Apelin-13 enhances BMSCs osteogenic differentiation and bone regeneration under hypoxia via upregulation of Ibsp.

BACKGROUND: Apelin-13 plays a critical role in promoting angiogenesis and regulating cellular metabolism. However, the short half-life of Apelin-13 in vivo limits its further applications. Stem cell transplantation has been widely applied in tissue regeneration, but the low survival rate of transplanted stem cells in vivo restricts its therapeutic efficacy. This study aims to combine Apelin-13 with stem cell transplantation to enhance the survival rate of transplanted stem cells and promote bone regeneration. METHODS: In this study, BMSCs and HUVEC were pretreated with different concentration

GsMTx4-loaded GelMA promotes tendon regeneration and suppresses heterotopic ossification via the Apelin signaling pathway.

Tendon injuries pose significant challenges in both athletes and the general population, often leading to prolonged healing, impaired functionality, and increased risk of re-injury. Current treatment options are limited and often yield unfavorable outcomes. Given that tendons are highly mechanosensitive tissues, recent studies highlight the crucial role of mechanotransduction in tissue repair. Piezo1, a mechanosensitive ion channel, has been recognized as a crucial factor attributing to many pathological processes in various tissues, but its specific role in tendon healing has not been previou

Apelin-13 protects against cisplatin-induced ototoxicity by inhibiting apoptosis and regulating STAT1 and STAT3.

The ototoxic side effect of cisplatin is a main cause of sensorineural hearing loss. This side effect limits the clinical application of cisplatin and affects patients' quality of life. This study was designed to investigate the effect of apelin-13 on cisplatin-induced C57BL/6 mice hearing loss model and explore the potential underlying molecular mechanisms. Mice were intraperitoneally injected with 100 μg/kg apelin-13 2 h before 3 mg/kg cisplatin injection for 7 consecutive days. Cochlear explants cultured in vitro were pretreated with 10 nM apelin-13 2 h prior to 30 μM cisplatin treatment fo

Adipokine Apelin/APJ Pathway Promotes Peritoneal Dissemination of Ovarian Cancer Cells by Regulating Lipid Metabolism.

Adipose tissue, which can provide adipokines and nutrients to tumors, plays a key role in promoting ovarian cancer metastatic lesions in peritoneal cavity. The adipokine apelin promotes ovarian cancer metastasis and progression through its receptor APJ, which regulates cell proliferation, energy metabolism, and angiogenesis. The objective of this study was to investigate the functional role and mechanisms of the apelin-APJ pathway in ovarian cancer metastasis, especially in context of tumor cell-adipocyte interactions. When co-cultured in the conditioned media (AdipoCM) derived from 3T3-L1 adi

Bioactive peptide apelin rescues acute kidney injury by protecting the function of renal tubular mitochondria.

Mitochondrial dysfunction in proximal tubular epithelial cells is a key event in acute kidney injury (AKI), which is a risk factor for the development of chronic kidney disease (CKD). Apelin is a bioactive peptide that protects against AKI by alleviating inflammation, inhibiting apoptosis, and preventing lipid oxidation, but its role in protecting against mitochondrial damage remains unknown. Herein, we examined the protective effects of apelin on mitochondria in cisplatin-stimulated human renal proximal tubular epithelial cells and evaluated its therapeutic efficacy in cisplatin-induced AKI m

Apelin-13 Inhibits Methylglyoxal-Induced Unfolded Protein Responses and Endothelial Dysfunction via Regulating AMPK Pathway.

It has been suggested that methylglyoxal (MGO), a glycolytic metabolite, has more detrimental effects on endothelial dysfunction than glucose itself. Recent reports showed that high glucose and MGO induced endoplasmic reticulum (ER) stress and myocyte apoptosis in ischemic heart disease was inhibited by apelin. The goal of the study is to investigate the molecular mechanism by which MGO induces endothelial dysfunction via the regulation of ER stress in endothelial cells, and to examine whether apelin-13, a cytoprotective polypeptide ligand, protects MGO-induced aortic endothelial dysfunction.

Association between plasma apelin levels and coronary collateral development in patients with stable angina pectoris.

Apelin is an endogenous ligand for the orphan G protein-coupled receptor (APJ receptor). Apelin is predominantly expressed in endocardial and vascular endothelial cells, while APJ receptor is localized to endothelial and smooth muscle cells, and cardiomyocytes. Apelin has recently attracted much attention due to its promotive effects on angiogenesis and its protective effects against mycardial infarction. In this issue of Atherosclerosis, Akboga et al. investigated plasma apelin levels in patients with stable angina and severe coronary artery stenosis and provided the first evidence that highe

Targeted delivery of apelin using a novel extracellular vesicle platform for pulmonary arterial hypertension treatment.

Pulmonary arterial hypertension (PAH) is a severe disease characterized by endothelial dysfunction, vascular remodeling, and pulmonary artery occlusion, culminating in right ventricular hypertrophy and heart failure. While apelin peptides are promising therapeutic candidates due to their critical role in vascular homeostasis, their efficacy as agonists is limited by insufficient lesion-specific targeting and suboptimal in vivo stability. Here, we developed an engineered extracellular vesicle (EV) platform for precise apelin delivery to PAH lesions, maximizing therapeutic impact. Using interfer

Screening apelin analogues and a small molecule agonist as effective cardiovascular therapeutics against reperfusion injury.

Ischemic reperfusion injury is a major global health threat, with the development of effective therapeutics still urgently needed. While both peptide therapeutics and small-molecule agonists have been extensively investigated to treat this condition, in this study, we explored the pharmacological potential of retro-inverso apelin peptides, including parent and modified forms, by evaluating their binding activity and physiological effects. We further assessed two metabolically stable apelin peptide analogues previously developed in our lab and compared them to a small-molecule agonist, BMS-9862

Apelin-13 alleviated ischemia reperfusion injury via KLF2/NF-κB signaling pathway after kidney transplantation.

Kidney transplantation (KT) represents the optimal treatment method for chronic kidney disease. However, various acute and chronic injuries seriously compromise the long-term survival rates of grafts and patients. Ischemia-reperfusion injury (IRI), which is inevitable in KT, initiates damage and inflammatory response in the allografts, leading to delayed graft function, and an increased risk of acute rejection, severely damage the transplanted kidney. The APJ/apelin system has been confirmed to exert an anti-inflammatory effect in multiple organ systems. In this study, a syngeneic orthotopic r

Protective effect of apelin-13 in lens epithelial cells via inhibiting oxidative stress-induced apoptosis.

BACKGROUND: It is widely accepted that glaucoma-induced oxidative stress expedites cataracts' process. Therefore, we examined the effects of apelin-13 against oxidative stress-induced damage in human lens epithelial cells (HLECs) and investigated the potential pathogenic mechanism of acute primary angle-closure glaucoma. METHODS: This experiment included five groups: control, H2O2, apelin-13 + H2O2, ML221 + H2O2, and apelin-13 + ML221 + H2O2. ML221 was employed in rescue experiments as an APJ antagonist. HLECs were pretreated with or without apelin-13 and subsequently exposed to H2O2. HLECs'

Apelin Receptor Homodimerisation Inhibits Hippocampal Neuronal Autophagy via G Protein-Dependent Signalling in Vascular Dementia.

Vascular dementia (VD), a progressive vascular cognitive impairment, is characterised by the presence of cerebral hypoperfusion, increased blood-brain barrier permeability, and white matter lesions. Although current treatment strategies primarily focus on risk factors such as hypertension, diabetes, and heart disease, efficient and targeted therapies are lacking and the underlying mechanisms of VD remain unclear. We previously discovered that Apelin receptors (APJ), which are G protein-coupled receptors (GPCRs), can homodimerize and generate signals that are distinct from those of APJ monomers

Maternal exercise improves epithelial development of fetal intestine by enhancing apelin signaling and oxidative metabolism.

Obesity in pregnancy is currently the leading cause of gestational complications for the mother and fetus worldwide. Maternal obesity (MO), common in western societies, impedes development of intestinal epithelium in the fetuses, which causes disorders in the nutrient absorption and intestine-related immune responses in offspring. Here, using a mouse model of maternal exercise (ME), we found that exercise during pregnancy protects the impairment of fetal intestinal morphometrical formation and epithelial development due to MO. MO decreased villus length and epithelial proliferation markers in

Apelin improves cardiac function mainly through peripheral vasodilation in a mouse model of dilated cardiomyopathy.

There is growing evidence that apelin plays a role in the regulation of the cardiovascular system by increasing myocardial contractility and acting as a vasodilator. However, it remains unclear whether apelin improves cardiac contractility in a load-dependent or independent manner in pathological conditions. For this purpose we investigated the cardiovascular effects of apelin in α-actin transgenic mice (mActin-Tg mice), a model of cardiomyopathy. [Pyr1]apelin-13 was administered by continuous infusion at 2 mg/kg/d for 3 weeks. Effects on cardiac function were determined by echocardiography an

Angiotensin (1-7) and Apelin co-therapy: New strategy for heart failure treatment of rats.

OBJECTIVE: Isoproterenol (ISO)-induced heart failure is a standardized model for the study of beneficial effects of various drugs. Both apelin and angiotensin 1-7 have a cardiac protective effect. We assumed that co-therapy with apelin and angiotensin 1-7 (Ang (1-7)) may have synergistic cardioprotective effects against isoproterenol-induced heart failure. Methods The animals were randomly assigned to one of eight groups of seven animals in each group as follows: (1) control I (saline; IP injection) (1) control II (saline; via mini-osmotic pump) (3) ISO (5 mg/ kg; IP), (4) Apelin (20μg/ kg; IP

Apelin-36 mitigates MPTP/MPP+-induced neurotoxicity: Involvement of α-synuclein and endoplasmic reticulum stress.

Parkinson's disease (PD) is characterized by the progressive loss of dopaminergic neurons within the substantia nigra compacta (SNpc) which leads to the behavioral dysfunction. In the present study, we investigated the effect of Apelin-36 on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridin (MPTP)/1-methyl-4-phenylpyridinium (MPP+)-induced neurotoxicity. The treatment with Apelin-36 significantly alleviated the MPTP-induced the behavioral dysfunction and dopaminergic neurodegeneration in the SNpc of mice, and also remarkably decreased the MPP+-induced cell death of SH-SY5Y cells. Furthermore, Apelin

Apelin-13 Regulates Vasopressin-Induced Aquaporin-2 Expression and Trafficking in Kidney Collecting Duct Cells.

BACKGROUND/AIMS: Apelin and its G protein-coupled receptor APJ (gene symbol Aplnr) are strongly expressed in magnocellular vasopressinergic neurons suggesting that the apelin/APJ system plays a key role at the central level in regulating salt and water balance by counteracting the antiduretic action of vasopressin (AVP). Likewise, recent studies revealed that apelin exerts opposite effects to those of vasopressin induced on water reabsorption via a direct action on the kidney collecting duct. However, the underlying mechanisms of the peripheral action of apelin are not clearly understood. Here

Apelin-13 promotes cardiomyocyte hypertrophy via PI3K-Akt-ERK1/2-p70S6K and PI3K-induced autophagy.

Apelin is highly expressed in rat left ventricular hypertrophy Sprague Dawley rat models, and it plays a crucial role in the cardiovascular system. The aim this study was to clarify whether apelin-13 promotes hypertrophy in H9c2 rat cardiomyocytes and to investigate its underlying mechanism. The cardiomyocyte hypertrophy was observed by measuring the diameter, volume, and protein content of H9c2 cells. The activation of autophagy was evaluated by observing the morphology of autophagosomes by transmission electron microscopy, observing the subcellular localization of LC3 by light microscopy, an

Altered glucose homeostasis is associated with increased serum apelin levels in type 2 diabetes mellitus.

BACKGROUND: Apelin is an adipokine that plays a role in the regulation of glucose homeostasis and in obesity. The relationship between apelin serum concentration and dysmetabolic conditions such as type 2 diabetes (T2D) is still controversial. Aims of our study are: 1) determine the circulating levels of apelin in a large cohort of Italian subjects with T2D, T1D and in non-diabetic controls; 2) identify putative metabolic determinants of modified apelin concentrations, in order to search possible mechanism of apelin control; 3) investigate changes in apelin levels in response to sharp modifica
