An impairment of cortical GABAergic neurons is involved in alkalosis-induced brain dysfunctions.

Acid-base imbalance leads to pathological cognition and behaviors in the clinical practices. In the comparison with acidosis, the cellular mechanisms underlying alkalosis-induced brain dysfunction remain unclear. By using electrophysiological approach, we investigated the influences of high extracellular pH environment on cortical GABAergic neurons in terms of their responsiveness to synaptic inputs and their ability to produce action potentials. Artificial cerebral spinal fluid in high pH impairs excitatory synaptic transmission and spike initiation in cortical GABAergic neurons. The alkalosi

Mapping membrane potential transients in crayfish (Procambarus clarkii) optic lobe neuropils with voltage-sensitive dyes.

Voltage-sensitive dyes NK 2761 and RH 155 were employed (in conjunction with a 12 x 12 photodiode array) to study membrane potential transients in optic lobe neuropils in the eye stalk of the crayfish Procambarus clarkii. By this means we investigated a pathway linking deutocerebral projection neurons, via hemiellipsoid body local interneurons, to an unidentified target (most likely neurons processing visual information) in the medulla terminalis. Rapid (10- to 20-ms duration), transient changes in absorption with the characteristics of action potentials were recorded from the optic nerve and

[Slow potentials of the olfactory bulb in the carp].

The slow negative potentials evoked in carp olfactory bulb (OB) by some odorants and slow positive potentials evoked by nonspecific irritation (water stream, NaCl solutions) of olfactory epithelium have been studied. The slow potentials of both types were not inverted in deep layers of OB and were resistant to blockade of synaptic transmission by manganese ions. The negative slow potentials were not also affected by hypoxia and associated with local increase of OB tissue resistance. Positive slow potentials were affected by hypoxia and associated with local decrease of OB tissue resistance. Th

Microglial CX3CR1 signaling mediates synaptic pruning in degenerative cervical myelopathy.

Degenerative cervical myelopathy (DCM) encompasses several conditions that cause compression of the cervical spinal cord. While the pathobiology underlying compression-induced neural degeneration remains incompletely understood, microglial synaptic pruning, driven by CX3CR1, is thought to be a main contributor. The current study builds on previous work demonstrating that Cx3cr1-/- mice exhibit improved gait following DCM and seeks to determine if CX3CR1-mediated synaptic pruning underlies this improvement. We compared C57BL/6 (WT, wild-type) and Cx3cr1-/- mice across 12-weeks of DCM. Locomotor

Defects in Neuromuscular Transmission May Underlie Motor Dysfunction in Spinal and Bulbar Muscular Atrophy.

UNLABELLED: Spinal and bulbar muscular atrophy (SBMA) in men is an androgen-dependent neuromuscular disease caused by expanded CAG repeats in the androgen receptor (AR). Whether muscle or motor neuron dysfunction or both underlies motor impairment in SBMA is unknown. Muscles of SBMA mice show significant contractile dysfunction, implicating them as a likely source of motor dysfunction, but whether disease also impairs neuromuscular transmission is an open question. Thus, we examined synaptic function in three well-studied SBMA mouse models-the AR97Q, knock-in (KI), and myogenic141 models-by re

Synaptic response patterns of neurons in the cortex of rat inferior colliculus.

The present study examined synaptic potentials of neurons in inferior colliculus (IC) cortex slice and the roles of GABA and glutamate receptors in generating these potentials. Multipolar (82%) and elongated (18%) cells were observed with intracellular biocytin staining. Electrical stimulation of the IC commissure (CoIC) elicited only inhibitory postsynaptic potentials (IPSPs) (10% of cells), only excitatory postsynaptic potentials (EPSPs) (51%), or both (38%). IPSPs were elicited at lower thresholds and shorter latencies than EPSPs (mean: 1.6+/-1.2 ms) and IPSPs were observed in all neurons f

Calcium sensing by the C2A domain of Synaptotagmin 7 is required through the linker for both synaptic facilitation and memory.

The earliest synaptic plasticity, called synaptic facilitation, is frequently observed at the beginning of any train of action potentials carrying information in the brain. During consecutive action potentials with short intervals, transmissions influence later ones, often enhancing neurotransmitter release. Facilitation is predicted to be controlled by a high-affinity Ca2+ sensor that responds to the presynaptic residual Ca2+ after the previous Ca2+ influx. While synapse strengthening via facilitation has been theoretically proposed to underlie instantaneously formed memory, the lack of exper

The action of general anaesthetic agents on root responses of the frog isolated spinal cord.

1. The action of volatile and barbiturate general anaesthetic agents on synaptic transmission in the frog isolated spinal cord has been studied by recording ventral root synaptic potentials and spike discharges evoked by volleys in a dorsal root and in the lateral column fibres.2. Some observations on the distribution of the lateral column fibres and the characteristics of the dorsal root potentials have been presented.3. Volatile agents depressed and eventually abolished all components of the ventral root responses. Failure of motoneurone discharge was the result of two factors, a decrease in

An intracellular analysis of dendrodendritic inhibition in the turtle in vitro olfactory bulb.

1. Intracellular recordings from an in vitro preparation of turtle olfactory bulb were used to determine the pathway responsible for producing synaptic inhibition of mitral cells.2. Inhibitory post-synaptic potentials (i.p.s.p.s) could be elicited in mitral cells by both olfactory nerve (orthodromic) and mitral cell axon (antidromic) stimulation and by suprathreshold depolarizing current pulses injected intracellularly through the recording micro-electrode. Reversing the chloride gradient by either intracellular injection of chloride or lowering the external chloride concentration reversed the

Emulating Bilingual Synaptic Response Using a Junction-Based Artificial Synaptic Device.

Excitatory and inhibitory postsynaptic potentials are the two fundamental categories of synaptic responses underlying the diverse functionalities of the mammalian nervous system. Recent advances in neuroscience have revealed the co-release of both glutamate and GABA neurotransmitters from a single axon terminal in neurons at the ventral tegmental area that can result in the reconfiguration of the postsynaptic potentials between excitatory and inhibitory effects. The ability to mimic such features of the biological synapses in semiconductor devices, which is lacking in the conventional field ef

Long-term synaptic plasticity in the spinal dorsal horn and its modulation by electroacupuncture in rats with neuropathic pain.

Our previous study has reported that electroacupuncture (EA) at low frequency of 2 Hz had greater and more prolonged analgesic effects on mechanical allodynia and thermal hyperalgesia than that EA at high frequency of 100 Hz in rats with neuropathic pain. However, how EA at different frequencies produces distinct analgesic effects on neuropathic pain is unclear. Neuronal plastic changes in spinal cord might contribute to the development and maintenance of neuropathic pain. In the present study, we investigated changes of spinal synaptic plasticity in the development of neuropathic pain and its

Effects of dithiothreitol, 5,5'-dithiobis(2-nitrobenzoic acid) and N-ethylmaleimide on synaptic transmission at sympathetic ganglion cells of frog.

The effects of disulfide bond and sulfhydryl group reagents on synaptic transmission were studied in the bullfrog sympathetic ganglion. Ten millimoles of dithiothreitol (DTT), a disulfide bond reducing agent, decreased the amplitude of the transmitted postganglionic compound action potential (CAP) to 41% of control. More than one hour wash-out of DTT with normal Ringer's solution did not reverse the block, but 1 mM 5,5'-dithiobis(2-nitrobenzoic acid) (DTNB), an oxidizing agent, rapidly restored the compound action potential amplitude. N-Ethylmalemide (NEM), an alkylating agent, added before DT

Functional and structural changes in mammalian sympathetic neurones following interruption of their axons.

The effects of interrupting the axons of principal neurones in the superior cervical ganglion of adult guinea-pigs were studied by means of intracellular recording, and light and electron microscopy. 1. Within 72 hr of axon interruption, the amplitude of exitatory postsynaptic potentials potentials (e.p.s.p.s) recorded in principal neurons in response to maximal preganglionic stimulation declined. E.p.s.p.s were maximally reduced (by more than 70% on average) 4-7 days following interruption, and failed to bring many cells to threshold. E.p.s.p.s. recorded in nearby neurones whose axons remaine

Contrasting forms of synaptic plasticity in monkey inferotemporal and primary visual cortices.

Activity-dependent synaptic plasticity was examined in vivo in two cortical areas of the adult monkey visual system, the primary visual and inferotemporal cortex, the first and late cortical stages essential for object recognition. Discontinuous high-frequency electrical stimulation of intrinsic horizontal connections in layer 2/3 caused contrasting forms of synaptic plasticity in the two areas. In the inferotemporal cortex, long-term potentiation of extracellular field potentials in layer 2/3 was induced, whereas in the same pathway of V1, identical stimulation protocol elicited long-term dep

In vivo electrophysiological investigations into the role of histamine in the dentate gyrus of the rat.

Drugs acting at the three known classes of histamine receptors were injected intracerebroventricularly into the rat. The effects of these drugs upon synaptic potentials recorded from the dentate gyrus of the freely-moving rat were determined. Population spikes and field excitatory postsynaptic potentials were recorded from the granule cell layer of the dentate gyrus following stimulation of the perforant path. Drugs, dissolved in 0.9% NaCl were applied into the lateral cerebral ventricle in a volume of 5 microl over a period of 6 min. The histamine H1 receptor antagonist mepyramine (0.4 or 0.8

Lambert Eaton Myasthenic Syndrome.

Lambert Eaton Myasthenic Syndrome (LEMS) is a pre-synaptic neuromuscular junction disorder characterised clinically by leg-predominant proximal weakness with spread of weakness distally and cranially with increasing severity as well as reduced reflexes and autonomic symptoms such as a dry mouth. Typical electrophysiological findings include small compound muscle action potentials at rest that augment following short exercise, decrement at low frequency (2-5 Hz) repetitive nerve stimulation, and increment at high frequency (20-50 Hz) repetitive nerve stimulation. Immunologically, antibodies to

[Auditory neuropathy (auditory neuropathy spectrum disorders): the approaches to diagnostics and rehabilitation].

Auditory neuropathies (auditory neuropathy spectrum disorders, ANSD) may be a consequence of dysfunction of inner hair cells and/or of synapses between these cells and auditory nerve fibers. Another cause of these disorders is supposed to be pathological changes in the auditory nerve itself. The outcome of the rehabilitative treatment of the patients presenting with this disorder depends on the quality of diagnosis and precise location of the pathological process. The present study involved 82 patients with auditory neuropathies. The audiological data obtained in the course of this work were c

Long-term plasticity of visually evoked potentials in humans is altered in major depression.

BACKGROUND: Long-term synaptic plasticity is a ubiquitous form of neuronal plasticity that regulates the strength of synaptic transmission in many brain areas. However, most data on long-term potentiation and long-term depression rely on research in animal brain slices. The role of synaptic plasticity in physiology and pathology of the functioning human brain remains obscure. Considering recent studies that provided evidence for a dysfunction of brain plasticity as the neurobiological basis of affective disorders, we assessed neural transmission and its plastic modulation in the visual pathway

Octopamine is the synaptic transmitter between identified neurons in the buccal feeding network of the pond snail lymnaea stagnalis.

We report the pharmacological properties of synaptic connections from the three octopamine-containing OC interneurons to identified buccal feeding neurons in the pond snail, Lymnaea stagnalis. Intracellular stimulation of an OC interneuron evokes inhibitory postsynaptic potentials in the B3 motoneurons and N2 (d) interneurons, while the synapse between OC and N3 (phasic) interneurons has two components: an initial electrical excitation followed by chemical inhibition. All synaptic responses persist in a saline with elevated calcium and magnesium suggesting that the connections are monosynaptic

Suppression of hippocampal synaptic transmission by the spider toxin omega-agatoxin-IV-A.

Effects of omega-agatoxin-IV-A (AGTX) on synaptic transmissions were examined in thin transverse sections of the guinea pig hippocampus. AGTX suppressed, in a dose-dependent manner, field potentials elicited by mossy fiber stimulation in CA3 pyramidal cell layer. AGTX also suppressed field potentials elicited by perforant fiber stimulation in granular layer without marked changes in antidromic responses. These results suggest that Ca2+ entry into presynaptic terminals via P-like calcium channels is at least partly responsible for release of glutamate.
