A TIR-NBS protein encoded by Arabidopsis Chilling Sensitive 1 (CHS1) limits chloroplast damage and cell death at low temperature.

Survival of plants at low temperature depends on mechanisms for limiting physiological damage and maintaining growth. We mapped the chs1-1 (chilling sensitive1-1) mutation in Arabidopsis accession Columbia to the TIR-NBS gene At1g17610. In chs1-1, a single amino acid exchange at the CHS1 N-terminus close to the conserved TIR domain creates a stable mutant protein that fails to protect leaves against chilling stress. The sequence of another TIR-NBS gene (At5g40090) named CHL1 (CHS1-like 1) is related to that of CHS1. Over-expression of CHS1 or CHL1 alleviates chilling damage and enhances plant

H2O2-triggered retrograde signaling from chloroplasts to nucleus plays specific role in response to stress.

Recent findings have suggested that reactive oxygen species (ROS) are important signaling molecules for regulating plant responses to abiotic and biotic stress and that there exist source- and kind-specific pathways for ROS signaling. In plant cells, a major source of ROS is chloroplasts, in which thylakoid membrane-bound ascorbate peroxidase (tAPX) plays a role in the regulation of H(2)O(2) levels. Here, to clarify the signaling function of H(2)O(2) derived from the chloroplast, we created a conditional system for producing H(2)O(2) in the organelle by chemical-dependent tAPX silencing using

Chlorophyll accumulation is enhanced by osmotic stress in graminaceous chlorophyllic cells.

We have developed a new chlorophyllic cell line ('TADH-XO') from the highly water stress tolerant grass Bouteloua gracilis (blue grama). When grown under normal (non-stress) conditions, this new cell line accumulates higher levels of chlorophyll (up to 368.1 microg total chlorophyll g(-1) FW) than a previously obtained cell line ('TIANSJ98'). Both cell lines are capable of developing substantially higher amounts of chlorophyll when subjected to osmotic stress. In order to explain these changes in the chlorophyll kinetics of the chlorophyllic cells, we analyzed the following population variable

Supramolecular organization of photosystem II and its light-harvesting antenna in partially solubilized photosystem II membranes.

We present an extended analysis of the organization of green plant photosystem II and its associated light-harvesting antenna using electron microscopy and image analysis. The analysis is based on a large dataset of 16 600 projections of negatively stained PSII-LHCII supercomplexes and megacomplexes prepared by means of three different pretreatments. In addition to our previous work on this system [Boekema, E.J., van Roon, H., Calkoen, F., Bassi, R. and Dekker, J.P. (1999) Biochemistry 38, 2233-2239], the following results were obtained. The rotational orientation of trimeric LHCII at the S, M

Is carbonic anhydrase activity of photosystem II required for its maximum electron transport rate?

It is known, that the multi-subunit complex of photosystem II (PSII) and some of its single proteins exhibit carbonic anhydrase activity. Previously, we have shown that PSII depletion of HCO3-/CO2 as well as the suppression of carbonic anhydrase activity of PSII by a known inhibitor of α‑carbonic anhydrases, acetazolamide (AZM), was accompanied by a decrease of electron transport rate on the PSII donor side. It was concluded that carbonic anhydrase activity was required for maximum photosynthetic activity of PSII but it was not excluded that AZM may have two independent mechanisms of action on

Hazardous gases (CO, NOx, CH4 and C3H8) released from CO2 fertilizer unit lead to oxidative damage and degrades photosynthesis in strawberry plants.

CO2 boilers/direct heating systems used in greenhouses often lead to incomplete combustion, which results in the formation of hazardous gases, such as carbon monoxide (CO), nitroxide (NOX) and other hydrocarbons. In this study, strawberry plants that were grown on rockwool cubes were transferred to airtight bottles and treated with CO, NOX, CH4 and C3H8 gases for 1-48 hours. Oxidative damage due to hazardous gases was observed, as indicated by H2O2 and [Formula: see text] determination. Photosynthetic pigments were reduced, and stomatal guard cells were damaged and remained closed compared to

Pigment-Structure Leaf Syndromes in Poinsettia (Euphorbia pulcherrima Willd. ex Klotzsch) Alter Chloroplast Ultrastructure, Biochemical Profiling, and Photosynthetic Quantum Yields.

Colored bracts and leaves are widespread in plants; however, how their pigments and structures jointly control the fate of absorbed light is poorly understood. We used contrasting albino and red Poinsettia (Euphorbia pulcherrima Willd. ex Klotzsch) phenotypes to test how selection for extreme color morphs reshapes chloroplast ultrastructure, photosynthetic quantum yield, and the spectral signatures of colored canopies. We combined hyperspectral and thermal imaging, pigment, biochemical and antioxidant profiling, gas exchange and chlorophyll a fluorescence, and light and electron microscopy acr

Surface functionalization of polystyrene nanoparticles modulates nanoparticle-induced phytotoxicity in Chlorella vulgaris.

Polystyrene nanoparticles (NPs) are increasingly released into aquatic systems, but how surface formulations shape their interactions with primary producers remains insufficiently understood. We examined the effects of nominally non-functionalized polystyrene NPs (PS-NPs) and amino- (PS-NH2-NPs) or carboxyl-functionalized NPs (PS-COOH-NPs) on Chlorella vulgaris after short-term 72 h exposure to a comparative high concentration (40 mg L-1). All polystyrene NPs remained stable in the culture medium and were detected in EPS-associated and cell-associated fractions, with stronger relative retentio

Proteomic and metabolic profiles of Cakile maritima Scop. Sea Rocket grown in the presence of cadmium.

Recent physiological reports have documented how Cakile maritima Scop. Sea Rocket could accumulate high doses of Cd without altering its physiological parameters. In the present study, we performed an integrated proteomics (2DE) and metabolomics (HPLC-MS) investigation to determine the molecular mechanisms underlying cadmium (Cd) tolerance of this halophyte. Peculiar features were observed: (i) up-regulation of thiol compound anabolism, including glutathione and phytochelatin homeostasis, which allows an intracellular chelation of Cd and its compartmentalization into vacuole by a significant u

Detecting hydrogen peroxide in leaves in vivo - a comparison of methods.

Four hydrogen peroxide detecting probes, 3,3'-diaminobenzidine (DAB), Amplex Red (AR), Amplex Ultra Red (AUR) and a europium-tetracycline complex (Eu(3)Tc) were infiltrated into tobacco leaves and tested for sensitivity to light, toxicity, subcellular localization and capacity to detect H(2)O(2) in vivo. In the absence of leaves, in water solutions, AUR was very much sensitive to strong light, AR showed slight light sensitivity, while DAB and Eu(3)Tc were insensitive to irradiation. When infiltrated into the leaves, the probes decreased the photochemical yield (Phi(PSII)) in the following orde

Solution structures of a cyanobacterial metallochaperone: insight into an atypical copper-binding motif.

The Atx1 copper metallochaperone from Synechocystis PCC 6803, ScAtx1, interacts with two P(1)-type copper ATPases to supply copper proteins within intracellular compartments, avoiding ATPases for other metals en route. Here we report NMR-derived solution structures for ScAtx1. The monomeric apo form has a betaalphabetabetaalpha fold with backbone motions largely restricted to loop 1 containing Cys-12 and Cys-15. The tumbling rate of Cu(I)ScAtx1 (0.1-0.8 mm) implies dimers. Experimental restraints are satisfied by symmetrical dimers with Cys-12 or His-61, but not Cys-15, invading the copper sit

Comparative physiological and transcriptomic analyses of differential mesotrione tolerance in foxtail millet seedlings.

Foxtail millet is an important drought-tolerant crop, yet its production is threatened by weed competition due to limited herbicide options. The 4-hydroxyphenylpyruvate dioxygenase (HPPD)-inhibiting herbicide mesotrione shows promise but causes differential responses among varieties. This study investigated the physiological and transcriptomic responses underlying differential mesotrione tolerance in two foxtail millet cultivars, Jingu 21 (JG21, sensitive) and Zhangzagu 10 (ZZG10, tolerant). Physiological analyses revealed that ZZG10 maintained higher net photosynthetic rate (Pn), photosystem

NpWRKY38 Negatively Regulates Drought Tolerance by Impairing ABA/SA Signaling as Well as Photosynthesis in Narenga porphyrocoma.

Narenga porphyrocoma, a wild relative of sugarcane with high drought tolerance, is a valuable germplasm resource for mining drought-tolerance genes. Weighted gene co-expression network analysis (WGCNA) identified NpWRKY38, a WRKY transcription factor that was significantly down-regulated under drought stress, a response that was further recapitulated by exogenous abscisic acid (ABA) treatment and consistent with the presence of ABA-responsive elements in its promoter. NpWRKY38 overexpression in rice resulted in a significant increase in endogenous ABA content, yet led to insufficient stomatal

Exploring the phytotoxicity mechanisms of PET nanoplastics and 6:2 FTSA in water hyacinth under individual and combined exposure scenarios.

Due to its similarity in hydrophobic properties to perfluorooctanesulfonic acid (PFOS), 6:2 fluorotelomer sulfonic acid (6:2 FTSA) has emerged as a key substitute for PFOS. Its presence in aquatic environments, along with the coexistence of polyethylene terephthalate (PET), may impact the growth of aquatic plants and ecosystem stability. This study explored the changes in antioxidant defense, photosynthetic system, and metabolic responses of water hyacinths (Eichhornia crassipes) under individual and combined exposure conditions. The results indicated that water hyacinth efficiently accumulate

Multiomics approach towards the detoxification of cadmium in Fritillaria cirrhosa using citrulline and zinc oxide nanoparticles in Qinghai-Tibet's ecosystems.

The Qinghai-Tibet Plateau, a biodiversity hotspot with escalating cadmium (Cd) pollution, demands urgent strategies to safeguard its endemic medicinal plants. Fritillaria cirrhosa, a critically endangered alpine species endemic to this region, confronts severe survival challenges due to habitat constraints and Cd-contaminated soils, leading to collapse of intrinsic detoxification systems. This study demonstrates that foliar application of citrulline and zinc oxide nanoparticles orchestrate altitude-specific physiological and molecular adaptations to counteract Cd toxicity while preserving medi

Efficient acclimation of the chloroplast antioxidant defence of Arabidopsis thaliana leaves in response to a 10- or 100-fold light increment and the possible involvement of retrograde signals.

Chloroplasts are equipped with a nuclear-encoded antioxidant defence system the components of which are usually expressed at high transcript and activity levels. To significantly challenge the chloroplast antioxidant system, Arabidopsis thaliana plants, acclimated to extremely low light slightly above the light compensation point or to normal growth chamber light, were moved to high light corresponding to a 100- and 10-fold light jump, for 6 h and 24 h in order to observe the responses of the water-water cycle at the transcript, protein, enzyme activity, and metabolite levels. The plants coped

Functional aspects of the photosynthetic light reactions in heat stressed Arabidopsis deficient in digalactosyl-diacylglycerol.

Plants are often submitted, in their natural environment, to various abiotic stresses such as heat stress. However, elevated temperature has a detrimental impact on overall plant growth and development. We have examined the physiological response of the dgd1-2 and dgd1-3 Arabidopsis mutants lacking 30-40% of digalactosyl-diacylglycerol (DGDG) exposed to heat constraint. These mutants, which grow similarly to wild type under normal conditions, were previously reported to be defective in basal thermotolerance as measured by cotyledon development. However their functional properties were not desc

Functional and structural study on chelator-induced suppression of S2/S1 FTIR spectrum in photosynthetic oxygen-evolving complex.

Chelating agents have been shown to induce characteristic changes in the light-minus-dark Fourier transform infrared (FTIR) difference spectrum for the S(2)/S(1) difference in the oxygen-evolving complex (OEC). Rafting kelp from California - is it alive? Physiological condition, ultrastructure and reaction to high UV treatment.

Kelp forests are essential marine ecosystems providing high-density habitats, nurseries, and food for countless species, including fish, invertebrates, and mammals. An increasing number of rafting thalli have been observed recently. This phenomenon may represent a natural dispersal strategy of kelp populations, or alternatively, it may be associated with rising ocean temperatures and the increased frequency and intensity of storm events. In the latter case, one can assume that thalli would be degrading during rafting and be associated with declining physiology. With this rationale in mind, we

TaZFP1, a C2H2 type-ZFP gene of T. aestivum, mediates salt stress tolerance of plants by modulating diverse stress-defensive physiological processes.

Salt stress suppresses plant growth, development, and crop productivity. In this study, we characterized the role of TaZFP1, a C2H2 type-zinc finger protein family member of T. aestivum, in salt stress tolerance. TaZFP1 possesses a conserved C2H2 motif (CX2-4CX12HX3-5H) shared by plant ZFP proteins, translocates to the nucleus after endoplasmic reticulum (ER) assortment, and displays a ZF 3-D structure similar to its eukaryote homologs. The transcripts of TaZFP1 were upregulated during salt stress condition and this effect was restored under normal conditions. Compared to wild type (WT), the t
