Gut Microbial Diversity Reveals Differences in Pathogenicity between Metarhizium rileyi and Beauveria bassiana during the Early Stage of Infection in Spodoptera litura Larvae.

Beauveria bassiana and Metarhizium rileyi are extensively utilized to investigate fungal pathogenic mechanisms and to develop biological control agents. Notwithstanding, notable distinctions exist in their pathogenicity against the same host insect. This study aimed to elucidate the pathogenic differences between M. rileyi and B. bassiana by examining the impact of various ratios of B. bassiana strain AJS91881 and M. rileyi strain SXBN200920 on fifth instar larvae of Spodoptera litura, focusing on early infection stages and intestinal microbial community structure. The lethal time 50 (LT50) fo

Characterization of ATP-binding cassette transporters associated with emamectin benzoate tolerance: from the model insect Drosophila melanogaster to the agricultural pest Spodoptera frugiperda.

BACKGROUND: Multiple families of detoxification genes, including the increasingly recognized family of ATP-binding cassette (ABC) transporters, work together to influence the toxicity of synthetic insecticides and thus their resistance. Effective management of insecticide resistance requires identification of all toxicity-affecting members from each family of toxicity-related genes. RESULTS: Here, we used emamectin benzoate (EB), ABC transporters and Spodoptera frugiperda as a working case to test whether the strategy of 'from the model insect Drosophila melanogaster to agricultural pests' ca

Involvement of small heat shock proteins (sHsps) in developmental stages of fall armyworm, Spodoptera frugiperda and its expression pattern under abiotic stress condition.

Fall armyworm (FAW), Spodoptera frugiperda a recent invasive pest in India is reported to cause significant damage by feeding voraciously on maize and other economic crops from tropical to temperate provinces. It is becoming an arduous challenge to control the pest as it can survive in a wide range of temperature conditions and is already said to develop resistance towards certain insecticides. The small Heat shock proteins (hereafter, sHsps) are known to play an important role in adaptation of insects under such stress conditions. Our present study involved characterization of the three sHsps

Simulation and Prediction of the Potential Distribution of Spodoptera frugiperda Under Current and Three Future Scenarios Based on the Biomod2 Model Under Global Climate Change.

Spodoptera frugiperda is a highly polyphagous pest that poses a serious threat to global agricultural production. Understanding its species distribution is of great importance under the current context of climate change. In this study, we used the biomod2 platform to integrate six models, namely XGBOOST, Maxnet, Maxent, MARS, GBM, and GLM, to construct an ensemble model. This ensemble model was used to simulate the potential distribution of S. frugiperda under current and three future climate scenarios, and to analyze the main environmental factors influencing its distribution and their change

Effects of feeding Spodoptera littoralis on lima bean leaves. II. Continuous mechanical wounding resembling insect feeding is sufficient to elicit herbivory-related volatile emission.

Herbivore feeding elicits defense responses in infested plants, including the emission of volatile organic compounds that can serve as indirect defense signals. Until now, the contribution of plant tissue wounding during the feeding process in the elicitation of defense responses has not been clear. For example, in lima bean (Phaseolus lunatus), the composition of the volatiles induced by both the insect caterpillar Spodoptera littoralis and the snail Cepaea hortensis is very similar. Thus, a mechanical caterpillar, MecWorm, has been designed and used in this study, which very closely resemble

The Fall Armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), Influences Nilaparvata lugens Population Growth Directly, by Preying on Its Eggs, and Indirectly, by Inducing Defenses in Rice.

The fall armyworm (FAW), Spodoptera frugiperda, has become one of the most important pests on corn in China since it invaded in 2019. Although FAW has not been reported to cause widespread damage to rice plants in China, it has been sporadically found feeding in the field. If FAW infests rice in China, the fitness of other insect pests on rice may be influenced. However, how FAW and other insect pests on rice interact remains unknown. In this study, we found that the infestation of FAW larvae on rice plants prolonged the developmental duration of the brown planthopper (BPH, Nilaparvata lugens

Two ABC transporters are differentially involved in the toxicity of two Bacillus thuringiensis Cry1 toxins to the invasive crop-pest Spodoptera frugiperda (J. E. Smith).

BACKGROUND: The fall armyworm Spodoptera frugiperda is a major agricultural pest that has invaded the East Hemisphere since 2016, generating a serious threat to food security worldwide including Africa and Asia. The Cry toxins produced by Bacillus thuringiensis (Bt) have been shown to be effective against this insect pest. In different insect ABC transporters (ABCC2 or ABCC3) have been shown to be involved as receptors of some Cry1 toxins. Here we analyzed the role of SfABCC2 and SfABCC3 in the toxicity of Cry1Fa and Cry1Ab toxins in this insect pest. RESULTS: Two S. frugiperda SfABCC2 and Sf

Evaluation of GS-omega/kappa-Hxtx-Hv1a and Bt toxins against Bt-resistant and -susceptible strains of Helicoverpa zea (Boddie) and Spodoptera frugiperda (J.E. Smith).

BACKGROUND: Helicoverpa zea and Spodoptera frugiperda are destructive insect pests of cotton and corn. Both have evolved practical resistance to transgenic crops producing Bacillus thuringiensis (Bt) toxins in the United States. GS-omega/kappa-Hxtx-Hv1a (ω/κ-Hv1a) is a novel biopesticide derived from spider venom with activity against lepidopteran pests. The efficacy of ω/κ-Hv1a is dependent on combining it with a facilitator product such as a Bt-based insecticide. However, it is unknown if ω/κ-Hv1a can be an alternative for the management of Bt resistance. In this study, we tested the effect

Elongation of the N-glycans of fowl plague virus hemagglutinin expressed in Spodoptera frugiperda (Sf9) cells by coexpression of human beta 1,2-N-acetylglucosaminyltransferase I.

Spodoptera frugiperda (Sf9)-cells differ markedly in their protein glycosylation capacities from vertebrate cells in that they are not able to generate complex type oligosaccharide side chains. In order to improve the oligosaccharide processing properties of these cells we have used baculovirus vectors for expression of human (beta 1,2-N-acetylglucosaminyltransferase I (hGNT-I), the enzyme catalysing the crucial step in the pathway leading to complex type N-glycans in vertebrate cells. One vector (Bac/GNT) was designed to express unmodified GNT-I protein, the second vector (Bac/tagGNT) to expr

The role of Desmodium intortum, Brachiaria sp. and Phaseolus vulgaris in the management of fall armyworm Spodoptera frugiperda (J. E. Smith) in maize cropping systems in Africa.

BACKGROUND: The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith) is a serious pest of maize. Farming systems such as push-pull or maize-legume intercropping have been reported to reduce FAW infestations significantly. However, the exact mechanisms involved in FAW management have not been practically elucidated. We therefore assessed larval host preference, feeding and survival rate when exposed to four host plants commonly used in push-pull and legume intercropping. We also compared adult moths' oviposition preference between maize and other grasses used as trap crops in push-pull. RES

Evaluation of extracellular subviral particles of dengue virus type 2 and Japanese encephalitis virus produced by Spodoptera frugiperda cells for use as vaccine and diagnostic antigens.

New or improved vaccines against dengue virus types 1 to 4 (DENV1 to DENV4) and Japanese encephalitis virus (JEV), the causative agents of dengue fever and Japanese encephalitis (JE), respectively, are urgently required. The use of noninfectious subviral extracellular particles (EPs) is an inexpensive and safe strategy for the production of protein-based flavivirus vaccines. Although coexpression of premembrane (prM) and envelope (E) proteins has been demonstrated to produce EPs in mammalian cells, low yields have hindered their commercial application. Therefore, we used an insect cell express

Effects of Different Spatial Extents of Occurrence Data on Biomod2-Based Species Distribution Modeling and Prediction: A Case Study of the Potential Distribution of Spodoptera frugiperda in China.

Climate change is reshaping species distributions worldwide, making reliable prediction of invasive species increasingly important for ecological risk assessment and pest management. However, species distribution models (SDMs) calibrated with regional occurrence records may underestimate potential suitable habitats because of niche truncation. Here, we evaluated the effects of occurrence data extent on SDM predictions using the globally invasive agricultural pest Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera: Noctuidae). Global occurrence records (1763 records) and China-specific occu

Toxicity assessment and transcriptomic analysis of Metarhizium rileyi (Hypocreales: Clavicipitaceae) against Spodoptera frugiperda (Lepidoptera: Noctuidae): identification of tyrosol as a key quorum sensing regulator of pathogenicity.

The fall armyworm, Spodoptera frugiperda, a severe crop pest, gained significant attention following its invasion of China in 2019. This study aimed to investigate the virulence and in vivo pathogenic transition of Metarhizium rileyi on S. frugiperda to understand biological control strategies. Using the immersion method, M. rileyi exhibited higher lethality toward third-instar larvae compared to fourth-instar larvae, with LC50 values of 3.15 × 107 conidia/ml and 1.04 × 108 conidia/ml, respectively. Microscopic observations revealed that M. rileyi initially existed as a yeast-like nonpathogeni

Mechanisms behind polyphagia in a pest insect: Responses of Spodoptera frugiperda (J.E. Smith) strains to preferential and alternative larval host plants assessed with gene regulatory networks.

A dataset of gene expression from Spodoptera frugiperda, a highly generalist pest moth, was used to understand how gene regulation is related to larval host plant preference. Transcriptomic data of corn and rice strains of S. frugiperda larvae, reared on different diets, were analysed with three different approaches of gene network inference, namely co-expression, weighted co-expression and Bayesian networks, since each methodology provides a different visualization of the data. Using these approaches, it was possible to identify two loosely interconnected co-expression networks, one of them r
