Suppression of transformation and tumorigenicity in interspecies hybrids of human SV40-transformed and mouse 3T3 cell lines.

Somatic cell hybrids, formed by fusion of human SV40-transformed fibroblast lines and mouse 3T3 cells, were isolated and analysed for expression of transformation phenotypes and tumorigenic potential in immunodeficient nude mice. Both tumorigenic and nontumorigenic SV40-transformants were used for these experiments. Regardless of whether or not the parental human transformed parent line was tumorigenic, the hybrid progeny-with rare exception-did not form tumors. The possibility that human SV40-transformed lines and the derived hybrid progeny were immunologically rejected in nude mice was unlik

Richter syndrome: a review of clinical, ocular, neurological and other manifestations.

Richter syndrome describes the development of high-grade non-Hodgkin lymphoma (NHL) or Hodgkin lymphoma in patients with chronic lymphocytic leukemia (CLL)/small lymphocytic lymphoma (SLL). Richter transformation occurs in 3.3 to 10.6% of patients with CLL. The large cell lymphoma clone occurs by transformation of the original CLL clone in the majority of patients, and as a separate and independent neoplasm in fewer cases. Richter transformation may be triggered by viral infections, such as Epstein-Barr virus infection, which are common in immunosuppressed patients. Trisomy 12 and chromosome 1

Report of a workshop on malignant lymphoma: a review of molecular and clinical risk profiling.

Molecular genetic analysis adds important information for lymphoma biology and classification, but the latter is challenged by recent improvement of combined chemo-immunotherapy. In aggressive lymphoma, molecular profiling identifies risk groups with certain genetic background but still the International Prognostic Index (IPI) remains the most important clinically applicable predictor of outcome. In follicular lymphoma (FL), the importance of the meshwork of bystander cells becomes increasingly evident. As combined immuno-chemotherapy improved the prognosis of the patients, several clinical tr

Induction of Embryogenic Callus, Protoplast Isolation, and PEG-Mediated Transformation Protocols in Eucommia ulmoides.

Eucommia ulmoides, a tree species native to China, holds considerable medicinal, ecological, and industrial importance. However, the absence of an efficient and stable genetic transformation system poses significant challenges to gene function studies and molecular breeding in E. ulmoides. Protoplasts, which lack cell walls, serve as effective receptors for transient transformation and are thus ideal for genetic engineering research. In this study, the optimal conditions for callus induction were identified, and formation of the embryogenic callus was confirmed by histological analysis. Furthe

Pathology of follicular lymphoma.

Follicular lymphoma (FL) is a heterogeneous disease, and there are many different subgroups, such as in terms of age of onset, involved organ (especially extranodal sites such as gastrointestinal tract) and genetic abnormality. Grade 3B is currently regarded as a distinct entity by molecular genetic analyses, but the independence of Grade 3A remains unclear. Variations of clinical course are known in FL. Some cases are very indolent, but others are not. The latter cases show histological transformation to diffuse large B-cell lymphoma (DLBCL) (high-grade transformation) and an aggressive cours

Soybean Hairy Root Transformation for the Analysis of Gene Function.

Soybean (Glycine max) is a valuable crop in agriculture that has thousands of industrial uses. Soybean roots are the primary site of interaction with soil-borne microbes that form symbiosis to fix nitrogen and pathogens, which makes research involving soybean root genetics of prime importance to improve its agricultural production. The genetic transformation of soybean hairy roots (HRs) is mediated by the Agrobacterium rhizogenes strain NCPPB2659 (K599) and is an efficient tool for studying gene function in soybean roots, taking only 2 months from start to finish. Here, we provide a detailed p

Evaluation of Parameters Affecting Agrobacterium-Mediated Transient Gene Expression in Industrial Hemp (Cannabis sativa L.).

Industrial hemp Cannabis sativa L. is an economically important crop mostly grown for its fiber, oil, and seeds. Due to its increasing applications in the pharmaceutical industry and a lack of knowledge of gene functions in cannabinoid biosynthesis pathways, developing an efficient transformation platform for the genetic engineering of industrial hemp has become necessary to enable functional genomic and industrial application studies. A critical step in the development of Agrobacterium tumefaciens-mediated transformation in the hemp genus is the establishment of optimal conditions for T-DNA g

SETBP1-R54P mutation promotes malignant transformation of cadmium-induced 16HBE cells by down-regulating circ_0007095 expression.

Some cancers are strongly associated with exposure to environmental carcinogens, and genetic and epigenetic alterations are crucial in carcinogenesis. However, the interaction between genetic mutations and epigenetic regulation in cancer development has not been sufficiently examined. Here, we investigated the roles of gene mutations and altered circRNA expression in the malignant transformation of human bronchial epithelial cells after continuous exposure to CdCl2 (10 µM). The circular RNA circ_0007095 was down-regulated, and the SETBP1-R54P single nucleotide variant (SNV) accumulated during

Transforming growth factor-beta pathway serves as a primary tumor suppressor in CD8+ T cell tumorigenesis.

Tumorigenesis in rodents, as well as in humans, has been shown to be a multistep process, with each step reflecting an altered gene product or gene regulatory process leading to autonomy of cell growth. Initial genetic mutations are often associated with dysfunctional growth regulation, as is demonstrated in several transgenic mouse models. These changes are often followed by alterations in tumor suppressor gene function, allowing unchecked cell cycle progression and, by genomic instability, additional genetic mutations responsible for tumor metastasis. Here we show that reduced transforming g

Neisseria gonorrhoeae secretes chromosomal DNA via a novel type IV secretion system.

The process of DNA donation for natural transformation of bacteria is poorly understood and has been assumed to involve bacterial cell death. Recently in Neisseria gonorrhoeae we found that mutations in three genes in the gonococcal genetic island (GGI) reduced the ability of a strain to act as a donor in transformation and to release DNA into the culture. To better characterize the GGI and the process of DNA donation, the 57 kb genetic island was cloned, sequenced and subjected to insertional mutagenesis. DNA sequencing revealed that the GGI has characteristics of a horizontally acquired geno

Vincristine sulfate-induced cell transformation, mitotic inhibition and aneuploidy in cultured Syrian hamster embryo cells.

Vincristine, a naturally occurring Vinca alkaloid and widely used anti-neoplastic agent, was examined for its ability to induce cell transformation, inhibition of growth and mitosis, and genetic effects in Syrian hamster embryo cells in culture. Treatment of the cells with doses of less than or equal to 1 ng/ml vincristine sulfate (VCR) had no effect on cell growth, while exposure to greater than or equal to 3 ng/ml reduced the growth rate and treatment with 30 ng/ml resulted in no detectable increase in cell number. At this latter dose the mitotic index of the cells increased significantly su

B-cell acute lymphoblastic leukemia following myelodysplastic syndromes: a case report and literature review.

Myelodysplastic syndromes (MDS) comprise clonal hematopoietic stem cell disorders characterized by heterogeneous clinical manifestations. Approximately 20-30% of MDS cases progress to acute myeloid leukemia, whereas transformation to acute lymphoblastic leukemia (ALL) is extremely rare. In this report, we present the case of a Chinese male patient who presented with MDS-refractory anemia with ringed sideroblasts, which developed into B-cell ALL. During disease transformation, the patient acquired novel gene mutations. By comparing the gene mutations identified at the initial MDS diagnosis with

Gastroesophageal reflux and Barrett's esophagus: a pathway to esophageal adenocarcinoma.

The incidence of esophageal adenocarcinoma (EAC) has increased significantly in Western countries. The rising prevalence of gastroesophageal reflux disease (GERD) and obesity has played a major role in the increasing numbers of EAC. Approximately 10-15% of patients with GERD will develop Barrett's esophagus (BE). This metaplastic transformation is due to the chronic injury produced by repeated reflux episodes and involves genetic mutations that can lead to a malignant transformation. Ultimately, the development of EAC is characterized by the sequence BE metaplasia-dysplasia-invasive carcinoma.

Boron/sodium co-doping enhances phase transformation in TiO2 nanofibers and amplifies photoelectrochemical nucleic acid oxidation detection.

BACKGROUND: Oxidative damage in genetic molecules disrupts cellular function and poses long-term risks to organismal health, underscoring the need for early, rapid, and sensitive detection methods. Early and precise detection of this oxidative damage in genetic molecules can enable necessary precautions to be taken and initiate the treatment process early, thereby saving the lives of patients. Oxidation detection and analysis using electrochemical methods are frequently employed. The properties of the electrodes used in electrochemical measurements are significant for sensor sensitivity. Evaluation of Potential Risk Factors that contribute to Malignant Transformation of Oral Lichen Planus: A Literature Review.

AIM: Many studies have suggested that a lesion originally diagnosed as oral lichen planus (OLP) has different possibilities of undergoing malignant transformation in time, although these findings remain a controversial issue; for example, some studies reported different values of potential malignancy of OLP. INTRODUCTION: World Health Organization (WHO) classifies OLP as a "potentially malignant disorder" with unspecified malignant transformation risk, and suggests that OLP patients should be closely monitored. Numerous studies have attempted to confirm the malignant transformation potential

The impact of category, cytopathology and cytogenetics on development and progression of clonal and malignant myeloid transformation in inherited bone marrow failure syndromes.

Inherited bone marrow failure syndromes are a group of rare, heterogeneous genetic disorders with a risk of clonal and malignant myeloid transformation including clonal marrow cytogenetic abnormalities, myelodysplastic syndrome and acute myeloid leukemia. The clinical characteristics, risk classification, prognostic factors and outcome of clonal and malignant myeloid transformation associated with inherited bone marrow failure syndromes are largely unknown. The aims of this study were to determine the impact of category, cytopathology and cytogenetics, the three components of the "Category Cyt

Natural transformation of Neisseria gonorrhoeae: from DNA donation to homologous recombination.

Gonococci undergo frequent and efficient natural transformation. Transformation occurs so often that the population structure is panmictic, with only one long-lived clone having been identified. This high degree of genetic exchange is likely necessary to generate antigenic diversity and allow the persistence of gonococcal infection within the human population. In addition to spreading different alleles of genes for surface markers and allowing avoidance of the immune response, transformation facilitates the spread of antibiotic resistance markers, a continuing problem for treatment of gonococc

Targeting the origins of multiple myeloma along hematopoietic stem cell lymphoid lineage differentiation.

The initiation and progression of multiple myeloma (MM) are intricate processes, and a critical challenge lies in understanding the mechanisms of malignant transformation in MM-initiating cells (MICs) and their driver genes. In this study, we used single-cell sequencing and genetic tracer analysis at each developmental stage, from hematopoietic stem cells to lymphoid lineage differentiation, to identify abnormal differentiation stages in patients with MM. We found that chromosome 1q amplification (1qAmp) originated from a specific subgroup of B cells, whereas chromosome 17p deletion occurred a
