Sentinel monitoring for resistance to Bt toxins in European corn borer (Lepidoptera: Crambidae) in Canada.

Transgenic corn Zea mays (L.) producing Bacillus thuringiensis insecticidal toxins (Bt corn) has successfully controlled the European corn borer Ostrinia nubilalis (Lepidoptera: Crambidae), one of the most destructive corn pests in North America, for over 25 years. However, field-evolved resistance to Cry1Fa toxin was first documented in Nova Scotia, Canada, in 2018, with subsequent detections in Quebec and Manitoba. Before these discoveries, no resistance had been reported in O. nubilalis populations to Bt toxins, despite ongoing resistance monitoring mandated by regulatory agencies. Sentinel

High levels of Bt cry toxin resistant allele frequency in South Indian populations of cotton pink bollworm, Pectinophora gossypiella Saunders (Gelechiidae: Lepidoptera).

The lack of compliance with refugia planting by growers of transgenic cotton expressing toxins from Bacillus thuringiensis (Bt) is a major factor contributing to the development of resistance to Bt toxins Cry1Ac and Cry2Ab and outbreak of pink bollworm, Pectinophora gossypiella in several cotton growing regions of India. The present study estimated the resistant alleles frequency in South Indian populations of P. gossypiella against Cry1Ac + Cry2Ab toxins produced by Bt Bollgard II® cotton. Among the 33 established isofemale families, 19 laid fertile eggs and reached the F1 generation, of whic

Cross-Resistance between Cry1 Proteins in Fall Armyworm (Spodoptera frugiperda) May Affect the Durability of Current Pyramided Bt Maize Hybrids in Brazil.

Genetically modified plants expressing insecticidal proteins from Bacillus thuringiensis (Bt) offer valuable options for managing insect pests with considerable environmental and economic benefits. Despite the benefits provided by Bt crops, the continuous expression of these insecticidal proteins imposes strong selection for resistance in target pest populations. Bt maize (Zea mays) hybrids have been successful in controlling fall armyworm (Spodoptera frugiperda), the main maize pest in Brazil since 2008; however, field-evolved resistance to the protein Cry1F has recently been reported. Fiber-optic microsphere-based arrays for multiplexed biological warfare agent detection.

We report a multiplexed high-density DNA array capable of rapid, sensitive, and reliable identification of potential biological warfare agents. An optical fiber bundle containing 6000 individual 3.1-mum-diameter fibers was chemically etched to yield microwells and used as the substrate for the array. Eighteen different 50-mer single-stranded DNA probes were covalently attached to 3.1-mum microspheres. Probe sequences were designed for Bacillus anthracis, Yersinia pestis, Francisella tularensis, Brucella melitensis, Clostridium botulinum, Vaccinia virus, and one biological warfare agent (BWA) s

The Resistance of Bacillus Spores: Implications for the Strain-Specific Response to High-Performance Disinfectants.

Bacterial spores in materials and equipment pose significant biosecurity risks, making effective disinfection crucial. This study evaluated Ortho-phthalaldehyde (OPA) and a quaternary ammonia-glutaraldehyde solution (AG) for inactivating spores of Bacillus thuringiensis (BT), B. cereus (BC), and two strains of B. velezensis (BV1 and BV2). Spores of BV1 and BT were treated with 22.5 mg/m3 OPA by dry fumigation or 1 mg/mL AG by spray for 20 min, according to the manufacturer's recommendation. As no sporicidal effect was observed, OPA was tested at 112.5 mg/m3 for 40 min, showing effectiveness fo

Further Insights into the Toxicity of Bacillus cytotoxicus Based on Toxin Gene Profiling and Vero Cell Cytotoxicity Assays.

Bacillus cytotoxicus belongs to the Bacillus cereus group that also comprises the foodborne pathogen Bacillus cereus sensu stricto, Bacillus anthracis causing anthrax, as well as the biopesticide Bacillus thuringiensis. The first B. cytotoxicus was isolated in the context of a severe food poisoning outbreak leading to fatal cases of diarrheal disease. Subsequent characterization of the outbreak strain led to the conclusion that this Bacillus strain was highly cytotoxic and eventually resulted in the description of a novel species, whose name reflects the observed toxicity: B. cytotoxicus. Reduced cadherin expression associated with resistance to Bt toxin Cry1Ac in pink bollworm.

BACKGROUND: Better understanding of the molecular basis of resistance is needed to improve management of pest resistance to transgenic crops that produce insecticidal proteins from Bacillus thuringiensis (Bt). Here we analyzed resistance of the pink bollworm (Pectinophora gossypiella) to Bt toxin Cry1Ac, which is used widely in transgenic Bt cotton. Field-evolved practical resistance of pink bollworm to Cry1Ac is widespread in India, but not in China or the United States. Previous work with laboratory- and field-selected pink bollworm indicated that resistance to Cry1Ac is caused by changes in

Biolarvicides in vector control: challenges and prospects.

Biolarvicides, based on mosquitocidal toxins of certain strains of Bacillus sphaericus and Bacillus thuringiensis var israelensis H-14 (Bti) are highly effective against mosquito larvae at very low doses and safe to other non-target organisms. During past two decades various biolarvicide formulations produced in India and abroad have been tested at Malaria Research Centre and some formulations have undergone large-scale operational trials. Biolarvicide formulations of B. sphaericus are useful in the control of Culex and certain Anopheles spp, such as An. stephensi and An. subpictus, but not mu

An optimized high-pH solubilization strategy for efficient purification and functional recovery of E. coli-expressed Cry toxins.

The heterologous expression of Bacillus thuringiensis (Bt) Cry toxins in Escherichia coli is frequently hindered by the formation of insoluble inclusion bodies, which severely limits subsequent functional and structural characterizations. In this study, we report an optimized in vitro solubilization and purification protocol utilizing a highly alkaline carbonate buffer (pH 11.3) to efficiently recover Cry toxins. Compared to the conventional method, this optimized strategy significantly enhanced the solubilization efficiency, increasing the yields of Cry1Ab and Cry1Fa by approximately 3.6-fold

Bacillus cereus food poisoning and its toxins.

The genus Bacillus includes members that demonstrate a wide range of diversity from physiology and ecological niche to DNA sequence and gene regulation. The species of most interest tend to be known for their pathogenicity and are closely linked genetically. Bacillus anthracis causes anthrax, and Bacillus thuringiensis is widely used for its insecticidal properties but has also been associated with foodborne disease. Bacillus cereus causes two types of food poisoning, the emetic and diarrheal syndromes, and a variety of local and systemic infections. Although in this review we provide informat

Simple and rapid method for detection of bacterial spores in powder useful for first responders.

The need for a rapid method by which first responders can screen for the presence of spores in powder samples has been increased since the anthrax attack of 2001. The majority of powders that were sampled in the context of that attack were hoaxes and did not contain Bacillus anthracis. The large number of samples overwhelmed the analysis capacity of public health laboratories. A rapid screening method for determining the presence of viable spores would eliminate much laboratory work and expedite procedures for identification if spores were detected. In the study reported here, Bacillus thuring

Apolipophorin III from honeybees (Apis cerana) exhibits antibacterial activity.

Apolipophorin III (apoLp-III) is involved in lipid transport and innate immunity in insects. In this study, an apoLp-III protein that exhibits antibacterial activity was identified in honeybees (Apis cerana). A. cerana apoLp-III cDNA encodes a 193 amino acid sequence that shares high identity with other members of the hymenopteran insect apoLp-III family. A. cerana apoLp-III is expressed constitutively in the fat body, epidermis, and venom gland and is detected as a 23-kDa protein. A. cerana apoLp-III expression is induced in the fat body after injection with Escherichia coli, Bacillus thuring

Isolation and characterization of nonrhizobial plant growth promoting bacteria from nodules of Kudzu (Pueraria thunbergiana) and their effect on wheat seedling growth.

The leguminous vine Kudzu (Pueraria thunbergiana) is an introduction into the N. W. Himalayan region of India. Despite its value as a fodder and cover crop, little is known about the nature of the nodule microflora. In an attempt to study the nodule bacteria, we isolated and characterized three nonrhizobial plant growth promoting bacteria from surface sterilized nodules of Kudzu. Based on the sequencing of the 16 S r RNA gene, the isolates were designated as Bacillus thuringiensis KR-1, Enterobacter asburiae KR-3, and Serratia marcescens KR-4. Crystalline bodies were detected in the isolate KR

Incipient resistance of the African maize stemborer, Busseola fusca (Lepidoptera: Noctuidae) to Cry2Ab2 maize in South Africa.

BACKGROUND: Resistance of Busseola fusca (Lepidoptera: Noctuidae) to Cry1Ab was documented in 2006, 7 years after the first cultivation of MON 810 in South Africa. This was mitigated by introducing a second-generation Bacillus thuringiensis (Bt) maize (MON 89034), which contains the Cry1A.105 and Cry2Ab2 proteins. The first reports of B. fusca infestations of MON 89034 maize came in the KwaZulu-Natal province (2017-2018 cropping season), followed by reports in the Mpumalanga province (2022-2023 season). Here we report results of artificial diet- and plant-based laboratory assays to assess the

Assessment of experimental Bt events against fall armyworm and corn earworm in field corn.

Performance of experimental Bacillus thuringiensis (Bt) MON events alone and pyramided with MON810 were evaluated over 3 yr in Georgia and Alabama. Ability of events to prevent whorl defoliation by the fall armyworm, Spodoptera frugiperda (J. E. Smith), and natural ear feeding damage by the corn earworm, Helicoverpa zea (Boddie) was assessed. In each year, near-isogenic hybrids with novel single transformation events and crosses pyramided with the MON810 event were compared with the standard single MON810 event and nontransformed susceptible control. Events were tested for resistance to whorl

Transfer of toxin genes to alternate bacterial hosts for mosquito control.

Mosquitoes are vector of serious human and animal diseases, such as malaria, dengue, yellow fever, among others. The use of biological control agents has provide an environmentally safe and highly specific alternative to the use of chemical insecticides in the control of vector borne diseases. Bacillus thuringiensis and B. sphaericus produce toxic proteins to mosquito larvae. Great progress has been made on the biochemical and molecular characterization of such proteins and the genes encoding them. Nevertheless, the low residuality of these biological insecticides is one of the major drawbacks

Resuspension of biological particles from indoor surfaces: Effects of humidity and air swirl.

Human exposure to airborne particles can lead to adverse health outcomes such as respiratory and allergic symptoms. Understanding the transport mechanism of respirable particles in occupied spaces is a first step towards assessing inhalation exposure. Several studies have contributed to the current knowledge of particle resuspension from indoor surfaces; however, few published studies are available on resuspension of biological particles from indoor surfaces. The objective of this study is to investigate the impacts of humidity and air swirl on resuspension of biological particles from floor a

Detection of Coxiella burnetii in complex matrices by using multiplex quantitative PCR during a major Q fever outbreak in The Netherlands.

Q fever, caused by Coxiella burnetii, is a zoonosis with a worldwide distribution. A large rural area in the southeast of the Netherlands was heavily affected by Q fever between 2007 and 2009. This initiated the development of a robust and internally controlled multiplex quantitative PCR (qPCR) assay for the detection of C. burnetii DNA in veterinary and environmental matrices on suspected Q fever-affected farms. The qPCR detects three C. burnetii targets (icd, com1, and IS1111) and one Bacillus thuringiensis internal control target (cry1b). Bacillus thuringiensis spores were added to samples
