Mapping neural representations of fine and gross upper-limb movements across dorsoventral subthalamic nucleus subregions in Parkinson's disease.

Objective. Deep brain stimulation (DBS) of the subthalamic nucleus (STN) is an established therapy for Parkinson's disease (PD), yet optimizing outcomes remains challenging due to the complexity of STN functional architecture and the subjectivity of intraoperative assessments. Furthermore, the encoding of fine versus gross movements across STN dorsoventral subregions remains poorly understood. This study aimed to characterize region- and movement-specific modulation of STN neuronal activity using synchronized electrophysiology and markerless motion tracking-derived kinematics.Approach. Human brain assembloids as a model of anaesthetic-induced neural dynamics in vitro.

BACKGROUND: The mechanisms by which anaesthetics alter neurophysiological states remain poorly understood. Although some hypotheses emphasise thalamic suppression, others propose that cortical (Cx) circuits alone, comprising interconnected excitatory and inhibitory neurones expressing gamma-aminobutyric acid type A (GABAA) receptors, are sufficient for anaesthetic-induced neural dynamics. METHODS: We generated human induced pluripotent stem cell-derived Cx-ganglionic eminence (GE) assembloids and characterised their cellular composition using immunohistochemistry and single-nucleus RNA sequen

Do small-conductance Ca2+-activated K+-channels contribute to ventricular repolarization in human heart failure?

Chronic heart failure constitutes a clinical syndrome characterized by substantial attenuation of repolarization reserve resulting from electrical remodeling. The small-conductance Ca2+-activated K+ channel (SK) has been reported to undergo upregulation in animal heart failure models and in human preparations; however, its exact function is not fully understood. This study aims to elucidate the functional role of SK channels in end-stage human heart failure. SK-protein expression of undiseased and failed human ventricular tissue was investigated by Western blot technique. Action potentials wer

Radiotherapy reduces ATP-mediated relaxation in human rectal smooth muscle.

Radiation therapy for rectal cancer often causes long-term bowel dysfunction. We investigated radiation-associated motility changes in human rectal smooth muscle, focusing on the role of platelet-derived growth factor receptor alpha-positive (PDGFRα+) cells and interstitial cells of Cajal (ICC). Human rectal tissues were obtained from patients undergoing surgery with (n = 61) or without (n = 51) preoperative concurrent chemoradiotherapy (CCRT). Isometric tension recordings were used to assess myogenic responses to potassium chloride (KCl), acetylcholine (ACh), sodium nitroprusside (SNP), and a

Dysregulation of protein SUMOylation networks in Huntington's disease R6/2 mouse striatum.

Huntington's disease is a neurodegenerative disorder caused by an expanded CAG repeat mutation in the Huntingtin (HTT) gene. The mutation impacts neuronal protein homeostasis and cortical/striatal circuitry. SUMOylation is a post-translational modification with broad cellular effects including via modification of synaptic proteins. Here, we used an optimized SUMO protein-enrichment and mass spectrometry method to identify the protein SUMOylation/SUMO interaction proteome in the context of Huntington's disease using R6/2 transgenic and non-transgenic mice. Significant changes in the enrichment

A transgenic mouse embryonic stem cell line for puromycin selection of V0V interneurons from heterogenous induced cultures.

BACKGROUND: Spinal interneurons (INs) relay sensory and motor control information between the brain and body. When this relay circuitry is disrupted from injury or disease, it is devastating to patients due to the lack of native recovery in central nervous system (CNS) tissues. Obtaining a purified population of INs is necessary to better understand their role in normal function and as potential therapies in CNS. The ventral V0 (V0V) INs are excitatory neurons involved in locomotor circuits and are thus of interest for understanding normal and pathological spinal cord function. To achieve scal

Assessment of Cd availability in rice cultivation (Oryza sativa): Effects of amendments and the spatiotemporal chemical changes in the rhizosphere and bulk soil.

Immobilization is widely used to decrease the availability of heavy metals, such as Cd and Pb, in contaminated soils. However, the spatial and temporal changes in the immobilization of soil by amendments combined with planting effects have not been studied well. In this study, unplanted and planted (with rice plants) pot experiments were used to assess the spatial and temporal changes in water-soluble Cd, Fe, Mn, and Ca. Soil properties, such as pH, redox potential (Eh), and dissolved organic carbon (DOC), were continuously recorded in both the rhizosphere and bulk soil using non-invasive rhiz

Carotid chemoreceptors tune breathing via multipath routing: reticular chain and loop operations supported by parallel spike train correlations.

We tested the hypothesis that carotid chemoreceptors tune breathing through parallel circuit paths that target distinct elements of an inspiratory neuron chain in the ventral respiratory column (VRC). Microelectrode arrays were used to monitor neuronal spike trains simultaneously in the VRC, peri-nucleus tractus solitarius (p-NTS)-medial medulla, the dorsal parafacial region of the lateral tegmental field (FTL-pF), and medullary raphe nuclei together with phrenic nerve activity during selective stimulation of carotid chemoreceptors or transient hypoxia in 19 decerebrate, neuromuscularly blocke

An ex vivo multi-electrode approach to evaluate endogenous hormones and receptor subtype pharmacology on evoked and spontaneous neuronal activity within the ventromedial hypothalamus; translation from female receptivity.

INTRODUCTION: The ventromedial hypothalamus (VMH) controls female rodent copulatory behavior, which can be modulated by injection of various compounds into the VMH. Aim. The aim was to determine whether evoked excitatory postsynaptic potentials (EPSPs) or single-unit activity within the VMH ex vivo is a better parameter to predict lordosis. METHODS: VMH slices were placed onto a 64 microelectrode chip and spontaneous single-unit activity was recorded or slices stimulated to evoke EPSPs. MAIN OUTCOME MEASURES: The sodium channel blocker, tetrodotoxin and alpha-amino-3-hydroxy-5-methyl-4-isoxa

Increased susceptibility of aged hearts to ventricular fibrillation during oxidative stress.

Oxidative stress with hydrogen peroxide (H(2)O(2)) readily promotes early afterdepolarizations (EADs) and triggered activity (TA) in isolated rat and rabbit ventricular myocytes. Here we examined the effects of H(2)O(2) on arrhythmias in intact Langendorff rat and rabbit hearts using dual-membrane voltage and intracellular calcium optical mapping and glass microelectrode recordings. Young adult rat (3-5 mo, N = 25) and rabbit (3-5 mo, N = 6) hearts exhibited no arrhythmias when perfused with H(2)O(2) (0.1-2 mM) for up to 3 h. However, in 33 out of 35 (94%) aged (24-26 mo) rat hearts, 0.1 mM H(

Topographical and electrophysiological characteristics of highly excitable S neurones in the myenteric plexus of the guinea-pig ileum.

1. Most intracellular electrical recordings from myenteric neurones have been made from the centre of large ganglia. In this study, we examined the electrophysiological properties of neurones at the corners of large ganglia close to internodal strands and in microganglia. 2. Of 150 neurones in these locations: 111 were tonic S neurones; 9 were phasic S neurones and 30 were AH neurones. 3. Tonic S neurones were characterized by: (i) low resting membrane potentials (-50 +/- 1 mV, mean +/- s.e.m.); (ii) high input impedance (522 +/- 23 MOmega); (iii) low threshold for action potential (AP) genera

Thyroid status and diabetes modulate regional differences in potassium currents in rat ventricle.

1. The rate dependence and recovery kinetics of the Ca(2+)-independent transient (I(t)) and steady-state or 'pedestal' (Iss) outward potassium (K+) currents were studied in single myocytes isolated from epicardial and endocardial regions of rat left ventricles. The whole-cell, suction microelectrode method was used to measure baseline (fully reactivated) I(t), as well as its rate-dependent attenuation. Results from a group of control animals were compared with data from three other groups having an experimentally altered hormonal status. 2. I(t) was significantly smaller in endocardial cells t

[Pontine control of the internal urethral sphincter activity on decerebrate dogs].

The present study was performed to investigate the pontine control of the internal urethral sphincter (IUS) activity using 13 supracolicular decerebrate female dogs. The lower urinary tract function was evaluated by measuring the bladder and the IUS pressure and the external urethral sphincter (EUS) EMG. Bilateral ureters were cannulated and urine was drained extracorporeally. The bladder neck was ligated to induce a isovolumetric bladder contraction, and the urethra was separated from the bladder to evaluate the function of the bladder and the urethra separately. Electrical stimulation (ES) (

Signal processing in brainstem auditory neurons which receive giant endings (calyces of Held) in the medial nucleus of the trapezoid body of the cat.

In the medial nucleus of the trapezoid body (MNTB), each principal neuron receives one large axonal ending (a calyx of Held) and many small endings. In this same region, microelectrode recordings show unusual 'unit' waveforms which have two components separated by about 0.5 ms. We show that the first component (C1) of such a waveform corresponds to a spike from the calyx of Held and that the second component (C2) corresponds to a spike from the MNTB principal neuron. There are two kinds of evidence for these correspondences. First, electrical stimulation of calyciferous axons in the contralate

The somatotopic organization of area 2 in macaque monkeys.

Area 2 is a traditional architectonic subdivision of anterior parietal cortex in macaque monkeys, but its overall somatotopic organization and responsiveness to different types of somatic stimuli are poorly understood, and there are uncertainties concerning its rostral and caudal extent. The goals of the present study were to define the rostral and caudal borders of area 2 better, and to describe its overall organization and responsiveness. Somatic receptive fields were defined for hundreds of closely spaced microelectrode recording sites in postcentral parietal cortex of individual macaque mo

The somatotopic organization of the ventroposterior thalamus of the squirrel monkey, Saimiri sciureus.

Multiunit microelectrode mapping techniques were used to investigate the organization of the somatosensory thalamus in squirrel monkeys. Receptive fields and response characteristics were determined for closely spaced recording sites along arrays of electrode penetrations that passed through the ventral thalamus dorsoventrally, rostrocaudally, or lateromedially. The results were related to thalamic architecture and led to the following conclusions: (1) A large, single, systematic representation of the body surface occupied most or all of the ventroposterior nucleus, VP. The nucleus was further

Evidence for coupled transport of bicarbonate and sodium in cultured bovine corneal endothelial cells.

Using intracellular microelectrode technique, the response of the voltage V across the plasma membrane of cultured bovine corneal endothelial cells to changes in sodium and bicarbonate concentrations was investigated. (1) The electrical response to changes in [HCO3-]o (depolarization upon lowering and hyperpolarization upon raising [HCO3-]o) was dependent on sodium. Lithium could fairly well be substituted for sodium, whereas potassium or choline were much less effective. (2) Removal of external sodium caused a depolarization, while a readdition led to a hyperpolarization, which increased with

A real-time spike classification method based on dynamic time warping for extracellular enteric neural recording with large waveform variability.

BACKGROUND: Computationally efficient spike recognition methods are required for real-time analysis of extracellular neural recordings. The enteric nervous system (ENS) is important to human health but less well-understood with few appropriate spike recognition algorithms due to large waveform variability. NEW METHOD: Here we present a method based on dynamic time warping (DTW) with high tolerance to variability in time and magnitude. Adaptive temporal gridding for "fastDTW" in similarity calculation significantly reduces the computational cost. The automated threshold selection allows for re

Voltammetric and mathematical evidence for dual transport mediation of serotonin clearance in vivo.

The neurotransmitter serotonin underlies many of the brain's functions. Understanding serotonin neurochemistry is important for improving treatments for neuropsychiatric disorders such as depression. Antidepressants commonly target serotonin clearance via serotonin transporters and have variable clinical effects. Adjunctive therapies, targeting other systems including serotonin autoreceptors, also vary clinically and carry adverse consequences. Fast scan cyclic voltammetry is particularly well suited for studying antidepressant effects on serotonin clearance and autoreceptors by providing real
