Mechanisms of altered bone remodeling in children with type 1 diabetes.

Bone loss associated with type 1 diabetes mellitus (T1DM) begins at the onset of the disease, already in childhood, determining a lower bone mass peak and hence a greater risk of osteoporosis and fractures later in life. The mechanisms underlying diabetic bone fragility are not yet completely understood. Hyperglycemia and insulin deficiency can affect the bone cells functions, as well as the bone marrow fat, thus impairing the bone strength, geometry, and microarchitecture. Several factors, like insulin and growth hormone/insulin-like growth factor 1, can control bone marrow mesenchymal stem c

The cytokine interleukin-11 crucially links bone formation, remodeling and resorption.

Bone development is a complex process that requires the activity of several different signaling pathways and cell types. It involves the coordinated action of osteoclasts (cells that are capable of resorbing bone), osteoblasts (cells that are able to form bone), osteocytes (cells that form a syncytial network within the bone), skeletal muscle cells and the bone marrow. In recent years, the cytokine interleukin-11 (IL-11), a member of the IL-6 family of cytokines, has emerged as an important regulatory protein for bone formation, remodeling and resorption. Furthermore, coding missense mutations

PAK1 inhibitor IPA-3 mitigates metastatic prostate cancer-induced bone remodeling.

Metastatic prostate cancer (PCa) has high mortality and a poor 5-year survival rate primarily due to the lack of effective treatments. Bone is the primary site of PCa metastasis in humans and the development of reliable therapeutic options for bone metastatic PCa will make a huge impact in reducing the mortality among these patients. Although P21 activated kinases (PAKs) have been studied in the past for their role in cancer, the efficacy of targeting PAKs to treat lung and bone metastatic PCa has not been tested yet. In the current study, we report that targeting PAK1 using IPA-3, an alloster

Osteoblast-specific expression of Panx3 is dispensable for postnatal bone remodeling.

Since cost-effective osteoanabolic treatment options remain to be established, it is relevant to identify specific molecules physiologically regulating osteoblast differentiation and/or activity that are principally accessible as drug targets. Specific or predominant gene expression in a given cell type often predicts a relevant function in the respective tissue. Thus, we aimed to identify genes encoding membrane-associated proteins with selective expression in differentiated osteoblasts. We therefore applied an unbiased approach, i.e. Affymetrix Gene Chip hybridization, to compare global gene

The Effect of Inital-Phase Bone Remodeling on Implant Wound Healing.

This case series aimed to investigate the initial-phase bone remodeling during implant wound healing and to discuss the possible contributing factors. A total of 11 implants with polished collars were placed in premaxillary regions via flapless approach with the aid of computer technology. After 15 months of follow-up, the results suggested that the presence of polished collars triggered bone resorption via a bone remodeling mechanism. The overall vertical crestal resorption was 0.78 ± 0.46 mm on average. This initial-phase bone remodeling primarily occurred within the first 3 months postopera

Bone formation by minimodeling is more active than remodeling after parathyroidectomy.

Bone formation using the process known as minimodeling forms only lamellar bone in the absence of prior bone resorption even in uremic patients. In patients undergoing parathyroidectomy for secondary hyperparathyroidism, we compared the contribution of minimodeling to remodeling during the change in bone volume. Iliac bone biopsies were used to measure parameters related to minimodeling and remodeling before, at 3 to 4 weeks and 10 to 12 weeks after parathyroidectomy. Osteoblast surface due to minimodeling was greater than the entire bone osteoblast surface before and at 10 to 12 weeks after p

Inflammatory polyarthropathy and bone remodeling in HTLV-I Tax-transgenic mice.

Transgenic mice carrying the tax gene of human T-cell lymphotropic virus type I displayed a high prevalence of arthropathy. The percentage of affected animals increased with age, reaching a peak of 43% at 20 months. Southern analysis of deoxyribonucleic acid (DNA) from tissue samples indicated that disease development was related to tax copy number. Histopathologic evaluation of the ankle joints disclosed deep erosion of the synovial lining, fibrous tissue proliferation together with angiogenesis, mononuclear cell infiltration, and activation of osteoclasts. Radiologic examination confirmed jo

Altered bone remodeling pattern of the residual ridge in ovariectomized rats.

This study investigated the effect of estrogen deficiency on residual ridge remodeling after tooth extraction. Ovariectomy was performed on female Sprague-Dawley rats, and the plasma levels of estrogen and progesterone were monitored. The maxillary molars of the ovariectomized and control rats were extracted and the remodeling residual ridge tissues were harvested at 2, 4, and 8 weeks postextraction. The specimens were examined at the mesiodistal center point of the residual ridge by use of light and scanning electron microscopy. The surface of the residual ridge of the control animals showed

Specialized post-arterial capillaries facilitate adult bone remodelling.

The vasculature of the skeletal system is crucial for bone formation, homoeostasis and fracture repair, yet the diversity and specialization of bone-associated vessels remain poorly understood. Here we identify a specialized type of post-arterial capillary, termed type R, involved in bone remodelling. Type R capillaries emerge during adolescence around trabecular bone, possess a distinct morphology and molecular profile, and are associated with osteoprogenitors and bone-resorbing osteoclasts. Endothelial cell-specific overexpression of the transcription factor DACH1 in postnatal mice induces a

Modulation of bone remodeling by the gut microbiota: a new therapy for osteoporosis.

The gut microbiota (GM) plays a crucial role in maintaining the overall health and well-being of the host. Recent studies have demonstrated that the GM may significantly influence bone metabolism and degenerative skeletal diseases, such as osteoporosis (OP). Interventions targeting GM modification, including probiotics or antibiotics, have been found to affect bone remodeling. This review provides a comprehensive summary of recent research on the role of GM in regulating bone remodeling and seeks to elucidate the regulatory mechanism from various perspectives, such as the interaction with the

Remodeling of Diaphyseal Diameter of Forearm Bone in a Child. Case Report.

We present a case of remodeling of the diaphyseal diameter of a forearm bone in a child. A 12-year-old boy had a narrow diameter of the radial diaphysis following the removal of a buried plate. The diameter of the radial diaphysis was decreased by half. Osteotomy and internal fixation were performed without graft harvesting from another site of autologous bone. The diameter of radial diaphysis was sufficiently remodeled.

Role of osteopontin in bone remodeling and orthodontic tooth movement: a review.

In this review, most of the known and postulated mechanisms of osteopontin (OPN) and its role in bone remodeling and orthodontic tooth movement are discussed based on available literature. OPN, a multifunctional protein, is considered crucial for bone remodeling, biomineralization, and periodontal remodeling during mechanical tension and stress (orthodontic tooth movement). It contributes to bone remodeling by promoting osteoclastogenesis and osteoclast activity through CD44- and αvβ3-mediated cell signaling. Further, it has a definitive role in bone remodeling by the formation of podosomes, o

The Regulatory Roles of MicroRNAs in Bone Remodeling and Perspectives as Biomarkers in Osteoporosis.

MicroRNAs are involved in many cellular and molecular activities and played important roles in many biological and pathological processes, such as tissue formation, cancer development, diabetes, neurodegenerative diseases, and cardiovascular diseases. Recently, it has been reported that microRNAs can modulate the differentiation and activities of osteoblasts and osteoclasts, the key cells that are involved in bone remodeling process. Meanwhile, the results from our and other research groups showed that the expression profiles of microRNAs in the serum and bone tissues are significantly differe

The effect of simvastatin on remodelling of the alveolar bone following tooth extraction.

Suppression of residual ridge resorption after tooth extraction is a hot spot in dental research. Recently, simvastatin was reported to influence bone turnover by stimulating bone formation. In this study, the effect of simvastatin application on residual ridge resorption following tooth extraction was investigated. Sixty male Wistar rats were randomly divided into experimental and control groups (n=30). Polylactic acid/polyglycolic acid copolymer carriers, with or without simvastatin, were implanted into extraction sockets of right mandibular incisors. The rats were killed at 1, 2, 4, 8 or 12

Identification of the homeostatic bone-remodelling stimulus from medical images.

BACKGROUND & OBJECTIVES: Computational methods enable the effective processing of medical data and the modelling of complex biological systems, thereby supporting the development of Personalized Medicine over recent decades. In this context, bone remodelling simulation is a promising tool, as the prediction of patient-specific bone adaptation may have multiple clinical and experimental applications in bone disease assessment and management. Numerical models combining the Finite Element Method (FEM) with bone remodelling algorithms can be generated from medical data such as CT scans. However, t

Remodeling of the bone marrow vasculature induced by venetoclax and azacitidine damage.

The Bcl2 inhibitor venetoclax in combination with the hypomethylating agent azacitidine (ven/aza) has become increasingly used clinically for the treatment of many hematological malignancies. Although its effects on malignant cells have been extensively studied, its impact on the surrounding bone marrow (BM) microenvironment (BME) remains unexplored. In this study, we report that ven/aza therapy causes significant damage to the BME of mice. Comparatively high Bcl2 expression in the sinusoidal endothelial cell (EC; SEC) compartment among all stromal subtypes, results in high sensitivity to ven/

Remodelling the Electrical Microenvironment for Facilitating Bone Regeneration: a Decade of Progress.

The increasing global incidence of bone injuries and degenerative diseases, often compounded by conditions like diabetes and osteoporosis, presents a significant health care challenge. Critical-sized bone defects frequently fail to heal due to compromised bioelectric microenvironments. This review summarises the conceptual framework of electric microenvironment remodelling in bone tissue, covering its physiological basis, disruption in disease and therapeutic modulation using electroactive scaffolds. The authors outline how the electric microenvironment of bone tissue is generated and continuo
