The role of Ca2+ in triggering inositol 1,4,5-trisphosphate receptor ubiquitination.

The IP3R (inositol 1,4,5-trisphosphate receptor) forms tetrameric Ca2+ channels in ER (endoplasmic reticulum) membranes, where channel activity is largely under the control of the co-agonists IP3 and Ca2+. In cells stimulated using extracellular ligands that persistently elevate phosphoinositidase C activity, IP3Rs are rapidly ubiquitinated and then degraded by the proteasome through as yet undefined mechanisms. Whereas binding of IP3 has been suggested to be a key event in the triggering of IP3R ubiquitination the role of Ca2+ in this process remains unknown. In the present study we use alpha

S-nitrosylation of parkin regulates ubiquitination and compromises parkin's protective function.

Parkin is an E3 ubiquitin ligase involved in the ubiquitination of proteins that are important in the survival of dopamine neurons in Parkinson's disease (PD). We show that parkin is S-nitrosylated in vitro, as well as in vivo in a mouse model of PD and in brains of patients with PD and diffuse Lewy body disease. Moreover, S-nitrosylation inhibits parkin's ubiquitin E3 ligase activity and its protective function. The inhibition of parkin's ubiquitin E3 ligase activity by S-nitrosylation could contribute to the degenerative process in these disorders by impairing the ubiquitination of parkin su

RNF34 restrains endometriosis progression by promoting PKM2 ubiquitination and suppressing glycolysis.

Metabolic reprogramming, particularly enhanced glycolysis, is an important feature of endometriosis, yet the upstream posttranslational mechanisms governing this metabolic phenotype remain poorly understood. Transcriptomic analysis and clinical tissue validation revealed reduced RNF34 expression in endometriotic tissues and an inverse association between RNF34 and pyruvate kinase M2 (PKM2). Mass spectrometry analysis revealed that PKM2 is an RNF34-interacting protein, and biochemical assays further demonstrated this direct association. Mechanistically, RNF34 promoted PKM2 ubiquitination and re

Mammalian DNA maintenance methylation depends on histone but not PAF15 ubiquitination.

In mammals, DNA methylation is an essential inheritable epigenetic modification maintained by the DNMT1-UHRF1 machinery in a UHRF1 E3 ubiquitin ligase-activity-dependent manner. UHRF1-catalyzed histone ubiquitination and PAF15 ubiquitination have been proposed to recruit DNMT1 to hemi-methylated CpGs. Here, we confirm that both PAF15 and histone H3 are robustly ubiquitinated by UHRF1 in a replication-dependent manner during S phase. However, loss of PAF15 or its ubiquitination, or overexpression of ubiquitination-deficient PAF15, does not alter DNMT1 recruitment to replicating DNA or global DN

Ubiquitination regulation in coronary microcirculation dysfunction: focus on the endothelial cells.

Basic and clinical research on coronary microvascular disease (CMD) has spanned half a century. As a pathogenesis independent of epicardial coronary stenosis, CMD has become a major hotspot in cardiovascular medicine. Ubiquitination, a vital post-translational modification in eukaryotic cells, precisely regulates target proteins via the E1-E2-E3 enzymatic cascade and plays a critical role in cardiovascular homeostasis and pathophysiology. Recent evidence indicates that the ubiquitin-proteasome system (UPS) is deeply involved in the development of CMD by modulating nitric oxide production, barr

RING E3 ligase TRIM8 is structurally selective for UBE2N-mediated ubiquitination.

RING domains catalyse ubiquitination by stabilising a closed conformation of E2-ubiquitin (Ub) conjugates, with TRIpartite Motif (TRIM) proteins representing the largest subfamily of RING-type E3 ligases. TRIM proteins are implicated in many physiological processes and diseases. For example, TRIM8 perturbation has been variously linked to neurodevelopmental defects and several cancer types, attributed to its ubiquitination of a myriad of putative substrates. However, the molecular mechanisms behind these modifications are not yet clear. Here we show that TRIM8 has preferential functionality wi

Ubiquitination of LMP-1 by UFD-2 regulates apoptotic cell clearance.

Apoptotic cell clearance, or efferocytosis, is essential for tissue homeostasis and preventing inflammation in multicellular organisms. Through a genome-wide RNAi screen targeting 169 E3 ubiquitin ligases in Caenorhabditis elegans, we identified six ligases involved in efferocytosis regulation, with UFD-2 deletion showing the most significant defect. UFD-2 was found to be critical for phagosome maturation and degradation during apoptotic cell clearance. We identified the lysosomal membrane protein LMP-1 as a direct substrate of UFD-2 and showed that UFD-2 cooperates with UBC-15 to catalyze K63

A20-mediated KEAP1 ubiquitination orchestrates hepatocyte ferroptosis to ameliorate autoimmune hepatitis.

BACKGROUND: Autoimmune hepatitis (AIH) is characterised by death of hepatocytes and chronic inflammation. Patients with deleterious variants in A20 are identified with AIH presentations and immune activation. However, it remains unclear how A20 dysregulation contributes to AIH pathogenesis. OBJECTIVE: This study elucidates the role and mechanism of A20 in AIH progression and its therapeutic potential. DESIGN: A20 expression was analysed in patient sample and AIH mouse model. A20 function was explored in mice with hepatocyte-specific A20 knockout and overactivation and wild type controls. Liv

HERC4-mediated ubiquitination licenses RIPK1 to initiate TNF-induced cell death.

TNF can activate both prosurvival and prodeath signaling downstream of tumor necrosis factor receptor 1 (TNFR1). Survival signaling originates from TNFR1-containing membrane-bound complex I, while death signaling is driven by cytosolic complex II. Receptor-interacting protein kinase 1 (RIPK1) is a central component of both complexes but the molecular switch converting RIPK1 from a prosurvival scaffold in complex I to a prodeath kinase in complex II has remained elusive. Here, we identify the E3 ligase HERC4 as the molecular determinant of prodeath signaling. We show that HERC4 binds complex I-

CHK1 activates mitophagy to attenuate cardiac aging via inhibiting AHSA1-ubiquitination.

With the acceleration of global population aging, the progressive deterioration of cardiac structure and function has become a critical determinant of cardiovascular health, presenting a significant public health challenge. Checkpoint kinase 1 (CHK1), a key cell cycle checkpoint protein, plays an essential role in various biological processes by mediating signaling cascades. While CHK1 has been shown to be important for heart regeneration, its role in the aging process of the heart remains unclear. In this study, we investigated the alterations in CHK1 expression in aging hearts and elucidated

WDR63 Promotes Pulmonary Fibrosis Through Facilitating K63-Linked Ubiquitination of p53.

BACKGROUND AND OBJECTIVE: Idiopathic pulmonary fibrosis (IPF) is a chronic and progressive interstitial lung disease driven by dysregulated alveolar epithelial cells (AECs) and fibroblasts. While aberrant p53 signalling contributes to IPF pathogenesis, the precise mechanism of p53 activation is not well understood. WDR63, a WD40-repeat protein implicated in lung cancer and male infertility, may be functionally relevant in IPF. METHODS: The mouse model of bleomycin-induced pulmonary fibrosis was used to investigate the role of WDR63 in IPF. Immunofluorescent staining and Western blotting assay

Ubiquitination in intervertebral disc degeneration: from mechanisms to potential therapeutic strategies.

Intervertebral disc degeneration (IDD) is the primary cause of low back pain, imposing a heavy economic burden on individuals and society. The pathogenesis of IDD involves complex pathological processes. The interaction of extracellular matrix metabolism, regulated cell death (such as apoptosis, ferroptosis, pyroptosis, and autophagy), cell senescence, oxidative stress, and inflammatory response collectively contribute to the development of IDD. As a widespread post-translational modification, ubiquitination modulates various biological processes by regulating the activity and stability of pro

PAQR3 Promotes Diabetic Tubulointerstitial Fibrosis by suppressing STUB1-mediated NOX4 ubiquitination.

Diabetic kidney disease (DKD) is a leading cause of chronic kidney disease, and tubulointerstitial fibrosis (TIF) represents its final pathological hallmark. Progestin and adipoQ receptor 3 (PAQR3) has been implicated in metabolism and inflammation, but its role in DKD remains largely unexplored. Here, we report that PAQR3 expression is markedly upregulated in the kidneys of STZ-induced diabetic mice and in high glucose-stimulated NRK-52E tubular epithelial cells, correlating with the onset of tubular epithelial-mesenchymal transition (EMT) and fibrosis. Functional studies demonstrated that PA

FBXO42 promotes hepatocellular carcinoma progression via mediating p57Kip2 ubiquitination and degradation.

BACKGROUND: Hepatocellular carcinoma (HCC), one of the most common malignancies, accounts for about 80% of all primary liver cancers. FBXO42 belongs to the F-box protein family and functions as a key component of the SCF (Skp1-Cullin-F-box) ubiquitin ligase complex, which is involved in the process of protein ubiquitination. The studies on the role of FBXO42 in different diseases are inadequate, especially in HCC. METHODS: The expression and the clinical features of FBXO42 in HCC were assessed using the HCCDB, TCGA, and ICGC databases. cBioPortal and SangerBox databases were used to analyze t

Conformation-specific Antibody Deciphers K27-linked Ubiquitination in Chaperone-Mediated Proteostasis.

UNLABELLED: Lysine 27 (K27)-linked polyubiquitination plays critical yet incompletely defined roles in proteostasis, innate immunity, and disease progression; however, investigations into this process have long been hindered by its extremely low abundance and the lack of conformation-specific enrichment tools. Herein, we describe the development of a long-sought conformation-specific antibody, K27-IgG, which can selectively recognize-among all ubiquitin chain types-the unique architecture of K27-linked polyubiquitin (K27-polyUb) characterized by a distinct buried K27-isopeptide bond, with high

Ubiquitination of PFKFB4 by CHIP regulates glycolysis and progression in endometriosis†.

Endometriosis is a common estrogen-dependent gynecological disorder characterized by chronic inflammatory responses and frequently associated with clinical infertility. Recent studies have demonstrated that the pathogenesis of this disease is closely linked to dysregulated post-translational modifications. This study focuses on the regulatory role of the ubiquitin-proteasome system (UPS) in endometriosis and, for the first time, reveals the molecular mechanism by which the E3 ubiquitin ligase STIP1 homology and U-box containing protein 1 (CHIP) specifically binds to and ubiquitinates the glyco
