Bacteria can migrate in groups of flagella-driven cells over semisolid surfaces. This coordinated form of motility is called swarming behavior. Swarming is associated with enhanced virulence and antibiotic resistance of various human pathogens and may be considered as favorable adaptation to the diverse challenges that microbes face in rapidly changing environments. Consequently, the differentiation of motile swarmer cells is tightly regulated and involves multi-layered signaling networks. Controlling swarming behavior is of major interest for the development of novel anti-infective strategies

The limited scope of DNA-compatible chemistry restricts the types of chemical features that can be incorporated into DNA-encoded libraries (DELs). Here, a method to synthesize DNA-conjugated polycyclic isoxazolidines via a [3 + 2] nitrone-olefin cycloaddition is described. The reaction is compatible with many olefin-containing substrates and diverse N-alkylhydroxylamines. The ability to perform subsequent DNA ligation and PCR amplification was also confirmed. This methodology facilitates the synthesis of DELs containing topographically complex compounds with underexplored chemical features.

Nanometer-scale pores have been developed for the detection, characterization, and quantification of a wide range of analytes (e.g., ions, polymers, proteins, anthrax toxins, neurotransmitters, and synthetic nanoparticles) and for DNA sequencing. We describe the key requirements that made this method possible and how the technique evolved. Finally, we show that, despite sound theoretical work, which advanced both the conceptual framework and quantitative capability of the method, there are still unresolved questions that need to be addressed to further improve the technique.

A major cause of the paucity of new starting points for drug discovery is the lack of interaction between academia and industry. Much of the global resource in biology is present in universities, whereas the focus of medicinal chemistry is still largely within industry. Open source drug discovery, with sharing of information, is clearly a first step towards overcoming this gap. But the interface could especially be bridged through a scale-up of open sharing of physical compounds, which would accelerate the finding of new starting points for drug discovery. In this work, the relationship between cyclophilin A (CypA) and EV71 prompted us to screen a series of small molecular CypA inhibitors which were previously reported by our group. Among them, compounds 1 and 2 were discovered as non-immunosuppressive anti-EV71 agents with an EC50 values of 1.07±0.17μM and 3.36±0.45μM in virus assay, respectively, which were desirably for the further study. The subsequent chemical modifications derived a novel class of molecules, among which compound 11 demonstrated the most potent anti-EV71 activity in virus assay (EC50=0.37±0.17μM), and low cytotoxicity (CC50

A solution-phase parallel synthesis of pharmaceutically important azaspiro[4.5]trienones has been developed by performing tandem Ugi four-component condensation (U4CC), involving substituted p-anisidines, aldehydes, 3-alkyl/aryl-propiolic acids, and isocyanides, and iodine-mediated ipso-iodocyclization in one-pot. This highly atom economical process produced functionalized azaspiro[4.5]trienones in good to excellent overall yields and products were easily isolated by precipitation followed by crystallization. These vinyl-iodide bearing azaspiro[4.5]trienones were utilized for further modificat

Staphylococcus aureus is a leading causative agent in sepsis, endocarditis, and pneumonia. An emerging concept is that prognosis worsens when the infecting S. aureus strain has the capacity to not only colonize tissue as an extracellular pathogen, but to invade host cells and establish intracellular bacterial populations. In previous work, we identified host CDC42 as a central regulator of endothelial cell invasion by S. aureus. In the current work, we report that ML 141, a first-in-class CDC42 inhibitor, decreases invasion and resultant pathogenesis in a dose-dependent and reversible manner.

A growing body of evidence suggests that misfolding of a mutant protein followed by its aggregation or premature degradation in the endoplasmic reticulum is one of the main mechanisms that underlie inherited neurodegenerative diseases, including lysosomal storage diseases. Chemical or pharmacological chaperones are small molecules that bind to and stabilize mutant lysosomal enzyme proteins in the endoplasmic reticulum. A number of chaperone compounds for lysosomal hydrolases have been identified in the last decade. They have gained attention because they can be orally administrated, and also b

Small-molecule drugs are utilized in a wide variety of clinical applications, however, many of these drugs suffer from one or more suboptimal properties that can hinder its delivery or cellular action in vivo, or even shelf an otherwise biologically tolerable drug. While high-throughput screening provides a method to discover drugs with altered chemical properties, directly engineering small-molecule bioconjugates provides an opportunity to specifically modulate drug properties rather than sifting through large drug libraries with seemingly 'random' drug properties. Herein, we propose that sel

DNA-encoded chemical libraries (DELs) accelerate ligand discovery by combining combinatorial synthesis with parallel affinity selection of DNA-tagged compounds. Conventional DEL selection protocols involve the immobilization of protein targets on a solid support such as magnetic beads. Several washes are performed on the beads after library incubation to remove non-binder library members and to enrich for binding compounds. However, excessive washing could also deplete weak or rapidly dissociating binders. Here, we systematically study the impact of the number of washing steps on DEL hit enric

Interleukin-17A (IL-17A) is a proinflammatory cytokine that occupies a central position at the interface of host defense, tissue homeostasis, and chronic inflammation. While IL-17A plays essential roles in antimicrobial immunity and epithelial barrier integrity, dysregulated IL-17A signaling drives the pathogenesis of autoimmune and inflammatory diseases. This functional duality underlies both the clinical success and the inherent limitations of IL-17A signaling-neutralizing therapies, highlighting the need for selective and context-dependent modulation. In this perspective, we provide an over

In today's pharmaceutical sector, innovation is not only the core driver of industry upgrading, but also closely tied to patient health and well-being. A drug's innovative mechanism of action, physiological effects, chemical structure, and indications collectively reflect both its clinical therapeutic value and the novelty of underlying basic research. This article presents the innovative advances of 31 small-molecule drugs (accounting for 67.4%) approved by the U.S. Food and Drug Administration (FDA) in 2025. These therapeutics span multiple therapeutic areas, including oncology, autoimmune d

Autophagy-dependent cell death (ADCD) is a regulated form of cell death that relies on the autophagic machinery. This review provides a systematic update on the landscape of small-molecule compounds that induce ADCD in cancer. We discuss core autophagy-related proteins and the role of autophagic flux in determining lethal outcomes. Compounds are classified by mechanism: inhibitors of the protein kinase B (AKT)/mammalian target of rapamycin (mTOR) axis, activators of the AMP-activated protein kinase (AMPK)/unc-51 like autophagy activating kinase 1 (ULK1) axis, endoplasmic reticulum stress induc

The interleukin (IL) family, a crucial class of cytokines in immune regulation, orchestrates diverse biological processes, including immune response modulation, lymphocyte proliferation/differentiation and pathological mechanisms underlying autoimmune disorders, inflammatory diseases and cancers. Building on this mechanistic foundation, biologic drugs such as monoclonal antibodies targeting ILs and their receptors have achieved significant success. Although developing low-molecular-weight compounds to target the protein-protein interaction interfaces of ILs and their receptors remains challeng

The continued emergence of viral pathogens underscores the need for novel antiviral therapeutics with improved efficacy. Small-molecule inhibitors of viral entry are attractive antiviral agents because they block the virus at its earliest step, preventing the virus from entering host cells and causing downstream damage. Some of the viruses of greatest concern for human health, such as human immunodeficiency virus, respiratory syncytial virus, influenza virus, Ebola virus, severe acute respiratory syndrome coronavirus 2, and Lassa virus, encode Class 1 fusion proteins which mediate membrane fus

Protein misfolding and aggregation are central pathological processes underlying amyloidosis, a group of disorders characterized by the deposition of insoluble amyloid fibrils. These toxic aggregates form due to conformational destabilization and exposure of hydrophobic protein regions, leading to aberrant interactions that disrupt normal cellular functions. Amyloid fibrillation is a key pathological event in both neurodegenerative and systemic disorders. In metabolic and systemic amyloidosis, such as lysozyme derived and insulin derived amyloidosis, peripheral fibril deposition leads to progr

Phosphoglycerate mutase 1 (PGAM1) is an important glycolytic enzyme that plays a significant role in cancer metabolism. It catalyzes the conversion of 3-phosphoglycerate (3-PG) to 2-phosphoglycerate (2-PG), facilitating energy release during glycolysis. PGAM1 is often upregulated in various cancers, including breast, lung, and prostate cancers, contributing to tumor growth and progression by coordinating glycolysis and anabolic processes which makes it an appealing target for drug development. In this study, we screened drugs, food, and natural compound libraries against PGAM1 by using structu

Polymyalgia rheumatica (PMR) and giant cell arteritis (GCA) are systemic inflammatory diseases deeply rooted in age-related immunosenescence and inflammaging. Conventional long-term glucocorticoid (GC) therapy poses significant metabolic and infectious risks for older adults, necessitating safer alternatives. This review critically evaluates the pathophysiological rationale and clinical efficacy of small-molecule drugs, including Janus kinase inhibitors (JAKi) and conventional synthetic disease-modifying antirheumatic drugs (csDMARDs), as steroid-sparing treatments for PMR and GCA. Dengue is no longer confined to tropical and subtropical regions; due to climate change, urbanization, and vector expansion, it has now spread to several Western countries, emerging as a significant global public health threat. A critical unmet need remains for the prevention and treatment of dengue and its complications. In addition to developing vaccines, substantial efforts are underway to discover prophylactic and therapeutic small-molecule inhibitors targeting viral proteins and host factors. This perspective article reviews discovery efforts from the past five years focused on inhibitors
