Technique for specimen collection of postsurgical exudate of wounds.

Postsurgical sepsis due to anaerobic microoganisms is often difficult to diagnose. The difficulty arises primarily because of inadequate specimen collection techniques and because of delays in the transport of the specimen to the microbiologic laboratory. Three acceptable techniques for anaerobic specimen collection and transportation have been offered. All of these techniques have in common the minimization of contact between the collected specimen and the atmosphere, a step which helps to preserve the viability of many of the fastidious anaerobic bacteria frequently implicated in surgical se

Nonspecific urethritis in females.

The possibility of nonspecific urethritis must be considered in females with persistent irritative symptoms of the lower urinary tract despite "negative" urine cultures. The diagnosis can be made only by the proper collections of urine specimens from both the urethra and the bladder. These specimens will reveal the presence of white blood cells in the urethral washings, while the midstream (bladder) specimen will be free of cells.

Assessing VirScan serosurvey epitope profiling variability between in-clinic venous blood draw and capillary blood self-sampling device.

Timely and ongoing in-clinic sample collections are a common logistical barrier to volunteer participation and retention in longitudinal clinical studies. To remove this barrier, clinical studies have recently begun to implement the use of at-home capillary blood self-sampling devices in place of in-clinic venous blood draws for participant blood sample collection. Thus, we assessed antibody responses to a broad library of over 300 viral, bacterial, and fungal pathogens using VirScan immunoassay testing on adult volunteer capillary (serum) and venous (plasma) blood samples collected by Tasso d

Evaluation of the impact of PDCA cycle management on heparin anticoagulation specimen quality rate in neurorespiratory care units.

BACKGROUND: The quality of heparin-anticoagulated specimens is of paramount importance for the delivery of highly accurate diagnostic results in neurorespiratory settings. However, pre-analytical factors such as hemolysis, clotting, and labeling errors often threaten this quality. This paper explores if the use of a Plan-Do-Check-Act cycle management would improve the quality of the heparin anticoagulation specimen. OBJECTIVES: To evaluate the effectiveness of managing the PDCA cycle to improve the quality of heparin anticoagulation specimen samples in a neurorespiratory care unit through err

Evidence-based standardization of infectious disease biobanking: aligning specimen preservation and clinical metadata with in vitro diagnostic clinical trials requirements.

BACKGROUND: Biobanks accelerate in vitro diagnostic (IVD) clinical trials by providing annotated, quality-controlled biospecimens. However, a persistent translational gap remains because the specific requirements of IVD trials-including sample matrices, preservation protocols, aliquot volumes, freeze-thaw tolerability, and clinical metadata-have not been systematically characterized. This study aims to establish an evidence-based framework to align infectious disease biobanking operations with regulatory-grade IVD trial demands by systematically characterizing the specimen preservation paramet

Using a standardized blood culture collection kit to reduce the rate of contamination in the emergency room.

Accurate blood culture collection is essential for guiding appropriate antibiotic therapy, with contamination posing a risk to diagnostic reliability. The American Society for Microbiology recommends contamination rates below 3% for optimal patient outcomes. In collaboration with the emergency and microbiology departments, a large rural hospital implemented a standardized blood culture collection kit as a quality improvement initiative that led to a reduction in contamination rates, reaching a low of 0.97% over several months.

Processing crime scenes: The influence of DNA success rate information on the collection and triage of traces at crime scenes.

Crime scene investigations require rapid decision-making under complex conditions, often without the opportunity for revision. Task-relevant information from similar cases can support Scene of Crime Officers (SoCOs) in making these decisions. Decision support can reduce human error and subjectivity, provide a sound basis for decision-making, and enhance transparency. What is not known is how and when to provide such decision support information so as to have the most improved outcomes. This study examines the influence of (1) providing information on the probability of recovering perpetrator D

A comparative analysis of three distinct rehydrating solutions for addressing liver tissue desiccation resulting from dehydration failure.

BACKGROUND: The tissue dehydration process is an important step in pathology specimen preparation. Dehydration failure, caused by equipment malfunction, incorrect program settings, or operator error, is a recognized problem in laboratory practice and can compromise the quality of hematoxylin and eosin (H&E) slides, thereby hindering subsequent pathological diagnosis. Common ad hoc responses include stepwise rehydration or short-term immersion in various buffers. However, information on repairing over-desiccated specimens is limited, and there is little systematic evidence comparing different r

Innovation in acid-fast staining methods: A comparative study of a modified staining rack, a conventional rack and an automated stainer.

Manual acid-fast staining often results in specimen slippage, inaccurate positioning, and increased biosafety risks. This study evaluated a modified staining rack incorporating a slide-positioning device and an adjustable tilt function to improve the stability and safety of manual acid-fast staining. A total of 142 clinical specimens (70 sputum, 30 bronchoalveolar lavage fluid, 32 pleural effusion and 10 stool specimens) were stained using the modified rack, a conventional rack, and a BSZ-GT116 fully automated stainer. The single-batch staining time with the modified rack was 935.33 ± 20.91 s,

Validation of rectal swabbing for total and aerobic gut microbiota study.

UNLABELLED: In microbiota research, whole stool sampling is the conventional approach but can be problematic or infeasible for certain patients. This study aims to validate the use of rectal swabbing as an alternative method for microbiota analysis and determine optimal storage conditions suitable for various clinical settings, including intensive care units. We evaluated different sampling techniques and storage temperatures. Our findings indicated that rectal swabs yield microbiota diversity comparable to whole stool samples. Notably, storage conditions significantly impacted microbiota prof

Holding time or fixative formulation has no obvious effect on histology of Porites evermanni and Montipora capitata.

Collection of coral for histologic examination requires holding of samples in seawater for a time before they are fixed for histologic processing. This could adversely affect the interpretation of morphologic changes during histologic examinations. We evaluated the microscopic morphology of Porites evermanni and Montipora capitata held (0-120 minutes) in seawater prior to fixation in Z-Fix formulated with raw or artificial seawater. We saw no evident effects of treatments on microscopic morphology. However, among 88 statistical comparisons, and after accounting for false discovery rate, holdin

Choosing the Best Formalin-Resistant Ink for Biopsy Specimen Labeling.

Accurate specimen identification is critical in the everyday practice of dermatology. Formalin, which is used to preserve tissue samples, also acts as a solvent and can cause fading of the ink used to label specimen containers. Herein, we identify commonly used pens and markers with various inks that are susceptible or resistant to accidental formalin exposure to ensure durability of specimen labeling.

Exploring improved sample recovery and processing methods targeting Bacillus anthracis spores from vegetation matrices.

Following an accidental or intentional outdoor release of an environmentally persistent biological agent, such as Bacillus anthracis (Ba), a sampling campaign involving the collection of bulk vegetation matrices (grass, ground litter, and leaves) to determine the scope of contamination might be part of a remediation plan. These vegetative materials contain fungi, bacteria, and/or other naturally present background microorganisms that will impact target analyte recovery. This study aimed to optimize recovery of surrogate Ba spores by exploring 1) methods to separate target spores from non-targe

Optimizing Tear Collection in Mice for mRNA and Protein Analysis.

The tear film is a highly dynamic biofluid capable of reflecting pathology-associated molecular changes, not only in the ocular surface but also in other tissues and organs. Molecular analysis of this biofluid offers a non-invasive way to diagnose or monitor diseases, assess medical treatment efficacy, and identify possible biomarkers. Due to the limited sample volume, collecting tear samples requires specific skills and appropriate tools to ensure high quality and maximum efficiency. Various tear sampling methodologies have been described in human studies. In this article, a comprehensive des

Optimizing Skin Surface Metabolomics: A Comprehensive Evaluation of Sampling Methods, Extraction Solvents, and Analytical Techniques.

Characterizing the metabolite fingerprint from the skin surface provides invaluable insights into skin biology and microbe-host interactions. To ensure data accuracy and reproducibility, it is essential to develop standard operating procedures for skin surface metabolomics. However, there is a notable lack of studies in this area. In this study, we thoroughly evaluated different sampling materials, extraction solvents, taping methods (frequency and number of tapes), and analytical techniques to optimize skin surface metabolomics. Our results showed that the combination of D-Squame D100 tape wi
