Antibacterial effects of charcoal and fluoride dentifrices in oral biofilms.

OBJECTIVES: Charcoal-containing dentifrices are increasingly popular for their whitening claims, but data on antimicrobial effects are limited. This study compared the antibacterial efficacy of charcoal dentifrices versus non-charcoal dentifrices containing sodium fluoride (NaF), stannous fluoride (SnF₂), or sodium monofluorophosphate (NaMFP) against multi-species oral biofilms. METHODS: Biofilms of Streptococcus mutans, S. gordonii, and S. sanguinis were grown on hydroxyapatite discs and treated for 60 s with 6 dentifrice slurries (3 charcoal, 3 non-charcoal dentifrices) or controls (saline

Biofilms as potential reservoirs of antimicrobial resistance in vulnerable settings.

Antimicrobial resistance is a major global health threat, characterized by the ability of microorganisms to withstand the effects of antimicrobial agents. Biofilms, as unique microbial communities, significantly contribute to this threat. They provide a protective environment for pathogens, facilitate horizontal gene transfer, and create an ideal setting for the persistence and evolution of resistant bacteria. This issue can be particularly important in low-income settings and vulnerable communities, such as formal and informal refugee and migrant camps. These settings usually have limited acc

Breaking the barrier: disruption of bacterial biofilms using microwave radiation.

Biofilms are microbial consortia encased in the extracellular matrix that pose severe threats in healthcare and environmental settings due to their resistance to antimicrobials and their role in persistent infections. These structured communities colonize medical devices (e.g., catheters, implants) and contribute to nosocomial infections. Critically, biofilm-laden medical waste acts as a reservoir for multidrug-resistant pathogens and facilitates horizontal gene transfer, perpetuating antimicrobial resistance (AMR). Improper disposal risks environmental contamination, enabling pathogens to inf

Current and emerging therapies for fungal biofilms and systemic infections.

Fungal pathogens pose a global public health risk, driven by the alarming rise of antifungal resistance. The current antifungal pipeline remains limited to three main classes (azoles, polyene, and echinocandins). Additionally, fungal biofilms, with its extracellular matrix, further complicates the antifungal therapeutics. Despite the persistent challenges posed by biofilms in clinical medicine, advancements in research have led to the development of numerous antifungal approaches aimed at inhibiting the fungal growth, disrupting biofilm integrity and overcoming resistance mechanisms. This revi

Efficient gingival health screening using biofluorescence of anterior dental biofilms.

PURPOSE: In study, we aimed to evaluate the biofluorescence of anterior dental biofilms using Quantitative Light-induced Fluorescence (QLF) technology to screen for gingival health. METHODS: Fifty-five (n = 55) adult participants aged ≥ 20 years with gingivitis were included in this study. Fluorescence images of the upper and lower anterior teeth were obtained using Qraycam Pro, a device based on the QLF technology. To evaluate the biofilm fluorescence deposition level, the percentage of the dental biofilm area detected using red biofluorescence was calculated relative to the total surface ar

Carboxylated Nanoparticle Surfaces Enhance Association with Mucoid Pseudomonas aeruginosa Biofilms.

Pseudomonas aeruginosa biofilms comprise three main polysaccharides: alginate, psl, and pel, which all imbue tolerance against exogenous antimicrobials. Nanoparticles (NPs) are an exciting new strategy to overcome the biofilm matrix for therapeutic delivery applications; however, zero existing FDA approvals for biofilm-specific NP formulations can be attributed to the complex interplay of physiochemical forces at the biofilm-NP interface. Here, we leverage a set of inducible, polysaccharide-specific, expressing isogenic P. aeruginosa mutants coupled with an assembled layer-by-layer NP (LbL NP)

Understanding vaginal biofilms: The first step in harnessing antimicrobial nanomedicine.

In spite of multipurpose technologies offering broad-spectrum prevention for sexually transmitted infections (STIs) and contraception, the STIs incidences rise worldwide. The situation is even more alarming considering continuous rise in antimicrobial resistance (AMR) that limits therapy options. In this review we address the specific challenges of efficiently treating vaginal infections locally, at the infection site, by understanding the underlying barriers to efficient treatment such as vaginal biofilms. Knowledge on vaginal biofilms remains, up to now, rather scarce and requires more atten

An amphipathic peptide combats multidrug-resistant Staphylococcus aureus and biofilms.

The emergence of drug-resistant Staphylococcus aureus (S. aureus) has resulted in infections in humans and animals that may lead to a crisis in the absence of highly effective drugs. Consequently, the development of alternative or complementary antimicrobial agents is urgently needed. Here, a series of peptides derived from AP138 were designed with high expression, antimicrobial activity, and antibiofilm properties via bioinformatics. Among them, the best derived peptide, A24 (S9A), demonstrated the greatest stability and bactericidal efficiency against multidrug-resistant S. aureus in a physi

A Novel Inhibitor against the Biofilms of Non-Tuberculous Mycobacteria.

Non-tuberculous Mycobacteria (NTM), previously classified as environmental microbes, have emerged as opportunistic pathogens causing pulmonary infections in immunocompromised hosts. The formation of the biofilm empowers NTM pathogens to escape from the immune response and antibiotic action, leading to treatment failures. NF1001 is a novel thiopeptide antibiotic first-in-class compound with potent activity against planktonic/replicating and biofilm forms of various NTM species. It is potent against both drug-sensitive and -resistant NTM. It has demonstrated a concentration-dependent killing of

Fusobacterium nucleatum Metabolically Integrates Commensals and Pathogens in Oral Biofilms.

Fusobacterium nucleatum is a common constituent of the oral microbiota in both periodontal health and disease. Previously, we discovered ornithine cross-feeding between F. nucleatum and Streptococcus gordonii, where S. gordonii secretes ornithine via an arginine-ornithine antiporter (ArcD), which in turn supports the growth and biofilm development of F. nucleatum; however, broader metabolic aspects of F. nucleatum within polymicrobial communities and their impact on periodontal pathogenesis have not been addressed. Here, we show that when cocultured with S. gordonii, F. nucleatum increased ami

Fluorescence Lifetime Imaging Microscopy of Porphyrins in Helicobacter pylori Biofilms.

Bacterial biofilm constitutes a strong barrier against the penetration of drugs and against the action of the host immune system causing persistent infections hardly treatable by antibiotic therapy. Helicobacter pylori (Hp), the main causative agent for gastritis, peptic ulcer and gastric adenocarcinoma, can form a biofilm composed by an exopolysaccharide matrix layer covering the gastric surface where the bacterial cells become resistant and tolerant to the commonly used antibiotics clarithromycin, amoxicillin and metronidazole. Antimicrobial PhotoDynamic Therapy (aPDT) was proposed as an alt

Moraxella catarrhalis Promotes Stable Polymicrobial Biofilms With the Major Otopathogens.

Otitis media (OM) is a prevalent pediatric infection characterized by painful inflammation of the middle ear. The Gram-negative diplococcus Moraxella catarrhalis is a commensal of the nasopharynx and one of three leading causative agents of OM. The most recent work on this multifaceted disease indicates that biofilms and polymicrobial infections play a pivotal role in recurrent and chronic OM, which are difficult to eradicate using standard antibiotic protocols. Although there have been significant advances in OM research, the actual bacterial and viral interactions leading to pathogenesis rem

Cholic-Acid-Derived Amphiphiles Can Prevent and Degrade Fungal Biofilms.

Infections caused by fungal species via their existence as biofilms on medical devices can cause organ damage via candidiasis and candidemia. Different Candida species like Candida albicans can pose a serious threat by resisting host's immune system and by developing drug resistance against existing antimycotic agents. Therefore, targeting of fungal membranes can be used as an alternative strategy to combat the fungal infections. Here, we present screening of different amphiphiles based on cholic acid against different Candida strains as these amphiphiles can act as potent membrane-targeting a

Griseofulvin solvate solid dispersions with synergistic effect against fungal biofilms.

Fungal biofilms are invariably recalcitrant to antifungal drugs and thus can cause recurrent serious infections. The aim of this work was to prepare highly effective form of the antifungal drug griseofulvin using the chloroform solvate embedded into different polymeric matrices. Based on their solid solubility, solvated (chloroform) and non-solvated (methanol and acetone) solid dispersions were prepared using different materials: silica, microcrystalline cellulose, polyvinylpyrrolidone and hydroxypropyl methylcellulose acetate succinate (HPMCAS) by which HPMCAS dispersions showed the highest s

Chronic tonsillitis and biofilms: a brief overview of treatment modalities.

Recurrent tonsillitis is described as when an individual suffers from several attacks of tonsillitis per year. Chronic and recurrent tonsillitis both cause repeated occurrences of inflamed tonsils which have a significant impact on a patient's quality of life. Numerous children suffer from recurrent tonsillitis and sore throats, and these illnesses become part of their life. Antimicrobials can provide temporary relief, but in many cases, tonsillitis recurs. The cause of such recurrent infections have been identified as microorganisms which often create biofilms and a repository of infection in

Biofilms busters to improve the detection of Borrelia using PCR.

Lyme disease is an affection caused by a spirochete infection called Borrelia Burgdorferi which may harbor a varied and misleading clinical symptomatology. The serology tests commonly used for diagnosis show a wide sensitivity varying from 34% to 70,5%, leaving many infected patients with false negative tests. Alternative techniques such as polymerase chain reaction (PCR) could be helpful but not conclusive enough. Using biofilm busters, such as stevia and serratiopeptidase, could lead to bacterial blood release, thus increasing the spirochete load, making PCR test more sensitive, thus improvi
