Effect of donor cornea preservation in tissue culture and in McCarey-Kaufmann medium on corneal graft rejection and visual acuity.

In a retrospective study the rate of rejection and clouding of the donor cornea for other reasons was investigated in 230 penetrating keratoplasties performed between 1984-1986. Donor corneas were stored in McCarey-Kaufmann Medium (MK) at 4 degrees C or in a modified Minimal Essential Medium (MEM) at 31 degrees C. No statistical differences in rejection rate, cloudiness due to other causes or visual acuity was found between MEM- and MK-stored donor corneas.

Evaluating effects of tissue type, preservation method, and decomposition on DNA quality to support genetic sampling in stranded small cetaceans.

Effective management decisions rely on accurate insights into species’ population structure, with high-quality genetic material being essential. Stranded cetaceans are an important source of samples, but meeting quality standards remains challenging. Despite their frequent use in conservation genetics, no controlled studies have compared DNA yield and quality across tissue types and preservation methods. This is further compounded by cetaceans uniquely derived physiology, meaning standards obtained from other mammals might not apply. In this study, we compared DNA concentration, purity, and in

Novel fixative solutions with superior preservation of structure and antigenicity are suitable for optical clearing, tissue staining, and flow cytometry.

Fixation of tissues and cells is an essential step in histological analysis; however, structural deformation and fluorescence signal attenuation caused by fixatives often pose challenges. This study aimed to evaluate the performance of a newly developed fixative, the FM Fix, by comparing it with conventional fixation methods in tissue staining, optical clearing, and flow cytometry analysis. FM Fix demonstrated staining properties comparable to those of standard fixatives and showed superiority with respect to tissue structure retention. Optical clearing allowed the visualization of fluorescent

A Novel Surgical Technique for Maxillary Alveolar Sealing: Use of a Tunneled Pedicled Connective Tissue Flap in Ridge Preservation Procedures.

This case report illustrates the use of a novel design for a tunneled connective tissue pedicled flap in socket sealing for guided bone regeneration procedures, specifically in the management of compromised sockets. The purpose of this flap is to optimize soft tissue healing and esthetics in these situations, permitting primary closure without buccal flap advancement. Initially, after creating the surgical access with a sulcular and a mesial releasing incision, extraction of the fractured tooth and immediate implant placement were performed. A xenograft and collagen membrane were then placed.

A Minimalistic Technique for Neural Tissue Preservation and Neuroanatomical Education: Quantitative Study of the Elnady Technique on Human Cadaveric Specimens.

Cadaveric shortages have been a challenge to anatomy education; as access remains low in many parts of the world, institutions are relying on plastinated specimens. Plastination typically requires the use of complex equipment and patented chemicals. While models solve cost and toxicity issues, in neuroanatomical education, the rigidity prevents deep-brain structure exploration and visual-spatial learning. The Elnady technique (ET) developed by Dr. Fawzy Elnady is an alternative method that solves the limitations of traditional plastination while maintaining the advantages previously developed

Use of a combination epithelized-subepithelial connective tissue graft for closure and soft tissue augmentation of an extraction site following ridge preservation or implant placement: description of a technique.

An esthetic implant-supported rehabilitation continues to be a major challenge in patients with a thin periodontium. Ridge preservation and immediate implant placement are intended to preserve the hard tissue volume and prevent preimplant bone loss following tooth extraction. Since these techniques are almost always combined with bone grafting, primary wound closure is indispensable. Therefore, a technique for reliable wound closure was developed. This technique employs a combined epithelized-subepithelial connective tissue graft, leaves the mucogingival line in its place, and has the added ad

Hard and soft tissue analysis of alveolar ridge preservation in esthetic zone using deproteinized bovine bone mineral and a saddle connective tissue graft: A long-term prospective case series.

AIM: Although alveolar ridge preservation (ARP) procedures appear to limit bone resorption after dental extraction, long-term outcomes remain limited. The objective of this prospective case series was to evaluate the long-term hard and soft tissue changes after ARP procedure in the aesthetic area, using deproteinized bovine bone mineral (DBBM) and saddle connective tissue graft. MATERIALS AND METHODS: Fifteen patients were subjected to ARP and impressions and CT scans were taken at baseline and 3 months. After 5 to 7 years, a secondary long-term clinical and radiological analysis was carried

Derivatization-Enabled Detection of Formalin From Preserved Tissues Using Coated Blade Spray Mass Spectrometry.

Formalin-fixed, fluid-preserved specimens in natural history collections offer invaluable scientific and cultural insight but pose genomic challenges due to molecular degradation and persistent chemical hazards. In particular, residual formaldehyde can impair downstream molecular workflows and present occupational safety risks for collection handlers. Formaldehyde modifies biological macromolecules by forming covalent crosslinks between nucleic acids and proteins and also by promoting DNA fragmentation, which lowers extraction efficiency and limits the recovery of amplifiable genetic material.

Talar Arthroscopic Reduction and Internal Fixation (TARIF): A Novel All-Inside Soft-Tissue-Preserving Technique.

BACKGROUND: Talar arthroscopic reduction and internal fixation (TARIF) is an alternative approach for the operative fixation of talar fractures that may be utilized instead of more traditional open approaches such as medial, lateral, or even dual anterolateral. The TARIF approach allows for nearly anatomic fracture reduction and fixation of talar neck, body, and posterior dome fractures while minimizing the soft-tissue stripping and vascular injury associated with the standard anterolateral approach. DESCRIPTION: Following initial closed fracture reduction and any associated procedures, we re

New insights on the systematics, palaeoecology and palaeobiology of a plesiosaurian with soft tissue preservation from the Toarcian of Holzmaden, Germany.

The Posidonienschiefer Formation (Toarcian) of Holzmaden, Baden-Württemberg in southwestern Germany has yielded several excellently preserved plesiosaurian specimens and received considerable research attention. The plesiosaurians found within these deposits are always significantly outnumbered by ichthyosaurs, and close examination of these rare specimens is crucial to a better understanding of the diversity and palaeoecology of Plesiosauria in this very peculiar ecosystem. The plesiosaurian specimen SMNS 51945 found in this area is a juvenile individual consisting of a partial, crushed skull

Ultrastructural preservation of rat embryonic dental tissues after rapid fixation and dehydration under microwave irradiation.

Adequate preservation of the cells and matrix of mineralising tissues remains difficult, as organic components and initial mineral deposits may be lost during conventional processing for electron microscopy. In this study, we have reduced significantly the processing time using microwave irradiation. Rat molar tooth germs were fixed in 4% glutaraldehyde + 4% formaldehyde with 0.1 M sodium cacodylate in a laboratory microwave oven for two periods of 20 s with a maximal temperature of 37 degrees C. After conventional washing and post-fixation, specimens were dehydrated in graded ethanols under m
