypanosoma brucei brucei. Within four days, infected rabbits developed fever, reduced food intake, and other signs of

Sleep Biomarkers Help Predict the Development of Alzheimer Disease.

Middle-aged or older adults who self-report sleep-wake disorders are at an increased risk for incident dementia, mild cognitive impairment, and Alzheimer disease. Dementia in people with mild cognitive impairment and Alzheimer disease who complain of sleep-wake disorders progress faster than those without sleep-wake disorders. Removal of amyloid-beta and tau tangles occurs preferentially in non-rapid eye movement 3 sleep and fragmented or insufficient sleep may lead to accumulation of these neurotoxins even in preclinical stages. Sleep reduces CSF concentrations of beta-amyloid and tau: a randomized crossover study in healthy adults.

BACKGROUND: Slow-wave sleep has been proposed to facilitate the removal of proteins, implicated in neurodegeneration, from the brain. While mechanistic evidence from animal models is accumulating, direct human data on how slow-wave sleep shapes cerebrospinal fluid (CSF) proteostasis remain limited, constraining our understanding of physiological resilience to neurodegenerative disease. METHODS: Twelve healthy adults (aged 20–40 years) underwent CSF sampling following three controlled sleep conditions in a randomized crossover design; (1) one night of sleep followed by afternoon CSF sampling, (
