Learned taste aversions induced by hypnotic drugs.

Taste aversions reflect the association of drug-produced noxious interoceptive stimuli with distinctive tastes. In the present experiments, taste aversions to sweetened condensed milk were induced in rats by a wide range of doses of the hypnotic drugs, hexobarbital, amobarbital, chloral hydrate, flurazepam and methaqualone and by anesthetization with ethyl ether. Observation of changes in motor behavior following drug administration could not be used to predict the development of taste aversions. Despite the fact that hypnotic drugs have punishing properties, which these paradox is discussed i

Human scalp hair follicles can 'taste': chemosensory signalling via the bitter taste receptor TAS2R4 inhibits hair growth ex vivo.

BACKGROUND: Taste receptors (TRs) exert many 'nongustatory' chemosensory functions beyond the sensation of taste. Recently, human keratinocytes have been found to express some bitter TRs, whose physiological functions remain unknown. As it has been discovered that human scalp hair follicles (HFs) use olfactory receptors to regulate their growth, we hypothesized that some bitter TRs may exert a similar function. OBJECTIVES: To explore whether human scalp HFs express the bitter TR TAS2R4 and whether its stimulation with cognate agonists or its selective knockdown affects key human HF functions

The formation of endoderm-derived taste sensory organs requires a Pax9-dependent expansion of embryonic taste bud progenitor cells.

In mammals, taste buds develop in different regions of the oral cavity. Small epithelial protrusions form fungiform papillae on the ectoderm-derived dorsum of the tongue and contain one or few taste buds, while taste buds in the soft palate develop without distinct papilla structures. In contrast, the endoderm-derived circumvallate and foliate papillae located at the back of the tongue contain a large number of taste buds. These taste buds cluster in deep epithelial trenches, which are generated by intercalating a period of epithelial growth between initial placode formation and conversion of

Sensory functions beyond taste: multifaceted role of solitary chemosensory cells and taste receptors in mucosal immune defense and disease pathogenesis.

Taste receptors are most commonly associated with the experience of flavor. However, these receptors are also crucial participants in wider physiological mechanisms, particularly immunological defense. Solitary chemosensory cells (SCCs) express taste receptors (e.g., TAS2Rs and TAS1Rs) and are distributed throughout many organs, including the oral cavity, respiratory system and gastrointestinal tract. These cells function as sensors for dangerous bacteria and trigger immunological responses to preserve the integrity of mucosal tissues and prevent disease. Recent research has revealed that SCCs

The power of taste: A scoping review about how taste experiences shapes dietary habits and health from pregnancy to adolescence.

Taste influences food preferences and consumption, potentially contributing to dietary choices across the lifespan. Hormonal changes during pregnancy and breastfeeding, as well as biological and cognitive development during childhood and adolescence, may alter taste perception, sensitivity, and preferences, with possible implications for diet and health. This scoping review aimed to map evidence on how taste experiences vary across critical life stages (pregnancy, breastfeeding, childhood, and adolescence) and how these changes affect dietary behaviours, food choices, and health outcomes. Involvement of multiple taste receptors in umami taste: analysis of gustatory nerve responses in metabotropic glutamate receptor 4 knockout mice.

KEY POINTS: The taste receptor T1R1 + T1R3 heterodimer and metabotropic glutamate receptors (mGluR) may function as umami taste receptors. Here, we used mGluR4 knockout (mGluR4-KO) mice and examined the function of mGluR4 in peripheral taste responses of mice. The mGluR4-KO mice showed reduced responses to glutamate and L-AP4 (mGluR4 agonist) in the chorda tympani and glossopharyngeal nerves without affecting responses to other taste stimuli. Residual glutamate responses in mGluR4-KO mice were suppressed by gurmarin (T1R3 blocker) and AIDA (group I mGluR antagonist). The present study not only

The effect of smell and taste of milk during tube feeding of preterm infants (the Taste trial): a protocol for a randomised controlled trial.

INTRODUCTION: Smell and taste of milk are not generally considered when tube feeding preterm infants. Preterm infants have rapid growth, particularly of the brain, and high caloric needs. Enteral feeding is often poorly tolerated which may lead to growth failure and long-term neurodevelopmental impairment. Smell and taste are strong stimulators of digestion and metabolism. We hypothesise that regular smell and taste during tube feeding will improve weight z-scores of very preterm infants at discharge from hospital. METHODS AND ANALYSIS: Taste is a randomised, unblinded two-centre trial. Infan

Chemosensory response to Pt-based chemotherapeutics via bitter taste receptors in vitro reveals a new mechanism for bitter taste disorders.

Chemotherapeutics like platinum (Pt)-based drugs cause bitter taste disorders, which impair patients’ food intake and quality of life. However, the role of bitter taste receptors (TAS2Rs) remains unexplored, limiting the development of targeted treatments for this clinically impactful side effect. By performing a TAS2R-sensitive bitterness assay based on human gastric parietal cells (HGT-1), we evaluated the bitterness of Pt-based agents and the counteracting potential of the bitter-masking sodium salt of the flavanone homoeriodictyol (Na-HED). Here, we report that carboplatin (50–750 µM) and

Deciphering Taste Disorders: Mechanistic Insights and Nonclinical Evaluation.

Drug-induced taste dysfunctions are an underrecognized yet clinically significant class of adverse effects that can substantially impair patients' quality of life and compromise adherence to therapeutic regimens. Despite the potential for many pharmacologic agents to alter taste perception, these effects are frequently overlooked in both clinical practice and drug development. Gustatory disorders are rarely assessed in toxicology studies, and current knowledge regarding the pathophysiology of dysgeusia, the relevant organ systems involved, and nonclinical evaluation methods remains limited. GLP-1 receptor agonists significantly impair taste function.

Over 10% of the US population are prescribed glucagon-like peptide-1 receptor agonists (GLP-1 RAs) to combat obesity. Although they decrease cravings for foods, their influence on chemosensory function is unknown. We employed state-of-the-art quantitative taste and smell tests to address this issue. The 53-item Waterless Empirical Taste Test (WETT®) and the 40-item University of Pennsylvania Smell Identification Test (UPSIT®) were completed by 46 persons taking GLP-1 RAs and 46 controls matched on age, sex, smoking behavior, and COVID-19 infection histories. Data were analyzed using analyses o

Neural correlates of rapid familiarization to novel taste.

The gustatory cortex (GC) plays a pivotal role in taste perception, with neural ensemble responses reflecting taste quality and influencing behavior. Recent work, however, has shown that GC taste responses change across sessions of novel taste exposure in taste-naïve rats. Here, we use single-trial analyses to explore changes in the cortical taste-code on the scale of individual trials. Contrary to the traditional view of taste perception as innate, our findings suggest rapid, experience-dependent changes in GC responses during initial taste exposure trials. Specifically, we find that early re

Smell, taste and COVID-19: testing is essential.

During the COVID-19 pandemic, it became clear that smell and taste (chemosensory) disturbance is very common in the early stages of disease. This article addresses (i) why COVID-19 specifically targets the modalities of smell and possibly taste and what is the mechanism, (ii) what is the frequency of smell and taste loss and (iii) what is the overall prognosis. It is suggested that mouth-breathers may be at particular risk of COVID-19. Symptom-based questionnaires are likely to under-estimate the prevalence of chemosensory impairment by as much as 50%. The prevalence of smell loss is so high t

Taste sensitivity in anorexia nervosa: A systematic review.

OBJECTIVE: There is evidence for altered processing of taste in anorexia nervosa, particularly in the areas of reward processing and hedonic sensitivity. However, research on whether people with anorexia nervosa identify taste stimuli accurately, known as taste sensitivity, has yielded mixed findings. The objective of this study was to synthesize the literature on taste sensitivity in this disorder to provide a basis for future discussion on whether altered taste sensitivity may be also implicated in wider atypical taste processing in anorexia. METHOD: Electronic databases were searched syste

[Two Cases of Invasive Thymoma with Taste Disorder].

Two 50s female patients with the taste disorder of sweet taste loss and stage IV a type B2 invasive thymoma underwent surgery at our hospital. One patient with myasthenia gravis (MG) developed postoperative myasthenic crisis and recovered by the treatment with plasma apheresis and steroid pulse therapy. Her taste disorder fully recovered together with her MG symptom. The taste disorder of the other patient without MG had persisted for 3 years after the surgery. The taste disorder of sweet taste loss was reported as one of non-motor symptoms caused by MG-related autoimmune mechanisms associated

Anesthesia-inducing drugs also induce conditioned taste aversions.

Animals learn to reduce their intake of a tastant when its ingestion is followed by the administration of an anesthesia-inducing drug. To determine the nature of this intake suppression, the current study examined whether ketamine/xylazine (Experiment 1) and pentobarbital (Experiment 2) also conditionally reduce taste palatability. Using lick pattern analysis, we found that pairing saccharin with either drug reduced total licks, lick cluster size, and initial lick rate. Given that both lick cluster size and initial lick rate are indices of palatability, this pattern of results indicates that a

Taste and Hypertension in Humans: Targeting Cardiovascular Disease.

The association between salty taste and NaCl intake with hypertension is well-established, although it is far from completely understood. Other taste types such as sweet, umami or bitter have also been related to alterations in blood pressure. Here, we review the mutual relationship between taste and hypertension to identify potential avenues to better control blood pressure. This review focuses on published data involving humans, with the exception of a section on molecular mechanisms. There is compelling evidence to suggest that changes in salty taste sensitivity can be used to predict the o

Age-dependent differences in morphine-induced taste aversions.

Adolescence is a developmental period of particular importance given the host of neurobiological changes that occur during this stage of development. Drug use and abuse is said to be a function of the balance of its rewarding and aversive effects, and any age-dependent differences in morphine's aversive effects could impact drug intake. The present experiments examined the ability of morphine sulfate (0, 3.2, 10, and 18 mg/kg) to induce taste aversions in adolescent and adult rats under high (20-min fluid access each day; Experiment 1A/B) and low (50% of ad libitum access; Experiment 2A/B) dep
