Liquid crystalline nanoparticles for co-delivery of gentamicin and cinnamaldehyde: enhancing antibiofilm efficacy against Pseudomonas aeruginosa via quorum sensing inhibition.

Biofilm-associated infections present a major therapeutic challenge due to reduced antibiotic susceptibility, altered bacterial physiology, and protection by the extracellular polymeric substance (EPS) matrix. In this study, liquid crystalline nanoparticles (LCNPs) were developed as a co-delivery platform for gentamicin (GEN) and the biofilm-modulating quorum sensing inhibitor cinnamaldehyde (CIN) to enhance antibiofilm activity against Pseudomonas aeruginosa PAO1. LCNPs were successfully fabricated using a hydrotrope dilution method, producing nanoparticles with mean diameters of approximatel

Biofilm sensitization to break Staphylococcus aureus tolerance to cold atmospheric plasma therapy.

Introduction. Biofilm-associated infections present a major therapeutic challenge due to their intrinsic tolerance to conventional antimicrobials. Cold atmospheric plasma (CAP) has shown promise as a non-antibiotic antimicrobial modality; however, some bacteria including Staphylococcus aureus can exhibit tolerance to plasma exposure.Gap Statement. Strategies that sensitize CAP-tolerant biofilms to plasma treatment may improve CAP efficacy, but suitable adjunctive compounds and mechanisms remain poorly defined.Aim. This study aimed to determine whether repurposed bioactive compounds could enhan

A step forward on the in vitro and in vivo validation of rifabutin-loaded liposomes for the management of highly virulent MRSA infections.

Staphylococcus aureus (S. aureus) infections, especially methicillin resistant (MRSA), constitute an alarming public health issue due to its association with high mortality, morbidity, and hospitalization costs. The increasing antibiotic resistance and biofilm-associated infections of MRSA prompted the discovery of novel and more effective therapeutic strategies. Our team has been working on alternative therapies against S. aureus infections. For this, we have been repurposing an existing antibacterial drug, rifabutin (RFB), through its association to a nanotechnological platform, liposomes, a

Marine resistome of a temperate zone: Distribution, diversity, and driving factors across the trophic gradient.

Marine and ocean environments are the most widespread habitats in the world but are still the least studied from the aspect of antibiotic resistance. The indigenous and tetracycline (TET)- and sulfamethoxazole (SXT)-resistant planktonic bacterial communities were simultaneously investigated for the first time along a trophic gradient of a temperate zone, regarding their taxonomic and functional structures as well as biotic and abiotic factors affecting their dynamics as vehicles of antibiotic resistance genes (ARGs), thus impacting the ARGs distribution at seasonal and spatial scales. The Membrane Activity of the Amphibian Temporin B Peptide Analog TB_KKG6K Sheds Light on the Mechanism That Kills Candida albicans.

Temporin B (TB) is a 13-amino-acid-long, cationic peptide secreted by the granular glands of the European frog Rana temporaria. We recently showed that the modified TB peptide analog TB_KKG6K rapidly killed planktonic and sessile Candida albicans at low micromolar concentrations and was neither hemolytic nor cytotoxic to mammalian cells in vitro. The present study aimed to shed light into its mechanism of action, with a focus on its fungal cell membrane activity. We utilized different fluorescent dyes to prove that it rapidly induces membrane depolarization and permeabilization. Studies on mod

Salmonella enterica Serovar Typhimurium Exploits Cycling through Epithelial Cells To Colonize Human and Murine Enteroids.

Enterobacterial pathogens infect the gut by a multistep process, resulting in colonization of both the lumen and the mucosal epithelium. Due to experimental constraints, it remains challenging to address how luminal and epithelium-lodged pathogen populations cross-feed each other in vivo Enteroids are cultured three-dimensional miniature intestinal organs with a single layer of primary intestinal epithelial cells (IECs) surrounding a central lumen. They offer new opportunities to study enterobacterial infection under near-physiological conditions, at a temporal and spatial resolution not attai

In vitro testing of a first-in-class tri-alkylnorspermidine-biaryl antibiotic in an anti-biofilm silicone coating.

Biofilm-related infection is among the worst complication to prosthetic joint replacement procedures; once established on the implant surface, biofilms show strong recalcitrance to clinical antibiotic therapy, frequently requiring costly revision procedures and prolonged systemic antibiotics for their removal. A well-designed active release coating might assist host immunity in clearing bacterial contaminants within the narrow perioperative window and ultimately prevent microbial colonization of the joint prosthesis. A first-in-class compound (CZ-01127) was tested as the active release agent i

Quinine Improves the Fungicidal Effects of Antimicrobial Blue Light: Implications for the Treatment of Cutaneous Candidiasis.

BACKGROUND AND OBJECTIVE: Candida albicans is an opportunistic fungal pathogen of clinical importance and is the primary cause of fungal-associated wound infections, sepsis, or pneumonia in immunocompromised individuals. With the rise in antimicrobial resistance, it is becoming increasingly difficult to successfully treat fungal infections using traditional antifungals, signifying that alternative non-traditional approaches must be explored for their efficacy. STUDY DESIGN/MATERIALS AND METHODS: We investigated the combination of antimicrobial blue light (aBL) and quinine hydrochloride (Q-HCL

A Case-Control Study to Determine the Prognostic Features of Salivary Epithelial Cells in Periodontitis.

Periodontitis-a biofilm-induced immunoinflammatory pathology-often progresses gradually, exhibiting periodic bursts and resolution. Exfoliating oral epithelial cells act as reservoirs for key periodontal pathogens, facilitating reinfection or infection of new sites. Since saliva is a rich source of oral epithelial cells, we hypothesized that the microbial and functional profile of salivary epithelial cells (SECs) will reflect the in situ host response and disease severity. We used a case-control study design. Unstimulated whole saliva was collected from 20 chronic periodontitis patients and 20

A review of the population biology and host-parasite interactions of the sea louse Lepeophtheirus salmonis (Copepoda: Caligidae).

Lepeophtheirus salmonis is a specific parasite of salmonids that occurs in the Atlantic and Pacific Oceans. When infestations are heavy fish mortality can occur although the factors that are responsible for causing epizootics, especially in wild salmonid populations are still largely unknown. Over the past 20 years this parasite has caused significant economic losses in farmed salmon production and possibly in wild salmonid populations locally. Understanding the connectivity between populations is crucial to an understanding of the epidemiology of infections and for management of infections in

Bacteria associated with deep, alkaline, anaerobic groundwaters in Southeast Washington.

The microbial diversity in two deep, confined aquifers, the Grande Ronde (1270 m) and the Priest Rapids (316 m), Hanford Reservation, Washington, USA, was investigated by sampling from artesian wells. These basaltic aquifers were alkaline (pH 8.5 to 10.5) and anaerobic (Eh -200 to -450 mV). The wells were allowed to free-flow until pH and Eh stabilized, then the microflora was sampled with water filtration and flow-through sandtrap methods. Direct microscopic counts showed 7.6 × 10(5) and 3.6 × 10(3) bacteria ml(-1) in water from the Grande Ronde and Priest Rapids aquifers, respectively. The s

Zinc-containing ionic liquid combined with carboxymethyl chitosan for vital pulp therapy in mature permanent teeth with carious pulp exposure.

BACKGROUND: The elimination of pulpal infection is critical for the success of vital pulp therapy (VPT) in mature permanent teeth with carious pulp exposure. However, commonly used pulp capping materials often possess inadequate antibacterial properties, which limits treatment success. This study aimed to develop a zinc-containing ionic liquid (IL-Zn)/carboxymethyl chitosan (CMCS) composite and evaluate its potential as an adjunctive pulp capping material for VPT. METHODS: IL-Zn was characterized by energy dispersive spectroscopy (EDS), Fourier transform infrared spectroscopy (FTIR), proton n

Preliminary species-dependent growth responses of selected oral bacteria to e-cigarette Liquids and aerosol extracts: a single-species in vitro study.

BACKGROUND: The increased use of electronic cigarettes (e-cigarettes) has raised concern regarding their potential impact on oral health. E-liquids contain a complex mixture of chemical constituents that may influence the growth of selected oral bacterial species under controlled in vitro conditions. However, comparative data evaluating commercial flavour categories, labelled nicotine concentrations, aerosol extracts, and undiluted e-liquids across both cariogenic and commensal oral bacteria remain limited. This study aimed to provide a preliminary assessment of growth responses of selected or

A wspA mutation in a clinical isolate of Pseudomonas aeruginosa results in rugose small colonies and enhances biofilm formation.

Pseudomonas aeruginosa is a major multidrug-resistant pathogen whose biofilm formation complicates treatment. Rugose small colony variants (RSCVs) exhibit enhanced biofilm production and resistance, posing significant clinical challenges; however, their formation mechanisms in clinical strains remain incompletely understood. Paired clinical isolates AR8023-1 (wild type) and AR8023-2 (RSCV) were chronologically collected from urine samples of a neurosurgery inpatient. Genome comparison via Breseq and plasmid complementation constructed the complemented strain AR8023-2RifR::wspAAR8023-1. All str

Biogenic Silver-Selenium nanocomposite with anticancer activity and potent efficacy against vancomycin-resistant Staphylococcus aureus.

The rapid emergence of multi-drug resistant (MDR) pathogenic bacteria as well as the continued burden of malignant diseases requires the safe development of novel and multifunctional therapeutic agents. The WHO designated vancomycin-resistant Staphylococcus aureus (VRSA) as "high priority" AMR pathogen. Thus, this study aimed to ecofriendly synthesis of silver-selenium nanocomposite (Ag-Se NC) and evaluate its in vitro anticancer effect and inhibitory activity against VRSA clinical isolates. Biogenic Ag-Se NC was successfully synthesized using the aqueous peel extract of Cucumis melo (C. melo)

Engineering multifunctional catheters with a nitric oxide-releasing ciprofloxacin conjugate to combat thrombosis and infection.

Indwelling medical devices, such as intravenous and urinary catheters, have significantly enhanced patient care and greatly improved treatment outcomes. Despite rapid advances in medical devices, infection and thrombosis continue to pose major challenges to device functionality and integrity. Given that current treatments often involve systemic administration of antibiotics and anticoagulants, there is an urgent need for medical devices that are inherently antibacterial and antithrombotic. We explore the chemical conjugation of the nitric oxide (NO) donor S-nitroso-N-acetylpenicillamine (SNAP)

Multidimensional characterization of a novel porcine Klebsiella pneumoniae phage Pkp-1.

BACKGROUND: In recent years, the inappropriate use of antibiotics has led to the widespread emergence of multidrug-resistant Klebsiella pneumoniae (MDR-K. pneumoniae), resulting in infections that are increasingly challenging to manage clinically. Bacteriophages (phages) are emerging as promising alternatives to antibiotics. This study aimed to isolate and characterize lytic phages targeting MDR-K. pneumoniae, providing biological resources and experimental data for phage-based control of MDR-K. pneumoniae infections. RESULTS: A lytic phage, designated vB_KpnA_Pkp-1 (Pkp-1), was successfully

The msf gene causes condition-specific shifts in global gene expression in Haemophilus influenzae.

UNLABELLED: Haemophilus influenzae is a diverse human-restricted bacterium that normally colonizes the healthy nasopharynx but also causes common infections. Comparisons of clinical isolate genomes previously identified a gene, msf, that contained Sel1-like repeats that were associated with clinical disease. Mutant analysis had further found that msf improved survival in macrophages and increased systemic infection in an animal model. However, the role of msf in other conditions and its molecular function remain unknown. To identify protein-protein interactions with Msf, a yeast two-hybrid scr
