Physiological increases in lactate inhibit intracellular calcium transients, acidify myocytes and decrease force in term pregnant rat myometrium.

Lactate is increased in myometrial capillary blood from women in slow or non-progressive labour (dystocia), suggesting that it is detrimental to uterine contractions. There are, however, no studies of the effect of lactate on the myometrium. We therefore investigated its effects and mechanism of action on myometrial strips from term pregnant rats. The effects on spontaneous and oxytocin-induced contractility in response to sodium lactate and other weak acids (1-20 mM) were studied. In some experiments, simultaneous force and intracellular Ca(2+) or pH (pH(i)) were measured with Indo-1 or Carbo

The anti-hypoxic effects of oat (Avena sativa L.) oligopeptides in mice.

OBJECTIVE: To explore the anti-hypoxic effects of oat oligopeptides (OOPs) in mice. METHODS: We randomly divided mice into six groups, including a vehicle control group, a whey protein group (0.50 g/kg), and four OOPs-treated groups (0.25, 0.50, 1.00, and 2.00 g/kg). The test substances were administered by gavage once a day for 30 days. The normobaric hypoxia, sodium nitrite toxicosis, and acute cerebral ischemia survival times were recorded. Also, the MDA content, the lactate levels, the LDH activity, and the mRNA levels of HIF-1α and VEGF in the brains were measured. We performed a whole b

Hypernatremic dehydration due to lactation failure in an exclusively breastfed neonate.

We report a 13-day-old exclusively breastfed neonate, admitted with a history of fever, poor feeding, lethargy and decreased urine output. The mother had history of lactation failure. The neonate had severe hypernatremia, acute renal failure and metabolic acidosis. Renal ultrasound was normal, but the sodium level in mother's milk was 96 mEq/L (normal 7 ± 2 mEq/L). The neonate required peritoneal dialysis on the second day of admission. The biochemical investigations gradually returned to normal and the neonate was discharged on day 12 of admission. We highlight the importance of lactation man

The physiological and ventilatory responses to repeated 60 s sprints following sodium citrate ingestion.

This study examined the influence of sodium citrate on changes in selected blood, ventilatory and performance variables in response to intermittent sprint exercise. Eight moderately active male students completed three tests over a 6 day experimental period. The first test involved incremental exercise to determine VO2 max, while the second and third tests were identical in nature and involved five 60 s sprints cycling against 0.075 kg kg-1 body mass (BM); each of the five sprints was separated by 5 min passive seated recovery. Three days separated the VO2 max test and first interval test, whi

An effective assessment of valproate sodium-induced hepatotoxicity with UPLC-MS and (1)HNMR-based metabonomics approach.

Valproate sodium is one of the most prescribed antiepileptic drugs. However, valproate sodium has various side effects, especially its toxicity on liver. Current markers for toxicity reflect mostly the late stages of tissue damage; thus, more efficient methods for toxicity evaluation are desired. To evaluate the toxicity of valproate sodium on liver, we performed both UPLC-MS and (1)HNMR-based metabonomics analysis of serum samples from 34 epileptic patients (age: 42.0±18.6, 18 male/16 female) after valproate sodium treatment. Compared to conventional markers, the serum metabolic profiles prov

Hyperosmotic oral replacement fluid for diarrheic calves.

Twenty-five diarrheic neonatal calves, none of which was recumbent, were used to determine oral efficacy of a commercial product for fluid replacement, either by suckle or intubation. Changes in plasma protein, glucose, sodium, potassium, and osmolality as well as blood lactate and PCV were measured. Core body temperature and surface temperature at the hock and fetlock were monitored. Data were analyzed for interaction with time and response according to clinical condition. Significant differences (P less than 0.05) following therapy were restricted to glucose and potassium changes. Beneficial

The effect of pre-exercise alkalosis on lactate/pH regulation and mitochondrial respiration following sprint-interval exercise in humans.

Purpose: The purpose of this study was to evaluate the effect of pre-exercise alkalosis, induced via ingestion of sodium bicarbonate, on changes to lactate/pH regulatory proteins and mitochondrial function induced by a sprint-interval exercise session in humans. Methods: On two occasions separated by 1 week, eight active men performed a 3 × 30-s all-out cycling test, interspersed with 20 min of recovery, following either placebo (PLA) or sodium bicarbonate (BIC) ingestion. Results: Blood bicarbonate and pH were elevated at all time points after ingestion in BIC vs PLA (p < 0.05). The protein c

Ferrous lactate enhances pressure ulcer healing and is associated with improved inflammatory, fibroblast, and angiogenic responses.

Pressure ulcers (PUs), also termed pressure injuries, are chronic inflammatory wounds associated with recurrent ischemia-reperfusion (I/R), impaired fibroblast repair, poor vascularization, and delayed extracellular matrix remodeling. We evaluated ferrous lactate in hypoxia/reoxygenation-injured human dermal fibroblasts (HDFs) and in a rabbit PU model generated by cyclic magnetic compression. In HDFs subjected to I/R-like stress, ferrous lactate increased viability, proliferation, and scratch migration, reduced TNF-α, IL-6, and IL-1β, and increased IL-10 and TGF-β. For in vivo treatment, ferro

Lactate attenuates astrocytic inflammation by inhibiting ubiquitination and degradation of NDRG2 under oxygen-glucose deprivation conditions.

BACKGROUND: Brain lactate concentrations are enhanced in response to cerebral ischemia and promote the formation of reactive astrocytes, which are major components of the neuroinflammatory response and functional recovery, following cerebral ischemia. NDRG2 is upregulated during reactive astrocyte formation. However, its regulation and function are unclear. We studied the relationship between lactate and NDRG2 in astrocytes under conditions of ischemia or oxygen-glucose deprivation (OGD). METHODS: We examined astrocytic NDRG2 expression after middle cerebral artery occlusion (MCAO) using west

Effects of sodium bicarbonate ingestion on anaerobic performance: a meta-analytic review.

Many researchers have investigated the effects of induced metabolic alkalosis, by ingestion of sodium bicarbonate, on anaerobic exercise performance. But the results have been inconsistent and often contradictory. The purpose of this review was to synthesize the varied findings using a meta-analytic approach. Twenty-nine investigations met our inclusion criteria. Results show that NaHCO3 ingestion clearly results in a more alkaline extracellular environment. The dosage, however, was only moderately related to the increase in pH and HCO3-. Overall, performance was enhanced but the range of effe

Metabolic changes in a case of malignant hyperthermia.

A patient with fulminant MH is presented. The metabolic changes in muscle tissue, whole blood and plasma are compared with those obtained from the porcine model. The samples were taken 1 h after treatment (including dantrolene sodium) had been commenced. Increased concentrations of lactate, pyruvate and citrate were found in muscle, whereas the concentrations of potassium, free inorganic phosphate and glycogen were reduced. The ATP, ADP and phosphoryl creatine concentrations remained unchanged. In blood, increased concentrations of alanine, lactate and glycerol were observed. An increase in ca

Successful Use of Hydroxocobalamin and Sodium Thiosulfate in Acute Cyanide Poisoning: A Case Report with Follow-up.

Hydroxocobalamin is an effective first-line antidote used mainly in monotherapy of cyanide poisonings, while the opinions are different on the effects of its combination with sodium thiosulfate. A 58-year-old male committed a suicide attempt by ingesting of 1200-1500 mg of potassium cyanide; he was unconscious for 1-1.5 min. after ingestion with the episode of generalized seizures. On admission to the ICU, the patient was acidotic (pH 7.28; HCO3 14.0 mmol/L, base excess -12.7 mmol/L, saturation O2 0.999) with high serum lactate (12.5 mmol/L). Hydroxocobalamin was administered 1.5 hr after inge

Intravenous and Oral Fluid Therapy in Neonatal Calves With Diarrhea or Sepsis and in Adult Cattle.

Optimal fluid therapy protocols in neonatal calves and adult cattle are based on consideration of signalment, history, and physical examination findings, and individually tailored whenever laboratory analysis is available. Measurement of the magnitude of eye recession, duration of skin tenting in the lateral neck region, and urine specific gravity by refractometry provide the best estimates of hydration status in calves and cattle. Intravenous and oral electrolyte solutions (OES) are frequently administered to critically ill calves and adult cattle. Application of physicochemical principles in

Differential responses to anxiogenic drugs in a mouse model of panic disorder as revealed by Fos immunocytochemistry in specific areas of the fear circuitry.

Sensitivity to pharmacological challenges has been reported in patients with panic disorder. We have previously validated transgenic mice overexpressing the neurotrophin-3 (NT-3) receptor, TrkC (TgNTRK3), as an engineered murine model of panic disorder. We could determine that TgNTRK3 mice presented increased cellularity in brain regions, such as the locus ceruleus, that are important neural substrates for the expression of anxiety in severe anxiety states. Here, we investigated the sensitivity to induce anxiety and panic-related symptoms by sodium lactate and the effects of various drugs (the

Hypoxia-driven HIF-1α-dependent glycolysis/lactate-associated signaling promotes autophagy-associated scleral remodeling in experimental myopia.

Myopic axial elongation is driven by scleral remodeling, yet the metabolic events linking scleral hypoxia to autophagy-associated remodeling remain incompletely defined. We examined whether HIF-1α-dependent glycolytic reprogramming and lactate-associated metabolic stress connect hypoxia to remodeling changes in human scleral fibroblasts (HSFs) and in a guinea pig model of form-deprivation myopia (FDM). RNA sequencing showed significant enrichment of HIF-1 signaling and glycolysis/gluconeogenesis in hypoxic HSFs. Hypoxia increased HIF-1α, HK2, PFKP, and LDHA expression and raised intracellular

Lactate induces renal lipid accumulation and aggravates renal fibrosis by inhibiting the PPARα signaling pathway and fatty acid oxidation.

Chronic kidney disease (CKD) is characterized by renal fibrosis as its core pathological feature, and lipid metabolism disorders are a key driver of disease progression. However, the specific pathological significance of elevated lactate levels in patients with CKD remains unclear. This study aimed to verify the hypothesis that lactate exacerbates renal fibrosis by inhibiting the PPARα/FAO pathway. A total of 15 healthy controls and 75 CKD patients were enrolled. Serum lactate levels were measured, and their correlations with Scr, BUN, eGFR, and lipid metabolism parameters (triglycerides [TG],

pH effects on lactate and excess lactate in relation to O2 deficit in hypoxic dogs.

Both hypoxemia and alkalemia increase arterial lactate levels, but excess lactate (XL) may be better related to the true O2 deficit and less subject to the pH effect upon glycolysis. To test this idea, 50 anesthetized (30 ml/kg pentobarbital sodium) and paralyzed dogs in five groups were ventilated with low O2 mixtures. Arterial pH was altered by CO2, hyperventilation, and HCO3- so that the average pH of the five groups were 6.99, 7.21, 7.34, 7.53, and 7.54 (+ propranolol). Pyruvate and lactate levels rose at rates roughly corresponding to pH. In all animals, rise of lactate (deltaL), XL and n
