Genomic Dissection of an Icelandic Epidemic of Respiratory Disease in Horses and Associated Zoonotic Cases.

Iceland is free of the major infectious diseases of horses. However, in 2010 an epidemic of respiratory disease of unknown cause spread through the country's native horse population of 77,000. Microbiological investigations ruled out known viral agents but identified the opportunistic pathogen Streptococcus equi subsp. zooepidemicus (S. zooepidemicus) in diseased animals. We sequenced the genomes of 257 isolates of S. zooepidemicus to differentiate epidemic from endemic strains. We found that although multiple endemic clones of S. zooepidemicus were present, one particular clone, sequence type

Group C streptococcal infection in a term newborn infant.

Group C streptococci were recovered from the cerebrospinal fluid of a term infant with symptoms whose mother had received intrapartum antibiotic therapy for chorioamnionitis. This case highlights that beta-hemolytic streptococci of Lancefield groups other than A or B can be isolated from a normally sterile site and should be considered as pathogens. It also underscores the potential importance of sampling cerebrospinal fluid in an infant with symptomatic suspected sepsis, especially in the setting of exposure to maternal antimicrobial agents.

Activation of the S100A8/A9 Alarmin Amplifies Inflammatory Pathways in Equine Ascending Placentitis.

Ascending placentitis is a significant cause of equine pregnancy loss, yet the upstream inflammatory triggers are poorly defined. Recently, we identified S100A8/S100A9 (S100A8/A9) alarmins as potential upstream regulators in a chronic equine placentitis model. The current study aimed to determine whether this upregulation is sustained in the acute model and in clinical cases, and to elucidate the expression of their downstream inflammatory mediators. Using an experimental model, we quantified S100A8/A9 mRNA expression in acute (n = 5) and chronic (n = 6) placentitis induced by Streptococcus eq

B-mode and Doppler ultrasonography in pony mares with experimentally induced ascending placentitis.

REASONS FOR PERFORMING STUDY: Early, accurate diagnosis of ascending placentitis in mares remains a key challenge for successful treatment of the disease. Doppler ultrasonography has shown promise as a tool to diagnose pregnancy abnormalities and is becoming more available to equine clinicians. However, to date, no studies have prospectively compared this technique to standard B-mode measurement of the combined thickness of the uterus and placenta (CTUP). OBJECTIVES: The objective of the current study was to compare Doppler and B-mode ultrasonography for the detection of experimentally-induce

Lipid profiling of the secondary infecting bacteria with porcine reproductive and respiratory syndrome virus, including Streptococcus suis serotype 2, and their hosts.

The lipid profile of bacteria associated with swine respiratory diseases remain unknown. This study analyzed the lipids of common bacteria, secondary to porcine reproductive and respiratory syndrome virus (PRRSV) infection, as well as lipid profile of the host. Streptococcus suis serotype 2 (SS2), Streptococcus equi subsp. zooepidemicus (SZ), Actinobacillus pleuropneumoniae serotype 7 (APP7), Haemophilus parasuis serotype 5 (HP5) and Pasteurella multocida type A (PM) were analyzed using the liquid chromatography-electrospray ionization tandem mass spectrometry (LC-MS-MS) method. Update and actual trends on bacterial infections following liver transplantation.

Recent advances in effective antimicrobial prophylactic strategies have led to a decline in the incidence of opportunistic infections in liver transplant recipients. However, morbidity and mortality due to infectious diseases remain as major problems. Bacterial infections occurring early after transplant are mainly related to the technical aspects of the procedure. By contrast, after the first postoperative days and beyond, the nature and variety of infectious complications change. Opportunistic bacterial infections are uncommon after 6 mo in patients receiving stable and reduced maintenance d

A streptococcal Fic domain-containing protein disrupts blood-brain barrier integrity by activating moesin in endothelial cells.

Streptococcus equi subsp. zooepidemicus (SEZ) is a zoonotic pathogen capable of causing meningitis in humans. The mechanisms that enable pathogens to traverse the blood-brain barrier (BBB) are incompletely understood. Here, we investigated the role of a newly identified Fic domain-containing protein, BifA, in SEZ virulence. BifA was required for SEZ to cross the BBB and to cause meningitis in mice. BifA also enhanced SEZ translocation across human Brain Microvascular Endothelial Cell (hBMEC) monolayers. Purified BifA or its Fic domain-containing C-terminus alone were able to enter into hBMECs,

Recovery of hyaluronic acid using thermosensitive polymer-salt aqueous biphasic system with polymer recycling.

Microbial fermentation emerges as a viable alternative to animal tissue-based extraction for industrial hyaluronic acid (HA) production. However, microbial HA requires rigorous separation and purification processes to meet high purity standards for cosmeceutical use. Aqueous biphasic system (ABS) presents a promising strategy for HA recovery due to its cost-effectiveness, high yield, purity and eco-friendly nature. Thermosensitive polymer, specifically ethylene oxide-propylene oxide (EOPO)-salt ABS, was proposed in the present study to recover microbial HA from Streptococcus zooepidemicus ferm

Neutrophil extracellular traps formation by bacteria causing endometritis in the mare.

Besides the classical functions, neutrophils (PMNs) are able to release DNA in response to infectious stimuli, forming neutrophil extracellular traps (NETs) and killing pathogens. The pathogenesis of endometritis in the mare is not completely understood. The aim was to evaluate the in vitro capacity of equine PMNs to secrete NETs by chemical activation, or stimulated with Streptococcus equi subspecies zooepidemicus (Szoo), Escherichia coli (Ecoli) or Staphylococcus capitis (Scap) strains obtained from mares with endometritis. Ex vivo endometrial mucus from mares with bacterial endometritis wer

Tumor necrosis factor signaling during equine placental infection leads to pro-apoptotic and necroptotic outcomes.

Ascending placentitis is the leading cause of abortion in the horse. The pleiotropic cytokine tumor necrosis factor (TNF) is an upstream regulator of this disease, but little is understood regarding its function in pregnancy maintenance or placental infection. To assess this, RNA sequencing was performed on chorioallantois and endometrium of healthy pregnant mares at various gestational lengths (n = 4/gestational age), in addition to postpartum chorioallantois, and diestrus endometrium to assess expression of TNF, TNFR-1, and TNFR-2. Additionally, ascending placentitis was induced via trans-ce

Interleukin-6 pathobiology in equine placental infection.

PROBLEM: Ascending placentitis is the leading cause of abortion in the horse. Interleukin (IL)-6 is considered predictive of placental infection in other species, but little is understood regarding its role in the pathophysiology of ascending placentitis. METHOD OF STUDY: Sub-acute ascending placentitis was induced via trans-cervical inoculation of S zooepidemicus, and various fluids/serum/tissues collected 8 days later. Concentrations of IL-6 were detected within fetal fluids and serum in inoculated (n = 6) and control (n = 6) mares. RNASeq was performed on the placenta (endometrium and chor

Moelerella wisconsensis: first isolation from lungs and spleen of a horse infected with Streptococcus dysgalactia subsp. equisimilis.

Moellerella wisconsensis is a Gram-negative, facultative anaerobic bacillus of Entero-bacteriaceae family, and it is an uncommon pathogen in domestic animals. To date, five cases were reported including two dogs, two cattle, and a goat. Streptococcus equisimilis is the second common bacterial agent after the S. equi subsp. zooepidemicus in equine pneumonia cases. The present report describes the isolation of M. wisconses from lungs and spleen of a 10-year-old Arabian horse (May 08, 2022) at post-mortem examination being co-infected with S. equisimilis. Clinical and pathological findings includ

Meningitis due to group C Streptococcus: a case report and review of the literature.

Group C streptococci (GCS) are common causes of veterinary diseases and may colonize humans. Human diseases due to GCS are uncommon and generally occur in the elderly and persons exposed to animals or animal products. We report a case of Streptococcus equi subsp. zooepidemicus meningitis in a horse trainer and review 36 cases of GCS meningitis reported in the literature. The median age was 48 y and the majority of patients were previously healthy. Thirty-one percent (11/36) of the reported cases followed equine exposure and 19% (7/36) ingestion of dairy products. Sixty-seven percent (24/36) we

Retrospective study of fatal pneumonia in racehorses.

Respiratory diseases have a major impact on racehorses in training and are often cited as the second most common reason of horses failing to perform. Cases were submitted by the California Horse Racing Board to the California Animal Health and Food Safety laboratory for postmortem examination between January 1, 2005 and December 31, 2014. We determined the demographics of racehorses with fatal pneumonia, characterized the pathologic findings in animals with a postmortem diagnosis of respiratory infection, and determined the most significant pathogens associated with lower respiratory tract dis

Gut virome derived bacteriophage endolysin Lys572 can effectively control Streptococcus suis in refrigerated and thawed pork.

Streptococcus suis (S. suis), a major zoonotic pathogen in pork production, contaminates meat products and processing facilities, posing risks to both animal and human health. Virome analysis enables the identification of novel antibacterial protein genes directly from environmental genetic material without microbial cultivation, offering a promising approach for discovering bacteriophage endolysins with applications in food safety and biopreservation. In this study, we identified a novel endolysin from porcine intestinal virome, named Lys572. Lys572 is a modular enzyme consisting of an N-term

Percutaneous puncture technique for treating persistent retropharyngeal lymph node infections in seven horses.

Between 1999 and 2001, seven horses with fever, dysphagia and a history of chronic upper respiratory tract infection lasting between three weeks and three months were examined. They had been treated unsuccessfully with a variety of antibiotics for three to four weeks. A deep abscess in a retropharyngeal lymph node was diagnosed in each case by clinical examination, endoscopy and echographic examination of the retropharyngeal region. The infected retropharyngeal lymph node of each horse was punctured with a spinal needle under ultrasound guidance. Pus was aspirated from four of the horses, and
