Dorsal raphe nucleus regulation of a panic-like defensive behavior evoked by chemical stimulation of the rat dorsal periaqueductal gray matter.

Electrical or chemical stimulation of the dorsal periaqueductal gray matter (DPAG) evokes escape, a defensive behavior that has been related to panic attacks. Injection of 5-HT(1A) or 5-HT(2A) receptor agonists into this midbrain area inhibits this response. It has been proposed that the impairment of 5-HT mechanisms controlling escape at the level of the DPAG may underlie the susceptibility to panic attacks that characterizes the panic disorder. In this study we evaluated the effects of the pharmacological manipulation of the dorsal raphe nucleus (DRN), which are the main source of 5-HT input

Effect of xanthone from Kielmeyera coriacea stems on serotonergic neurons of the median raphe nucleus.

Kielmeyera coriacea Mart. (Clusiaceae), known as "Pau Santo", is used to treat several tropical diseases. The hydroethanolic extract (HE) of Kielmeyera coriacea stems and its semi-pure dichloromethane constituent (DCM) produced an anti-immobility effect in rats submitted to the forced swimming test (FST), suggesting a antidepressant-like profile. This study evaluated the effect of intra-median raphe nucleus (MRN) microinjection of 1,3,7-trihydroxy-2-(3-methylbut-2-enyl)-xanthone, present in large quantity in the HE from Kielmeyera coriacea stems, on immobility behaviour in the FST in rats. The dopamine transporter: a vigilant border control for psychostimulant action.

Neurotransmission within the mesocorticolimbic dopamine system has remained the central focus of investigation into the molecular, cellular and behavioral properties of psychostimulants for nearly three decades. The primary means by which dopamine transmission in the synapse is terminated is via the dopamine transporter (DAT), the presynaptic plasmalemmal protein that is responsible for the reuptake of released dopamine. Numerous abused as well as clinically important drugs have important pharmacological interactions with DAT. In general, these compounds fall into two categories: those that bl

Acetylcholine contributes to hypoxic chemotransmission in co-cultures of rat type 1 cells and petrosal neurons.

The neurotransmitter mechanisms that mediate chemosensory transmission in the mammalian carotid body (CB), i.e. the primary arterial P(O2) detector, are controversial. Given the inherent difficulty of recording from afferent terminals in situ, the authors have adopted an alternative approach based on co-culture of dissociated CB receptor (type 1) cell clusters and petrosal neurons (PN) from 8-14-day-old rat pups. Electrophysiological, perforated patch recordings from petrosal somas, juxtaposed to type 1 clusters, revealed the development of a high incidence of functional 'synapses' in vitro. R

Neuropharmacology and neurochemistry of canine narcolepsy.

It is believed that narcolepsy involves abnormalities of rapid eye movement (REM) sleep, especially of REM sleep atonia. Compelling evidence suggests that the regulation of REM sleep and REM sleep atonia involves a reciprocal interaction of cholinergic and monoaminergic systems. Using our canine model of narcolepsy and a pharmacological approach, we have previously demonstrated a similar interaction in the regulation of cataplexy. Global activation of cholinergic or monoaminergic transmission aggravates or suppresses canine cataplexy, respectively. We have also identified the subtypes of monoa

Current use of atypical antipsychotics.

In terms of the phenomenology of schizophrenia, there are four targets for drug treatments: positive symptoms, negative symptoms, affective dysfunction, and cognitive dysfunction. Because of the side-effects of both conventional antipsychotics and the new atypicals, there still is a need to search for better-tolerated antipsychotics. Conventional antipsychotics have two principal limitations: 30-40% of patients have an insufficient response to them, and they have a large variety of adverse effects. Side-effects will reduce patients' compliance with treatment, as well as their immediate quality

Opposing Estrous Cycle-Dependent Norepinephrine and Dopamine Regulation in Response to Methamphetamine.

The central catecholamine systems, norepinephrine and dopamine, play a critical role in encoding the valence of environmental stimuli to promote engagement in behaviors that potentiate an organism's survival. Furthermore, both neurochemicals in limbic brain areas such as the nucleus accumbens (NAc) and bed nucleus of the stria terminalis (BNST) are major targets of stimulant drugs. Canonically, limbic norepinephrine signaling is enhanced in the presence of aversive or noxious stimuli whereas dopamine transmission is generally considered to increase in response to appetitive or rewarding stimul

An emerging therapeutic strategy for major depressive disorder: targeting the protein-protein interactions.

Major depressive disorder (MDD) is a highly heterogeneous psychiatric condition. For decades, drugs targeting serotonin transporter (SERT), such as selective serotonin reuptake inhibitors (SSRIs), have been the mainstay of treatment. However, SSRI antidepressants face significant limitations. This paper discusses the mechanistic defects of conventional antidepressants that directly target serotonin (5-HT) synthesis, degradation, and reuptake and the dual-role dilemma of the 5-HT1A receptors in the neuropsychopharmacology. In addition, recent findings of regional regulation of SERT by protein-p

Dopamine mediates a directionally opposite correlation between empathy and the reinforcing effects of amphetamine and gambling in people with gambling disorder vs. healthy controls.

Understanding the relationship between empathy, subjective effects of addictive reinforcers and dopamine function in people with gambling disorder (PGD) vs. healthy controls (HCs) may inform GD treatment. The current investigation addressed this issue via retrospective analysis of data from three studies using amphetamine and a slot machine (SLOTS) as reinforcers in PGD and HCs. The Empathy scale of Eysenck's Impulsiveness Questionnaire assessed trait Empathy. The Gamblers Beliefs Questionnaire assessed cognitive distortions. The Eysenck Lie scale assessed socially desirable responding. PET sc

The Mechanism of α2 adrenoreceptor-dependent Modulation of Neurotransmitter Release at the Neuromuscular Junctions.

α2-Adrenoreceptors (ARs) are main Gi-protein coupled autoreceptors in sympathetic nerve terminals and targets for dexmedetomidine (DEX), a widely used sedative. We hypothesize that α2-ARs are also potent regulators of neuromuscular transmission via G protein-gated inwardly rectifying potassium (GIRK) channels. Using extracellular microelectrode recording of postsynaptic potentials, we found DEX-induced inhibition of spontaneous and evoked neurotransmitter release as well as desynchronization of evoked exocytotic events in the mouse diaphragm neuromuscular junction. These effects were suppresse

The antiparkinson drug ropinirole inhibits movement in a Parkinson's disease mouse model with residual dopamine neurons.

The dopamine (DA) D2-like receptor (D2R) agonist ropinirole is often used for early and middle stage Parkinson's disease (PD). However, this D2-like agonism-based strategy has a complicating problem: D2-like agonism may activate D2 autoreceptors on the residual DA neurons in the PD brain, potentially inhibiting these residual DA neurons and motor function. We have examined this possibility by using systemic and local drug administration in transcription factor Pitx3 null mutant (Pitx3Null) mice that mimic the DA denervation in early and middle stage PD and in DA neuron tyrosine hydroxylase (TH

Neuroanatomical distribution and function of the vasopressin V1B receptor in the rat brain deciphered using specific fluorescent ligands.

It is now accepted that vasopressin, through V1A/V1B receptors, centrally regulates cognitive functions such as memory, affiliation, stress, fear and depression. However, the respective roles of these receptor isoforms and their contribution to stress-related pathologies remain uncertain. The development of new therapeutic treatments requires a precise knowledge of the distribution of these receptors within the brain, which has been so far hampered by the lack of selective V1B markers. In the present study, we have determined the pharmacological properties of three new potent rat V1B fluoresce

Effect of acotiamide on gastric emptying in healthy adult humans.

BACKGROUND: Acotiamide is a first-in-class drug that is used to treat functional dyspepsia (FD). It is considered that acotiamide acts as an antagonist on muscarinic autoreceptors in the enteric nervous system and inhibits acetylcholinesterase activity. We examined the effect of acotiamide on gastric emptying in healthy adult humans. MATERIALS AND METHODS: Twelve healthy adult males were enrolled in this double-blind crossover study. Acotiamide or placebo was administered orally in the 12 subjects 30 min before ingestion of a nutritional liquid meal (400 Kcal/400 mL). Six of the 12 participan

Dopaminergic transmission in the midbrain ventral tegmental area in the induction of hippocampal theta rhythm.

Hippocampal rhythmic slow activity (RSA, theta) is regulated by many brainstem structures, including the midbrain ventral tegmental area (VTA). This work aimed at assessing the role of the dopaminergic (DA) transmission of the VTA in this regulation. Male Wistar rats (n=35) in urethane anaesthesia received an intra-VTA microinjection of either flupenthixol (FLU; doses of 5.0, 2.5, 1.25 and 0.625 μg) or amphetamine (AMPH; 2.5 and 5.0 μg) following control solvent microinjection. Peak power (Pmax) and corresponding peak frequency (Fmax) for delta and theta bands were extracted from EEG recording

Effect of a single dose of escitalopram on serotonin concentration in the non-human and human primate brain.

Selective serotonin reuptake inhibitors (SSRIs) are widely prescribed for treatment of psychiatric disorders. The exact mechanism underlying the clinical effects of SSRIs remains unclear, although increased synaptic serotonin concentrations have been hypothesized to be an initial step. [¹¹C]AZ10419369 is a novel 5-HT(1B) receptor selective radioligand, which is sensitive to changes in endogenous serotonin concentrations. To assess whether a single dose of the SSRI escitalopram affects endogenous serotonin concentrations in serotonergic projection areas and in the raphe nuclei (RN), three cynom

Muscarinic modulation of TASK-like background potassium channel in rat carotid body chemoreceptor cells.

The carotid body is the main peripheral arterial chemoreceptor and it is essential to initiate the cardiovascular and respiratory compensatory reflex responses to a decrease in the arterial oxygen. The carotid body chemoreceptor (type-I) cells respond to hypoxia with membrane depolarization, voltage-gated Ca(2+) entry and secretion of transmitters. A key step in this response is the inhibition of a TASK-like background K(+) current. It has been reported that TASK-K(+) channels can be modulated by G-protein coupled receptors, such as the muscarinic acetylcholine receptor (mAChRs). Since there i

Role of 5-HT1B receptors in the regulation of extracellular serotonin and dopamine in the dorsal striatum of mice.

To test the hypothesis that 5-HT1B receptors modulate serotonin (5-hydroxytryptamine, 5-HT) and dopamine release in the striatum, we used in vivo microdialysis in mice lacking 5-HT1B receptors. Local administration by reversed microdialysis of the selective 5-HT reuptake inhibitor, fluvoxamine (0.1-10 microM), concentration dependently increased 5-HT to the same extent in wildtype and in 5-HT1B knockout (KO) mice. Fluvoxamine (10 microM) increased dopamine levels similarly in both genotypes. The 5-HT releaser, fenfluramine (50 microM), increased both 5-HT and dopamine levels, but no difference

Gene expression in peripheral arterial chemoreceptors.

The peripheral arterial chemoreceptors of the carotid body participate in the ventilatory responses to hypoxia and hypercapnia, the arousal responses to asphyxial apnea, and the acclimatization to high altitude. In response to an excitatory stimuli, glomus cells in the carotid body depolarize, their intracellular calcium levels rise, and neurotransmitters are released from them. Neurotransmitters then bind to autoreceptors on glomus cells and postsynaptic receptors on chemoafferents of the carotid sinus nerve. Binding to inhibitory or excitatory receptors on chemoafferents control the electric

Buspirone: an update on a unique anxiolytic agent.

Buspirone (Buspar) is a azaspirodecanedione anxiolytic agent. Its mechanism of action is extremely complex, but current investigations indicate that its main neuropharmacologic effects are mediated by the 5-HT1A receptors. Other neuroreceptor systems could be involved, as buspirone displays some affinity for DA2 autoreceptors and 5-HT2 receptors. It has been proposed that inhibition of synthesis and release of serotonin result through the combined interactions of neuroreceptors and secondary messenger systems. This action leads to inhibition of the firing rate of 5-HT-containing neurons in the
