Frequent loss of KAI1 expression in squamous and lymphoid neoplasms. An immunohistochemical study of archival tissues.

The metastasis suppressor gene KAI1 was identified by its ability to inhibit the formation of pulmonary metastases in experimental models for prostatic carcinoma. Down-regulation of this gene may be correlated with the invasive phenotype in melanomas and colon and bladder carcinomas and with the metastatic phenotype in carcinomas of the lung, breast, prostate, and pancreas. The goal of our study was to establish an immunohistochemical method to detect KAI1 expression in archival tissues. Using cell lines with known KAI1 levels and paraffin-embedded KAI1 positive tissues as controls, we observe

Characterization of DNA extracted from archival celloidin-embedded human temporal bone sections.

The focus of the present investigation was to study, via molecular biology techniques, the character of the DNA present in individual archival celloidin-embedded human temporal bone sections. Polymerase chain reaction (PCR) amplification of 92 base pair (bp), 121 bp, and 471 bp regions of mitochondrial DNA (mtDNA) extracted from a single archival celloidin-embedded human temporal bone section was used to assess the length of the template DNA extracted. The effects of digestion time and sample motion during the extraction method on DNA concentration was also studied. These data are crucial to d

Applications of experimental techniques to epidemiological studies of non-Hodgkin's lymphoma: use of archival tissues.

Archival tissues, particularly formalin-fixed paraffin-embedded tumors, have become increasingly valuable in studies of the etiology of cancer. In non-Hodgkin's lymphoma, subclassification of tumors by immunophenotyping and identification of oncogenic viruses has allowed more accurate interpretation of associated epidemiological information. One such example is adult T-cell leukemia/lymphoma, which is not a single histopathological entity and usually is associated with human T-cell lymphotropic virus, type I. In addition to confirming the diagnosis, the pattern of virus distribution, utilized

Dual Nd isotopic archives in fossil teeth reveal bottom-current control of rare-earth element enrichment in deep-sea sediments.

The global emergence of deep-sea sediments enriched in rare-earth elements and yttrium (REY) after the Late Eocene cooling coincides spatiotemporally with Antarctic bottom water (AABW) expansion, yet the underlying mechanism remains unresolved. Here, we present in situ elemental and neodymium isotopic analyses of apatites extracted from Oligocene-Miocene sediments along the North Pacific AABW pathway. Results show ∼90% of the REY budget resides in inner dentin of fossil teeth. The dentin and coexisting authigenic fluorapatites display similar εNd values (∼-5) matching North Pacific seawater, w

From the archives of MD Anderson Cancer Center: Composite mantle cell lymphoma and lymphoplasmacytic lymphoma involving bone marrow at presentation.

Composite lymphoma, defined as two or more distinct well-defined entities involving the same anatomic site, is rare. Here we report a 79-year-old woman with composite mantle cell lymphoma (MCL) and lymphoplasmacytic lymphoma (LPL) involving bone marrow at the time of initial diagnosis. The patient presented with splenomegaly and lymphadenopathy and laboratory studies showed an elevated serum IgM level and IgM kappa paraprotein. Bone marrow evaluation showed concurrent involvement by MCL and LPL, supported by immunophenotypic studies that revealed two distinct aberrant B-cell populations. Next-

From the archives of MD Anderson Cancer Center: Aleukemic T-prolymphocytic leukemia, a rare presentation and review of the literature.

T-prolymphocytic leukemia (T-PLL) is a rare, aggressive T-cell leukemia, and patients typically present with marked peripheral blood lymphocytosis. Approximately 15-20 % of patients may present with moderate and relative stable lymphocytosis and an indolent clinical course that can persist for a few years. However, eventually these patients go on to develop marked lymphocytosis and rapidly progressive disease. We report a 72-year-old man who presented with multicompartmental lymphadenopathy and a normal complete blood count. Excision of left and right cervical lymph nodes showed replacement of

From the archives of MD Anderson Cancer Center: Primary effusion lymphoma with simultaneous involvement of the retroperitoneum and pleural cavity.

Primary effusion lymphoma (PEL) is a rare neoplasm that arises in the context of severe immunosuppression. Acquired immunodeficiency syndrome (AIDS) as a result of human immunodeficiency virus (HIV) infection is the most common cause of immunodeficiency in patients who develop PEL. These neoplasms usually involve one or more body cavities, so-called classic PEL. The pleural cavity is most often involved, followed by the peritoneal and pericardial cavities. Involvement of the cerebrospinal fluid (CSF) and meninges is rare. A subset of patients can present with a tissue-based mass, known as the

Changes and challenges of the archives: researching early-twentieth century lesbianism in United States prisons during the COVID-19 pandemic.

"Changes and Challenges of the Archives" is based on a still-in-progress master's thesis that I will complete by May 2021 at Sarah Lawrence College. The purpose of this article is not to come to any conclusions about the challenges of researching lesbian history during the COVID-19 pandemic but, rather, to explore how these world circumstances have further complicated the labor required of a lesbian historian. Many elements of this thesis and research are still in flux, including my investigation of the role race played in creating and developing a sexually deviant, criminalized definition of

From the archives of AFIP. Imaging of giant cell tumor and giant cell reparative granuloma of bone: radiologic-pathologic correlation.

The radiologic features of giant cell tumor (GCT) and giant cell reparative granuloma (GCRG) of bone often strongly suggest the diagnosis and reflect their pathologic appearance. At radiography, GCT often demonstrates a metaepiphyseal location with extension to subchondral bone. GCRG has a similar appearance but most commonly affects the mandible, maxilla, hands, or feet. Computed tomography and magnetic resonance (MR) imaging are helpful in staging lesions, particularly in delineating soft-tissue extension. Cystic (secondary aneurysmal bone cyst) components are reported in 14% of GCTs. Howeve

Genetic Screening for Hereditary Transthyretin Amyloidosis in the Population of Cammarata and San Giovanni Gemini Through Red Flags and Registry Archives.

Introduction: Hereditary transthyretin amyloidosis (ATTRv) is a severe, multisystemic, autosomal dominant disease with variable penetrance caused by mutations in the TTR gene generating protein misfolding and accumulation of amyloid fibrils. The diagnosis is usually challenging because ATTRv may initially manifest with nonspecific multisystemic symptoms. Conversely, an early diagnosis is needed to start timely appropriate therapy. Hence, screening models have been proposed to improve ATTRv diagnosis. In this study, we propose a genetic screening model based on predefined "red flags" followed b

Availability and quality of paraffin blocks identified in pathology archives: a multi-institutional study by the Shared Pathology Informatics Network (SPIN).

BACKGROUND: Shared Pathology Informatics Network (SPIN) is a tissue resource initiative that utilizes clinical reports of the vast amount of paraffin-embedded tissues routinely stored by medical centers. SPIN has an informatics component (sending tissue-related queries to multiple institutions via the internet) and a service component (providing histopathologically annotated tissue specimens for medical research). This paper examines if tissue blocks, identified by localized computer searches at participating institutions, can be retrieved in adequate quantity and quality to support medical re

From the archives of the AFIP: Pediatric orbit tumors and tumorlike lesions: neuroepithelial lesions of the ocular globe and optic nerve.

Tumors and tumorlike lesions of the globe and optic nerve in children represent a different histologic spectrum than in adults; the imaging appearances of these lesions reflect their pathologic features. Retinoblastoma is a tumor of infancy and the most common intraocular tumor in children. There are heritable and nonheritable forms. The most common clinical finding is leukocoria. The differential diagnoses of this sign include several nonneoplastic lesions: Persistent hyperplastic primary vitreous is a congenital persistence of an embryonic structure causing a retrolental mass. The primitive

Active nuclease inhibition over passive freezing secures the genomic archive of fish eye lenses.

Fish eye lenses offer a chronological archive of individual life histories and preserve genomic DNA. However, integrating ecological and genetic data is hindered by post-mortem nuclease degradation, and standardized DNA preservation protocols for this tissue remain undefined. We evaluated chemical and thermal preservation methods to maximize DNA yield and integrity from fish lenses. Using a paired experimental design across three marine species, we demonstrate that chemical stabilization with TRIzol yields significantly higher DNA quantities (2654.7 ± 331.5 ng) than ultra-low temperature freez

Molecular Profiling of Archival Tonsillar Squamous Cell Carcinoma in a Korean Pre-HPV-vaccination Cohort.

BACKGROUND/AIM: Tonsillar squamous cell carcinoma (TSCC) is a biologically heterogeneous malignancy in which human papillomavirus (HPV) infection, epidermal growth factor receptor (EGFR) alteration, and chromosomal instability represent major oncogenic mechanisms. This study evaluated HPV genotyping, EGFR mutation, and loss of heterozygosity (LOH) at chromosomal loci 17p13 and 9q22 in archival TSCC using conventional polymerase chain reaction (PCR)-based methodologies reproducible without next-generation sequencing infrastructure. The cohort, collected between 2000 and 2006 prior to the implem

Virological success despite archived INSTI drug resistance mutations on INSTI-based antiretroviral regimens.

OBJECTIVES: The clinical relevance of archived drug resistance mutations (DRMs) detected by HIV-1 proviral genotypic resistance testing (GRT) in virologically suppressed (VS) people with HIV (PWH) remains unclear, especially since these mutations may reside in defective proviruses. We assessed the impact of archived full-class integrase strand transfer inhibitor (INSTI) DRMs in a real-life setting. DESIGN: This was a retrospective study conducted in Paris, France, evaluating archived INSTI DRMs in VS PWH receiving INSTI-based regimens. METHODS: We included VS PWH receiving INSTI-based regime
