Switch of regioselectivity in palladium-catalyzed silaboration of terminal alkynes by ligand-dependent control of reductive elimination.

The regioselectivity in the addition of silylboronic esters to terminal alkynes can be switched by the choice of phosphorus ligands on the palladium catalysts. The silaboration proceeds with normal regioselectivity in the presence of (eta(3)-C(3)H(5))Pd(PPh(3))Cl (1.0 mol %) to give 1-boryl-2-silyl-1-alkenes in high yields. In sharp contrast, selective formation of the inverse regioisomers, 2-boryl-1-silyl-1-alkenes, takes place when the reaction is carried out with a palladium catalyst bearing P(t-Bu)(2)(biphenyl-2-yl). A reaction mechanism for the change of regioselectivity that involves rev

meta-Chloroperbenzoic-Acid-Promoted Oxidative Annulations of Alkynes and Nitriles into Oxazoles via Putative β-Hydroxy Vinyl Cations.

Electrophile-promoted alkyne transformations are fundamental to organic synthesis, yet the utilization of oxygen-centered electrophiles compared to carbon- or other heteroatom-centered ones has remained challenging due to the fundamental mismatch between the low nucleophilicity and electron density of alkynes and the high electronegativity of oxygen which hinders the formation of oxygen-centered cations or highly polarized oxy-electrophiles. Herein, we address this long-standing challenge by reporting the DFT-supported electrophilic hydroxylation of alkynes using meta-chloroperbenzoic acid as

Benzothiadiazole Ligand Modulation of Cu(I)-Based Coordination Polymers to Regulate Catalytic Properties for the Hydroboration of Alkynes.

In this work, four novel Cu(I)-based coordination polymers with the formulas of [CuI(BIBT)]n (CuI-BIBT, BIBT = 4,7-di(1H-imidazole-1-yl)benzo[c][1,2,5]thiadiazole), [CuI(BPT)]n (CuI-BPT, BPT = 1,4-bis(1H-imidazole-1-yl)benzene), [CuI(BTDB)]n (CuI-BTBD, BTBD = 4,7-bis(1H-1,2,4-triazol-1-yl)benzo[c][1,2,5]thiadiazole), and [CuI(BTD)0.5]n (CuI-BTD, BTD = benzothiadiazole) were synthesized by the reactions of the corresponding ligand with CuI. All four Cu(I)-based coordination polymers featured two-dimensional (2D) coordination networks built from the organic ligands and Cu2I2 clusters or CunIn ch

Pd/Cu Dual Metal-Catalyzed Regioselective [2 + 2 + 2] Cycloaddition of Malononitriles with Alkynes to Densely Substituted Pyridines.

Transition metal-catalyzed [2 + 2 + 2] cycloaddition of nitriles and two alkynes is an efficient method for assembling pyridines. However, examples employing palladium catalysis have rarely been disclosed, and the processes of reactivity and selectivity remain unclear. We report here a palladium/copper dual metal-catalyzed [2 + 2 + 2] cycloaddition of diynyl-tethered malononitriles and terminal alkynes to synthesize densely substituted pyridines. This method features a good substrate scope, synthetically useful yields, and perfect regioselectivity. The derivatization of the pyridine products d

Copper catalyzed photoredox synthesis of α-keto esters, quinoxaline, and naphthoquinone: controlled oxidation of terminal alkynes to glyoxals.

Herein, we report a facile visible light induced copper catalyzed controlled oxidation of terminal C[triple bond, length as m-dash]C alkynes to α-keto esters and quinoxalines via formation of phenylglyoxals as stable intermediates, under mild conditions by using molecular O2 as a sustainable oxidant. The current copper catalysed photoredox method is simple, highly functional group compatible with a broad range of electron rich and electron poor aromatic alkynes as well as aliphatic alcohols (1°, 2° and 3° alcohols), providing an efficient route for the preparation of α-keto esters (43 examples

Copper- or phosphine-catalyzed reaction of alkynes with isocyanides. Regioselective synthesis of substituted pyrroles controlled by the catalyst.

The copper-catalyzed reaction of isocyanides (CNCH2EWG1) 1 with electron-deficient alkynes (RC[triple bond]CEWG2) 2 gave the 2,4-di-EWG-substituted pyrroles 3 selectively, whereas the phosphine-catalyzed reaction of 1 with 2 afforded the 2,3-di-EWG-subsituted pyrroles 4. Accordingly, regioselective synthesis of substituted pyrroles has been achieved by merely choosing the catalyst.

Sterically Engineered Mo─V Cluster Catalyst for Switchable Z/E-Selective Alkyne Hydrophosphorylation.

Stereodivergent alkyne hydrophosphorylation represents a formidable challenge in synthetic chemistry, particularly for selective and tunable synthesis of (Z)- and (E)-alkenylphosphine oxides. Herein, we report a steric hindrance-mediated strategy that achieves switchable Z/E selectivity via rational design of a metal-organic-functionalized phosphovanadomolybdate [PMo8V4O40(VO)2][(VO)(IM)4]3·5.5H2O (IM = 1-methylimidazole), which features a highly congested steric environment. Steric repulsion between the cluster framework and substrate enables highly Z-selective hydrophosphorylation, affording

Hydrolytic, Thermal, and Electrochemical Stability of Thiol- and Terminal Alkyne-Based Monolayers on Gold: A Comparative Study.

The terminal alkyne-Au interaction is emerging as a promising adsorbing bonding motif for organic monolayers, allowing it to be used for installing antifouling layers and/or recognition elements on gold surfaces for biosensing applications. In contrast to the well-known thiol-on-gold monolayers, the long-term hydrolytic, thermal, and electrochemical stability of the alkyne-Au bond remains relatively unexplored. Insight into these is, however, essential to deliver on the promise of the alkyne-Au bond for (bio)sensing applications, and to see under which conditions they might replace thiolate-go

Diversity-oriented synthesis of stereodefined tetrasubstituted alkenes via a modular alkyne gem-addition strategy.

Stereocontrolled construction of tetrasubstituted olefins has been an attractive issue yet remains challenging for synthetic chemists. In this manuscript, alkynyl selenides, when treated with ArBCl2, are subject to an exclusive 1,1-carboboration, affording tetrasubstituted alkenes with excellent levels of E-selectivity. Detailed mechanistic studies, supported by DFT calculations, elucidates the role of selenium in this 1,1-addition process. Coupled with subsequent C-B and C-Se bond transformations, this 1,1-addition protocol constitutes a modular access to stereodefined all-carbon tetrasubstit

Enantioselective Construction of Spirocyclohexenones via Ni-Catalyzed Double Arylative Cyclization of Alkyne-Tethered Malononitriles.

We report a nickel-catalyzed double arylative cyclization of bis(alkyne)-tethered malononitriles with aryl boronic acids, delivering spirocyclohexenones in moderate yields and high enantioselectivities (up to 97:3 er). The method exhibits a broad substrate scope, is amenable to gram-scale synthesis, and enables downstream transformations through stereocontrolled 1,2-reduction of the ketones to diols. A complementary reductive arylative cyclization employing aryl iodides was also developed. This strategy provides a direct route to enantioenriched spirocycles from simple precursors, offering con

Cu-Catalyzed Azide-Alkyne-Thiol Reaction Forms Ubiquitous Background in Chemical Proteomic Studies.

We report here a Cu-catalyzed azide-alkyne-thiol reaction forming thiotriazoles as the major byproduct under widely used bio-orthogonal protein labeling "click" conditions. The development of Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) had a tremendous impact on many biological discoveries. However, the considered chemoselectivity of CuAAC is hampered by the high reactivity of cysteine free thiols, yielding thiotriazole protein conjugates. The reaction byproducts generate false-positive protein hits in functional proteomic studies. The reported detail investigation of conjugates between

An easy and simple method for the immobilization of dyes through click reactions: activated alkyne, copper not needed.

The copper-free azide-alkyne click reaction has shown to be a successful alternative to immobilize covalently a fluorescente compound onto poly(-l-lactic) acid (PLLA) surfaces. Proceded by basic hydrolysis and amidation reaction, typical surface characterization techniques have validated each functionaliztion step and the success of the conjugation. This method offers a catalyst-free option for various surface conjugations, extremely demanded in biomedical and biosensory fields.

Bench-Stable Dinuclear Mn(I) Catalysts in E-Selective Alkyne Semihydrogenation: A Mechanistic Investigation.

Dinuclear manganese hydride complexes of the form [Mn2 (CO)8 (μ-H)(μ-PR2 )] (R=Ph, 1; R=iPr, 2) were used in E-selective alkyne semi-hydrogenation (E-SASH) catalysis. Catalyst speciation studies revealed rich coordination chemistry and the complexes thus formed were isolated and in turn tested as catalysts; the results underscore the importance of dinuclearity in engendering the observed E-selectivity and provide insights into the nature of the active catalyst. The insertion product obtained from treating 2 with (cyclopropylethynyl)benzene contains a cis-alkenyl bridging ligand with the cyclop

Ribosome-Templated Azide-Alkyne Cycloadditions: Synthesis of Potent Macrolide Antibiotics by In Situ Click Chemistry.

Over half of all antibiotics target the bacterial ribosome-nature's complex, 2.5 MDa nanomachine responsible for decoding mRNA and synthesizing proteins. Macrolide antibiotics, exemplified by erythromycin, bind the 50S subunit with nM affinity and inhibit protein synthesis by blocking the passage of nascent oligopeptides. Solithromycin (1), a third-generation semisynthetic macrolide discovered by combinatorial copper-catalyzed click chemistry, was synthesized in situ by incubating either E. coli 70S ribosomes or 50S subunits with macrolide-functionalized azide 2 and 3-ethynylaniline (3) precur

DNA-Alkyne-Chain Probes Enable N-Glycosite-Resolved GlycoBarcoding of Cell-Surface Proteins.

Cell surface glycoproteins are vital mediators of intercellular communication and primary targets for therapeutic intervention. However, the comprehensive profiling of the "surface glycome" on living cells remains hindered by the low abundance of these proteins and the inherent complexity of glycosylation. Here, we present GlycoCSP, a site-specific glycoproteomics strategy that utilizes alkyne-functionalized DNA scaffolds to achieve high-resolution GlycoBarcoding of cell surface proteins. By leveraging the spatial reach of extended DNA chains and a high-density alkyne array, GlycoCSP ensures s
