Identification of Livestock Ticks and Molecular Characterization of Tick-Associated Microorganisms in Senegal.

Ticks are obligate hematophagous arthropods at all developmental stages, primarily feeding on terrestrial vertebrates. They serve as vectors for numerous pathogens affecting both humans and animals. Accurate tick species identification currently requires an integrated approach combining morphological, molecular, and proteomic techniques. As part of a tick-borne disease surveillance program conducted in 2023 in the Kaffrine and Tambacounda regions of Senegal, ticks were collected from livestock in domestic environments to investigate the presence of Rickettsia spp. and other associated microorg

Immunohistochemical Assays to Visualize Tick-Borne Flaviviruses at the Tick Feeding Site.

Traditional immunohistochemistry (IHC) involves chromogenic detection of target antigens by antibodies that are conjugated to enzymes. Such immunostaining of thin tissue sections enables visualization of the distribution and localization of target antigens within their proper histological context. In this chapter, we describe how tick-borne flaviviruses can be visualized at the tick feeding site via IHC. We use antibodies specific to the North American tick-borne flavivirus, Powassan virus (POWV), to demonstrate how viral antigen can be visualized in the skin of a vertebrate host that is fed u

Yezo Virus Infection in Tick-Bitten Patient and Ticks, Northeastern China.

We identified Yezo virus infection in a febrile patient who had a tick bite in northeastern China, where 0.5% of Ixodes persulcatus ticks were positive for viral RNA. Clinicians should be aware of this potential health threat and include this emerging virus in the differential diagnosis for tick-bitten patients in this region.

Parasites of wombats (family Vombatidae), with a focus on ticks and tick-borne pathogens.

Ticks (Arachnida: Acari) are vectors for pathogens and the biggest threat to animal health. Many Australian ticks are associated with pathogens that impact humans, domestic animals and livestock. However, little is known about the presence or impact of tick-borne pathogens in native Australian wildlife. Wombats are particularly susceptible to the effects of the ectoparasite Sarcoptes scabiei which causes sarcoptic mange, the reason for which is unknown. Factors such as other ectoparasites and their associated pathogens may play a role. A critical understanding of the species of ectoparasites t

Tick-Borne Pathogens Shape the Native Microbiome Within Tick Vectors.

Ticks are blood-feeding arthropods and transmit a variety of medically important viral, bacterial, protozoan pathogens to animals and humans. Ticks also harbor a diverse community of microbes linked to their biological processes, such as hematophagy, and hence affect vector competence. The interactions between bacterial and/or protozoan pathogens and the tick microbiome is a black-box, and therefore we tested the hypothesis that the presence of a protozoan or bacterial pathogen will alter the microbial composition within a tick. Hence, this study was designed to define the microbial compositio

Systematic Review of Ticks and Tick-Borne Pathogens of Small Ruminants in Pakistan.

Ticks and tick-borne diseases (TTBDis) are a major constraint to the health and production of small ruminants in Pakistan. Despite being the subject of intermittent studies over the past few decades, comprehensive information on the epidemiology and control of TTBDis is lacking. Herein, we have systematically reviewed the current knowledge on TTBDis of small ruminants in Pakistan. Critical appraisal of the selected 71 articles published between 1947 to 2020 revealed that morphological examination had been the most widely used method for the identification of TTBDis in Pakistan. The Tick Project: Testing Environmental Methods of Preventing Tick-borne Diseases.

Prevention of tick-borne diseases in humans is challenging. To date, no prevention strategies have been shown to be consistently effective. Here, we describe the design of a new large-scale study, involving hundreds of households in Dutchess County, New York, testing whether environmental interventions, applied intensively and over 4 years, can prevent human cases.

Borrelia Diversity and Co-infection with Other Tick Borne Pathogens in Ticks.

Identifying Borrelia burgdorferi as the causative agent of Lyme disease in 1981 was a watershed moment in understanding the major impact that tick-borne zoonoses can have on public health worldwide, particularly in Europe and the USA. The medical importance of tick-borne diseases has long since been acknowledged, yet little is known regarding the occurrence of emerging tick-borne pathogens such as Borrelia spp., Anaplasma phagocytophilum, Rickettsia spp., Bartonella spp., "Candidatus Neoehrlichia mikurensis", and tick-borne encephalitis virus in questing ticks in Romania, a gateway into Europe

Tick iron and heme metabolism - New target for an anti-tick intervention.

Ticks are blood-feeding parasites and vectors of serious human and animal diseases. Ixodes ricinus is a common tick in Europe, transmitting tick-borne encephalitis, Lyme borreliosis, anaplasmosis, or babesiosis. Immunization of hosts with recombinant tick proteins has, in theory, the potential to interfere with tick feeding and block transmission of pathogens from the tick to the host. However, the efficacy of tick antigens has, to date, not been fully sufficient to achieve this. We have focused on 11 in silico identified genes encoding proteins potentially involved in tick iron and heme metab

Survival dynamics of tick-borne encephalitis virus in Ixodes ricinus ticks.

Biotic factors contributing to the survival of tick-borne viruses in nature are poorly understood. Using tick-borne encephalitis virus (TBEV) and its principal European vector, Ixodes ricinus, we examined the relative roles of salivary gland infection, co-feeding transmission, and moulting in virus survival. Virus titres in the salivary glands increased after blood-feeding in a time- and dose-dependent manner. This was observed in ticks infected by inoculation but not in ticks infected by the natural route of co-feeding. Amplification of infection prevalence occurred via co-feeding. However, w

Tick-borne encephalitis virus in ticks in Finland, Russian Karelia and Buryatia.

Tick-borne encephalitis (TBE) is a central nervous system infection caused by a flavivirus [tick-borne encephalitis virus (TBEV)], transmitted by Ixodes ticks and endemic in a large region in Eurasia. We collected 2411 ticks from Finland and Russia in 2003-2008, screened them for TBEV by RT-PCR and isolated and analysed eight strains belonging to all three TBEV subtypes; in addition, we obtained two European-subtype strains from human serum samples. TBEV RNA prevalence in unengorged ticks was approximately 1 % both in the northernmost TBE-endemic areas of Europe in Finland and Russian Karelia,

Molecular evidence for tick-borne encephalitis virus in ticks in South Korea.

To investigate whether tick-borne encephalitis viruses (TBEVs) are present in South Korea, Korean ixodid ticks were tested for TBEV RNA. Ticks (n = 2460) were collected from wild and domestic animals or by flagging at forest and grassland in 12 regions of five provinces in 2005-06. Four species in two genera were identified, yielding 197 sample pools (1-20 ticks per pool); from these, 12 envelope protein gene fragments of TBEV were amplified by reverse transcriptase-nested polymerase chain reaction (RT-nested PCR). Ten of the 2104 adult ticks (0.4%) and two of the 356 nymph ticks (0.2%) were p

Spread of ticks and tick-borne diseases in Germany due to global warming.

Tick-transmitted diseases like tick-borne encephalitis and Lyme Borreliosis have been well known in Germany for decades. Global climate changes may influence the emergence and reemergence of diseases. Ongoing research now gives an additional focus on other tick-borne pathogens such as Coxiella burnetii, Rickettsia conorii, Anaplasma phagocytophilum and Babesia spp., the causative agents of Q-fever, Mediterranean spotted fever, Anaplasmosis and Babesiosis, respectively. The epidemiology of these pathogens was investigated on ticks as well as on rodents, the main hosts. Therefore adults of Derma

Larval tick infestation: a case report and review of tick-borne disease.

Tick-borne disease in the United States continues to be a threat as people interact with their natural surroundings. We present a case of an 8-year-old boy with a larval tick infestation. Ticks within the United States can carry Lyme disease, Rocky Mountain spotted fever (RMSF), ehrlichiosis, babesiosis, tularemia, tick-borne relapsing fever, and tick paralysis. These preventable diseases are treatable when accurately recognized and diagnosed; however, if left untreated, they can cause substantial morbidity and mortality. This article highlights the knowledge necessary to recognize, treat, and

Ticks, tick-borne rickettsiae, and Coxiella burnetii in the Greek Island of Cephalonia.

Domestic animals are the hosts of several tick species and the reservoirs of some tick-borne pathogens; hence, they play an important role in the circulation of these arthropods and their pathogens in nature. They may act as vectors, but, also, as reservoirs of spotted fever group (SFG) rickettsiae, which are the causative agents of SFG rickettsioses. Q fever is a worldwide zoonosis caused by Coxiella burnetii (C. burnetii), which can be isolated from ticks. A total of 1,848 ticks (954 female, 853 male, and 41 nymph) were collected from dogs, goats, sheep, cattle, and horses in 32 different lo

Quantitative ecology of ticks as a basis for transmission models of tick-borne pathogens.

Tick population ecology is the basis of spatiotemporal variation in the risk of infection by tick-borne pathogens. The continental distribution of tick-borne encephalitis (TBE) in Europe is statistically associated with a specific pattern of the seasonal dynamics of Ixodes ricinus, and a particular characteristic of the seasonal land surface temperature profile. A tick population model would allow us to move from this statistical analysis to a biological, process-based model for TBE virus and the many other pathogens vectored by Ixodes spp. ticks in Europe and the United States.
