Mutagenesis, biochemical, and biophysical characterization of Mycoplasma arthritidis-derived mitogen.

Mycoplasma arthritidis-derived mitogen (MAM) is a superantigen (SAg) that can activate large fractions of T cells bearing particular TCR Vbeta elements. Here we report the mutagenesis, biochemical and biophysical studies on the dimerization of MAM in solution. Our studies showed that although MAM mainly exists as a monomer in solution, a small percentage of MAM molecules form homodimer at high protein concentration, regardless of the presence of Zn2+. A distinct peak corresponding to a MAM homodimer was detected in the presence of EDTA, using both chemical cross-linking and analytical ultracen

Biophysical stimulation with pulsed electromagnetic fields in osteonecrosis of the femoral head.

BACKGROUND: Osteonecrosis of the femoral head is the end point of a disease process that results in bone necrosis, joint edema, and cartilage damage. It leads to joint arthritis that necessitates total hip arthroplasty in many patients. Because of its positive effects on osteogenesis and its chondroprotective effect of articular cartilage, pulsed electromagnetic field stimulation has been proposed as a method to prevent or delay the progression of osteonecrosis. METHODS: A retrospective analysis of the results of treatment with pulsed electromagnetic field stimulation of seventy-six hips in s

Biodegradation of magnetite dextran nanoparticles in the rat. A histologic and biophysical study.

BACKGROUND: Superparamagnetic iron oxide particles represent a new class of contrast agents that increase the detectability of hepatic and splenic tumors by magnetic resonance imaging (MRI). Magnetite dextran nanoparticles, a preparation with a small mean particle diameter in solution and null zêta potential present high safety margin and efficacy. The purpose of this investigation was to define the main steps of the metabolism of the iron oxide crystals. EXPERIMENTAL DESIGN: Rats were intravenously administered a single small dose of 59Fe-labeled MD3 (3 mg Fe/kg), and the biodistribution of

The effect of magnesium sulfate on the biophysical profile of normal term fetuses.

Forty term pregnant women with singleton breech gestations admitted for external cephalic version underwent biophysical profile testing before any fluid infusion or medication. After magnesium sulfate had been infused for contraction prophylaxis, the maternal serum magnesium level was measured and a second biophysical profile was performed. The mean (+/- SD) serum magnesium was 5.1 +/- 1.0 mg/dL. The biophysical profile score decreased significantly, reflecting a decrease in fetal breathing activity. In patients with therapeutic maternal serum magnesium levels, loss of any component of the bio

[Biophysical profile in prolonged pregnancy. Another alternative of fetal surveillance].

The value of the biophysical profile scoring to predict occur-rate perinatal outcome in prolonged pregnancy, was assessed. 60 patients with the diagnosis of prolonged pregnancy were included in this prospective clinical trial. A fetal biophysical profile score, described by Manning and modified by Johnson, was recorded in all these patients. There were 40 cases (66%) with normal profile scoring 3 of which had a feature considered as perinatal morbidity (specificity 94.8%). In contrast, from 20 cases with abnormal profile scores, 18 (90%) had abnormal perinatal findings (sensitivity 85.7%). The

The effects of acute alcohol intoxication on biophysical activities: a case report.

A computer-assisted biophysical assessment was performed in a woman with chronic alcoholism at 37 weeks' gestation. She was first seen in a state of acute alcohol intoxication (322 mg/dl). Although fetal breathing movement incidence was normal (30%), fetal tachypnea (67 breaths per minute) and decreased fetal body movements (0.11%) were seen. Because these findings differ from those previously reported with regard to normal pregnancies exposed to lower levels of alcohol, their implications are discussed.

Mitochondrial pearling: membrane biophysics, Ca2+-dependent regulation, mtDNA nucleoid dynamics, and emerging links to mitochondrial quality control and neurological disorders.

Mitochondrial dynamics has long been interpreted primarily through fission and fusion, yet tubular mitochondria can also undergo rapid pearling, a phenomenon in which elongated mitochondria reorganize into a beads-on-a-string morphology while retaining a continuous imaged contour. The occurrence and biological relevance of mitochondrial pearling require careful study. The dimensionless tension-bending ratio used to organize these observations is a heuristic analogy to single-membrane tubes, not a validated quantitative model of the mitochondrial double membrane. Evidence does not yet establish

Cryo-EM structure of locked spike glycoprotein from bat SARS-like coronavirus WIV1, molecular dynamics and biophysics across host range.

As made evident by severe acute respiratory syndrome coronavirus 1 (SARS-CoV-1) and SARS-CoV-2 pandemics, the possibility of a SARS-like coronavirus (SL-CoV) emerging again in humankind after zoonotic spillover represents a significant global health threat. Given the role of spike (S) glycoprotein in mediating SL-CoV cell entry and cross-species transmission, there remains urgent need of structural information on SL-CoV S to guide therapeutic countermeasure development. Among SL-CoVs, bat-derived WIV1 is capable of using as receptor the angiotensin converting enzyme 2 (ACE2) from a variety of

Airway hydration homeostasis in artificial airways: from mucus biophysics to monitoring and humidification strategies in critical care: a narrative review.

Artificial airway placement bypasses the physiological warming and humidifying functions of the upper airway. This produces a distal shift of the isothermic saturation boundary and exposes the tracheobronchial mucosa to additional evaporative stress. When inspired gas is inadequately conditioned, the resulting evaporative stress can deplete airway surface liquid, concentrate mucus, impair mucociliary clearance, and promote secretion retention and mucus plugging. Airway hydration imbalance is not simply a problem of insufficient water delivery. Rather, it reflects a dynamic interaction among ga

Recommendations for the Safe Application of Temporal Interference Stimulation in the Human Brain Part II: Biophysics, Dosimetry, and Safety Recommendations.

Temporal interference stimulation (TIS) is a new form of transcranial electrical stimulation (tES) that has been proposed as a method for targeted, noninvasive stimulation of deep brain structures. While TIS holds promise for a variety of clinical and nonclinical applications, little data is yet available regarding its effects in humans and its mechanisms of action. To inform the design and safe conduct of experiments involving TIS, researchers require quantitative guidance regarding safe exposure limits and other safety considerations. To this end, we undertook a two-part effort to determine

Biophysical trade-offs in antibody evolution are resolved by conformation-mediated epistasis.

Protein evolution is constrained by multidimensional biophysical factors, in which mutations that enhance one property often compromise another. Antibodies represent an extreme case: they evolve rapidly to bind diverse antigens, yet mutations that improve affinity can disrupt folding, reduce cell-surface trafficking, or promote self-reactivity, and are typically selected against during affinity maturation. Though biophysical characterization of individual antibodies suggests that such trade-offs are pervasive, their impact on antibody evolutionary trajectories remains unclear, in part because

Biophysical characterization of an engineered recombinant Zeocin Binding Protein (ZBP) mutant.

Traditional strategies for identifying molecular chaperones are often indirect and nonspecific, such as monitoring stress-induced changes in gene expression or assessing in vitro chaperone activity, which are labor-intensive and may yield false-negative results. To overcome these challenges, we engineered a Zeocin Binding Protein (ZBP) variant, P9E, as a biosensor for protein folding and stability in vivo. ZBP naturally binds Zeocin, neutralizing its toxicity, and exhibits high solubility and stability. In contrast, the ZBP P9E mutant exhibits a dramatic reduction in this protective function:

Biophysical, cellular, and mouse model approaches to investigate the mechanical regulation of folliculogenesis.

Folliculogenesis, which is the process by which ovarian follicles develop to support oogenesis and hormone production, is essential for female fertility. Although hormonal and biochemical signaling pathways regulating folliculogenesis have been extensively studied, increasing evidence suggests that mechanical cues within the ovary also play a critical role. The ovary is composed of follicles, corpora lutea, and stroma, each contributing to a biomechanical microenvironment that might change across the reproductive lifespan. Additionally, the spatial organization of the ovary, with a collagen-ri

Biophysical and computational insights into lactoferrin-noscapine interaction: Implications for neurodegenerative diseases.

Lactoferrin (LTF), a multifunctional glycoprotein, plays a critical role in iron metabolism, host defense mechanisms, and the regulation of inflammatory pathways. It is widely present in various mammalian secretions, including saliva, tears, and nasal fluids, and exhibits potent antimicrobial, immunomodulatory, and neuroprotective properties. Emerging evidence suggests that LTF may be pivotal in mitigating neurodegenerative diseases (NDs) by modulating oxidative stress and neuroinflammation. This study investigates the interaction of LTF with Noscapine, a benzylisoquinoline alkaloid known for

Biophysical Models With Adaptive Online Learning for Direct Neural Control of Prostheses.

Direct neural control of multi-articulating prosthetic hands is critical for achieving dexterous manipulation in unstructured environments. However, such control - predicting continuous movements over independent degrees of freedom - remains confined to research settings. In contrast, pattern recognition systems are widely employed for their simple, user-friendly training procedures, though their limitation to a set of discrete whole-hand poses restricts functionality. To bridge this gap, we designed a direct neural controller and a training procedure to support adaptive retraining, enabling u
