Recent advances in venom pharmacology reshaping venom-to-drug discovery.

Animal venoms represent rich sources of pharmacologically active molecules, yet their translation into clinical therapeutics has historically progressed slowly. Recent advances in AI-driven venomics, cryo-electron microscopy, and computational peptide engineering are helping to overcome long-standing barriers in venom-based drug discovery and accelerate therapeutic translation.

Constructing comprehensive venom proteome reference maps for integrative venomics.

OBJECTIVE: Understanding the molecular basis of complex adaptive traits, such as snake venom, demands qualitative and quantitative comparisons of the temporal and spatial patterns of venom variation. Here, we assessed the proof-of-concept that locus-resolved reference venom proteome maps can be achieved through efficient pre-MS venom proteome decomplexation, peptide-centric MS/MS analysis and species-specific database searching. METHODS: Venom proteome components were fractionated and quantified by RP-HPLC, SDS-PAGE and 2DE prior to LC-MS/MS matching against a species-specific transcriptomic

Venomic and pharmacological activity of Acanthoscurria paulensis (Theraphosidae) spider venom.

In the present study we conducted proteomic and pharmacological characterizations of the venom extracted from the Brazilian tarantula Acanthoscurria paulensis, and evaluated the cardiotoxicity of its two main fractions. The molecular masses of the venom components were identified by mass spectrometry (MALDI-TOF-MS) after chromatographic separation (HPLC). The lethal dose (LD(50)) was determined in mice. Nociceptive behavior was evaluated by intradermal injection in mice and the edematogenic activity by the rat hind-paw assay. Cardiotoxic activity was evaluated on in situ frog heart and on isol

Understanding and utilising mammalian venom via a platypus venom transcriptome.

Only five mammalian species are known to be venomous, and while a large amount of research has been carried out on reptile venom, mammalian venom has been poorly studied to date. Here we describe the status of current research into the venom of the platypus, a semi-aquatic egg-laying Australian mammal, and discuss our approach to platypus venom transcriptomics. We propose that such construction and analysis of mammalian venom transcriptomes from small samples of venom gland, in tandem with proteomics studies, will allow the identification of the full range of mammalian venom components. Tityus serrulatus venom peptidomics: assessing venom peptide diversity.

MALDI-TOF-TOF and de novo sequencing were employed to assess the Tityus serrulatus venom peptide diversity. Previous works has shown the cornucopia of molecular masses, ranging from 800 to 3000Da, present in the venom from this and other scorpions species. This work reports the identification/sequencing of several of these peptides. The majority of the peptides found were fragments of larger venom toxins. For instance, 28 peptides could be identified as fragments from Pape proteins, 10 peptides corresponded to N-terminal fragments of the TsK beta (scorpine-like) toxin and fragments of potassiu

Soluble copolymer of wasp venom with human albumin for venom immunotherapy.

Polymerization of allergens decreases allergenicity while retaining immunogenicity, as we have demonstrated for ragweed, grass, and tree pollens. We have also polymerized bee venom with human albumin to form soluble, high-molecular-weight copolymers that are immunogenic in rabbits. We now have prepared a soluble wasp venom-albumin polymer (WVAP), molecular weight greater than or equal to 240,000 daltons, by glutaraldehyde treatment and Sephacryl S-300 column fractionation. Rabbits immunized with WVAP produced IgG to both WVAP and wasp venom (WV), as measured by ELISA. IgG against WVAP was tota

Venom gland size and venom complexity-essential trophic adaptations of venomous predators: A case study using spiders.

Specialized predators possess a variety of adaptations. In venomous predators, this may include the size of the venom gland and venom composition. It is expected that due to different foraging strategies, predators with a wide trophic niche (generalists) should possess larger venom glands that contain more diversified components than predators with a narrow niche (specialists). We focused on spiders, as the most diversified group of venomous predators, in which a wide variety of trophic strategies have evolved. We conducted a comparative analysis using 40 spider species, in which we measured t

A multi-species lipidomic analysis of viperid snake venoms from the Americas.

Snake venoms are multilayered and chemically diverse mixtures with important ecological and medical relevance, particularly in many regions of the Americas. Although venom research has largely focused on proteinaceous toxins, venoms also contain low-molecular-weight components that remain poorly characterized. Among these, the lipidome represents one of the least explored layers of venom composition, and comparative information across species is limited. In this study, we applied an untargeted high-resolution mass spectrometry-based lipidomics approach to characterize and compare lipid signatu

Pharmacological characterization and nephroprotective potential of venoms from three egyptian scorpion species.

INTRODUCTION: Scorpion venoms are complex natural mixtures rich in bioactive molecules with emerging pharmacological relevance. Drug-induced kidney injury, particularly from chemotherapy agents like Taxol® (paclitaxel), remains a significant clinical challenge. This study investigates the molecular composition, enzymatic activities, and potential nephroprotective effects of venoms from three Egyptian scorpion species (Leiurus quinquestriatus, Buthacus leptochelys, and Scorpio maurus palmatus) against Taxol®-induced kidney injury. METHODS: Comprehensive molecular mass profiling of the three ve

Past, Present, and Future of Naturally Occurring Antimicrobials Related to Snake Venoms.

This review focuses on proteins and peptides with antimicrobial activity because these biopolymers can be useful in the fight against infectious diseases and to overcome the critical problem of microbial resistance to antibiotics. In fact, snakes show the highest diversification among reptiles, surviving in various environments; their innate immunity is similar to mammals and the response of their plasma to bacteria and fungi has been explored mainly in ecological studies. Snake venoms are a rich source of components that have a variety of biological functions. Among them are proteins like lec

Biologically Active Peptides from Venoms: Applications in Antibiotic Resistance, Cancer, and Beyond.

Peptides are potential therapeutic alternatives against global diseases, such as antimicrobial-resistant infections and cancer. Venoms are a rich source of bioactive peptides that have evolved over time to act on specific targets of the prey. Peptides are one of the main components responsible for the biological activity and toxicity of venoms. South American organisms such as scorpions, snakes, and spiders are important producers of a myriad of peptides with different biological activities. In this review, we report the main venom-derived peptide families produced from South American organism

Hyaluronidase-like enzymes are a frequent component of venoms from theraphosid spiders.

Theraphosid spider venoms are extremely complex mixtures, composed mainly by low molecular compounds, peptides, and enzymes. The large size of these spiders and their ability to breed in captivity permits access to rather large amounts of venom and an easier venom extraction. In the present study, we conducted a comparative investigation about the content of hyaluronidase-like enzymes in the venoms from several theraphosid spiders, with a special focus on the Poecilotheria species, which are considered as underestimated theraphosids of medical importance. The following species were analyzed: P

Enter the Dragon: The Dynamic and Multifunctional Evolution of Anguimorpha Lizard Venoms.

While snake venoms have been the subject of intense study, comparatively little work has been done on lizard venoms. In this study, we have examined the structural and functional diversification of anguimorph lizard venoms and associated toxins, and related these results to dentition and predatory ecology. Venom composition was shown to be highly variable across the 20 species of Heloderma, Lanthanotus, and Varanus included in our study. While kallikrein enzymes were ubiquitous, they were also a particularly multifunctional toxin type, with differential activities on enzyme substrates and also

Disintegrins from snake venoms and their applications in cancer research and therapy.

Integrins regulate diverse functions in cancer pathology and in tumor cell development and contribute to important processes such as cell shape, survival, proliferation, transcription, angiogenesis, migration, and invasion. A number of snake venom proteins have the ability to interact with integrins. Among these are the disintegrins, a family of small, non-enzymatic, and cysteine-rich proteins found in the venom of numerous snake families. The venom proteins may have a potential role in terms of novel therapeutic leads for cancer treatment. Disintegrin can target specific integrins and as such

Snake venoms: A brief treatise on etymology, origins of terminology, and definitions.

The ancient perceptions of "venomous" and "poisonous snakes", as well as the Indo-European (IE) etymological origins of the term "venom" specifically associated with snakes are considered. Although several ancient cultures perceived snakes as symbols of fecundity and renewal, concurrent beliefs also associated venomous snakes with undesirable human characteristics or as portending non-propitious events. The respective IE roots of the terms "venom" and "poison", "wen" and "poi" refer to desire or the act of ingesting liquids. The origin of the term, "venom", is associated with polytheistic cult

Sensitization to Hymenoptera venoms is common, but systemic sting reactions are rare.

BACKGROUND: Sensitization to Hymenoptera venom without systemic sting reactions (SSRs) is commonly observed in the general population. Clinical relevance for a future sting has not yet been investigated. OBJECTIVE: We aimed to evaluate the effect of these debatable sensitizations with deliberate sting challenges and to monitor serologic changes for up to 2 years. METHODS: One hundred thirty-one challenges with bees and wasps were performed in 94 subjects with a hitherto irrelevant sensitization. The clinical outcome was recorded, and results of specific IgE (sIgE) determinations, skin tests,

SjAPI, the first functionally characterized Ascaris-type protease inhibitor from animal venoms.

BACKGROUND: Serine protease inhibitors act as modulators of serine proteases, playing important roles in protecting animal toxin peptides from degradation. However, all known serine protease inhibitors discovered thus far from animal venom belong to the Kunitz-type subfamily, and whether there are other novel types of protease inhibitors in animal venom remains unclear. PRINCIPAL FINDINGS: Here, by screening scorpion venom gland cDNA libraries, we identified the first Ascaris-type animal toxin family, which contains four members: Scorpiops jendeki Ascaris-type protease inhibitor (SjAPI), Scor
