Pulmonary-route-dominant retention and exposure burden of biodegradable PLA microplastics: Radiotracer-based toxicokinetics and development of the TRACE exposure-response framework.

Despite the growing adoption of polylactic acid (PLA) as a biodegradable alternative to conventional plastics, its biological fate following human exposure remains poorly defined. No study has systematically compared the toxicokinetics of PLA microplastics (MPs) across inhalation and oral exposure routes within a unified experimental framework. Here, we developed a radiotracer-based platform combining 125I radiolabeling and high-resolution SPECT/CT imaging to quantify route-dependent biodistribution and clearance kinetics of PLA MPs in mice. Intratracheal administration produced greater and mo

Toxicokinetics of urinary 2-ethylhexyl salicylate and its metabolite 2-ethyl-hydroxyhexyl salicylate in humans after simulating real-life dermal sunscreen exposure.

Chemical UV filters are common components in sunscreens and cosmetic products. The question of adverse health risks is not completely resolved, partly owing to lacking human data from dermal exposure, which are essential for sound risk assessment. Therefore, we investigated the urinary toxicokinetics of 2-ethylhexyl salicylate (EHS) after a 1-day dermal real-life sunscreen application scenario. Twenty human volunteers were dermally exposed to a commercial sunscreen for 9 h under real-life conditions (2 mg/cm2 body surface area; double re-application; corresponding to 3.8 g EHS). Urine samples

Methyl mercuric chloride toxicokinetics in mice. II: Sexual differences in whole-body retention and deposition in blood, hair, skin, muscles and fat.

This article reports the time course for deposition of methyl mercury administered as a single oral dose in whole body, total carcass, liver, kidneys, brain, blood, fat, muscle, bone, skin and hair of male and female Bom:NMRI mice. The whole-body elimination initially approximated first order kinetics with half-times around 7 days and 12 days for males and females respectively, although a decreased elimination rate was observed during the last 10 days. The elimination of mercury from carcass was slower than the elimination from the whole-body, causing an increasing relative carcass deposition

Low level nose-only exposure to the nerve agent soman: toxicokinetics of soman stereoisomers and cholinesterase inhibition in atropinized guinea pigs.

In order to initiate a quantitative basis for the toxicology of low level exposure to nerve agents, the toxicokinetics of soman stereoisomers during nose-only exposure for 5 h to 20 ppb (160 microg/m3) of C(+/-)P(+/-)-soman in air were studied in restrained, anesthetized, and atropinized guinea pigs. The concentrations of the toxic C(+/-)P(-)-soman stereoisomers in blood increased according to a biexponential function, after an initial lag time of ca. 30 min for C(+)P(-)-soman, with final concentrations </= 36 pg/ml. It is hypothesized that the lag time is due to binding to carboxylesterases (

Does a carboxamide moiety alter the toxicokinetics of synthetic cannabinoids? A study after pulmonary and intravenous administration of cumyl-5F-P7AICA to pigs.

Synthetic cannabinoids (SCs) are consumed as an alternative to cannabis. Novel compounds are developed by minor modifications in their chemical structure, e.g. insertion of a carboxamide moiety as a linker, which can potentially lead to altered toxicokinetics (TK). Knowledge on the TK data of SCs, especially structural modified substances, is scarce. Hence, interpretation of toxicological results is challenging. Therefore, the aim of the present study was to evaluate the TK of cumyl-5F-P7AICA in a pig model, which was shown to be suitable for TK studies of SCs. A 200 µg/kg body weight dose of

Toxicokinetics and internal exposure of acrylamide: new insight into comprehensively profiling mercapturic acid metabolites as short-term biomarkers in rats and Chinese adolescents.

Acrylamide is classified as a probable carcinogen to humans and generated from Maillard reaction. Currently, the short-term exposure to acrylamide was evaluated via external diet sources in vitro or two main mercapturic acid metabolites: N-acetyl-S-(2-carbamoylethyl)-L-cysteine (AAMA) and N-acetyl-S-(2-carbamoyl-2-hydroxyethyl)-L-cysteine (GAMA) in vivo. In the present work, we comprehensively profiled four mercapturic acid metabolites and evaluated their internal exposure in rats and Chinese adolescents. The cumulative excretion of mercapturic acid metabolites contributes 38.4-73.0 and 43.8-6

Toxicokinetic Characterization of Isopropyl Glycidyl Ether in Rat by a Validated LC-APCI-MS/MS Method Using In-Source Derivatization.

Isopropyl glycidyl ether (IPGE) is a member of the large glycidyl ether family frequently used as a reactive diluent during the epoxy resin manufacturing process. Although the toxicity induced by this type of chemical has been investigated in many studies of different aspects (acute, subchronic, genotoxic, reproduction, etc.), there is still little known about their toxicokinetics. To gain information about the attainable systemic concentration, a liquid chromatography─tandem mass spectrometry (LC-MS/MS) method was developed and validated for the quantification of IPGE in rat plasma using its

The role of roughage provision on the absorption and disposition of the mycotoxin deoxynivalenol and its acetylated derivatives in calves: from field observations to toxicokinetics.

A clinical case in Belgium demonstrated that feeding a feed concentrate containing considerable levels of deoxynivalenol (DON, 1.13 mg/kg feed) induced severe liver failure in 2- to 3-month-old beef calves. Symptoms disappeared by replacing the highly contaminated corn and by stimulating ruminal development via roughage administration. A multi-mycotoxin contamination was demonstrated in feed samples collected at 15 different veal farms in Belgium. DON was most prevalent, contaminating 80% of the roughage samples (mixed straw and maize silage; average concentration in positives: 637 ± 621 µg/kg

Importance of Toxicokinetics to Assess the Utility of Zebrafish Larvae as Model for Psychoactive Drug Screening Using Meta-Chlorophenylpiperazine (mCPP) as Example.

The number of new psychoactive substances (NPS) increases rapidly, harming society and fuelling the need for alternative testing strategies. These should allow the ever-increasing number of drugs to be tested more effectively for their toxicity and psychoactive effects. One proposed strategy is to complement rodent models with zebrafish (Danio rerio) larvae. Yet, our understanding of the toxicokinetics in this model, owing to the waterborne drug exposure and the distinct physiology of the fish, is incomplete. We here explore the toxicokinetics and behavioral effects of an NPS, meta-chloropheny

Sex differences in urinary levels of several biological indicators of exposure: a simulation study using a compartmental-based toxicokinetic model.

Toxicokinetic modeling is a useful tool to describe or predict the behavior of a chemical agent in the human or animal organism. A general model based on four compartments was developed in a previous study to quantify the effect of human variability on a wide range of biological exposure indicators. The aim of this study was to adapt this existing general toxicokinetic model to three organic solvents--methyl ethyl ketone, 1-methoxy-2-propanol, and 1,1,1,-trichloroethane--and to take into account sex differences. In a previous human volunteer study we assessed the impact of sex on different bio

Evaluation of a mother-child dyad toxicokinetic model paired with environmental doses for estimation of serum per- and polyfluoroalkyl substances in early life.

Contact-based exposure estimation paired with empirically evaluated toxicokinetic (TK) modeling can complement direct measurements of serum concentrations for characterizing individual exposures and provide a proxy for risk assessment. This study examined whether a mother-child dyad TK model paired with PFAS exposure estimates provide reasonable predictions of serum PFAS concentrations for children. A previously published mother-child dyad TK model was used to estimate the child serum concentrations. We modified the model by updating with recent TK parameter estimates. Background intake rates

Toxicokinetics and analytical toxicology of four N, N-dimethyltryptamine derivatives studied in human in vitro systems and in vivo by means of zebrafish embryos.

N,N-Dimethyltryptamine (DMT) is a naturally occurring substituted tryptamine used as a psychedelic drug for ritual purposes by various cultures. Derivatives were already found on the drug of abuse market, such as N1-tert-butoxycarbonyl-N,N-dimethyltryptamine (DMT-Boc). The aim of the current study was to investigate the toxicokinetics of DMT-Boc and three related DMT derivatives, by name DMT-isopropylcarbamate, DMT-pivaloylamide, and DMT-THP (N,N-dimethyl-2-[1-(oxan-2-yl)indol-3-yl]ethanamine). Benchmark Dose for Dioxin Based on Gestational Diabetes Mellitus Using Coexposure Statistical Methods and an Optimized Physiologically Based Toxicokinetic Model.

Dioxins are ubiquitous endocrine-disrupting substances, but determining the effects and benchmark doses in situations of coexposure is highly challenging. The objective of this study was to assess the relationship between dioxin andgestational diabetes mellitus (GDM), calculate the benchmark dose (BMD) of dioxin in coexposure scenarios, and derive a daily exposure threshold using an optimized physiologically based toxicokinetic (PBTK) model. Based on a nested case-control study including 77 cases with GDM and 154 controls, serum levels of 29 dioxin-like compounds (DLCs) along with 10 perfluoro

Tissue-specific accumulation and integrated toxicological responses of the Pacific oyster, Crassostrea gigas, to chronic polyethylene microplastic exposure: An empirical toxicokinetic interpretation.

Microplastic (MP) bioaccumulation is increasingly recognized as a critical determinant of toxicological outcomes in aquatic organisms; however, the relationships between tissue-specific accumulation patterns and subsequent biological responses remain insufficiently understood. This study investigated polyethylene microplastic (PE-MP) bioaccumulation and associated toxicological responses in the Pacific oyster, Crassostrea gigas, under chronic exposure conditions (0-1000 μg/L) for 2 and 4 weeks. PE-MPs accumulated in a concentration-dependent manner and were consistently retained at higher leve

Evidence Supporting the Use of Optically Pure S-(+)-Indoxacarb: Discovery and Toxicokinetic Characterization of Two New Pairs of Metabolite Enantiomers in Rats.

Indoxacarb is a typical chiral pesticide composed of two enantiomers, of which only S-(+)-indoxacarb exhibits strong insecticidal activity. Growing interest in its enantioselective toxicology and metabolites has highlighted the need for more data on mammalian metabolism to assess human health risks. In this study, two new pairs of enantiomeric metabolites were identified from indoxacarb metabolism in rats: S-IN-RM493 and S-IN-RM541 derived from S-(+)-indoxacarb, and R-IN-RM493 and R-IN-RM541 derived from R-(-)-indoxacarb.
