Poly(alkylene oxide) copolymers for nucleic acid delivery.

The advancement of gene-based therapeutics to the clinic is limited by the ability to deliver physiologically relevant doses of nucleic acids to target tissues safely and effectively. Over the last couple of decades, researchers have successfully employed polymer and lipid based nanoassemblies to deliver nucleic acids for the treatment of a variety of diseases. Results of phase I/II clinical studies to evaluate the efficacy and biosafety of these gene delivery vehicles have been encouraging, which has promoted the design of more efficient and biocompatible systems. Research has focused on desi

"Dual-Key"-Controlled Activatable Semiconducting Polymer Nanoprobe for Highly Precision Phototheranostics Under NIR-II Light Excitation.

The development of activatable optical probes has proved to be a promising approach to improve the accuracy of phototheranostics. However, most such probes are usually responsive to single pathological factor and visible-near-infrared-I light upon activation, which restricts the theranostic specificity and tissue penetration depth, respectively. Herein, we develop a nitric oxide (NO) and acidity dual-stimuli responsive semiconducting polymer nanoprobe for activatable near-infrared-II (NIR-II) photoacoustic imaging (PAI) and photothermal therapy (PTT). Specifically, a weak electron acceptor (be

Mechanistic insights into the calcium-mediated gelation of poly(4-tert-butylstyrene-alt-maleic acid)-block-poly(N-acryloylmorpholine) double hydrophilic block copolymers.

Stimuli responsive double hydrophilic block copolymers (DHBCs) are ubiquitous in water-based applications, such as drug/gene delivery, nanoreactor and sensor development, photocatalysis, 3D inkjet printing, etc. DHBC-based gels, cross-linked with calcium ions, are particularly valuable for the development of biomedically relevant materials (such as wound dressings or injectable formulations), due to the benignity of Ca2+ in biological systems. Therefore, gels formed via the calcium-mediated crosslinking of poly(4-tert-butylstyrene-alt-maleic acid)-block-poly(N-acryloylmorpholine) (PtBuSMA-b-PN

Natural Polymer-Based Hemostatic Hydrogels with Advanced Material and Structural Designs for Functional Applications.

Uncontrolled hemorrhage remains a major challenge in trauma care and surgical interventions, where rapid hemostasis and wound sealing are essential for improving patient survival. Natural polymer-based hydrogels have emerged as promising hemostatic materials owing to their excellent biocompatibility, biodegradability, and biomimetic properties. However, their clinical translation remains limited by insufficient mechanical robustness, wet adhesion, and functional responsiveness. To address these challenges, considerable progress has been achieved through rational material design and structural

Emulsion Stabilized by Pickering Emulsion Polymerization Technique: A State-of-the-Art Review.

Pickering emulsion is an attractive approach for developing stabilized emulsions. It has proved its potential use not only in the food industry but also in the pharmaceutical industry. The use of surfactants in ocular and other parenteral drug delivery systems is limited due to their potential to cause allergic reactions, thereby limiting the application of emulsions as drug delivery routes in the pharmaceutical industry. Pickering emulsion is known for imparting better stability to emulsions with respect to Ostwald ripening and destabilization under varying environmental conditions. The techn

A Nanosystem of Amphiphilic Oligopeptide-Drug Conjugate Actualizing Both αvβ3 Targeting and Reduction-Triggered Release for Maytansinoid.

To design a prodrug-based self-assembling nanosystem with both ligand targeting and stimuli-responsive features, and elucidate the superiority of each targeting strategy and the synergistic effect between them, we synthesized four small molecule amphiphilic peptide-drug conjugates (APDCs) using maytansinoid (DM1) as a cytotoxic agent, cRGDfK as a homing peptide, and disulfide (SS) or thioether (SMCC) as linker. Owing to their amphiphilicity, the APDCs could self-assemble into nanoparticles (APDC@NPs) which were evaluated in vitro in three different cell lines and in vivo in tumor-bearing C57BL

Elastoviscous substances with analgesic effects on joint pain reduce stretch-activated ion channel activity in vitro.

Activation by noxious mechanical stimuli of sensory nerve fibres that signal joint pain takes place through stretch-activated ion channels, which open in response to increased membrane tension. It has been suggested that the analgesic effect of hyaluronan solutions used for intra-articular treatment of joint pain in humans are mediated by a reduction of the sensitivity of mechanosensory ion channels of nociceptive nerve terminals. We have investigated whether cross-linked hyaluronan solutions (hylans) of different elastoviscosities modify the response characteristics of stretch-activated ion c

Breaking the delivery barrier: magnetic nanoparticles at the Nexus of imaging and theranostics.

Cancer is still one of the major causes of death globally, and the lack of specificity of current chemotherapeutic and imaging agents still remains to be a major issue in the therapy of cancer, which lead to the lack of efficacy and high systemic toxicity. Magnetic nanoparticles (MNPs) especially Superparamagnetic Iron Oxide Nanoparticles (SPION) and Ultrasmall Superparamagnetic Iron Oxide Nanoparticles (USPION) have shown potential as theranostic platforms due to their magnetic responsiveness, biocompatibility, and multiple functionalities. The advantages of iron oxide-based nanomaterials suc

Applications of mesoporous silica in biosensing and controlled release of insulin.

The development of new oral insulin delivery systems could bring significant benefits to insulin-dependent patients due to the simplicity of the method, avoidance of pain caused by parenteral administration and maintenance of optimal therapeutic levels for a longer period. However, administration of such therapeutic proteins orally remains a challenge because insulin (Ins) is a very sensitive molecule and can be easily degraded under the existing pH conditions in the stomach and intestines. Moreover, due to the large size of insulin, intestinal epithelium permeability is very low. This could b

Intranasal tetrandrine temperature-sensitive in situ hydrogels for the treatment of microwave-induced brain injury.

The brain is the most sensitive organ to microwave radiation. However, few effective drugs are available for the treatment of microwave-induced brain injury due to the poor drug permeation into the brain. Here, intranasal tetrandrine (TET) temperature-sensitive in situ hydrogels (ISGs) were prepared with poloxamers 407 and 188. Its characteristics were evaluated, including rheological properties, drug release in vitro, and mucosal irritation. The pharmacodynamics and brain-targeting effects were also studied. The highly viscous ISGs remained in the nasal cavity for a long time with the sustain

Harnessing multifunctional collagen mimetic peptides to create bioinspired stimuli responsive hydrogels for controlled cell culture.

The demand for synthetic soft materials with bioinspired structures continues to grow. Material applications range from in vitro and in vivo tissue mimics to therapeutic delivery systems, where well-defined synthetic building blocks offer precise and reproducible property control. This work examines a synthetic assembling peptide, specifically a multifunctional collagen mimetic peptide (mfCMP) either alone or with reactive macromers, for the creation of responsive hydrogels that capture aspects of soft collagen-rich tissues. We first explored how buffer choice impacts mfCMP hierarchical assemb

Metal Ion-Specific Modulation of Network Connectivity and Defects in Poly(ethylene glycol)-Peptide Conjugate Assemblies and Hydrogels.

Self-assembling peptide-polymer conjugates offer a versatile platform to engineer nanostructures with tunable morphology and functions. Here we show that alternating phenylalanine-histidine pentapeptide units, conjugated to a short linear poly-(ethylene glycol), show pH-induced assembly into β-sheet nanofibers that act as multifunctional cross-links in the resulting hydrogels. Circular dichroism spectra demonstrate that the self-assembly is enthalpy driven at low concentrations, while rheological results suggest that the network connectivity at high concentrations is compromised by the entropi

A molecularly defined polymeric platform for environmentally responsive activation of STING to enhance cancer immunotherapy.

The stimulator of interferon genes (STING) pathway is a promising immuno-oncology target. Despite their potential, STING agonists have yielded underwhelming results in clinical trials due to pharmacological barriers that limit their safety and efficacy. Herein, we describe the design and pre-clinical evaluation of a polymeric dimeric amidobenzimidazole (diABZI) STING agonist delivery platform for cancer immunotherapy. Central to our technology is a stimuli-responsive diABZI-functionalized reversible addition-fragmentation chain transfer (RAFT) polymerization chain transfer agent (CTA) that all

Recent progress of redox-responsive polymeric nanomaterials for controlled release.

Redox-responsive polymeric nanomaterials (PNMs) have been attractive research targets for drug delivery systems because disturbed levels of redox molecules are associated with the progression of various diseases. To enable PNMs to target biorelevant redox molecules, including reactive oxygen species (ROS), glutathione (GSH) and hydrogen sulfide (H2S), appropriate responsive moieties have to be installed within the polymer structure. Upon application of redox stimuli, redox-responsive PNMs undergo structural changes to release encapsulated payloads. Chalcogen ether, thioketal and arylboronic es

A Multi-Responsive Film from Polymerizable Cholesteric Liquid Crystal Microcapsules for Advanced Anti-Counterfeiting.

Cholesteric liquid crystals (CLCs) can be encapsulated within liquid crystal microcapsules (CLCMs) via ultrasonic emulsification, and this microcapsule structure offers advantages in liquid crystal (LC) orientation control and stability. However, CLCMs still face challenges in multiple stimulus responsiveness. In this study, polymerizable LC monomers are introduced as the core filler of CLCMs and polymerized under UV irradiation to form polymer networks. As a result, the structural color of the film changes, and the patterned coloration of the final prepared film gradually disappears upon expo

Evaluation of a Photopolymerized Gelatin Hydrogel Network with bFGF-Loaded Alginate Microspheres for Tympanic Membrane Perforation Repair in Rats.

Tympanic membrane perforation (TMP) often leads to hearing loss and requires effective repair strategies. However, existing surgical options are invasive and lack ideal biomaterials for scaffold-based healing. Herein, we present a custom-engineered mechano-acoustic responsive hydrogel incorporating bFGF-loaded sodium alginate microspheres, designed for controlled drug release and tissue regeneration under dual stimulation: vibrational simulation and low-frequency acoustic waves (generated by radio devices) - representing its first transition from in vitro characterization to in vivo regenerati

The synthesis of switch-off fluorescent water-stable copper nanocluster Hg2+ sensors via a simple one-pot approach by an in situ metal reduction strategy in the presence of a thiolated polymer ligand template.

The fabrication of stable fluorescent copper nanoclusters (CuNCs) in aqueous media is still challenging, despite the low price and potential biomedical applications. Herein, we report a facile and efficient strategy for assembling CuNCs using multifunctional thiolated copolymers with pH and thermoresponsive features. The new nanohybrids are formed via a simple one-pot approach through the reduction of a copper salt with hydrazine in the presence of a multithiolated polymer, which provides a template during nanocluster assembly and further efficient protection against oxidation and aggregation.

Thermoresponsive copolymer microgels.

Colloidal aqueous microgels have attracted notable attention due to their possible applications in both polymer chemistry and the (bio)material sector. Due to their soft internal network structure and their response to different external stimuli, they are suitable as carrier systems for different kinds of molecules (e.g. nanoparticles, drugs, diagnostic agents, and enzymes), as optically active materials or even as model systems for fundamental research on soft condensed matter. Within the class of microgels, a variety of molecular structures is possible. The aim of this manuscript is to revie

Stimuli-Responsive, Shape-Transforming Nanostructured Particles.

Development of particles that change shape in response to external stimuli has been a long-thought goal for producing bioinspired, smart materials. Herein, the temperature-driven transformation of the shape and morphology of polymer particles composed of polystyrene-b-poly(4-vinylpyridine) (PS-b-P4VP) block copolymers (BCPs) and temperature-responsive poly(N-isopropylacrylamide) (PNIPAM) surfactants is reported. PNIPAM acts as a temperature-responsive surfactant with two important roles. First, PNIPAM stabilizes oil-in-water droplets as a P4VP-selective surfactant, creating a nearly neutral in

Implementation of Highly Stable Memristive Characteristics in an Organic-Inorganic Hybrid Resistive Switching Layer of Chitosan-Titanium Oxide with Microwave-Assisted Oxidation.

This study proposes a high-performance organic-inorganic hybrid memristor for the development of neuromorphic devices in the memristor-based artificial synapse. The memristor consists of a solid polymer electrolyte (SPE) chitosan layer and a titanium oxide (TiOx) layer grown with a low-thermal-budget, microwave-assisted oxidation. The fabricated Ti/SPE-chitosan/TiOx/Pt-structured memristor exhibited steady bipolar resistive switching (BRS) characteristics and demonstrated excellent endurance in 100-cycle repetition tests. Compared to SPE-chitosan memristors without a TiOx layer, the proposed o
