Unilateral renal agenesia associated with partial epididymis and vas deferens agenesia in a patient with abdominal testicle.

This study considers a unilateral renal agenesia associated with agenesia of the epididymis body and tail and the vas deferens and non-palpable left testicle in a 20-month-year-old patient. During laparoscopic procedure, the testicle was positioned at approximately 5 cm above the inguinal ring. The size was appropriate for the age and the head of the epididymis was situated in its normal position. The decision was made to perform the first step of the Fowler-Stephens' surgery and the patient presented a good evolution. The association of male duct system agenesia with unilateral renal agenesia

Cystic fibrosis phenotype evaluation and paternity outcome in 50 males with congenital bilateral absence of vas deferens.

BACKGROUND: Most infertile males with congenital bilateral absence of vas deferens (CBAVD) carry mutations on the cystic fibrosis transmembrane conductance regulator gene and may express mild cystic fibrosis (CF) symptoms. Barriers to paternity for these men can now be overcome by assisted reproduction. Our aims were to investigate the CF-related phenotype and clinical outcome for 50 patients with CBAVD seen at a CF adult centre between 1992 and 1999. METHODS AND RESULTS: The investigation of the patients included screening for 22 CF mutations and identification of the poly-T variant of intro

Unilateral renal agenesis associated with congenital bilateral absence of the vas deferens: phenotypic findings and genetic considerations.

An association between congenital bilateral absence of the vas deferens (CBAVD), normal renal anatomy and cystic fibrosis (CF) gene mutations is well established (CF/CBAVD). We postulate that unilateral renal agenesis (URA) and CBAVD (URA/CBAVD) may have a non-CF mutation-mediated genetic basis that leads to abnormal development of the entire mesonephric duct at a very early stage in embryo development (< or =7 weeks). The physical, laboratory and radiographic findings of men with URA/CBAVD (n = 17) and CF/CBAVD (n = 97) were compared; the fertilization and pregnancy rates in the URA/CBAVD pop

[Male infertility caused by bilateral agenesis of the vas deferens: a new clinical form of cystic fibrosis?].

Congenital bilateral absence of vas deferens causes male excretory infertility and represents 1 to 2% of male infertility. Because of a genotypic similarity with cystic fibrosis, the possible in vitro fertilization with epididymal sperm requires careful genetic counselling. We studied genotype, sweat chloride concentration, respiratory function tests, sinus abnormalities, pancreatic and hepatic functions in 22 subjects with congenital bilateral absence of vas deferens. Among them, four were compound heterozygotus, all of them with the R117H mutation. Ten had a positive sweat test, one of them

Involvement of calcium and cyclic AMP in the K+-induced rhythmic contraction of isolated rat vas deferens.

Mechanisms involved in the K+-induced rhythmic contraction were investigated in the isolated rat vas deferens. Isolated vas deferens is mechanically quiescent in normal Krebs-bicarbonate solution, but addition of KCl (10 to 35 mM) to the solution induces a rhythmic contraction. The K+-induced rhythmic contraction was not inhibited by tetrodotoxin, prazosin and indomethacin as well as by in vivo pretreatment with reserpine, but was abolished by verapamil or by Ca++ removal from the solution. TMB-8, a calcium release blocker, blocked the K+-induced rhythmic contraction and, instead, sustained to

Morphiceptin ( beta-casomorphin (1-4) amide): a peptide opioid antagonist in the field stimulated rat vas deferens.

The effect of Morphiceptin (H-Tyr-Pro-Phe-Pro-NH2) on opioid inhibition of field stimulated rat vas deferens (RVD) was studied. The morphiceptin was found to produce a weak inhibition of the twitch response of the RVD. In addition morphiceptin antagonized full agonists acting on the same receptor system (i.e. beta-endorphin selective antagonist for the delta receptor system of RVD by virtue of being a weak partial agonist.

Comparison of effects of low temperature and metabolic inhibition on high-K induced contracture of vas deferens.

The effects of temperature on high-K induced contracture of guinea-pig vas deferens were studied and compared with those of metabolic inhibitions. The rate of rise and fall of the phasic contraction decreased with the decrease in temperature. At 5 degrees C, the time to peak tension and half relaxation time increased to 25 times and to 38 times of those observed at 35 degrees C, respectively. The height of the phasic contraction increased by lowering of the temperature. The height of the tonic contraction also increased by lowering the temperature down to 15 degrees C but then decreased. Docosahexaenoic Acid Targets TRPC3/6 Channels to Suppress Noradrenaline-Induced Contraction in Rat Vas Deferens Smooth Muscle.

We investigated the effects of docosahexaenoic acid (DHA) on noradrenaline (NA)-induced contractile responses in rat vas deferens smooth muscle (VDSM). DHA (3 × 10-6-3 × 10-5 M) significantly inhibited phasic contractions induced by NA (3 × 10-5 M) in a concentration-dependent manner. In contrast, under Ca2+-free conditions, DHA (3 × 10-5 M) slightly enhanced NA-induced contractile responses. DHA did not inhibit NA-induced contractile responses in the prostate, which are mediated by α1A-adrenoceptor stimulation, similar to those in VDSM. Although verapamil (10-5 M), an L-type Ca2+ channel inhi

Compound heterozygous mutations in CFTR causing congenital bilateral absence of the vas deferens in a Chinese pedigree.

BACKGROUND: Cystic fibrosis (CF) is an autosomal recessive disorder rarely found in Asian populations. Most males with CF are infertile because of obstructive azoospermia (OA) caused by congenital bilateral absence of the vas deferens (CBAVD). Compound heterozygous mutations of cystic fibrosis transmembrane conductance regulator (CFTR) are among the most common pathogenic factors in CBAVD. However, few genealogical analyses have been performed. METHODS: In this study, whole-exome sequencing and cosegregation analysis were performed in a Chinese pedigree involving two siblings with CBAVD. More

The relaxation effect of endocannabinoid anandamide on isolated rat bladder and vas deferens tissues and possible mechanisms.

BACKGROUND: The endocannabinoid system plays important roles in various systems, including the genitourinary system; however, its mechanism of action is not fully understood. OBJECTIVES: This study aimed to investigate the direct relaxant effects of anandamide and its possible mechanisms in isolated rat bladder and vas deferens tissues. METHODS: Twenty-one adult male Wistar albino rats were used. Bladder and vas deferens (prostatic and epididymal portions) tissues were mounted in 10 mL of organ baths. Relaxation responses to anandamide were recorded at 3 and 10 μM concentrations. After the r

Genetic analysis and intracytoplasmic sperm injection outcomes of Chinese patients with congenital bilateral absence of vas deferens.

PURPOSE: Congenital bilateral absence of the vas deferens (CBAVD) is a major cause of obstructive azoospermia and male factor infertility. CBAVD is mainly caused by mutations in the genes encoding CFTR (cystic fibrosis transmembrane conductance regulator) and ADGRG2 (adhesion G protein-coupled receptor G2). This study aimed to describe CFTR and ADGRG2 variations in 46 Chinese CBAVD patients and evaluated sperm retrieval and assisted reproductive technology outcomes. METHODS: The CFTR and ADGRG2 genes were sequenced and analyzed by whole-exome sequencing (WES), and variations were identified b

Undernutrition affects cell survival, oxidative stress, Ca2+ handling and signaling pathways in vas deferens, crippling reproductive capacity.

BACKGROUND: The aim of this work was to investigate the mechanisms by which chronic malnutrition (CM) affects vas deferens function, leading to compromised reproductive capacity. Previous studies have shown that maternal malnutrition affects the reproductive tracts of adult male offspring. However, little is known about the effects of CM, a widespread life-long condition that persists from conception throughout growth to adult life. METHODOLOGY/PRINCIPAL FINDINGS: Young adult male rats, which were chronically malnourished from weaning, presented decreased total and haploid cells in the vas de

Vas deferens smooth muscle responses to the nitric oxide-independent soluble guanylate cyclase stimulator BAY 41-2272.

The nitric oxide-cGMP signaling pathway modulates the ejaculatory functions. The nitric oxide (NO)-independent soluble guanylate cyclase haem-dependent stimulator BAY 41-2272 potently relaxes different types of smooth muscles. However, no study investigated its effects in vas deferens smooth muscle. Therefore, we designed experiments to evaluate the in vitro relaxing responses of vas deferens to BAY 41-2272. The effects of prolonged oral intake with BAY 41-2272 in vas deferens contractions of rats treated chronically with the NO synthase inhibitor N(ω)-nitro-L-arginine methyl ester (L-NAME) we

Cystic fibrosis and infertility caused by congenital bilateral absence of the vas deferens and related clinical entities.

The condition of congenital bilateral absence of the vas deferens (CBAVD) is, in the majority of patients, related to defects in the cystic fibrosis transmembrane conductance regulator (CFTR) gene. CBAVD patients either are compound heterozygotes (carrying different mutations in their two CFTR genes) or carry a mutation in one of their CFTR genes and an intron 8 5T splice variant, associated with low levels of functional CFTR protein, in their second gene. The relationship between cystic fibrosis (CF) and CBAVD requires a proper clinical examination of the patient, a CFTR mutation analysis for

Do H3-receptors participate in the effects of histamine on electrically-evoked contractions of rat vas deferens?

1. The specific effects of histamine were studied on rat vas deferens contractions induced by electrical field stimulation (0.1 Hz, 1 pulse, 1 msec duration, supramaximal voltage). 2. Histamine (5 x 10(-7)-10(-4) M) inhibited the electrically-evoked contractions (EEC). This effect was unchanged after the H1-receptor antagonist diphenhydramine (5 x 10(-6) M) and was decreased after the H2-receptor antagonist cimetidine (10(-5) M). 3. Betahistine, described as H1-agonist (in guinea pig ileum) and H3-autoreceptor antagonist (in rat cerebral cortex) slightly decreased the EEC, but competitively in
