The localization, origin, and impact of platelets in the tumor microenvironment are tumor type-dependent.

BACKGROUND: How platelets interact with and influence the tumor microenvironment (TME) remains poorly characterized. METHODS: We compared the presence and participation of platelets in the TME of two tumors characterized by highly different TME, PyMT AT-3 mammary tumors and B16F1 melanoma. RESULTS: We show that whereas firmly adherent platelets continuously line tumor vessels of both AT-3 and B16F1 tumors, abundant extravascular stromal clusters of platelets from thrombopoietin-independent origin were present only in AT-3 mammary tumors. We further show that platelets influence the angiogeni

Demethylases in tumors and the tumor microenvironment: Key modifiers of N6-methyladenosine methylation.

RNA methylation modifications are widespread in eukaryotes and prokaryotes, with N6-methyladenosine (m6A) the most common among them. Demethylases, including Fat mass and obesity associated gene (FTO) and AlkB homolog 5 (ALKBH5), are important in maintaining the balance between RNA methylation and demethylation. Recent studies have clearly shown that demethylases affect the biological functions of tumors by regulating their m6A levels. However, their effects are complicated, and even opposite results have appeared in different articles. Here, we summarize the complex regulatory networks of dem

Enhanced manipulation of tumor microenvironments by nanomotor for synergistic therapy of malignant tumor.

Tumor microenvironments (TME) play critical roles in the growth and metastasis of tumor tissue, which provide a promising way to treat malignant tumor via manipulation of TME. However, developing proper strategy to effectively control TME is still a challenge. Herein, a Ce6@AT-PEG-MSN-Pt (CAPMP) nanomotor is fabricated to spontaneously move in tumor tissue and concurrently perform the enhanced manipulation of various tumor microenvironments including copper levels, hypoxia, local temperature and reactive oxygen species (ROS) for effective tumor therapy. The CAPMP nanomotor consists of a janus

Tumor-associated macrophages in the tumor immune microenvironment of non-small cell lung cancer.

BACKGROUND: Non-small cell lung cancer (NSCLC) remains the leading cause of cancer mortality, and resistance to immune checkpoint inhibitors (ICIs) is largely driven by the tumor immune microenvironment (TIME), in which tumor-associated macrophages (TAMs) are major regulators. OBJECTIVES: This review summarizes recent advances in understanding TAM heterogeneity, their contribution to immune resistance in NSCLC, and emerging therapeutic strategies targeting TAMs. METHODS: A literature search was conducted using PubMed and Google Scholar, focusing on studies published between 2021 and 2025. The Osteosarcoma Tumor Microenvironment: Supportive of Tumor Progression, Hostile to Immunity, but Therapeutically Targetable.

Osteosarcoma (OSA) remains the most common primary malignant bone tumor of children and young adults, with limited survival improvements in patients with relapsed or metastatic disease despite multimodal chemotherapeutic treatment. Beyond the intrinsic heterogeneity and malignancy of tumor cells, OSA progression and therapeutic resistance are strongly influenced by the specialized bone tumor microenvironment (TME). Interactions among tumor, immune and stromal cells generate a complex ecosystem characterized by immune suppression, hypoxia and metabolic adaptation, representing a major challenge

Inhibition of KDEL Receptors Remodels the Tumor Microenvironment for T Cell Independent Tumor Regression.

Tumor immunotherapy is supported by low-grade inflammatory conditions in the microenvironment, triggered by immunogenic cell death (ICD). However, ICD is dampened when tumors acquire resistance, affecting immune recognition. KDEL receptors (KDELRs), through a retrograde Golgi-to-ER transport, prevent spontaneous secretion of KDEL proteins. We report that inhibition of a single KDELR in a minor fraction of tumor cells, primarily KDELR2, provokes robust infiltration of macrophages and neutrophils into the tumor microenvironment, resulting in regression of both immunogenic and non-immunogenic tum

Phenotypic Plasticity of Tumor-Associated Neutrophils in the Solid Tumor Microenvironment and Emerging Interventions.

Tumor-associated neutrophils (TANs) are a highly dynamic and key component of the tumor microenvironment (TME) in solid tumors and are characterized by an exceptional degree of phenotypic plasticity and functional heterogeneity. TANs play an important role in the control of tumor evolution, metastasis, immune suppression, and therapeutic resistance. The heterogeneity of TANs is reflected in their protumor (TAN2) or antitumor (TAN1) behaviors, which depend on the signals provided by the TME, such as cytokines, metabolism, and epigenetic changes. New developments in single cell RNA sequencing an

Colorectal Cancer Progression and Bone Metastasis: Molecular Mechanisms, Tumor Microenvironment, and Tumor-Bone Crosstalk.

Colorectal cancer (CRC) ranks as the third most common malignancy globally and represents one of the main causes of cancer-related death. This narrative review provides a comprehensive synthesis of recent advances in the molecular mechanisms underlying CRC bone metastasis, with an emphasis on key signaling pathways, tumor microenvironment interactions, and potential therapeutic targets. Bone is a relatively uncommon metastatic site in advanced CRC (1.2-12% of patients), but it frequently coexists with hepatic or pulmonary metastases and triggers skeletal-related events (SREs) such as pathologi

Tumor-Associated Macrophages: Key Players in the Non-Small Cell Lung Cancer Tumor Microenvironment.

BACKGROUND: Lung cancer is among the most common and deadliest malignant tumors worldwide. It is often detected at late stages, resulting in unfavorable outcomes, with tumor cell heterogeneity and medication resistance. Tumor-associated macrophages are among the key cells contributing to cancer progression. They are categorized into two primary phenotypes: Proinflammatory (M1) and anti-inflammatory (M2) which are involved in the onset and progression of NSCLC. The role of common cytokines secreted by macrophages in the progression of lung cancer are described, and the effects of various substa

Metastatic tumor growth in steatotic liver is promoted by HAS2-mediated fibrotic tumor microenvironment.

Steatotic liver enhances liver metastasis of colorectal cancer (CRC), but this process is not fully understood. Steatotic liver induced by a high-fat diet increases cancer-associated fibroblast (CAF) infiltration and collagen and hyaluronic acid (HA) production. We investigated the role of HA synthase 2 (HAS2) in the fibrotic tumor microenvironment in steatotic liver using Has2ΔHSC mice, in which Has2 is deleted from hepatic stellate cells. Has2ΔHSC mice had reduced steatotic liver-associated metastatic tumor growth of MC38 CRC cells, collagen and HA deposition, and CAF and M2 macrophage infil

Tumor-neutrophil cross talk orchestrates the tumor microenvironment to determine the bladder cancer progression.

The immune landscape of bladder cancer progression is not fully understood, and effective therapies are lacking in advanced bladder cancer. Here, we visualized that bladder cancer cells recruited neutrophils by secreting interleukin-8 (IL-8); in turn, neutrophils played dual functions in bladder cancer, including hepatocyte growth factor (HGF) release and CCL3highPD-L1high super-immunosuppressive subset formation. Mechanistically, c-Fos was identified as the mediator of HGF up-regulating IL-8 transcription in bladder cancer cells, which was central to the positive feedback of neutrophil recrui

Tumor Biology and Microenvironment of Vestibular Schwannoma-Relation to Tumor Growth and Hearing Loss.

Vestibular schwannoma is the most common benign neoplasm of the cerebellopontine angle. It arises from Schwann cells of the vestibular nerve. The first symptoms of vestibular schwannoma include hearing loss, tinnitus, and vestibular symptoms. In the event of further growth, cerebellar and brainstem symptoms, along with palsy of the adjacent cranial nerves, may be present. Although hearing impairment is present in 95% of patients diagnosed with vestibular schwannoma, most tumors do not progress in size or have low growth rates. However, the clinical picture has unpredictable dynamics, and there

Ultrasound-Targeted Microbubble Destruction: Modulation in the Tumor Microenvironment and Application in Tumor Immunotherapy.

Tumor immunotherapy has shown strong therapeutic potential for stimulating or reconstructing the immune system to control and kill tumor cells. It is a promising and effective anti-cancer treatment besides surgery, radiotherapy and chemotherapy. Presently, some immunotherapy methods have been approved for clinical application, and numerous others have demonstrated promising in vitro results and have entered clinical trial stages. Although immunotherapy has exhibited encouraging results in various cancer types, however, a large proportion of patients are limited from these benefits due to speci

WNT5A inhibition alters the malignant peripheral nerve sheath tumor microenvironment and enhances tumor growth.

Malignant peripheral nerve sheath tumors (MPNST) are aggressive soft-tissue sarcomas that cause significant mortality in adults with neurofibromatosis type 1. We compared gene expression of growth factors in normal human nerves to MPNST and normal human Schwann cells to MPNST cell lines. We identified WNT5A as the most significantly upregulated ligand-coding gene and verified its protein expression in MPNST cell lines and tumors. In many contexts WNT5A acts as an oncogene. However, inhibiting WNT5A expression using shRNA did not alter MPNST cell proliferation, invasion, migration, or survival

Tumor-draining lymph nodes shape the tumor microenvironment through enrichment of tumor-reactive effector-exhausted CD8+ T cells.

Tumor-draining lymph nodes are central to anti-tumor immunity and influence the response to immunotherapy. Colorectal cancers with microsatellite instability and microsatellite stability differ substantially in tumor biology, prognosis and sensitivity to immune checkpoint blockade. However, the differences in tumor-draining lymph nodes between these two subtypes and their contribution to distinct anti-tumor immune responses remain poorly understood. Here, we performed an integrative multi-omics analysis to systematically compare tumor-draining lymph nodes and tumor microenvironments in these t

Immunostimulation of tumor microenvironment by targeting tumor-associated macrophages with hypoxia-responsive nanocomplex for enhanced anti-tumor therapy.

Tumor-associated macrophages (TAMs), which dampen the therapeutic efficacy of cancer immunotherapy, are the key players in the immunosuppressive tumor microenvironment (TME). Therefore, reprogramming TAMs into tumoricidal M1 macrophages possesses considerable potential as a novel immunotherapy. However, the low bioavailability of polarization agents and limited accumulation of TAMs restrict their anti-tumor efficacy. In this study, we developed a polymer-based hypoxia-responsive nanocomplex to target TAMs in hypoxia for enhanced cancer immunotherapy. We synthesized a hypoxia-cleavable polymer

TIME to Target Sarcomas: Lessons from the Tumor Immune Microenvironment.

The tumor immune microenvironment (TIME) has emerged as a crucial regulator of antitumor immune responses with important therapeutic, prognostic, and predictive implications. Despite the success of immunotherapies in select solid tumors, notably melanoma and non-small cell lung cancer, their impact in soft-tissue sarcomas (STSs) is limited, often attributed to an immune-cold TIME passively driven by low neoantigen burden. Herein, we review the current understanding of the STS TIME, its variability across STS subtypes, and its implications for STS immunotherapy. We propose that the prevailing v

Tumor microenvironment dynamics in gastric cancer pathogenesis and therapeutic resistance.

Gastric cancer remains a significant global health challenge due to its high incidence and mortality, and limited treatment options in advanced stages. Notably, gastric cancer exhibits a complex tumor microenvironment (TME) with substantial cellular and spatial heterogeneity, which profoundly impacts disease pathogenesis and therapeutic resistance. Genetic mutations and chronic inflammation contribute to its development by promoting abnormal cell proliferation and creating an immunosuppressive TME. The TME comprises various cellular and acellular components—including tumor-infiltrating lymphoc

Tumor-associated CD19+ macrophages induce immunosuppressive microenvironment in hepatocellular carcinoma.

Tumor-associated macrophages are a key component that contributes to the immunosuppressive microenvironment in human cancers. However, therapeutic targeting of macrophages has been a challenge in clinic due to the limited understanding of their heterogeneous subpopulations and distinct functions. Here, we identify a clinically relevant CD19+ subpopulation of macrophages that is enriched in many types of cancer, particularly in hepatocellular carcinoma (HCC). The CD19+ macrophages exhibit increased levels of programmed cell death 1 ligand 1 (PD-L1) and CD73, enhanced mitochondrial oxidation, an

Tumor Microenvironment: Insights from Multiparametric MRI in Pancreatic Ductal Adenocarcinoma.

Background/Objectives: The tumor microenvironment (TME) of pancreatic ductal adenocarcinoma (PDAC) is characterized by an enriched stroma, hampering the effectiveness of therapy. This co-clinical study aimed to (1) provide insight into early post-treatment changes in the TME using multiparametric magnetic resonance imaging (mpMRI)-derived quantitative imaging biomarkers (QIBs) in a preclinical PDAC model treated with radiotherapy and correlate these QIBs with histology; (2) evaluate the feasibility of obtaining these QIBs in patients with PDAC using clinically approved mpMRI data acquisitions.
