Neuropsychopharmacology : official publication of the American College of Neuropsychopharmacology2015eng
Neuroimaging has shown promise as a tool to predict likelihood of treatment response in adult anxiety disorders, with potential implications for clinical decision-making. Despite the relatively high prevalence and emergence of anxiety disorders in youth, very little work has evaluated neural predictors of response to treatment. The goal of the current study was to examine brain function during emotional face processing as a predictor of response to treatment in children and adolescents (age 7-19 years; N=41) with generalized, social, and/or separation anxiety disorder. Prior to beginning treat

Recent research has examined the effects of oxytocin (OT) and vasopressin (AVP) on human social behavior and brain function. However, most participants have been male, while previous research in our lab demonstrated sexually differentiated effects of OT and AVP on the neural response to reciprocated cooperation. Here we extend our previous work by significantly increasing the number of participants to enable the use of more stringent statistical thresholds that permit more precise localization of OT and AVP effects in the brain. In a double-blind, placebo-controlled study, 153 men and 151 wome

UNLABELLED: In patients with social anxiety disorder (SAD) it has been reported that selective serotonin reuptake inhibitors (SSRIs) and placebo induce anxiolytic effects by attenuating neural activity in overlapping amygdala subregions, i.e. left basolateral and right ventrolateral amygdala. However, it is not known whether these treatments inhibit amygdala subregions via similar or distinct brain pathways. As anxiolytic treatments may alter amygdala-frontal couplings we investigated differences and similarities in amygdala-frontal functional co-activation patterns between responders and nonr

Social isolation in rodents may interfere in their behavioural responses on paradigms used to test anxiety- and depressive-like states. Herein we study the influence of social isolation upon the behavioural responses of olfactory bulbectomized mice (OBX). In the open-field test (OFT), social isolation enhanced OBX-induced hyperactivity and exploratory behaviour. However, OBX-induced anxiety in the OFT (central activity) was less apparent after isolation, due to the increased level of anxiety showed by the sham-isolated counterparts. In the novelty-suppressed feeding (NSF), isolation derived in

The amygdala is a key structure in the pathophysiology of anxiety disorders, and a putative target for anxiolytic treatments. Selective serotonin reuptake inhibitors (SSRIs) and placebo seem to induce anxiolytic effects by attenuating amygdala responsiveness. However, conflicting amygdala findings have also been reported. Moreover, the neural profile of responders and nonresponders is insufficiently characterized and it remains unknown whether SSRIs and placebo engage common or distinct amygdala subregions or different modulatory cortical areas. We examined similarities and differences in the

BACKGROUND: Generalized Social Anxiety Disorder (GSAD) is characterized by excessive fear and avoidance of several types of social and performance situations. The pathophysiology is not well understood, but research in animals and humans has provided evidence that oxytocin helps regulate normal social affiliative behavior. Previous work in healthy male subjects demonstrated a rise in plasma oxytocin after receiving a high trust signal. To examine the oxytocin system in GSAD, we measured plasma oxytocin in GSAD patients and controls, before and after the social "Trust Game," a neuroeconomic tes

According to the Stress Response Dampening model, problem drinking develops after learning that alcohol limits the stress response in anxiety-provoking situations. However, laboratory-based studies testing alcohol's effects on social anxiety have yielded mixed results. The current study was the first to examine stress response dampening across two contexts: a performance-based (a speech) and an interaction-based (a conversation) social situation. Undergraduates (N = 62; M(age) = 22.85; 31% women; 81% Caucasian) were randomly assigned to consume an alcoholic (target BAC = .08%; n = 22), placebo

Natural selection should favour parents that are able to adjust their offspring's life-history strategy and resource allocation in response to changing environmental and social conditions. Pathogens impose particularly strong and variable selective pressure on host life histories, and parental genes will benefit if offspring are appropriately primed to meet the immunological challenges ahead. Here, we investigated transgenerational immune priming by examining reproductive resource allocation by female mice in response to direct infection with Babesia microti prior to pregnancy. Female mice pre

We compared the response to repeated social defeat in rats selected as high (HR) and low (LR) responders to novelty. In experiment 1, we investigated the behavioral and neuroendocrine effects of repeated social defeat in HR-LR rats. By the last defeat session, HR rats exhibited less passive-submissive behaviors than LR rats, and exhibited higher corticosterone secretion when recovering from defeat. Furthermore, in the forced swim test, while HR defeated rats spent more time immobile than their undefeated controls, LR rats' immobility was unaffected by defeat. In experiment 2, we compared the e

Generalized social anxiety disorder (GSAD) is characterized by excessive fears of scrutiny and negative evaluation, but neural circuitry related to scrutiny in GSAD has been little studied. In this study, 16 unmedicated adults with GSAD and 16 matched healthy comparison (HC) participants underwent functional magnetic resonance imaging to assess neural response to viewed images of faces simulating movement into eye contact versus away from eye contact. GSAD patients were then treated for 8 weeks with paroxetine, and 15 patients were re-imaged. At baseline, GSAD patients had elevated neural resp

Prior research has confirmed a casual path between social rejection and aggression, but there has been no clear explanation of why social rejection causes aggression. A series of experiments tested the hypothesis that social exclusion increases the inclination to perceive neutral information as hostile, which has implications for aggression. Compared to accepted and control participants, socially excluded participants were more likely to rate aggressive and ambiguous words as similar (Experiment 1a), to complete word fragments with aggressive words (Experiment 1b), and to rate the ambiguous ac

CONTEXT: Generalized social phobia (GSP) is characterized by fear/avoidance of social situations. Previous studies have examined the neural responses in GSP to one class of social stimuli, facial expressions. However, studies have not examined the neural response in GSP to another equally important class of social stimuli, the communication of praise or criticism.
OBJECTIVE: To examine the neural response to receipt of praise or criticism in GSP; specifically, to determine whether patients with GSP show an increased response to the receipt of both praise and criticism and whether self-relevan

OBJECTIVES: Hypomania in bipolar disorders is characterized by disinhibited, fearless and reward-seeking behavior. This behavioral pattern suggests that early and automatic responding to socio-emotional cues such as facial expressions might be aberrant in hypomania. The present study tested the predictions that participants selected on hypomania-like trait would show hypovigilant responses to facial cues of danger and increased responses to facial cues of reward.
METHODS: From a group of 513, the 16 most trait-hypomanic individuals were selected by use of a shortened version of an established

This study investigated the joint influence of trait social anxiety and evaluative threat on psychological and cardiovascular responses to active coping situations. Fifty-two normotensive female students characterized as either high or low in trait social anxiety performed a mental arithmetic task and a speech task requiring persuasive behavior in a context of high or low evaluative threat. Trait social anxiety exerted a substantial influence on cardiovascular reactivity. High socially anxious individuals overall exhibited greater heart rate reactivity. For systolic and diastolic blood pressur

Seventy-three agoraphobic and 31 social phobic women, all rated unsuitable for insight-oriented psychotherapy, were compared regarding family and personal history, intelligence, personality and factors pertaining to the disorder. The same patients, with 11 men included in the social phobic group, were also compared regarding response to four randomly assigned types of treatment given over a 3 month period, with a 9 months' follow-up. The following differences were revealed: 1) Social phobias were associated with a higher social class of the parental home, higher education, higher scores on ver

BACKGROUND: There is enough evidence to believe that young children's social-emotional problems can have a long-term effect if extra support is not given early. Therefore, early identification of such problems and any differences between boys and girls are of importance. We utilized the 36-month interval of the Ages and Stages Questionnaires: Social-Emotional (ASQ:SE) among 3-year-olds aiming: 1) to report the normative values of social-emotional problems for Swedish boys and girls; 2) to identify ASQ:SE items that are most commonly endorsed by children with high level of social-emotional prob

BACKGROUND: Social dysfunction is a putative risk and maintaining factor for Eating Disorders (EDs). We assessed biological, emotional, and cognitive responses to a psychosocial stressor, in order to provide a multilevel investigation of the RDoC social process system in EDs.
METHODS: Patients were recruited among those seeking treatment for an ED. Cortisol response to Trier Social Stress Test (TSST) was measured in 105 subjects: 35 women with anorexia nervosa (AN), 32 with bulimia nervosa (BN) and 38 healthy women. Anxiety, hunger, and desire to eat throughout TSST were rated in a subgroup o

Many neural systems are undergoing marked development over adolescence, which may heighten an animal's vulnerability to stressors. One consequence may be altered sensitivity to drugs of abuse. We previously reported that social stressors in adolescence increased behavioral sensitization to nicotine in adulthood in female, but not male, rats. Here we examined whether social stressors in adolescence alter the functioning of the hypothalamic-pituitary-adrenal (HPA) axis by examining corticosterone release in response to restraint in adulthood. To further assess effects of social stressors on beha
