Regulation of HTLV-1 tax stability, cellular trafficking and NF-κB activation by the ubiquitin-proteasome pathway.

Human T-cell leukemia virus type 1 (HTLV-1) is a complex retrovirus that infects CD4+ T cells and causes adult T-cell leukemia/lymphoma (ATLL) in 3%-5% of infected individuals after a long latent period. HTLV-1 Tax is a trans-activating protein that regulates viral gene expression and also modulates cellular signaling pathways to enhance T-cell proliferation and cell survival. The Tax oncoprotein promotes T-cell transformation, in part via constitutive activation of the NF-κB transcription factor; however, the underlying mechanisms remain unknown. Ubiquitination is a type of post-translational

The tumor marker Fascin is strongly induced by the Tax oncoprotein of HTLV-1 through NF-kappaB signals.

Oncogenic transformation of CD4(+) T cells by human T-cell lymphotropic virus type 1 (HTLV-1) is understood as the initial step to adult T-cell leukemia/lymphoma, a process that is mainly initiated by perturbation of cellular signaling by the viral Tax oncoprotein, a potent transcriptional regulator. In search of novel biomarkers with relevance to oncogenesis, we identified the tumor marker and actin-bundling protein Fascin (FSCN1) to be specifically and strongly up-regulated in both HTLV-1-transformed and adult T-cell leukemia/lymphoma patient-derived CD4(+) T cells. Fascin is important for m

HTLV-1 Tax transgenic mice develop spontaneous osteolytic bone metastases prevented by osteoclast inhibition.

One in 20 carriers of human T-cell leukemia virus type 1 (HTLV-1) will develop adult T-cell leukemia/lymphoma (ATL), a disease frequently associated with hypercalcemia, bone destruction, and a fatal course refractory to current therapies. Overexpression of the HTLV-1-encoded Tax oncoprotein under the human granzyme B promoter causes large granular lymphocytic leukemia/lymphomas in mice. We found that Tax+ mice spontaneously developed hypercalcemia, high-frequency osteolytic bone metastases, and enhanced osteoclast activity. We evaluated Tax tumors for the production of osteoclast-activating fa

HTLV-I Tax directly binds the Cdc20-associated anaphase-promoting complex and activates it ahead of schedule.

Expression of the human T lymphotropic virus type I (HTLV-I) transactivator/oncoprotein, Tax, leads to faulty mitosis as reflected by chromosome aneuploidy, cytokinesis failure, and formation of micro- and multinucleated cells. Here we show that HTLV-I-transformed T cells progress through S/G(2)/M phases of the cell cycle with a delay. This delay is correlated with a decrease in the levels of cyclin A, cyclin B1, and securin. In tax-expressing cells, the Cdc20-associated anaphase promoting complex (APC(Cdc20)), an E3 ubiquitin ligase that controls metaphase to anaphase transition, becomes acti

Paradoxical effects of a stress signal on pro- and anti-apoptotic machinery in HTLV-1 Tax expressing cells.

Adult T-cell leukemia (ATL) and HTLV-1 associated myelopathy/tropical spastic paraparesis (HAM/TSP) are associated with Human T-cell lymphotropic virus type 1 (HTLV-1) infection. The viral transactivator, Tax is able to mediate the cell cycle progression by targeting key regulators of the cell cycle such as p21/waf1, p16/ink4a, p53, cyclins D1-3/cdk complexes, and the mitotic spindle checkpoint MAD apparatus, thereby deregulating cellular DNA damage and checkpoint control. Genome expression profiling of infected cells exemplified by the development of DNA microarrays represents a major advance

IL-15 plays a major role in the persistence of Tax-specific CD8 cells in HAM/TSP patients.

IL-15 is a critical cytokine for the maintenance of memory-phenotype CD8 cells in mice. Here, we investigated the role of IL-15 in the neurological disease termed human T cell lymphotropic virus I-associated myelopathy/tropical spastic paraparesis (HAM/TSP). The high number of viral-specific CD8 cells in these patients is associated with inflammatory responses in the central nervous system. Because IL-15 is overexpressed in these patients, we asked whether IL-15 contributes to the persistence of human T cell lymphotropic virus I viral-specific CD8 cells. Using ex vivo cultures of HAM/TSP perip

HTLV-I Tax transrepresses the human c-Myb promoter independently of its interaction with CBP or p300.

The c-Myb proto-oncogene is preferentially expressed in hematopoietic lineages, and highly expressed in several leukemia types. The Human T-cell Leukemia Virus Type I (HTLV-I) is the etiological agent of Adult T-cell Leukemia/Lymphoma (ATLL). A previous report suggested that Tax, the viral transactivator, is able to suppress the transactivation potential of c-Myb protein by competing for recruitment of CBP. We tested whether such a competition could affect transcription from the c-Myb promoter in Tax expressing T-cells. Using several c-Myb promoter reporter constructs carrying mutations in var

The Expanded Child Tax Credit: Highest Burdens, Smallest Improvements Among Low-Income, Female-Headed Households.

The 2021 expansion of the Child Tax Credit provided advance monthly cash transfers to most US households with children. Although prior evaluations documented overall improvements in household well-being, less is known about variation by household structure. This study examined policy-period changes in parental mental health and material hardship among female-headed households. Using data from 1.3 million respondents to the Census Bureau's Household Pulse Survey (2020-25) and an intersectional framework, we applied difference-in-differences and multilevel models to estimate changes in parental

Effects of refundable state earned income tax credits on access to medical and dental services of low-income mothers.

BACKGROUND: Low-income women face constraints to timely access to medical and dental services. There is little evidence on whether refundable state earned income tax credit (EITC) programs affect access to care. We examine the effects of refundable state EITC levels on accessing medical and dental care among low-income mothers and potential interactions with state Medicaid eligibility levels. METHODS: We use data from 1996-2019 Behavioral Risk Factor Surveillance System (BRFSS). We focus on single mothers aged 18-44 with high school or less education and two or more children as the group that

Moderating Effects of the Low-Income Housing Tax Credit on Associations Between Race and Elevated Blood Pressure in Chicago.

While there is growing awareness of discriminatory housing policies and their adverse impacts on health, little is known about how housing policy may promote health equity. We focused on the Low-Income Housing Tax Credit (LIHTC), the largest affordable housing program in the United States, and examined how living in neighborhoods with LIHTC housing may moderate associations between elevated blood pressure and race/ethnicity in Chicago, IL. A retrospective cohort included 15,339 adult patients at an academic medical center from 2018 to 2019. We used mixed-effects hierarchal logistic regression

How can regional inequality caused by taxing non-CO2 greenhouse gases be mitigated? An economy-wide analysis.

To mitigate the risk of warming beyond the warming tipping point, countries need to focus on non-CO2 greenhouse gases (GHGs) emission reductions in addition to action on CO2. Considering that climate policies may lead to regional disparities and thus affect the achievement of emission reduction targets, ex ante assessment of the regional distributional impacts of global unified carbon pricing mechanism is critical. To this end, this study uses a global multi-regional computable general equilibrium model to assess the regional distributional impacts of including non-CO2 GHGs in the global unifi

Community-Based Intervention to Promote Awareness and Implementation Accuracy of Food and Beverage Tax Policies in Stores on Navajo Nation.

BACKGROUND: Unhealthy food and beverage taxes are gaining global momentum, but implementation complexities at the retailer level are poorly understood. The only such policy in the United States, a 2014 Navajo Nation legislation, applies a 2% tax to unhealthy foods and beverages, while exempting healthy foods and beverages from their 6% Navajo sales tax. In 2019, approximately half of small stores had not implemented both taxes correctly, primarily the tax exemption. Therefore, this study aimed to provide culturally-informed promotional materials to enhance implementation accuracy in these stor

TAX gene expression is associated with western blot patterns and high viral loadin HTLV-1 infected individuals.

BACKGROUND: Western blot (WB) is a conventional confirmatory test for evaluating infection with HTLV-1; however, indeterminate WB patterns remain a significant concern. TAX is a key regulatory protein of the HTLV-1 virus that increases the expression of viral gene products. This study aimed to compare the TAX expression profile in individuals with WB-positive and WB-indeterminate and evaluate its association with HTLV-1 proviral load MATERIALS AND METHODS: This study was conducted on individuals with WB-positive and WB-indeterminate HTLV-1 who had been referred to the Iranian Blood Transfusion

Tax or subsidy? The impact assessment of environmental policies on carbon allocation and emissions abatement of prefabricated construction supply chain.

The construction sector, as the prime culprit that aggravates greenhouse gas emissions, has initiated transitions in construction methods for emissions abatement. In such a context, prefabricated construction has been widely promoted in developing countries. This paper assesses the effects of three environmental policies: carbon tax, green subsidy, and unit subsidy on carbon allocation and emissions abatement of prefabricated construction supply chain (PCSC) consisting of a prefab manufacturer (PM) and a project developer (PD). In our game model, the PD determines a prefabricated ratio, the ra

Affordable housing through the Low-Income Housing Tax Credit program and opioid overdose emergency department visits.

INTRODUCTION: The United States continues to experience an opioid overdose crisis. As a key social determinant of health, housing insecurity may contribute to initiation of substance use and can threaten outcomes for those with substance use disorders by increasing stress, risky substance use, discontinuity of treatment, and return to use, all of which may increase the risk of overdose. The Low-Income Housing Tax Credit (LIHTC) program supports access to rental housing for low-income populations. By facilitating access to affordable housing, this program may improve housing security, thereby r

[Tax-targeted dendritic cell vaccine therapy for long-term remission of adult T-cell leukemia-lymphoma].

Adult T-cell leukemia-lymphoma (ATL) is a highly aggressive peripheral T-cell neoplasm caused by human T-cell leukemia virus type 1 (HTLV-1) infection occurring in approximately 5% of patients after prolonged latent period. ATL relapses within a short period despite its transient response to multiagent chemotherapy and the prognosis is extremely poor due to anticancer drug resistance and immunodeficiency. Although novel agents with different mechanisms, such as molecular targeted agents, have improved the prognosis, the number of cured patients remains limited. Hematopoietic stem cell transpla

A dichotomy of smokers in the Philippines following sin tax reform: Distinguishing potential quitters from those unlikely to quit.

The Philippine government significantly raised cigarette excise taxes in 2013, following passage of the landmark Sin Tax Reform Act of 2012. As a result, cigarette prices increased substantially. Given varying smokers' responses to the price increase, we examined underlying typologies of Filipino smokers and assessed how these typologies determine smoking intensity. We used cross-sectional data from the 2015 wave of the Philippine Global Adult Tobacco Survey (N = 1,651). To uncover typologies, random effects latent class modelling was used on six individual smoker responses (attempting to stop
