House dust mite sensitization shapes the clinical expression of shellfish allergy.

Background. Cross-reactivity among phylogenetically distant invertebrates-such as mites, crustaceans, and mollusks-is mainly driven by conserved panallergens including tropomyosin, arginine kinase, and troponin C. However, the clinical impact and molecular basis of these cross-reactivities remain incompletely defined. Methods. A multicenter observational study was conducted in 3,777 patients reactive to at least one arthropod or invertebrate allergen component. Serum IgE profiles were assessed using the ALEX²® multiplex platform, including allergens from mites, crustaceans, mollusks, and relat

The insight into house dust mites: which Dermatophagoides pteronyssinus proteins and cross-reactive allergens play pivotal role in sensitization among Polish patients with suspicion of allergy?

PURPOSE: The profiles of house dust mite (HDM) sensitization may differ in various geographical regions, which could affect the course of allergic diseases. The identification of these discrepancies could help to introduce improved methods of HDM allergy management, more suited to the local needs. The study aimed to analyze the pattern of sensitization to Dermatophagoides pteronyssinus (DP) allergen components and allergens that may cross-react with the allergen components among Polish children and adults with suspicion of allergy. METHODS: The sensitization to DP allergens and homologous pro

Shrimp sensitization in house dust mite allergic patients.

Shrimp tropomyosin has a similar structure to house dust mite (HDM) tropomyosin. In this research, 232 adult patients with symptoms of persistent allergic rhinitis were randomly selected. In the group, 59% were sensitized to Dermatophagoides pteronyssinus and 57.8% to Dermatophagoides farinae. In total, 128 (55.2%) patients were sensitized to both HDM species and 143 (61.6%) to at least one. Slightly over a quarter (25.4%) of patients were sensitized to shrimp. Of the 35 shrimp-sensitized patients, the sensitization to Der p 10 and Pen a 1 was found in 11 cases (31.4%). There was a strong corr

Changes in the soluble bone proteome of reared white seabream (Diplodus sargus) with skeletal deformities.

One of the main constrains for commercial aquaculture production of white seabream (Diplodus sargus) is the high incidence of skeletal malformations in reared fish. The purpose of this study was to obtain a better understanding of the mechanisms involved in the development of these types of skeletal malformations by comparative proteomic analysis of the vertebral column of normal and deformed fish using 2DE for protein separation and MS for protein identification. We observed a 3.2 and 3.4-fold increase in the expression of two tropomyosin isoforms, one of which (tropomyosin-4) is essential fo

Helminth Allergens, Parasite-Specific IgE, and Its Protective Role in Human Immunity.

The Th2 immune response, culminating in eosinophilia and IgE production, is not only characteristic of allergy but also of infection by parasitic worms (helminths). Anti-parasite IgE has been associated with immunity against a range of helminth infections and many believe that IgE and its receptors evolved to help counter metazoan parasites. Allergens (IgE-antigens) are present in only a small minority of protein families and known IgE targets in helminths belong to these same families (e.g., EF-hand proteins, tropomyosin, and PR-1 proteins). During some helminth infection, especially with the

Shrimp Extract Exacerbates Allergic Immune Responses in Mice: Implications on Clinical Diagnosis of Shellfish Allergy.

Tropomyosin has been identified as the major cross-reactive shellfish allergen, but recent studies showed the presence of other clinically relevant allergens. This study aims at determining the allergic immune responses of mice sensitized with raw and boiled shrimp extracts in comparison to recombinant tropomyosin (rTM). Female Balb/c mice were intragastrically sensitized and challenged with raw, boiled shrimp or rTM. Systemic, cellular and humoral allergic responses were compared, while allergenicity of the extracts was also compared by skin prick test (SPT) and immunoblot on shrimp allergic

Identification and characterization of novel allergen candidates in blood clams (Anadara broughtonii and Tegillarca granosa) using proteomics and immunoinformatics.

Blood clams (Anadara broughtonii and Tegillarca granosa) are widely consumed in Southeast Asia and can trigger allergic reactions. Whereas tropomyosin and arginine kinase are established allergens, the broader allergen profile and cross-reactivity with shrimp remain poorly defined. Using gel and LC-MS-based proteomics, a novel immunoinformatic approach combining three algorithms and IgE immunoassays, we examined IgE-reactivity in shrimp-sensitized patients. Among 2580 identified proteins, 35 were from blood clams, while 9 were predicted as allergens. From IgE-reactive 2D gel spots, known aller

Sensitization pattern of crustacean-allergic individuals can indicate allergy to molluscs.

This study investigated the sensitization pattern of crustacean-allergic patients according to tolerance to molluscs. Thirty-one patients with anaphylaxis to crustaceans (14 with mollusc allergy and 17 with mollusc tolerance) were studied using skin prick tests (SPTs), specific IgEs (sIgEs) and SDS-PAGE immunoblotting. IgE-reactive shrimp proteins were identified by proteomic analyses. Patients with mollusc allergy presented more frequently SPTs positive to molluscs and higher sIgE titres in response to both molluscs and crustaceans. Shrimp-sIgE and rPen a1-sIgE values of 1.57 kUA /l and 4.38

Effect of enzymatic hydrolysis on bioactive properties and allergenicity of cricket (Gryllodes sigillatus) protein.

Food-derived bioactive peptides have gained attention for their role in preventing chronic diseases. Edible insects are viable sources of bioactive peptides owing to their high protein content and sustainable production. In this study, whole crickets (Gryllodes sigillatus) were alcalase-hydrolyzed to a degree of hydrolysis (DH) ranging from 15 to 85%. Antioxidant activity, angiotensin converting enzyme (ACE), and dipeptidyl peptidase-4 (DPP-IV)- inhibition of the cricket protein hydrolysates (CPH) were evaluated before and after simulated gastrointestinal digestion (SGD). Antioxidant activity

Partial TrkB receptor activation suppresses cortical epileptogenesis through actions on parvalbumin interneurons.

Post-traumatic epilepsy is one of the most common and difficult to treat forms of acquired epilepsy worldwide. Currently, there is no effective way to prevent post-traumatic epileptogenesis. It is known that abnormalities of interneurons, particularly parvalbumin-containing interneurons, play a critical role in epileptogenesis following traumatic brain injury. Thus, enhancing the function of existing parvalbumin interneurons might provide a logical therapeutic approach to prevention of post-traumatic epilepsy. The known positive effects of brain-derived neurotrophic factor on interneuronal gro

Loss of crossbridge inhibition drives pathological cardiac hypertrophy in patients harboring the TPM1 E192K mutation.

Hypertrophic cardiomyopathy (HCM) is an inherited disorder caused primarily by mutations to thick and thinfilament proteins. Although thin filament mutations are less prevalent than their oft-studied thick filament counterparts, they are frequently associated with severe patient phenotypes and can offer important insight into fundamental disease mechanisms. We have performed a detailed study of tropomyosin (TPM1) E192K, a variant of uncertain significance associated with HCM. Molecular dynamics revealed that E192K results in a more flexible TPM1 molecule, which could affect its ability to regu

A TPM2 mutation causes congenital myopathy with fibre-type disproportion.

We report a 9-year-old girl with delayed motor milestones and respiratory difficulty since birth. She presented as a floppy infant, with generalised muscle wasting, dysphagia and facial weakness. The muscle biopsy of the biceps brachii revealed congenital fibre-type disproportion (CFTD) and Sanger sequencing detected a pathogenic variant in the beta-tropomyosin (TPM2) gene (c.415_417delGAG; p.Glu139del). There has been only one previous report of CFTD associated with p.Glu139del in the TPM2 gene.

Reduced Allergenicity of Shrimp (Penaeus vannamei) by Altering the Protein Fold, Digestion Susceptibility, and Allergen Epitopes.

The mechanism by which thermal/pressure processing influences the allergenicity of shrimp (Penaeus vannamei) was explored by anaphylaxis in mice, the protein structure, gastrointestinal digestion, and linear epitopes. Roasting induced the unfolding of the structure, which may reduce the allergenicity, but it made more linear epitopes to be exposed, causing mice to exhibit similar systemic anaphylaxis as mice fed with the raw shrimp protein (p > 0.05). However, the roasted + reverse-pressure-sterilized shrimp can significantly reduce specific antibodies, mast cell degranulation, vascular permea

The Mechanisms of Thin Filament Assembly and Length Regulation in Muscles.

The actin containing tropomyosin and troponin decorated thin filaments form one of the crucial components of the contractile apparatus in muscles. The thin filaments are organized into densely packed lattices interdigitated with myosin-based thick filaments. The crossbridge interactions between these myofilaments drive muscle contraction, and the degree of myofilament overlap is a key factor of contractile force determination. As such, the optimal length of the thin filaments is critical for efficient activity, therefore, this parameter is precisely controlled according to the workload of a gi

Evaluation of Der p 10 in a Cohort of European Children: Role of Molecular Diagnostics and Clinical Features.

BACKGROUND: Allergy toward the dust mite is steadily increasing on the European continent. This sensitization may be a risk factor for developing sensitization to other mite molecules such as tropomyosin Der p 10. This molecule often correlates with food allergy and the risk of anaphylaxis after ingesting mollusks and shrimps. MATERIALS AND METHODS: We analyzed the sensitization profiles by ImmunoCAP ISAC of pediatric patients from 2017 to 2021. The patients under investigation were being followed for atopic disorders such as allergic asthma and food allergies. The study aimed to analyze the

Modifications of myofilament protein phosphorylation and function in response to cardiac arrest induced in a swine model.

Cardiac arrest is a prevalent condition with a poor prognosis, attributable in part to persistent myocardial dysfunction following resuscitation. The molecular basis of this dysfunction remains unclear. We induced cardiac arrest in a porcine model of acute sudden death and assessed the impact of ischemia and reperfusion on the molecular function of isolated cardiac contractile proteins. Cardiac arrest was electrically induced, left untreated for 12 min, and followed by a resuscitation protocol. With successful resuscitations, the heart was reperfused for 2 h (IR2) and the muscle harvested. In

Shellfish allergy.

Seafood plays an important role in human nutrition and health. The growing international trade in seafood species and products has added to the popularity and frequency of consumption of a variety of seafood products across many countries. This increased production and consumption of seafood has been accompanied by more frequent reports of adverse health problems among consumers as well as processors of seafood. Adverse reactions to seafood are often generated by contaminants but can also be mediated by the immune system and cause allergies. These reactions can result from exposure to the seaf
