A stochastic analysis of iliac trabecular bone dynamics.

Published normal histomorphometric data were used to derive distributions of thicknesses of trabecular plates and completed bone remodelling units (the basic multicellular unit carrying out bone remodelling, the BMU, when completed is termed a structural unit BSU). A stochastic model was set up to investigate the predictions of current BMU theory. Each of 100 trabecular "thicknesses" was drawn from the appropriate normal distribution using a pseudorandom number generator. Each day, each of its two surfaces when quiescent was assumed to have a 1:900 chance of initiating a remodelling cycle. Res

[Mathematical models in epidemiological analysis. II. Typhoid fever: time series analysis (author's transl)].

In this paper the systematic analysis of oro-faecal infectious diseases is continued. In order to propose a dynamic model for the evolution of the endemic typhoid fever, we have studied the behaviour of the number of cases of this disease in the town of Bari (Italy) during the years 1955-1978, by the methods of time series analysis. The analysis has pointed out the typical seasonal character of such a disease, and in particular the presence of a main winter cycle followed by a less evident summer cycle. The stochastic model which has been identified on the basis of the data from 1964 to 1978 a

Photochromic Fluorophores Enable Imaging of Lowly Expressed Proteins in the Autofluorescent Fungus Candida albicans.

Fluorescence microscopy is a standard research tool in many fields, although collecting reliable images can be difficult in systems characterized by low expression levels and/or high background fluorescence. We present the combination of a photochromic fluorescent protein and stochastic optical fluctuation imaging (SOFI) to deliver suppression of the background fluorescence. This strategy makes it possible to resolve lowly or endogenously expressed proteins, as we demonstrate for Gcn5, a histone acetyltransferase required for complete virulence, and Erg11, the target of the azole antifungal ag

Anthropogenic disturbances on antibiotic resistome along the Yarlung Tsangpo River on the Tibetan Plateau: Ecological dissemination mechanisms of antibiotic resistance genes to bacterial pathogens.

Human activities can accelerate the antibiotic resistome prevalence and pose threats to ecological safety and public health globally. However, antibiotic resistance gene (ARG) mobility and dissemination into bacterial pathogens under anthropogenic disturbances are still poorly understood. Here, we used a metagenomic approach to profile the biogeography of ARGs and pathogenic antibiotic resistant bacteria (PARB) under anthropogenic disturbances along the Yarlung Tsangpo River. Results showed the ARGs was dominated by bacA gene along the Yarlung Tsangpo River on the Tibetan Plateau. The ARG comp

Modeling cellular compartmentation in one-carbon metabolism.

Folate-mediated one-carbon metabolism (FOCM) is associated with risk for numerous pathological states including birth defects, cancers, and chronic diseases. Although the enzymes that constitute the biological pathways have been well described and their interdependency through the shared use of folate cofactors appreciated, the biological mechanisms underlying disease etiologies remain elusive. The FOCM network is highly sensitive to nutritional status of several B-vitamins and numerous penetrant gene variants that alter network outputs, but current computational approaches do not fully captur

Reconstructing the early spatial spread of pandemic respiratory viruses in the United States.

Understanding the geographic spread of emerging respiratory viruses is critical for pandemic preparedness, yet the early spatiotemporal dynamics of the 2009 H1N1 pandemic influenza and SARS-CoV-2 in the United States (US) remain unclear. While mobility and genomic data have revealed important aspects of pandemic spatial spread, several key questions remain: Did the two pandemics follow similar spatial transmission routes? How rapidly did they spread across the US? What role did stochastic processes play in early spatial transmission? To address these questions, we integrated high-resolution di

Dynamics and Sensitivity of the Lifecycle of Hepatitis B Virus.

A detailed mathematical model has been developed for the dynamics of hepatitis B virus (HBV) infection in a single cell. It provides a platform for a better quantitative understanding of the biochemical kinetics of the HBV lifecycle. The model is used to study the sensitivity of virus growth, providing a clear ranking of intracellular virus replication processes with respect to their contribution to net viral production. The stochastic formulation of the model enables the quantification of the variability characteristics in viral production, the probability of productive infection and the secr

Detecting stochasticity in discrete signals via nonparametric excursion theorem.

We develop a practical framework for distinguishing diffusive stochastic processes from deterministic signals using only a single discrete time series. Our approach is based on classical excursion and crossing theorems for continuous semimartingales, which correlates number Nε of excursions of magnitude at least ε with the quadratic variation [X]T of the process. The scaling law holds universally for all continuous semimartingales with finite quadratic variation, including general Ito diffusions with nonlinear or state-dependent volatility, but fails sharply for deterministic systems-thereby p

Automatic skin lesion segmentation via iterative stochastic region merging.

An automatic method for segmenting skin lesions in conventional macroscopic images is presented. The images are acquired with conventional cameras, without the use of a dermoscope. Automatic segmentation of skin lesions from macroscopic images is a very challenging problem due to factors such as illumination variations, irregular structural and color variations, the presence of hair, as well as the occurrence of multiple unhealthy skin regions. To address these factors, a novel iterative stochastic region-merging approach is employed to segment the regions corresponding to skin lesions from th

Inferring Adaptation at Community Composition Level From the Relationship Between Succession Rate and Global Change Intensity.

In response to anthropogenic environmental changes, biological communities often undergo compositional changes, which are traditionally thought to represent adaptation. However, this hypothesis and its mechanism remain surprisingly underexplored. In this study, we conducted a 10-year nitrogen deposition experiment in the Eurasian steppe, manipulating nine intensities under two management strategies (fencing or mowing). As the intensity increased from 0-20 to 50 g N m-2 yr-1, the succession rates of both plant and soil microbial communities first increased but then decreased, implying that high

Stochastic galvanic vestibular stimulation improves kinetic performance in adolescent idiopathic scoliosis during obstacle negotiation.

Vestibular dysfunction has been reported as a potential cause in adolescent idiopathic scoliosis (AIS). However, it remained unclear how stochastic galvanic vestibular stimulation (GVS) affected kinetic performance of patients with AIS. This study aimed to investigate the effect of stochastic GVS on ground reaction forces (GRF) measures during obstacle negotiation among patients with AIS. Fifteen patients with AIS and 15 age/sex-matched healthy controls (HC) participated in this study. Stochastic GVS was applied via electrodes placed over bilateral mastoid process with the intensity of 80% of

Abundant and equipotent founder cells establish and maintain acute lymphoblastic leukaemia.

High frequencies of blasts in primary acute lymphoblastic leukaemia (ALL) samples have the potential to induce leukaemia and to engraft mice. However, it is unclear how individual ALL cells each contribute to drive leukaemic development in a bulk transplant and the extent to which these blasts vary functionally. We used cellular barcoding as a fate mapping tool to track primograft ALL blasts in vivo. Our results show that high numbers of ALL founder cells contribute at similar frequencies to leukaemic propagation over serial transplants, without any clear evidence of clonal succession. These f

Fate of a biodegradable plastic in forest soil: Dominant tree species and forest types drive changes in microbial community assembly, influence the composition of plastisphere, and affect poly(butylene succinate-co-adipate) degradation.

Poly(butylene succinate-co-adipate) (PBSA) degradation and its plastisphere microbiome in cropland soils have been studied; however, such knowledge is limited in the case of forest ecosystems. In this context, we investigated: i) the impact of forest types (conifer and broadleaved forests) on the plastisphere microbiome and its community assembly, ii) their link to PBSA degradation, and iii) the identities of potential microbial keystone taxa. We determined that forest type significantly affected microbial richness (F = 5.26-9.88, P = 0.034 to 0.006) and fungal community composition (R2 = 0.38

An analytical stochastic stage-structured model for pest population dynamics: a case study on Cydia pomonella in Italy.

In this work, we propose a novel phenological model to describe population dynamics based on a compound Poisson process driven development. The model offers an alternative to other well-established approaches founded on systems of ordinary or partial differential equations, in which, the stochasticity is driven by the Brownian motion. A key advantage of the proposed framework is that it prevents the age regression of individuals and allows for the analytical derivation of the number of individuals in each developmental stage into which the population is structured. The model is applied to Cydi

Economic effects of automated health monitoring based on sensors versus visual observation for dairy cows in early lactation.

The objective of this study was to compare the cash flow of lactating Holstein cows randomized to health monitoring strategies that relied exclusively on automated health alerts or visual observation for selecting cows for clinical examination from 3 to 21 DIM. Data from a randomized controlled trial were used to estimate total cash flow per cow and per slot up to 100 DIM. We expected that greater milk yield in early lactation for the group monitored with automation would result in greater cash flow than for the group including visual observation despite greater health monitoring and treatment

Individual and population level uncertainty interact to determine the performance of outbreak surveillance systems.

BACKGROUND: Outbreak detection frequently relies on imperfect individual-level case diagnosis. Both outbreaks and cases are discrete events that can be misclassified and uncertainty at the case level may impact the performance of outbreak alert and detection systems. Here, we describe how the performance of outbreak detection depends on individual-level diagnostic test characteristics and population-level epidemiology, and describe settings where imperfect individual-level tests can achieve consistent performance comparable to "perfect" diagnostic tests. METHODOLOGY: We generated a stochastic

Optimization of sequential therapies to maximize extinction of resistant bacteria through collateral sensitivity.

Antimicrobial resistance (AMR) threatens global health. A promising and underexplored strategy to tackle this problem is sequential therapies exploiting collateral sensitivity (CS), whereby resistance to one drug increases sensitivity to another. Here, we develop a four-genotype stochastic birth-death model with two bacteriostatic antibiotics to identify switching periods that maximize bacterial extinction under subinhibitory concentrations. We show that extinction probability depends nonlinearly on switching period, with stepwise increases aligned to discrete switch events: fast sequential th

Numerical computation of the stochastic hepatitis B model using feed forward neural network and real data.

Hepatitis B is a global health burden and can persist for years, with nearly two billion infections worldwide, where its spread is influenced by environmental heterogeneity, host-pathogen interactions, and vaccination-induced immune variability. Proper understanding and developing models with a suitable framework is essential to accurately capture the complexity of the hepatitis B virus (HBV) and its transmission. In this work, we present a novel framework of a stochastic model and a forward neural network that combines neural networks and stochastic differential equations to analyze the dynam

Development and validation of diverse active human models for simulating stochastic occupant responses during pre-crash vehicle maneuvers.

Occupant kinematics in abrupt vehicle maneuvers are highly variable, yet previous active human body models provided only deterministic predictions for a limited range of body sizes. This study bridges the gap by developing and validating an efficient tool capable of stochastic predictions, thereby capturing behavioral variability across diverse occupant characteristics during pre-crash maneuvers. A computationally efficient version of the midsize male GHBMC simplified model (GHBMCsi-pre) was first developed by rigidizing non-deformable body components in vehicle maneuvers while preserving key
