Previous investigations concerning the biologic effects of heterologous antisera raised against rat chorioallantoic placenta demonstrated that the antisera were nephrotoxic, abortifacient, and teratogenic. It is important to determine if maternal nephritis is associated with abnormal embryonic development induced by antiplacenta sera. In this report, a soluble glycoprotein fraction was isolated from rat chorioallantoic placentas of day 16 of gestation by concanavalin A affinity chromatography after solubilizing the saline-washed placental sediment by sodium deoxycholate. Antiserum raised again

Monoclonal antibodies activate immune effector cells through Fc-Fcγ receptor interactions, which are strongly influenced by Fc region glycosylation. In this study, three anti-SARS-CoV-2 IgG1 human monoclonal antibodies (hmAbs1-3) derived from the B-cell clones of vaccinated (hmAbs1-2) and convalescent (hmAb3) individuals were investigated, with hmAb3 showing the strongest neutralization activity. Glycoform analysis revealed that hmAb1 predominantly contained ∼60% combined G1F and G2F glycoforms, while hmAb2 consisted of ∼63% G1F, G2F, and G2FS1. In contrast, hmAb3 displayed the greatest glycan

Bluetongue disease remains a global economic threat, necessitating the development of rapid, high-precision diagnostics. The genome of the Bluetongue virus (BTV) encodes seven structural proteins, VP1-VP7. Among these proteins, VP7 is the major immunodominant structural protein conserved across BTV serotypes and is therefore targeted for the development of serogroup-specific immunodiagnostic assays. This study aimed to identify an immunodominant VP7 epitope recognized by BTV-specific monoclonal antibodies (MAbs) and to evaluate its utility in a peptide-based ELISA. The gene encoding VP7 protei

The aim of this study was to evaluate the diagnostic potential of human papillomavirus type 18 (HPV18) E6/E7-specific T cell receptor (TCR)-like antibodies for the early detection of HPV-associated cervical cancer. An integrated in silico and in vitro approach was employed, in which three fully human TCR-like antibodies (H11\Ab, E5\Ab, and G8\Ab) were analysed by molecular docking to assess antibody-peptide-major histocompatibility complex (P-MHC) class I HLA-A24:02 interactions, followed by experimental validation using SiHa and C33A cervical cancer cell lines. Docking analyses demonstrated s

A 110-kDa type II transmembrane glycoprotein with dipeptidyl peptidase IV (DPPIV) activity in its extracellular region, CD26 has a multitude of biological functions and plays an important role in the regulation of inflammatory responses and tumor biology. Our work has focused on CD26 as a novel therapeutic target for various tumors and immune disorders, and we have recently developed a humanized anti-CD26 monoclonal antibody (mAb), YS110, which has promising safety profile and clinical activity in patients with malignant pleural mesothelioma. The development of an anti-human CD26 mAb that can

Vaccines against drugs of abuse have induced antibodies in animals that blocked the biological effects of the drug by sequestering the drug in the blood and preventing it from crossing the blood-brain barrier. Drugs of abuse are too small to induce antibodies and, therefore, require conjugation of drug hapten analogs to a carrier protein. The efficacy of these conjugate vaccines depends on several factors including hapten design, coupling strategy, hapten density, carrier protein selection, and vaccine adjuvant. Previously, we have shown that 1 (MorHap), a heroin/morphine hapten, conjugated to

Inconsistent results obtained with published methods for the elution of antibodies from tissue sections prompted the assessment of both old and new methods in combination with monoclonal rabbit antibodies of known, increased affinity (above 1×10(-9) KD). We tested an acidic (pH 2) glycine buffer, a 6 M urea hot buffer and a 2-Mercaptoethanol, SDS buffer (2-ME/SDS). Some antibodies were not removed by the glycine pH 2 or 6 M urea hot buffers, indicating that antibodies survive much harsher conditions than previously believed. We found that the elution is dependent upon the antibody affinity and

Tissues from atrophic large plaque parapsoriasis were examined using the indirect immunoperoxidase technique with a monoclonal T cell antibody as primary antibody, and affinity isolated peroxidase conjugated goat anti-mouse IgG as second stage antibody. The "T" cell nature of the dermal infiltrate and of single exocytic epidermal lymphocytes was demonstrated. The results obtained suggest that the method utilized will be useful for the demonstration of lymphoid cell subpopulations in a variety of cutaneous disorders in which a lymphocytic infiltrate forms a significant component of the histopat

BACKGROUND: Death receptor 3 (DR3; also termed tumor necrosis factor receptor superfamily member 25 [TNFRSF25]) is the sole functional receptor for tumor necrosis factor (TNF)-like cytokine 1A (TL1A) and is constitutively expressed on effector lymphoid cells in peripheral blood and in both inflamed and adjacent noninflamed tissues. Excessive DR3 signaling contributes to chronic inflammation in inflammatory bowel disease (IBD). While clinical trials of anti-TL1A antibodies have demonstrated efficacy in ulcerative colitis and Crohn disease, the stable expression pattern of DR3 suggests that rece

Sepsis, a lethal organ dysfunction caused by a dysregulated host response to infection, is the leading cause of worldwide in-hospital mortality. However, the early diagnostic methods for sepsis are still urgent for guiding accurate antibiotic usage and improving the survival rate of the patients. Herein, we constructed a PD-L1 antibody affinity microfluidic (PAAM) chip for early sepsis diagnosis and severity assessment. The chip was used to capture PD-L1-expressing leukocytes from whole blood samples obtained from healthy control (HC) volunteers (n = 15) and sepsis patients on day 1 (D1) and d

Cluster of differentiation 47 (CD47) is a transmembrane protein highly expressed in tumor cells that interacts with signal regulatory protein alpha (SIRPα) and triggers a "don't eat me" signal to the macrophage, inhibiting phagocytosis and enabling tumor escape from immunosurveillance. The CD47-SIRPα axis has become an important target for cancer immunotherapy. To date, the advancement of CD47-targeted modalities is hindered by the ubiquitous expression of the target, often leading to rapid drug elimination and hematologic toxicity including anemia. To overcome those challenges a bispecific ap

α-Galactosidase (αGal) is a lysosomal enzyme that hydrolyses the terminal α-galactosyl moiety from glycosphingolipids. Mutations in the encoding genes for αGal lead to defective or misfolded enzyme, which results in substrate accumulation and subsequent organ dysfunction. The metabolic disease caused by a deficiency of human α-galactosidase A is known as Fabry disease or Fabry-Anderson disease, and it belongs to a larger group known as lysosomal storage diseases. An effective treatment for Fabry disease has been developed by enzyme replacement therapy (ERT), which involves infusions of purifie

Nerve growth factor (NGF) and its low-affinity receptor, p75 neurotrophin receptor (p75 NTR), are important mediators of pain. To explore further the mechanisms involved, we examined suppression of pain behavior and expression of neuropeptides such as calcitonin gene-related peptide (CGRP) using a p75 NTR inhibitory antibody, in a mouse sciatic nerve crush model. In the nerve-injured model, 150 microg of a p75 NTR inhibitory antibody or 10 microl of saline were applied. The sciatic nerve in the sham-operated group was uninjured. Mechanical allodynia was measured for 2 weeks. CGRP and p75 NTR e

PURPOSE: In patients with cancer who have an abnormal biomarker finding, the source of the biomarker in the bloodstream must be located for confirmation of diagnosis, staging, and therapy planning. We evaluated if immuno-PET with the radiolabeled high-affinity antibody HuMab-5B1 (MVT-2163), binding to the cancer antigen CA19-9, can identify the source of elevated biomarkers in patients with pancreatic cancer.
PATIENTS AND METHODS: In this phase I dose-escalating study, 12 patients with CA19-9-positive metastatic malignancies were injected with MVT-2163. Within 7 days, all patients underwent a

This phase 2 study assessed the safety and efficacy of ocaratuzumab, a humanized anti-CD20 monoclonal antibody. Fifty patients with previously treated follicular lymphoma (FL) and a low-affinity genotype of FcγRIIIa received ocaratuzumab 375 mg/m(2) weekly for 4 weeks. Grade 3/4/5 adverse events (AEs) were reported in 11/1/1 patients, respectively. Serious AEs were reported by 11/50 patients, and three discontinued due to AEs. One patient died from aspiration pneumonia due to possibly drug-related nausea and vomiting. Investigator-assessed response rate was 30% (15/50), including four complete

Multiple SARS-CoV-2 variants of concern have emerged and caused a significant number of infections and deaths worldwide. These variants of concern contain mutations that might significantly affect antigen-targeting by antibodies. It is therefore important to further understand how antibody binding and neutralization are affected by the mutations in SARS-CoV-2 variants. We highlighted how antibody epitope specificity can influence antibody binding to SARS-CoV-2 spike protein variants and neutralization of SARS-CoV-2 variants. We showed that weakened spike binding and neutralization of Beta (B.1

Fever is a typical symptom of most infectious diseases. While prolonged fever may be clinically undesirable, mild reversible fever (<39℃, 312 K) can potentiate the immune responses against pathogens. Here, using molecular dynamics and free energy calculations, we investigated the effect of febrile temperatures (38℃ to 40℃, 311 K to 313 K) on the immune complexes formed by the SARS-CoV-2 spike protein with two neutralizing monoclonal antibodies. In analyzing the conformational dynamics of the interactions between the antibodies and the spike protein under different thermal conditions, we found

Rozanolixizumab (UCB7665), a humanized high-affinity anti-human neonatal Fc receptor (FcRn) monoclonal antibody (IgG4P), has been developed to reduce pathogenic IgG in autoimmune and alloimmune diseases. We document the antibody isolation and compare rozanolixizumab with the same variable region expressed in various mono-, bi- and trivalent formats. We report activity data for rozanolixizumab and the different molecular formats in human cells, FcRn-transgenic mice, and cynomolgus monkeys. Rozanolixizumab, considered the most effective molecular format, dose-dependently and selectively reduced

The involvement of gangliosides in processes related to nerve regeneration and sprouting has been demonstrated recently. The type of interaction by which gangliosides may influence neuronal sprouting was investigated in the present work. Affinity-purified rabbit anti-GM1 antibodies were found to block the sprouting from dorsal root ganglia (DRG) of chicken embryo induced by nerve growth factor (NGF). Only a moderately inhibitory effect was produced by antibodies directed to GM2, suggesting a specific interaction between NGF and GM1. In spite of the low inhibitory effect of anti-GM2 antibodies

In a previously reported phase I clinical trial, subjects vaccinated with two doses of an unadjuvanted H7N9 virus like particle (VLP) vaccine responded poorly (15.6% seroconversion rates with 45μg hemagglutinin (HA) dose). In contrast, 80.6% of subjects receiving H7N9 VLP vaccine (5μg HA) with ISCOMATRIX™ adjuvant developed hemagglutination-inhibition (HI) responses. To better understand the role of adjuvant, complete antibody epitope repertoires of post-vaccination sera were investigated using Whole Genome Fragment Phage Display Library (GFPDL).
