Intrahepatic bacterial metataxonomic signature in non-alcoholic fatty liver disease.

OBJECTIVE: We aimed to characterise the liver tissue bacterial metataxonomic signature in two independent cohorts of patients with biopsy-proven non-alcoholic fatty liver disease (NAFLD) diagnosis, as differences in the host phenotypic features-from moderate to severe obesity-may be associated with significant changes in the microbial DNA profile. DESIGN AND METHODS: Liver tissue samples from 116 individuals, comprising of 47 NAFLD overweight or moderately obese patients, 50 NAFLD morbidly obese patients elected for bariatric surgery and 19 controls, were analysed using high-throughput 16S rR

Congenital Tick Borne Diseases: Is This An Alternative Route of Transmission of Tick-Borne Pathogens In Mammals?

Tick-borne diseases (TBDs) have become a popular topic in many medical journals. Besides the obvious participation of ticks in the transmission of pathogens that cause TBD, little is written about alternative methods of their spread. An important role is played in this process by mammals, which serve as reservoirs. Transplacental transfer also plays important role in the spread of some TBD etiological agents. Reservoir species take part in the spread of pathogens, a phenomenon that has extreme importance in synanthropic environments. Animals that accompany humans and animals migrating from wil

Isolation of Aureimonas altamirensis, a Brucella canis-like bacterium, from an edematous canine testicle.

Microbiological and histological analysis of a sample from a swollen testicle of a 2-year-old Border Collie dog revealed a mixed infection of the fungus Blastomyces dermatitidis and the Gram-negative bacterium Aureimonas altamirensis. When subjected to an automated microbial identification system, the latter isolate was provisionally identified as Psychrobacter phenylpyruvicus, but the organism shared several biochemical features with Brucella canis and exhibited agglutination, albeit weakly, with anti-B. canis antiserum. Unequivocal identification of the organism was only achieved by 16S ribo

Brain microbial populations in HIV/AIDS: α-proteobacteria predominate independent of host immune status.

The brain is assumed to be a sterile organ in the absence of disease although the impact of immune disruption is uncertain in terms of brain microbial diversity or quantity. To investigate microbial diversity and quantity in the brain, the profile of infectious agents was examined in pathologically normal and abnormal brains from persons with HIV/AIDS [HIV] (n = 12), other disease controls [ODC] (n = 14) and in cerebral surgical resections for epilepsy [SURG] (n = 6). Deep sequencing of cerebral white matter-derived RNA from the HIV (n = 4) and ODC (n = 4) patients and SURG (n = 2) groups reve

Electroacupuncture alleviates osteoarthritis and is associated with changes in gut microbiota and steroid biosynthesis-related metabolites.

BACKGROUND: Electroacupuncture (EA) is widely used for osteoarthritis (OA), but its mechanisms remain unclear. This study investigated the therapeutic effects of EA on OA in rats and its potential mechanisms. METHODS: A rat model of knee OA was established via intra-articular injection of sodium monoiodoacetate. The effects of EA on behavioral indices (paw withdrawal threshold (PWT), body weight, knee width), cartilage pathology (HE staining, Safranin O-fast green staining, TUNEL), and inflammatory factors (IL-6, IL-1β, TNF-α) were assessed. Gut microbiota and metabolites were analyzed using

Viral lysis accelerates microbial succession patterns resembling diatom senescence.

Diatom blooms influence carbon cycling through organic matter production and its deposition or remineralization-processes mediated by the microbial community. Viruses can influence diatom bloom dynamics and even terminate blooms, yet interactions between diatoms, their viruses, and associated bacteria remain poorly resolved. Here, we examined how infection of the toxigenic diatom Pseudo-nitzschia galaxiae by its ssRNA virus PnGalRNAV reshapes host physiology, microbiome structure, and organic-matter processing in non-axenic batch cultures. Using epi-fluorescence microscopy, 16S rRNA amplicon s

Light-driven carbon-carbon bond formation via CO2 reduction catalyzed by complexes of CdS nanorods and a 2-oxoacid oxidoreductase.

Redox enzymes are capable of catalyzing a vast array of useful reactions, but they require redox partners that donate or accept electrons. Semiconductor nanocrystals provide a mechanism to convert absorbed photon energy into redox equivalents for enzyme catalysis. Here, we describe a system for photochemical carbon-carbon bond formation to make 2-oxoglutarate by coupling CO2 with a succinyl group. Photoexcited electrons from cadmium sulfide nanorods (CdS NRs) transfer to 2-oxoglutarate:ferredoxin oxidoreductase from Magnetococcus marinus MC-1 (MmOGOR), which catalyzes a carbon-carbon bond form

Taxonogenomic Investigation of Yoonia aquicola sp. nov., Isolated from Coastal Seawater.

An alphaproteobacterium, KMU-150T, was isolated from a coastal seawater sample collected in the Republic of Korea and investigated using a taxonogenomic approach. Cells were light beige in color, oval- to short rod-shaped, Gram-stain-negative, strictly aerobic, and non-motile. It was able to grow at 0-7.0% NaCl concentrations (w/v), pH 5.5-9.0, and 20-37 °C. A phylogenetic tree based on 16S rRNA gene sequences showed that strain KMU-150ᵀ clustered with the genus Yoonia within the class Alphaproteobacteria and was closely related to Yoonia litorea DPG-5ᵀ, with a sequence similarity of 98.4%. Intersecting threats: Brucella spp. and nematodes in small cetaceans of the California coast.

Brucella is a genus of zoonotic, intracellular alphaproteobacteria known to cause disease in marine mammals as well as livestock and companion animals. Specifically, Brucella ceti may be associated with severe neurological, respiratory, and reproductive disease in small cetaceans. Nematodes, particularly members of the Pseudaliidae, are common in small cetaceans and may harbor Brucella ceti, although the nature of this relationship remains unclear. This study investigated the prevalence of Brucella spp. in nematodes collected at necropsy from 64 small cetaceans that stranded on the central Cal

Comparative Metagenomic Study of Rhizospheric and Bulk Mercury-Contaminated Soils in the Mining District of Almadén.

Soil contamination by heavy metals, particularly mercury (Hg), is a problem that can seriously affect the environment, animals, and human health. Hg has the capacity to biomagnify in the food chain. That fact can lead to pathologies, of those which affect the central nervous system being the most severe. It is convenient to know the biological environmental indicators that alert of the effects of Hg contamination as well as the biological mechanisms that can help in its remediation. To contribute to this knowledge, this study conducted comparative analysis by the use of Shotgun metagenomics of

The composition of the lung microbiome differs between patients with dermatomyositis and rheumatoid arthritis associated with interstitial lung disease.

Dermatomyositis and rheumatoid arthritis are inflammatory diseases that affect the skeletal muscles and joints, respectively. A common systemic complication of these diseases is interstitial lung disease (ILD), which leads to a poor prognosis and increased mortality. However, the mechanism for the initiation and development of ILD in patients with dermatomyositis is currently unknown. In the present study, we used 16S rRNA high-throughput sequencing to profile the bacterial community composition of bronchoalveolar lavage fluid of patients with dermatomyositis associated with ILD (DM-ILD; short

Synergistic interactions between biogenic organic matter and microbial dynamics during simulated senescent cyanobacterial blooms in freshwater mesocosms.

One of the major consequences of a phytoplanktonic bloom is the massive release of autochthonous organic matter (OM) into the water column, stimulating heterotrophic microbial activity and disrupting the trophic web. To better understand the consequences of such biotic stress, the senescent phase of a bloom was simulated under semi-controlled conditions through the addition of cyanobacterial-derived OM from Microcystis aeruginosa and Aphanizomenon gracile (30% enrichment as eq C) into lake water mesocosms. By following both the autochthonous OM and the microbial communities during 28 days, we

Stratified Bacterial Diversity along Physico-chemical Gradients in High-Altitude Modern Stromatolites.

At an altitude of 3,570 m, the volcanic lake Socompa in the Argentinean Andes is presently the highest site where actively forming stromatolite-like structures have been reported. Interestingly, pigment and microsensor analyses performed through the different layers of the stromatolites (50 mm-deep) showed steep vertical gradients of light and oxygen, hydrogen sulfide and pH in the porewater. Given the relatively good characterization of these physico-chemical gradients, the aim of this follow-up work was to specifically address how the bacterial diversity stratified along the top six layers o

Influence of hydraulic regimes on bacterial community structure and composition in an experimental drinking water distribution system.

Microbial biofilms formed on the inner-pipe surfaces of drinking water distribution systems (DWDS) can alter drinking water quality, particularly if they are mechanically detached from the pipe wall to the bulk water, such as due to changes in hydraulic conditions. Results are presented here from applying 454 pyrosequencing of the 16S ribosomal RNA (rRNA) gene to investigate the influence of different hydrological regimes on bacterial community structure and to study the potential mobilisation of material from the pipe walls to the network using a full scale, temperature-controlled experimenta

Fencing off critical source areas reduces faecal contamination and reshapes microbial communities in agricultural headwater catchments.

Agricultural headwater catchments are critical sources of microbial contamination to surface waters, yet the effectiveness of mitigation practices in these small, dynamic systems is not well defined. A paired-catchment experimental design was used to assess the effects of permanent fencing of critical source areas (CSAs) in ephemeral streams within deer-grazed pastures. Faecal, soil, and freshwater samples were collected over four years, comprising two years of baseline monitoring and two years following mitigation. Viable Escherichia coli concentrations were measured in faeces, soil and water

Bacterial extracellular vesicle as a predictive biomarker for postoperative delirium status after spinal surgery: a prospective cohort study.

BACKGROUND: Prognostic factors significantly associated with postoperative delirium (POD) have been reported discordantly, possibly due to heterogeneous cohorts. Here, bacteria extracellular vesicles (BEVs) were introduced to predict the POD status of a unique patient cohort. METHODS: One hundred twenty-eight patients who underwent spinal surgery participated in this prospective cohort study. Significant preoperative factors (i.e., baseline characteristics, and sequences of 16s rRNA genes from bloods and stools) between patients with and without delirium were subjected to random forest classi

Causal relationship between gut microbiome and sex hormone-binding globulin: A bidirectional two-sample Mendelian randomization study.

PROBLEM: Currently, there is a variety of evidence linking the gut microbiota to changes in sex hormones. In contrast, the causal relationship between SHBG, a carrier of sex hormones, and the gut microbiota is unclear. METHOD OF STUDY: Bidirectional two-sample Mendelian randomization (MR) analysis was used to detect the causal effect between SHBG and the gut microbiome. Summary statistics of genome-wide association studies (GWASs) for the gut microbiome and SHBG were obtained from public datasets. Inverse-variance weighting (IVW), weighted median, weighted mode, MR-Egger and simple mode metho

Role of immune cells in mediating the effect of gut microbiota on Hashimoto's thyroiditis: a 2-sample Mendelian randomization study.

PURPOSE: Hashimoto's thyroiditis (HT) is one of the most commonly encountered types of autoimmune thyroid disorders (AITDs), influenced by environmental factors, genetics, and the immune system. Previous research has shown a correlation between gut microbiota and HT, as well as the involvement of immune cells in its onset and progression. We aimed to investigate whether immune cells act as intermediaries in the causal relationship between gut microbiota and HT. METHODS: In this study, we conducted bidirectional two-sample Mendelian randomization (MR) analyses to explore the relationship betwe
