Role of Sphingolipids in Multiple Myeloma Progression, Drug Resistance, and Their Potential as Therapeutic Targets.

Multiple myeloma (MM) is an incapacitating hematological malignancy characterized by accumulation of cancerous plasma cells in the bone marrow (BM) and production of an abnormal monoclonal protein (M-protein). The BM microenvironment has a key role in myeloma development by facilitating the growth of the aberrant plasma cells, which eventually interfere with the homeostasis of the bone cells, exacerbating osteolysis and inhibiting osteoblast differentiation. Recent recognition that metabolic reprograming has a major role in tumor growth and adaptation to specific changes in the microenvironmen

Deregulation of signalling in genetic conditions affecting the lysosomal metabolism of cholesterol and galactosyl-sphingolipids.

The role of lipids in neuroglial function is gaining momentum in part due to a better understanding of how many lipid species contribute to key cellular signalling pathways at the membrane level. The description of lipid rafts as membrane domains composed by defined classes of lipids such as cholesterol and sphingolipids has greatly helped in our understanding of how cellular signalling can be regulated and compartmentalized in neurons and glial cells. Genetic conditions affecting the metabolism of these lipids greatly impact on how some of these signalling pathways work, providing a context t

Placental Production of Eicosanoids and Sphingolipids in Women Who Developed Preeclampsia on Low-Dose Aspirin.

Low-dose aspirin, which selectively inhibits thromboxane synthesis, is now standard of care for the prevention of preeclampsia in at risk women, but some women still develop preeclampsia despite an aspirin regimen. To explore the "aspirin failures," we undertook a comprehensive evaluation of placental lipids to determine if abnormalities in non-aspirin sensitive lipids might help explain why some women on low-dose aspirin develop preeclampsia. We studied placentas from women with normal pregnancies and women with preeclampsia. Placental villous explants were cultured and media analyzed by mass

Mass and relative elution time profiling: two-dimensional analysis of sphingolipids in Alzheimer's disease brains.

Current lipidomic profiling methods rely mainly on MS to identify unknown lipids within a complex sample. We describe a new approach, involving LC×MS/MS (liquid chromatography×tandem MS) analysis of sphingolipids based on both mass and hydrophobicity, and use this method to characterize the SM (sphingomyelin), ceramide and GalCer (galactosylceramide) content of hippocampus from AD (Alzheimer's disease) and control subjects. Using a mathematical relationship we exclude the influence of sphingolipid mass on retention time, and generate two-dimensional plots that facilitate accurate visualization

Bioactive Sphingolipids of Cordyceps sinensis Modulate Tumor Immunity Through the SphK1-Mediated Cer/S1P Axis.

Cordyceps sinensis is a traditional medicinal fungus known for its immunomodulatory properties. Its bioactive sphingolipids (SPLs) exhibit antitumor potential, though their mechanisms remain poorly understood. This study aimed to identify the active SPLs from C. sinensis and investigate their synergistic effects with αPD-1 (anti-programmed death-1 antibody) therapy. SPLs were isolated from C. sinensis via UPLC-MS/MS-guided purification. The antitumor efficacy and immunomodulatory synergy with αPD-1 were assessed using a Lewis lung carcinoma (LLC) tumor-bearing mouse model and in vitro co-cultu

Very long chain sphingolipids govern brain myelination by regulating oligodendrocyte differentiation and membrane microdomain integrity.

BACKGROUND: Myelin abnormalities, which have no effective treatment to-date, underlie numerous debilitating neurological disorders like Multiple Sclerosis (MS). While lipids constitute ~70% of myelin, the specific lipids critical for establishment of myelin sheath, and the mechanisms linking lipid metabolism to myelin assembly, have remained poorly defined, hindering therapeutic development. METHODS: We generated central nervous system-specific (cKO-nestin) and oligodendrocyte-specific (cKO-OL) conditional knockout mice for Ceramide Synthase 2 (CerS2), which synthesizes very long-chain (VLC; C

Reduced circulating sphingolipids and CERS2 activity are linked to T2D risk and impaired insulin secretion.

Gestational diabetes mellitus (GDM), a transient form of diabetes that resolves postpartum, is a major risk factor for type 2 diabetes (T2D) in women. While the progression from GDM to T2D is not fully understood, it involves both genetic and environmental components. By integrating clinical, metabolomic, and genome-wide association study (GWAS) data, we identified associations between decreased sphingolipid biosynthesis and future T2D, in part through the rs267738 allele of the CERS2 gene in Hispanic women shortly after a GDM pregnancy. To understand the impact of the CERS2 gene and risk alle

Urothelial Urinary Bladder Cancer Is Characterized by Stage-Dependent Aberrations in Metabolism of Bioactive Sphingolipids.

Although dysregulated sphingolipid metabolism was observed in many malignant tumors, bladder cancer has not yet been examined in this regard. This study aims to investigate the metabolism of bioactive sphingolipids across different stages of urothelial urinary bladder cancer (UBC). Forty-eight patients with UBC were included in this study. The neoplasms were classified as either non-muscle-invasive (NMIBC, n = 24) or muscle-invasive (MIBC, n = 24). Samples of the healthy bladder tissue were taken from the patients who underwent radical cystectomy. The content of sphingolipids was measured usin

The Role of Sphingolipids and Sphingosine-1-phosphate-Sphingosine-1-phosphate-receptor Signaling in Psoriasis.

Psoriasis is a long-lasting skin condition characterized by redness and thick silver scales on the skin's surface. It involves various skin cells, including keratinocytes, dendritic cells, T lymphocytes, and neutrophils. The treatments for psoriasis range from topical to systemic therapies, but they only alleviate the symptoms and do not provide a fundamental cure. Moreover, systemic treatments have the disadvantage of suppressing the entire body's immune system. Therefore, a new treatment strategy with minimal impact on the immune system is required. Recent studies have shown that sphingolipi

Fatty Acid 2-Hydroxylase and 2-Hydroxylated Sphingolipids: Metabolism and Function in Health and Diseases.

Sphingolipids containing acyl residues that are hydroxylated at C-2 are found in most, if not all, eukaryotes and certain bacteria. 2-hydroxylated sphingolipids are present in many organs and cell types, though they are especially abundant in myelin and skin. The enzyme fatty acid 2-hydroxylase (FA2H) is involved in the synthesis of many but not all 2-hydroxylated sphingolipids. Deficiency in FA2H causes a neurodegenerative disease known as hereditary spastic paraplegia 35 (HSP35/SPG35) or fatty acid hydroxylase-associated neurodegeneration (FAHN). FA2H likely also plays a role in other diseas

Circular Network of Coregulated Sphingolipids Dictates Chronic Hypoxia Damage in Patients With Tetralogy of Fallot.

Background: Tetralogy of Fallot (TOF) is the most common cyanotic heart disease. However, the association of cardiac metabolic reprogramming changes and underlying molecular mechanisms in TOF-related chronic myocardial hypoxia damage are still unclear. Methods: In this study, we combined microarray transcriptomics analysis with liquid chromatography tandem-mass spectrometry (LC-MS/MS) spectrum metabolomics analysis to establish the metabolic reprogramming that occurs in response to chronic hypoxia damage. Analysis of sphingolipids in extracted human plasma using liquid chromatography electrospray ionization tandem mass spectrometry.

We have developed an analytical method used to quantify sphingolipids, including deoxysphingoid bases, in lipid extracts prepared from human plasma. In total, 39 analytes were identified and analyzed in a single chromatographic run in less than 5 min. The new method is 4-8 times faster and more sensitive than previously published methods. We also describe a simple sample preparation method that allows medium-throughput screening of human plasma samples. Mass spectrometric analyses were performed online using liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS)

Molecular insights into amyloid regulation by membrane cholesterol and sphingolipids: common mechanisms in neurodegenerative diseases.

Alzheimer, Parkinson and other neurodegenerative diseases involve a series of brain proteins, referred to as 'amyloidogenic proteins', with exceptional conformational plasticity and a high propensity for self-aggregation. Although the mechanisms by which amyloidogenic proteins kill neural cells are not fully understood, a common feature is the concentration of unstructured amyloidogenic monomers on bidimensional membrane lattices. Membrane-bound monomers undergo a series of lipid-dependent conformational changes, leading to the formation of oligomers of varying toxicity rich in beta-sheet stru

Deciphering the role of ergosterol and sphingolipids in the antifungal mode of action of rhamnolipids on Sclerotiniaceae.

Rhamnolipids (RLs) are natural glycolipids very promising to reduce the use of pesticides to protect crops. They have efficient antifungal properties against Sclerotiniaceae fungi. They are membranotropic compounds permeabilizing fungal cells. We have previously proposed that a lower ergosterol amount could be linked to a lower antifungal sensitivity. The main objective of this paper was to gain insights into the role of plasma membrane lipids in the antifungal mode of action of RLs on Sclerotiniaceae. We obtained ergosterol biosynthesis pathway mutants of Botrytis cinerea and evaluated their

Dysregulation of sphingolipid and cholesterol homeostasis imposes oxidative stress in human spermatozoa.

Infertility is a significant public health concern, affecting one in six couples globally, with male-factor infertility representing half of all cases. Obesity is an important health problem and is increasingly linked to poor reproductive outcomes as it induces metabolic disturbances and dyslipidemia, both of which impair male fertility. Dyslipidemia, characterized by abnormal lipid profiles, is associated with declining sperm quality and alters cholesterol and sphingolipid abundances in the seminal plasma. Sperm capacitation enables spermatozoa to acquire fertilizing competence by promoting c

Sphingolipid de novo synthesis is upregulated in a macrophage model of Gaucher disease.

Gaucher disease (GD) is an inborn error of sphingolipid metabolism characterised by a block in the lysosomal degradation of glucosylceramide (GlcCer), which consequently accumulates in the lysosomes of affected cells. The product of GlcCer degradation is ceramide, and it is reasonable to expect that there would be a reduction in ceramide in GD, but there is conflicting evidence in different cells and tissues. Using a stable isotope 13C16-palmitate labelling method, we sought to investigate the impact of the block in GlcCer degradation on the sphingolipid metabolic pathway in a conduritol B epo

Tezacaftor is a direct inhibitor of sphingolipid delta-4 desaturase enzyme (DEGS).

BACKGROUND: We recently demonstrated that 48 h exposure of primary human bronchial epithelial (hBE) cells, obtained from both CF (F508del homozygous) and non-CF subjects, to the triple drug combination Elexacaftor/Tezacaftor/Ivacaftor (ETI) results in a CFTR genotype-independent modulation of the de novo synthethic pathway of sphingolipids, with an accumulation of dihydroceramides (dHCer). Since dHCer are converted into ceramides (Cer) by the action of a delta-4 sphingolipid desaturase (DEGS) enzyme, we aimed to better understand this off-target effect of ETI (i.e., not related to CFTR rescue)

Blood sphingolipid as a novel biomarker in patients with neuromyelitis optica spectrum disorder.

BACKGROUND: Sphingolipids are signaling molecules and structural components of the axolemma and myelin sheath. Plasma sphingolipid levels may reflect disease status of neuromyelitis optica spectrum disorder (NMOSD). We aimed to examine plasma sphingolipids as disease severity biomarkers for NMOSD and compare their characteristics with those of serum neurofilament light chain (sNfL) and glial fibrillary acidic protein (sGFAP). METHODS: We measured plasma sphingolipids, sNfL, and sGFAP levels in NMOSD cases with anti-aquaporin-4-antibody. An unbiased approach, partial least square discriminant

Sphingolipid diversity in Candida auris: unraveling interclade and drug resistance fingerprints.

In this study, we explored the sphingolipid (SL) landscape in Candida auris, which plays pivotal roles in fungal biology and drug susceptibility. The composition of SLs exhibited substantial variations at both the SL class and molecular species levels among clade isolates. Utilizing principal component analysis, we successfully differentiated the five clades based on their SL class composition. While phytoceramide (PCer) was uniformly the most abundant SL class in all the isolates, other classes showed significant variations. These variations were not limited to SL class level only as the prop
